Abstract
Se describen los métodos para producir una proteína en cultivo celular que comprende un compuesto anti-senectud, tal como el antioxidante carnosina. De acuerdo a las enseñanzas de la presente invención, las células desarrolladas en un medio de cultivo celular que comprende un compuesto anti-senectud muestran viabilidad y productividad incrementadas. Además, los cultivos celulares desarrollados en presencia de un compuesto anti-senectud muestran niveles disminuidos de agregados de alto peso molecular en el medio de cultivo celular. 1.- Un método para producir una proteína de fusión TNFR en cultivo celular, caracterizado porque comprende los pasos de: cultivar las células de mamífero que contienen un gen que codifica para la proteína de fusión TNFR, cuyo gen es expresado bajo condiciones de cultivo celular, en un medio de cultivo celular que comprende un compuesto anti-senectud; y mantener el cultivo bajo condiciones y por un tiempo suficiente para permitir la expresión de la proteína;en donde el cultivo celular muestra una característica mejorada de cultivo celular que difiere de una característica correspondiente de cultivo celular que pudiera ser observada bajo condiciones de otro modo idénticas en un medio de otro nodo idéntico, que carece del compuesto anti-senectud; en donde la característica de cultivo mejorada se selecciona del grupo que consiste de: título incrementado, productividad específica celular incrementada, viabilidad celular incrementada, densidad incrementada de células viables integradas, acumulación disminuida de agregados de alto peso molecular, acumulación disminuida de especies ácidas, y combinaciones de las mismas. 2.- El método de conformidad con la reivindicación 1, caracterizado porque el compuesto anti-senectud se selecciona del grupo que consiste de carnosina, acetil-carnosina, homocarnosina, anserina, y beta-alanina y combinaciones de las mismas. 3.- El método de conformidad con la reivindicación 1, caracterizado porque el compuesto anti-senectud comprende carnosina. 4.- El método de conformidad con la reivindicación 1, 2 ó 3, caracterizado porque el compuesto anti-senectud está presente en el medio de cultivo celular a una concentración de entre aproximadamente 5 mM y aproximadamente100 mM.
