Abstract
Se revelan células huéspedes recombinantes adecuadas para degradar un oligosacárido, que se han optimizado para el crecimiento y produccion de alto rendimiento de etanol y métodos de fabricacion y empleo de estas células. La presente también proporciona medios mínimos, que comprenden compuestos similares a urea para la produccion economica de etanol por microorganismos recombinantes. Las células huéspedes recombinantes de acuerdo con la presente se modifican por mutacion génica para eliminar genes responsables de la produccion de productos no deseados diferentes de etanol, por lo que aumentan el rendimiento de etanol producido a partir de los oligosacáridos, en relacion con las cepas progenitoras no mutadas. Las cepas nuevas y mejoradas de la bacterias recombinantes son capaces de un productividad y rendimiento de etanol superiores, cuando se cultivan en condiciones adecuadas para la fermentacion en medios de crecimiento mínimos que contienen reactivos economicos. Los sistemas optimizados para la produccion de etanol combinan un medio mínimo optimizado seleccionado con una célula huésped recombinante optimizada para su uso en el medio seleccionado. Los sistemas preferidos son adecuados para la produccion eficiente de etanol por medio de la sacarificacion y fermentacion simultáneas (SSF) empleando lignocelulosa como fuente de oligosacáridos. La presente también proporciona nuevas secuencias de polinucleotidos aisladas, secuencias de polipéptidos, vectores y anticuerpos.
