oder Dimethylsulfoxid in Form einer Dispersion oder Suspension in einem inerten organischen Lösungsmittel bei -20 bis -40° C
durchführt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man das Verfahren ausgehend von
4-(2-Tetrahydropyranyloxy)phenyl-essigsäure-4'-methoxybenzylester
durchführt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man das Verfahren ausgehend von
4-(4"-Methoxybenzoyloxy)-phenylessigsäure-4'-methoxybenzyl-ester
durchführt.
4. Verfahren nach Anspruch 1, dadurch gekennzeichnet,
daß man bei ungefähr — 30° C arbeitet.
Die Erfindung betrifft ein Verfahren zur Herstellung von Phenylmalonsäuremonoestern durch Carboxylierung
von Phenylessigsäureestern mit Kohlendioxid.
Phenyimalonsäuremonoester sind wertvolle Ausgangssubstanzen zur Herstellung von Arzneimitteln,
wie synthetischen Penizillinen und Cephalosporin. Von diesen Malonsäuremonoestern ist 2-[4-(2-Tetrahydropyranyloxy)phenyl]:malonsäure-4'-methoxybenzyimoncvester
ein wichtiges Ausgangsmaterial zur Herstellung von Penicillinen und Cephalosporinen mit 4-Hydroxyphenylmalonsäuremonoamidoresten
in 3-Stellung des /J-Lactomringes.
In der JP-OS 54-106 447 ist ein Verfahren zur Herstellung von Phenylmalonsäuremonoestern angegeben.
Dabei wird ein Wasserstoffatom in «-Stellung von Phenylessigsäureestern durch ein Alkaliatom ersetzt
ι ο durch Umsetzung mit einem Alkalisalz von Hexaalkyldisilazan.
Dann werden die durch Alkali substituierten Phenylessigsäureester mit Kohlendioxid in einem
inerten Lösungsmittel unter Feuchtigkeitsausfchluß bei
niederer Temperatur umgesetzt, wobei Alkalisalze von Phenylmalonsäuremonoestern entstehen.
Bei diesem Verfahren muß das sehr teure Hexaalkyldisilazan angewandt werden und die Reaktionstemperatur
muß im Falle der Herstellung von 2-(4-(2-Tetrahydropyrany!oxy)phenyl)malonsäure-4'-methoxybenzyl-
mono-ester auf —60° C oder darunter gehalten werden.
In Journal Amer. Chem. Soc, Bd. 66 (1944), S.
1037 — 1038 ist angegeben, daß Malonsäure- und Dimethylmalonsäureester durch Carboxylierung der
entsprechenden Essigsäure- bzw. Isobuttersäureester in Gegenwart von Triphenylmethannatrium erhalten werden
können.
Es ist Aufgabe der vorliegenden Erfindung, ein wirtschaftlicheres Verfahren zur Herstellung von
Phenylmalonsäuremonoestern zu entwickeln.
Diese Aufgabe wird durch das im Hauptanspruch gekennzeichnete Verfahren gelöst.
2-[4-(2-Tetrahydropyranyloxy)phenyl]-malonsäure-4'-methoxybenzyi-mono-ester
kann bei wirtschaftlicheren Reaktionstemperaturen, d.h. bei —20 bis —40°C
hergestellt werden, im Vergleich mit einer Temperatur von —60° C oder darunter nach dem bekannten
Verfahren.
Die erfindungsgemäß verwendeten, mit Natrium substituierten Verbindungen, die als Aktivatoren für die
Carboxylierungsreaktion angewandt werden, sind leicht zugänglich. Natriumacetylen kann leicht hergestellt
werden durch Einleiten von Acetylen in eine Dispersion von metallischem Natrium in einem organischen
Lösungsmittel wie Kerosin, Toluol und Xylol bei Raumtemperatur. Alle anderen Aktivatoren werden
hergestellt durch Zutropfen einer Mischung von Monochlorbenzol und einer der genannten Verbindungen
außer Acetylen in ungefähr äquimolarem Verhältnis in eine Natriumdispersion, enthaltend zwei bis drei
vorzugsweise 2,3 bis 2,7 mol-Äquivalent Natrium,
bezogen auf Monochlorbenzol, in einem inerten Lösungsmittel bei Raumtemperatur und anschließendes
Erwärmen. Natriumsulfoxide können auch leicht hergestellt werden durch Umsetzung von Sulfoxiden mit
Natriumamid oder Natriumhydrid bei 50 bis 70° C.
Die Phenylessigsäureester werden bei dem erfindungsgemäßen Verfahren mit einer Dispersion oder
Suspension des Aktivators in einem inerten organischen Lösungsmittel wie Kerosin, Toluol und Xylol vermischt
und das Gemisch wird auf eine Temperatur von —20 bis -40°C vorzugsweise ungefähr -30°C gekühlt. Die
Natriumsalze der Phenyimalonsäuremonoester werden durch Einleiten von Kohlendioxid in Form von
Kohlendioxidgas oder.Zugabe von Trockeneis zu dem
b5 Gemisch bei einer Temperatur wie oben angegeben
gebildet. Alle die oben genannten Verfahrensstufen müssen unter Ausschluß von Feuchtigkeit durchgeführt
werden.
Zu dem so erhaltenen Reaktionsgemisch werden entsprechende Mengen einer gesättigten wäßrigen
Natriumchloridlösung und eines organischen Lösungsmittels wie Toluol, η-Hexan oder einer Mischung davon
bei einer Temperatur von etwa —10 bis +100C zugegeben. Dabei fallen die Natriumsalze der Phenylmalonsäuremonoester
in Form von Kristallen aus dem Gemisch aus. Diese Kristalle werden abfiltriert und
dann einer Mischung aus Wasser und einem organischen Lösungsmittel wie Alkylacetat zugesetzt und
durch eine anorganische Säure neutralisiert oder schwach angesäuert. Nach der Neutralisation wird die
organische Schicht von dem Gemisch abgetrennt und mit Wasser gewaschen. Die gewünschten Phenylmalonsäuremonoester
werden erhalten durch Abdampfen des , Lösungsmittels von der organischen Schicht
Das erfindungsgemäße Verfahren stellt ein industriell günstiges und wirtschaftliches Verfahren zur Herstellung
von Phenylmalonsäuremonoestern dar, da die Ausbeuten der gewünschten Produkte gut sind und die
Preise der Ausgangsmaterialien für die Aktivatoren niedrig sind und diese Substanzen auf dem Markt leicht
erhalten werden können und weil die Reaktionstemperatur nicht so niedrig sein muß, was zur Energieeinsparung
führt.
Die Erfindung wird durch die folgenden Beispiele näher erläutert. Beispie, ,
26 g (216mmol) Cumol wurden zu einer 40%igen Dispersion von Natrium in Kerosin, enthaltend 4,7 g
(205 mmol) metallisches Natrium, gegeben und das Gemisch gleichmäßig gerührt Dann wurden 9 g
(80 mmol) Monochlorbenzol zu dem Gemisch zugetropft, wobei die Temperatur auf 10 bis 20°C gehalten
wurde und das Gemisch wurde anschließend auf 1100C erhitzt und ungefähr 2 h auf dieser Temperatur
gehalten.
In dieses Gemisch wurden 30 ml Toluol-Lösung, enthaltend 7,12 g (20 mmol) 4-(2-Te*.rahydropyranyloxy)phenyl-essigsäure-4'-methoxybenzylester
bei
Raumtemperatur gegeben und das Gemisch 30 min gerührt anschließend 30 ml Toluol zugegeben und auf
-300C gekühlt. 20 g Trockeneis wurden langsam zu dem gekühlten Gemisch gegeben, das auf der gleichen
Temperatur gehalten wurde. Nach vollständiger Umsetzung wurde das Gemisch auf 00C erwärmt und dann
100 ml gesättigte wäßrige Natriumchloridlösung und 90 ml η-Hexan zugeben. Das Natriumsalz von 2-[4-(2-Tetrahydropyranyloxy)phenyl]malonsäure-4'-methoxy-
benzyl-mono-ester fiel in Form von Kristallen in der wäßrigen Schicht aus. Die Kristalle wurden abfiltriert
und dann mit einer Mischung von Toluol und n-Hexan (1 :1) gewaschen.
Die Kristalle wurden in 200 ml Wasser dispergiert und 150 ml Äthylacetat zugegeben und das Gemisch
durch Zugabe von 10%iger wäßriger Phosphorsäure-Lösung auf einen pH-Wert von 3 bis 4 gebracht.
Die organische Schicht wurde von der wäßrigen Schicht abgetrennt, mit Wasser gewaschen und über
wasserfreiem Natriumsulfat getrocknet.
60 Man erhielt 6,7 g 2-[4-(2-Tetrahydropyranyloxy)phenyl]malonsäure-4'-methoxybenzyl-mono-ester
durch Verdampfen des organischen Lösungsmittels. Fp. 115 bis 1170C, Ausbeute 83%.
In eine 30%ige Dispersion von Natrium in Kerosin, enthaltend 4,7 g (205 mmol) metallisches Natrium,
wurde bei Raumtemperatur Acetylen geleitet Zu dieser Dispersion wurden 60 ml Toluol-Lösung, enthaltend
14,24 g (40 mmol) 4-(2-Tetrahydropyranyloxy)phenylessigäsure-4'-methoxybenzylester,
gegeben. Anschließend wurde das Gemisch entsprechend Beispiel 1 behandelt mit der Ausnahme, daß die doppelten
Mengen Trockeneis, gesättigte wäßrige Natriumchloridlösung. η-Hexan, Wasser, Äthylacetat Lösungsmitteln
usw. verwendet wurden.
Man erhielt 13,5 g 2-[4-(2-Tetrahydropyranyloxy)phenyl]-malonsäure-4'-methoxybenzyl-mono-ester,
Fp. 114° C bis 117° C, Ausbeute 84%.
30 ml Toluol-Lösung, enthaltend 9 g (80 mmol) Monochlorbenzol, wurden zu einer 40%igen Dispersion
von Natrium in Kerosin, enthaltend 4,7 g (205 mmol) metallisches Natrium, gegeben und zu dieser Dispersion
wurden 30 ml Toluol-Lösung enthaltend 80 mmol Methyl-methylthiomethylsulfoxid zugegeben.
Zu dem oben angegebenen Gemisch wurden 30 ml
Toluol-Lösung, enthaltend 7,12 g (20 mmol) 4-(2-Tetrahydropyranyloxy)phenylessigsäure-4'-methoxybenzyl-
ester gegeben und das Gemisch anschließend wie in Beispiel 1 angegeben behandelt.
Man erhielt die gewünschte Verbindung in einer Ausbeute von 82% (Fp. 115 bis 117° C).
Es wurde wie in Beispiel 3 gearbeitet mit der Ausnahme, daß Dimethylsulfoxid anstelle von Methylmethylthiomethylsulfoxid
wurde.
Man erhielt die gewünschte Verbindung in einer Ausbeute von 81,5% (Fp. 115 bis 117°C).
Eine Dispersion von 7,2 g (65 mmol) durch Natrium substituertes Dimethylsulfoxid wurde hergestellt durch
Umsetzung von 3,1 g (80 mmol) Natriumamid mit 9 g (115 mmol) Dimethylsulfoxid in Toluol bei 700C unter
Argonatmosphäre. 7,12 g (20 mmol) 4-(2-Tetrahydropy-
ranyloxy)phenyl-essigsäure-4'-methoxybenzylester wurden in 30 ml der Dispersion von Natriumdimethylsulfoxid
gegeben und anschließend 30 ml Toluol zugesetzt. Daraufhin wurde das Gemisch wie in Beispiel
1 angegeben behandelt.
Man erhielt 6,5 g 2-[4-(2-Tetrahydropyranyloxy)phenyl]-malonsäure-4'-methoxybenzyl-mono-ester,
Fp. 115° C bis 117° C, Ausbeute 81%.
