Description
- 1) C1-10-Alkanoylreste;
- 2) C1-5-Halogenalkanoylreste;
- 3) die Azidoacetyl- und Cyanacetylgruppe;
- 4) Acylreste der allgemeinen Formel
Ar-CQQ′-CO-
in der Q und Q′ Wasserstoffatome oder Methylgruppen darstellen und Ar eine Phenyl- oder Dihydrophenylgruppe oder einen monocyclischen heteroaromatischen Rest mit 1 bis 4 Stickstoff-, Sauerstoff- und/oder Schwefelatomen als Ringatomen bedeutet, wobei diese Reste gegebenenfalls durch einen inerten Rest substituiert sind, beispielsweise einen C1-5-Alkyl-, C1-3-Alkoxy-, C1-10-Acyloxy- oder C7-10-Aralkoxyrest, ein Fluor-, Chlor-, Brom- oder Jodatom, eine Trifluormethyl-, Cyano-, Aminomethyl-, gegebenenfalls geschützte Carboxymethylthio-, Hydroxyl- oder Nitrogruppe; - 5) die (4-Pyridon-1-yl)-acetyl- und (2-Iminothiazolin-4-yl)- acetylgruppe;
- 6) Acylreste der allgemeinen Formel
Ar-G-CQQ′-CO-
in der Ar, Q und Q′ die vorstehend angegebene Bedeutung haben und G ein Sauerstoff- oder Schwefelatom darstellt; - 7) Acylreste der allgemeinen Formel
Ar-CHT-CO-
in der Ar die vorstehend angegebene Bedeutung hat und T entweder- a) eine Hydroxylgruppe oder einen C1-10-Acyloxyrest oder
- b) eine Carboxylgruppe, einen C2-7-Alkoxycarbonyl-, C2-15- bzw. C8-15-Aralkoxycarbonyl-, C1-12- bzw. C7-12-Aryloxycarbonyl- oder C1-7-Alkanoyloxy-C1-3-alkoxycarbonylrest, eine Cyano- oder Carbamoylgruppe oder
- c) eine Sulfonsäuregruppe oder einen C1-7-Alkoxysulfonylrest bedeutet;
- 8) Acylreste der allgemeinen Formel in der Ar die vorstehend angegebene Bedeutung hat und W und W′ jeweils Wasserstoffatome oder einen Aminosubstituent darstellen, beispielsweise einen C2-10-Alkoxycarbonyl-, C3-10-Cycloalkyl-C2-3-alkoxycarbonyl-, C5-8-Cycloalkoxycarbonyl-, C1-4-Alkylsulfonyl-C1-4-alkoxycarbonyl-, Halogen-C1-3-alkoxycarbonyl-, C1-15- bzw. C8-15-Aralkoxycarbonyl-, C1-10-Alkanoyl- oder C2-15- bzw. C6-15-Aroylrest, jeweils gegebenenfalls substituiert durch einen inerten Rest, beispielsweise eine Hydroxylgruppe, einen C1-5-Alkyl-, C1-10-Alkanoyloxy- oder C1-3-Hydroxyalkylrest, eine Trifluormethylgruppe oder ein Halogenatom, eine Pyroncarbonyl-, Thiopyroncarbonyl-, Pyridoncarbonyl-, Carbamoyl- oder Guanidinocarbonylgruppe oder eine gegebenenfalls substituierte Ureidocarbonylgruppe, beispielsweise eine 3-Methyl- 2-oxoimidazolidin-1-ylcarbonyl-, 3-Methansulfonyl-2-oxoimidazolidin- 1-ylcarbonyl-, 3-Methylureidocarbonyl- oder 1-Methylureidocarbonylgruppe, eine gegebenenfalls substituierte Aminooxalylcarbamoylgruppe, beispielsweise eine 4-Methyl-2,3-dioxopiperazin-1-ylcarbonyl- oder 4-Ethyl- 2,3-dioxopiperazin-1-ylcarbonylgruppe, gegebenenfalls substituierte Thioureidocarbonyl-Äquivalente der vorgenannten Ureidocarbonyl- oder Aminooxalylcarbamoylgruppe, oder W und W′ bilden zusammen mit dem Stickstoffatom, an das sie gebunden sind, eine Phthalimido-, Maleinimido- oder Enaminogruppe, die sich von enolisierbaren Carbonylverbindungen ableitet, beispielsweise einem C5-10-Acetessigsäureester, einem C4-10-Acetessigsäureamid, Acetoacetanilid, Acetylaceton, Acetoacetonitril, α-Acetyl-γ- butyrolacton oder 1,3-Cyclopentandion;
- 9) Acylreste der allgemeinen Formel in der Ar die vorstehend angegebene Bedeutung hat und E ein Wasserstoffatom oder einen C1-5-Alkylrest darstellt;
- 10) eine 5-Aminoadipoylgruppe oder eine N-geschützte 5-Aminoadipoylgruppe, die beispielsweise durch einen C1-10-Alkanoyl-, C1-10- bzw. C8-10-Aralkanoyl-, C2-11- bzw. C7-11-Aroyl-, C1-5- Halogenalkanoyl- oder C2-11-Alkoxycarbonylrest geschützt ist, oder eine an der Carboxylgruppe geschützte 5-Aminoadipoylgruppe, die beispielsweise durch einen C1-5-Alkyl-, C2-21- bzw. C7-21-Aralkyl-, C1-10- bzw. C7-10-Aroyl-, C2-10-Trialkylsilyl- oder C2-5-Dialkyl-C1-5-alkoxysilylrest geschützt ist, und wobei die Amino- oder Carboxylschutzgruppe gegebenenfalls durch einen C1-5-Alkyl- oder C1-5-Alkoxyrest, ein Halogenatom oder eine Nitrogruppe substituiert ist, und
- 11) Acylreste der allgemeinen Formel
L-O-CO
in der L eine leicht abspaltbare und gegebenenfalls substituierte C1-10-Kohlenwasserstoffgruppe, beispielsweise eine tert.-Butyl-, 1,1-Dimethylpropyl-, Cyclopropylmethyl-, 1-Methylcyclohexyl-, Isobornyl-, 2-C1-2-Alkoxy- tert.-butyl-, 2,2,2-Trichlorethyl-, Benzyl-, Naphthylmethyl-, p-Methoxybenzyl-, Pyridylmethyl- oder Diphenylmethylgruppe bedeutet.
(b) Eine Lösung von 50 g 7β-Phenoxyacetamidodesacetoxycephalosporansäurebenzylester in 1,5 Liter Benzol wird bei 50°C unter Stickstoff als Schutzgas mit 16,05 ml Pyridin und 35,6 mg Phosphorpentachlorid versetzt. Nach 2stündigem Rühren wird das Reaktionsgemisch mit Eiswasser abgekühlt und mit 750 ml kaltem Isobutanol versetzt. Nach 1stündigem Rühren bei Raumtemperatur wird das Gemisch auf 10°C abgekühlt, mit 750 ml kaltem Wasser verdünnt und 30 Minuten bei Raumtemperatur gerührt. Danach wird das Reaktionsgemisch eingedampft, der Rückstand in einem Gemisch von 400 ml Benzol und 100 ml Wasser gelöst und durchgeschüttelt. Die Benzolphase wird zweimal mit 50 ml 2 n Salzsäure angesäuert. Die wäßrige Phase wird abgetrennt und mit 200 ml Benzol gewaschen. Sodann wird die wäßrige Phase mit 1 Liter Ethylacetat überschichtet, mit 2 n Natronlauge auf einen pH-Wert von 6,5 eingestellt und durchgeschüttelt. Danach werden die Schichten getrennt. Die organische Phase wird unter vermindertem Druck eingedampft, der Rückstand mit 70 ml Toluol versetzt und filtriert. Es werden 32,06 g (92,37% d. Th.) 7β-Aminodesacetoxycephalosporansäurebenzylester vom F. 129 bis 130°C erhalten.
