Description
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen basischen Äthern der allgemeinen Formel I EMI1.1 in der R eine Alkylgruppe mit maximal 4 C-Atomen, R1 Wasserstoff, einen niederen Alkylrest, einen Allylrest oder einen Benzylrest, R2 eine Alkenyl-, Aralkenyl-, Haloalkenyl-, Alkinyl-, Cycloalkyl- oder Cycloalkylidenmethylgruppe und R3 Wasserstoff oder eine Methylgruppe bedeuten und von deren Salzen. Die erfindungsgemäss hergestellten Verbindungen sind Substanzen mit guter analgetischer und gleichzeitig morphinantagonistischer Wirkung. Bekanntlich erzeugen Morphin und andere starke narcotische Analgetica bei regelmässiger Anwendung Sucht, die zur physischen und psychischen Abhängigkeit von der Droge führt und die therapeutische Anwendung dieser Stoffe stark einschränkt. Verbindungen vom Typ der Morphinantagonisten sind in der Lage, die Wirkungen des Morphins aufzuheben und daher bei morphinsüchtigen Tieren Entziehungserscheinungen hervorzurufen. Da die erfindungsgemässen Verbindungen neben der morphinantagonistischen Wirkung auch eine analgetische Wirkung besitzen, können sie als Analgetica verwendet werden, wobei man infolge ihrer der Wirkung des Morphins entgegengesetzten Eigenschaften der allgemeinen Lehrmeinung zufolge schliessen kann, dass sie bei ihrer Anwendung keine Sucht erzeugen. Es sind bisher schon zahlreiche Verbindungen mit morphinantagonistischen Eigenschaften bekannt geworden, die teilweise auch gleichzeitig analgetische Wirkung besitzen. Dabei handelte es sich jedoch durchwegs um kompliziertere heterocyclische Verbindungen, also solche, die das Stickstoffatom als basisches Zentrum in einen Ring eingebaut enthalten. Demgegenüber besitzen die erfindungsgemäss herstellbaren Verbindungen die einfache Struktur eines rein aliphatischen Amins. Ferner sind schon Verbindungen hergestellt worden, welche die Struktur der allgemeinen Formel I besitzen, in denen jedoch die an den Stickstoff gebundenen Gruppen keine ungesättigten Reste und keine Cycloalkylgruppen enthielten (österreichische Patentschrift Nr. 270617, Nr. 255400, Nr. 255401, Nr. 264501 und Nr, 266085). Diese Verbindungen besitzen wohl auch teilweise eine analgetische Wirkung, jedoch fehlen ihnen die morphinantagonistischen Eigenschaften, die den erfindungsgemäss herstellbaren Stoffen überdies zukommen und die ihre Unbedenklichkeit im Hinblick auf eine Entstehung von Sucht gewährleisten. In der österreichischen Patentschrift Nr. 266085 ist unter anderem auch die Herstellung einer solchen Verbindung mit einem Benzylrest am Stickstoff in der Seitenkette durch Reduktion der entsprechenden Benzoylverbindung mit Lithiumaluminiumhydrid beschrieben. Das Verfahren zur Herstellung der Verbindungen der Formel I und von deren Salzen ist dadurch gekennzeichnet, dass die neuen Säureamide der allgemeinen Formel EMI1.2 in der R, R1, R2 und R3 wie in Formel I definiert sind, mit komplexen Hydriden des Aluminiums, vorzugsweise in einem inerten Lösungsmittel, reduziert und dabei anfallende Basen nach Entfernen des überschüssigen Reduktionsmittels als solche isoliert in Salze übergeführt werden. Als komplexes Hydrid des Aluminiums sind vornehmlich Lithiumaluminiumhydrid und Natriumdihydro bis (methoxy äthoxy)-aluminat zu nennen. Als Lösungsmittel kommen vor allem Diäthyläther oder Tetrahydrofuran in Betracht. Die Reduktion wird zweckmässig in der Wärme, vorzugsweise unter Sieden des Lösungsmittels vorgenommen. Das überschüssige Reduktionsmittel kann mit Wasser zersetzt und das Reaktionsprodukt durch Eindampfen in relativ reiner Form gewonnen werden. Es kann als Base weiter gereinigt werden oder auch in Salze übergeführt werden. Als solche Salze können beispielsweise genannt werden: Hydrohalogenide, Sulfate, Tartrate, Mandelate, Fumarate und Cyclohexylsulfamate, die wegen der physiologischen Unbedenklichkeit der zugrundeliegenden Säuren pharmazeutisch anwendbar sind. Die als Ausgangsprodukt verwendeten Säureamide der Formel II sind neu. Sie können z.B. auf einfache Weise erhalten werden, wenn man die gegebenenfalls monosubstituierten Aminoverbindungen der allgemeinen Formel EMI1.3 in der R, R1 und R3 wie in Formel I definiert sind, mit reaktionsfähigen Derivaten von Carbonsäuren der Formel R2COOH IV in der R2 wie in Formel I definiert ist, vornehmlich mit deren Säurechloriden, umsetzt. Die Umsetzung kann in einem inerten organischen Lösungsmittel erfolgen, wobei die Gegenwart säurebindender Mittel wie Pyridin, Triäthylamin oder Natriumcarbonat zu empfehlen ist, damit verhindert wird, dass ein Teil des Amins der Formel III als Säureacceptor dient und somit der Reaktion entzogen wird. Beispiel 1: 10,6 g 1,1 -Diphenyl- 1 -propoxy-3-cyclopropylcarbonylaminopropan (Fp 144-148 C) werden in einen Soxhlet-Extraktor gefüllt und zu einer vorgelegten Suspension von 8,0 g Lithiumaluminiumhydrid in 300 ml Äther zuextrahiert. Anschliessend wird noch 20 Stunden gekocht. Der Überschuss des Reduktionsmittels wird mit Wasser zersetzt, die Ätherlösung abgetrennt und eingedampft. Es werden 10 g 1,1-Diphenyl-1-propoxy-3-(cyclopropyl- methyl)-amino-propan als zähes Öl erhalten (88% der Theorie). Das daraus gefällte Hydrochlorid hat nach Umkristallisieren aus Äthylacetat einen Schmelzpunkt von 170- 174 C. Das als Ausgangsmaterial verwendete l,l-Diphenyl-l-pro- poxy-3-cyclopropylcarbonylaminopropan wird wie folgt erhalten: 10 g 3,3-Diphenyl-3-propoxy-propylamin (vom Schmelzpunkt 45 bis 50"C), 7,5 g Triäthylamin, 4,7 g Cyclopropylcarbonsäurechlorid und 160 ml Benzol werden 1 Stunde lang auf 40 C erwärmt. Dann wird Wasser zugesetzt und die Schichten werden getrennt. Die organische Schicht wird mit Sodalösung und Wasser gewaschen, eingedampft und der Rückstand aus Cyclohexan umkristallisiert. Es werden 10,6 g 1,1-Diphenyl-1-propoxy-3- cyclopropylcarbonylaminopropan vom Schmelzpunkt 144-148 C erhalten. Beispiel 2: 12,1 gN-(3,3-Diphenyl-3-methoxypropyl)-ss,ss-dimethylacryla- mid werden wie in Beispiel 1 beschrieben in Äther als Lösungs- mittel mit Lithiumaluminiumhydrid reduziert. Man erhält so 11,0 g 1,1 -Diphenyl- 1 -methoxy-3-(γ,γ-dimethyl-ally]-aminopropan als zähes Öl entsprechend einer Ausbeute von 95% der Theorie. Das daraus gefällte Hydrochlorid hat nach dem Umkristallisieren aus Wasser einen Schmelzpunkt von 85-88' C. Die Herstellung des als Ausgangsmaterial benötigten N-(3,3 Diphenyl-3-methoxy-propyl) mit ss,ss-dimethylacrylamid gelingt durch Umsetzen von 3,3-Diphenyl-3-methoxy-propylamin mit ss,ss- Dimethylacrylsäurechlorid nach der in Beispiel 1 beschriebenen Methode. Beispiel 3: 6,0 g N-(3,3-Diphenyl-3-methoxy-propyl-ss,ss-dimethylacrylsäureamid werden in 100 ml Benzol, dem 7,0 ml einer 70%igen benzolischen Lösung von Natriumdihydro-bis-(methoxy-äthoxy) aluminat zugesetzt war, 60 Stunden lang gekocht. Dann wird 20 ml 4 n Natronlauge zugesetzt, die Schichten getrennt und die organische Schicht eingedampft. Es verbleiben 5,5 g Ö1 (96% der Theorie), welches in Äther gelöst und mit Chlorwasserstoff als Hydrochlorid gefällt wird. Das erhaltene l,l-Diphenyl-l-me thoxy-3-(y,r-dimethyl-allyl)-aminopropan-hydrochlorid hat nach dem Umkristallisieren aus Wasser den Schmelzpunkt 85 bis 88 C. Beispiel 4: 0,75 g Lithiumaluminiumhydrid werden in 100 ml Tetrahydrofuran suspendiert und eine Lösung von 3,7 g N-(3,3-Diphenyl-3methoxy-propyl)-zimtsäureamid in 40 ml Tetrahydrofuran zugetropft. Dann wird das Gemisch 24 Stunden lang gekocht, gekühlt, 4 ml Wasser zugetropft, die Salze abgetrennt und die verbleibende Lösung eingedampft. Es werden 3,4 g l,l-Diphenyl-l-methoxy-3- cinnamylamino-propan in öliger Form entsprechend 95% der Theorie als Rückstand erhalten. Das daraus durch Fällen mit Chlorwasserstoff in ätherischer Lösung erhältliche Hydrochlorid hat den Schmelzpunkt 195 bis 198 C. Das als Ausgangsmaterial verwendete N-(3,3-Diphenyl-3methoxy-propyl)-zimtsäureamid wird wie folgt hergestellt: 5,0 g 1,1-Diphenyl-1-methoxy-3-aminopropan. 4,2 g Triäthylamin und 4,0 g Zimtsäurechlorid werden in 200 ml Benzol wie in Beispiel 1 umgesetzt und aufgearbeitet. Nach dem Umkristallisieren aus Cyclohexan werden 7,55 g (entsprechend 98% der Theorie) N-(3,3-Diphenyl-3-methoxypropyl)-zimtsäureamid vom Schmelzpunkt 118 bis 122"C erhalten. Beispiel 5: 10,6 g 1,1 -Diphenyl- 1 -methoxy-2-methyl-3-cyclobutylcarbonylamino-propan werden wie in Beispiel 1 zu einer Suspension von 5,3 g Lithiumaluminiumhydrid in einem Gemisch aus 200 ml Äther und 75 ml Benzol zuextrahiert und ebenso aufgearbeitet. Es werden 10,2 g 1,1-Diphenyl-1-methoxy-2-methyl-3-(cyclo- butyl-methyl)-aminopropan als zähes Öl erhalten (100% der Theorie). Das daraus gefällte Hydrochlorid hat einen Schmelzpunkt von 198 bis 200"C. Das als Ausgangsmaterial verwendete Amin wird wie folgt erhalten: 9,7 g 1,1-Diphenyl-1-methoxy-2-methyl-3-aminopropan, 7,7 g Triäthylamin und 5,4 g Cyclobutancarbonsäurechlorid werden in 150 ml Benzol 1 Stunde lang auf 400 C erwärmt und wie in Beispiel 1 beschrieben aufgearbeitet. Nach dem Umkristallisieren aus Cyclokexan werden 10,6 g (entsprechend 83% der Theorie) 1,1 - Diphenyl- l-methoxy-2-methyl-3-cyclobutylcarbonylamino-propan vom Schmelzpunkt 139 bis 140 C erhalten. Beispiel 6: 26,7 g 1,1-Diphenyl-1-methoxy-3-cyclobutylcarbonylaminopropan werden wie in Beispiel 1 beschrieben mit 13,0 g Lithiumaluminiumhydrid in 400 ml Äther und 150 ml Benzol reduziert. Man erhält 24,7 g (97% der Theorie) 1 ,l-Diphenyl-l-methoxy-3- (cyclobutyl-methyl)-aminopropan vom Schmelzpunkt 60 bis 63 C. Das daraus gefällte Hydrochlorid hat den Schmelzpunkt 194 bis 196 C. Das Ausgangsmaterial wird wie folgt erhalten: 24,1 g 1,1-Diphenyl-1-methoxy-3-aminopropan, 20,2 g Tri äthylamin, 14,3 g Cyclobutancarbonsäurechlorid werden in 250 ml Benzol wie in Beispiel 1 beschrieben umgesetzt und aufgearbeitet. Man erhält nach Umkristallisieren aus Cyclohexan 26,9 g (83% der Theorie) 1,1-Diphenyl-1-methoxy-3-cyclobutyl- carbonylamino-propan vom Schmelzpunkt 110 bis 112 C. Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von neuen basischen Äthern der allgemeinen Formel EMI4.1 in der R eine Alkylgruppe mit maximal 4 C-Atomen, R1 Wasserstoff, einen niederen Alkylrest, einen Allylrest oder einen Benzylrest, R2 eine Alkenyl-, Aralkenyl-, Haloalkenyl-, Alkinyl, Cycloalkyl- oder Cycloalkylidenmethylgruppe und R3 Wasserstoff oder eine Methylgruppe bedeuten und von deren Säureadditions Salzen, dadurch gekennzeichnet, dass Säureamide der Formel EMI4.2 in der R, R1, R2 und R3 wie in Formel I definiert sind, mit komplexen Hydriden des Aluminiums reduziert und die dabei anfallenden Basen nach Entfernen des überschüssigen Reduktionsmittels isoliert werden.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man die Reduktion in einem inerten Lösungsmittel durchführt.2. Verfahren nach Patentanspruch oder Unteranspruch 1, dadurch gekennzeichnet, dass man die erhaltenen Verbindungen in ihre Säureadditionssalze überführt.
