Abstract
Η παρούσα εφεύρεση παρέχει αμφιειδικά αντισώματα που δεσμεύονται σε αντιγόνα CD3 και όγκων, καθώς και μεθόδους χρήσης αυτών. Σύμφωνα με ορισμένες υλοποιήσεις, τα αμφιειδικά αντισώματα της εφεύρεσης εμφανίζουν μειωμένες δραστικές λειτουργίες τελεστή και έχουν μοναδικό προφίλ δέσμευσης αναφορικά με τους υποδοχείς Fcγ. Τα αμφιειδικά αντισώματα είναι τροποποιημένα ώστε να επάγουν αποτελεσματικά τη μεσολαβούμενη από τα Τ κύτταρα θανάτωση κυττάρων όγκου. Σύμφωνα με ορισμένες υλοποιήσεις, η παρούσα εφεύρεση παρέχει αμφιειδικά αντιγονοδεσμευτικά μόρια που περιλαμβάνουν έναν πρώτο αντιγονοδεσμευτικό τομέα ο οποίος δεσμεύει ειδικά το ανθρώπινο CD3, ένα δεύτερο αντιγονοδεσμευτικό μόριο το οποίο δεσμεύει ειδικά το ανθρώπινο CD20 και έναν τομέα Fc ο οποίος δεσμεύει τους υποδοχείς Fcγ με συγκεκριμένο μοτίβο δέσμευσης. Σε ορισμένες υλοποιήσεις, τα αμφιοδικά αντιγονοδεσμευτικό μόρια της παρούσας εφεύρεσης είναι ικανά να αναστέλλουν την ανάπτυξη Β-κυτταρικών όγκων ή όγκων μελανώματος που εκφράζουν το CD20.
