Description
4-Cyano-4-phenyl-aminocyclohexane
Aus dem südafrikanischen Patent 66/5399 ist bekannt, dass basische
Cyano-phenyl-cyclohexyl-Verbindungen Morphin und Mescalin
antagonisieren, antitussive Eigenschaften aufweisen und analgetische
Wirkung zeigen. Aus dem südafrikanischen Patent 65/63OI
ist weiterhin bekannt, dass 4,4-Diphenyl-cyclohexylamine spasmolytische
und parasympathicolytische Wirkungen zeigen sowie den Blutdruck senken.
Die deutschen Patente 1 154 8IO und 1 I58 O83 und die deutschen
Patentanmeldungen P I5 93 921. 6 und P 16 43 429. 0 betreffen Verfahren zur Herstellung basisch substituierter Phenylacetonitrile,
welche infolge ihrer pharmakologischen Wirksamkeit wertvolle Arzneimittel darstellen und als Herz-Sympathicolytica
und als Coronardilatatoren Verwendung finden können.
Gegenstand der Erfindung sind basisch substituierte 4-Cyano-4-phenyl-aminocyclohexane
der allgemeinen Formel

sowie deren Salze, worin A, B und C Wasserstoff- oder Chloratome,
Methyl-, Trifluormethyl-, Methoxy- oder Äthoxy-Gruppen,
R ein Wasserstoffatom oder die Methylgruppe und Alk eine Äthylen-,
n-Propylen-, a-Methyläthylen- oder ce-Methyl-ß-hydroxyäthylengruppe
bedeuten. .
Die neuen Verbindungen zeigen spasmolytische, blutdrucksenkende,
neuroleptische und coronarwirksame Eigenschaften und sollen als Heilmittel Verwendung finden.
Gegenstand der Erfindung ist weiterhin ein Verfahren zur Herstellung
der neuen Verbindungen. Das erfindungsgemässe Verfahren
ist dadurch gekennzeichnet, dass man 4—Cyano-4-phenyl-cyclohexanone
der allgemeinen Formel
in an sich bekannter Weise mit Arylalkylaminen der allgemeinen

Die reduktive Kondensation der 4-Cyano-4-phenyl-cyclohexanone
der allgemeinen Formel II mit den Aminen der allgemeinen Formel III kann nach allen bekannten Verfahren durchgeführt werden,
beispielsweise mit Wasserstoff in Gegenwart von Edelmetall- j Nickel- oder Gobalt-Katalysatoren bei Normaldruck oder
erhöhtem Druck oder in Gegenwart von aktiviertem metallischem
Aluminium. Man kann die Reduktion weiterhin mit Natriumborhy-

drid durchführen* wobei man zweekmässigerweise durch Wasserabspaltung
erst das Azomethin herstellt und dieses dann anschliessend
reduziert. Die erhaltenen Amine können entweder durch Destillation oder durch Überführung in ihre Hydrochloride gereinigt
werden.
Die sekundären Amine können gegebenenfalls nach allen bekannten
Methoden methyliert werden, zweckmässigerweise verwendet
man Formaldehyd und AmeisensäureV

Die erfindttngsgeatässen Verbindungen sind mit anorganischen oder
organischen Säuren in wasserlösliche Salze überführbar.
Die als Äusgangsmaterial eingesetzten 4-Cyano-4-phenyl-cyclohexanoneder
allgemeinen Formel III sind beispielsweise erhältlich
nach der von E. C. Horning et al., J.Amer.ehem.Soc. 74,
773, (1952) beschriebenen Methode durch Esterkondensation von

-ft-(3, 4-dimethoxyphenyl)-äthyl-aminocyclohexan
19?9 g (X)?l Mol) 4-Cyana~4-phenyl-cyclohexanon (hergestellt
aus y-CyanQ-y-phenyi-pimelinsäure-dimethylester) und 18,1 g
(O3X MqI) Homaveratrylamin werden in 100 ecm trockenem Toluol
gelöst und zum Sieden erhitzt. Das sich bildende Reaktionswasser
wird azeotrop entfernt. Danach destilliert man das Lösungsmittel ab, versetzt den Rückstand mit 150 ecm Methanol und hydriert
durch portionsweise Zugabe von 2,5 g Natriumborhydrid.
Nach einer Stunde wird die Reaktionsmischung zur Trockne eingeengt
undmit 200 ecm 2n Kaliumhydroxydlösung und 200 ecm Äther
versetzt,. Danach wird die wässrige Schicht noch zweimal mit
Äther ausgeschüttelt und die vereinigten Ätherlösungen getrocknet.
In die Äther!σsung wird bis zur Sättigung trockener Chlorwasserstoff
eingeleitet. Das ausgefallene Hydroehlorid wird abgesaugt und aus Isopropanol umkristallisiert. Man erhält 26,7 S
(73$ der Theorie) 4-Cyano-4*phenyl-0-(3>4'-«iieiethoxyphenyl)-äthyl-aminoeyclohexanhydrochlorid,
F 221 - 222° C.
In analoger Weise wurden die Verbindungen Nr. 1 - 23 der Tabelle 1 hergestellt.
