Abstract
Se proporcionan métodos y procesos para la producción de proteína recombinante. Los métodos son útiles para la producción de factores de crecimiento, en particular los de la superfamilia de TGF-b, que incluyen. proteínas morfogenéticas óseas (BMP), tales como BMP-2. Las células huésped adecuadas se cultivan en un medio en donde el hierro está presente en una concentración de al menos 2,25 mM y si el piridoxal está presente, constituye menos de 55% de la concentración molar de vitamina B6 del medio. Reivindicación 1: Un método de producción de BMP-2 que comprende las etapas de: i) cultivar una célula huésped adecuada que comprende una molécula de ADN que codifica una BMP-2 en un medio de cultivo que comprende hierro en una concentración de al menos 2,25 mM, un compuesto polianiónico y si está presente piridoxal, constituye menos de 55% de la concentración molar de vitamina B6 en el medio de cultivo; y ii) recuperar la proteína de interés, en donde la célula huésped se cultiva en un biorreactor que tiene una capacidad mayor de 3 L.
