(2)D-グルコースに対する反応性を100%とした場合に、マルトース、D-ガラクトース、およびD-キシロースに対する反応性がいずれも2%以下である上記(1)記載のフラビン結合型GDH。
(3)以下の(a)から(c)の糖化合物:
(a)マルトース
(b)D-ガラクトース
(c)D-キシロース
の1以上が存在する場合のD-グルコースに対する反応性が、前記(a)~(c)が存在しない場合のD-グルコースに対する反応性を100%とした場合の96%~104%である、上記(1)~(2)に記載のフラビン結合型GDH。

本発明のフラビン結合型GDHは、基質特異性に優れ、D-グルコースに対する選択性が極めて高いことを特徴とする。具体的には、本発明のフラビン結合型GDHは、マルトース、D-ガラクトース、D-キシロースに対する反応性が極めて低い。具体的には、D-グルコースに対する反応性を100%とした場合に、マルトース、D-ガラクトースおよびD-キシロースに対する反応性がいずれも2%以下であることを特徴とする。本発明のフラビン結合型GDHは、このような高い基質特異性を有するため、マルトースを含む輸液の投与を受けている患者や、ガラクトース負荷試験およびキシロース吸収試験を実施中の患者の試料についても、測定試料に含まれるマルトース、D-ガラクトース、D-キシロース等の糖化合物の影響を受けることなく、正確にD-グルコース量を測定することが可能となる。
本発明のフラビン結合型GDHとして好ましい酵素の例としては、以下の酵素化学的特徴を有するものが挙げられる。
(1)作用:電子受容体存在下でGDH活性を示す
(2)分子量:タンパク質のポリペプチド鎖部分の分子量が約80kDaである
(3)基質特異性:D-グルコースに対する反応性と比較して、マルトース、D-ガラクトース、D-キシロースに対する反応性が低い
(4)至適pH:pH6.5~7.0
(5)至適温度:37~40℃
(6)安定pH範囲:pH3.5~7.0
(7)熱安定性:40℃、15分間の熱処理後に80%以上の残存活性を有する
(8)フラビン化合物を補酵素とする
(9)Km値:D-グルコースに対するKm値が26~33mMである
上記のような酵素化学的特徴を有するGDHであれば、測定試料に含まれるマルトース、D-ガラクトース、D-キシロース等の糖化合物の影響を受けることなく、正確にD-グルコース量を測定することが可能となる。また、血糖値の測定等の臨床診断に応用するために好適なpH範囲、温度範囲で良好に作用するので、診断用測定試薬等の用途に好適に使用することができる。
なお、上記の諸性質パラメータは典型的な例であるが、所定の測定条件においてD-グルコースの測定を行う際に本発明の効果を達成可能である範囲で、上記パラメータのうちには、許容可能な変動の幅を有するものがある。例えば、安定pH範囲や至適pH範囲、至適温度範囲等のパラメータは、所定の測定条件を含む範囲で、上記の典型的な範囲よりやや広くてもよいし、逆に、上記の典型的な範囲よりやや狭い場合でも、測定条件において十分な活性および/または安定性が確保されていればよい。Km値は一般的には小さくなるほど基質特異性が良いとされるが、本発明の酵素としては、所定の測定条件において実質的に十分な基質の選択が実現される範囲の値を有していればよい。
精製酵素とは、当該酵素以外の成分、特に当該酵素以外のタンパク質(夾雑タンパク質)を実質的に含まない状態に分離された酵素をいう。具体的には、例えば、夾雑タンパク質の含有量が重量換算で全体の約20%未満、好ましくは約10%未満、更に好ましくは約5%未満、より一層好ましくは約1%未満である。なお、本明細書中に後述する「MpGDH」、「MjGDH」および「McGDH」は、特に断りの無い限り、精製酵素をいう。
本発明のフラビン結合型GDHとして好ましい酵素の例としては、D-グルコースに対するKm値が26~33mMであるフラビン結合型GDHが挙げられる。
本発明のフラビン結合型GDHは、電子受容体存在下でグルコースの水酸基を酸化してグルコノ-δ-ラクトンを生成する反応を触媒する。
したがって、この原理を利用して、例えば、電子受容体としてフェナジンメトサルフェート(PMS)および2,6-ジクロロインドフェノール(DCIP)を用いた以下の測定系により、本発明のフラビン結合型GDHの活性を測定することができる。
(反応1) D-グルコ-ス + PMS(酸化型)
→ D-グルコノ-δ-ラクトン + PMS(還元型)
(反応2) PMS(還元型) + DCIP(酸化型)
→ PMS + DCIP(還元型)
具体的には、本発明において、フラビン結合型GDHの活性は、以下の手順に従って測定する。100mM リン酸緩衝液(pH7.0) 1.79mL、1.25M D-グルコース溶液 0.08mLおよび20mM DCIP溶液 0.01mLを混合し、37℃で5分間保温する。次いで、20mM PMS溶液 0.02mLおよび酵素サンプル溶液0.1mLを添加し、反応を開始する。反応開始時、および、経時的な吸光度を測定し、酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量(ΔA600)を求め、次式に従いフラビン結合型GDH活性を算出する。この際、フラビン結合型GDH活性は、37℃において濃度50mMのD-グルコース存在下で1分間に1μmolのDCIPを還元する酵素量を1Uと定義する。
上記の特徴を有する本発明のフラビン結合型GDHは、ケカビ亜門、好ましくは、ケカビ綱、より好ましくは、ケカビ目、さらに好ましくは、ケカビ科に分類される微生物から得ることができる。ケカビ亜門、好ましくは、ケカビ綱、より好ましくは、ケカビ目、さらに好ましくは、ケカビ科に分類される微生物としては、例えば、Mucor属、Absidia属、Actinomucor属等が挙げられる。Mucor属に分類される微生物であって、本発明のフラビン結合型GDHを生産する具体的な好ましい微生物の例としては、Mucor prainii、Mucor javanicusもしくはMucor circinelloides f. circinelloidesが挙げられる。より具体的には、Mucor prainii NISL0103、Mucor javanicus NISL0111もしくはMucor circinelloides f. circinelloides NISL0117が挙げられる。Absidia属に分類される微生物であって、本発明のフラビン結合型GDHを生産する具体的な好ましい微生物の例としては、Absidia cylindrospora、Absidia hyalosporaを挙げることができる。より具体的には、Absidia cylindrospora NISL0211、Absidia hyalospora NISL0218を挙げることができる。Actinomucor属に分類される微生物であって、本発明のフラビン結合型GDHを生産する具体的な好ましい微生物の例としては、Actinomucor elegansを挙げることができる。より具体的には、Actinomucor elegans NISL9082を挙げることができる。なお、上記の菌株はNISL(財団法人 野田産業科学研究所)の保管菌株であり、所定の手続きを経ることにより、分譲を受けることができる。
本発明のフラビン結合型GDHは、配列番号1又は配列番号3で示されるアミノ酸配列、又は該アミノ酸配列と80%以上相同なアミノ酸配列、又は該アミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列を有することを特徴とする。配列番号1又は配列番号3で示されるアミノ酸配列を有するフラビン結合型GDHは、上述の各種の性質を有する。また、配列番号1又は配列番号3で示されるアミノ酸配列と80%以上の相同性、好ましくは85%、より好ましくは90%、最も好ましくは95%以上の相同性を有するアミノ酸配列を有し、配列番号1又は配列番号3で示されるアミノ酸配列を有するフラビン結合型GDHと同様な諸性質を有するGDHも、本発明のフラビン結合型GDHに含まれる。
本発明のフラビン結合型GDHをコードする遺伝子とは、配列番号1又は配列番号3で示されるアミノ酸配列、又は該アミノ酸配列と80%以上相同なアミノ酸配列、又は該アミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列を有するフラビン結合型GDHをコードするDNAをいう。または、本発明のフラビン結合型GDHをコードする遺伝子とは、配列番号2又は配列番号4で示される塩基配列からなるDNAをいう。あるいは、本発明のフラビン結合型GDHをコードする遺伝子とは、配列番号2又は配列番号4で示される塩基配列と80%以上の相同性、好ましくは85%、より好ましくは90%、最も好ましくは95%以上の相同性を有する塩基配列を有し、且つフラビン結合型GDH酵素活性をもつタンパク質をコードするDNAをいう。
本発明のフラビン結合型GDHをコードする遺伝子は、適当な公知の各種ベクター中に挿入することができる。さらに、このベクターを適当な公知の各宿主に導入して、フラビン結合型GDH遺伝子を含む組換え体DNAが導入されている形質転換体を作製することができる。これらの遺伝子の取得方法や、遺伝子配列、アミノ酸配列情報の取得方法、各種ベクターの製造方法や形質転換体の作製方法は、当業者にとって公知であり、一例を後述する。
上述のように得られたフラビン結合型GDH遺伝子を、常法により、バクテリオファージ、コスミド、または原核細胞若しくは真核細胞の形質転換に用いられるプラスミド等のベクターに組み込み、各々のベクターに対応する宿主を常法により形質転換または形質導入することができる。宿主としては、例えば、Escherichia属に属する微生物、例えば、大腸菌K-12、好ましくは大腸菌JM109、DH5α(ともにタカラバイオ社製)等が挙げられ、これらの宿主を形質転換して、またはそれらに形質導入してそれぞれの菌株を得る。こうして得られた上記形質転換体を培養することによって、フラビン結合型GDHを大量に生産することができる。
本発明のフラビン結合型GDHは、各種公知の酵素生産方法を用いて製造することができる。例えば、前述のフラビン結合型GDH生産微生物を培地中で培養して目的とするフラビン結合型GDHを産生させ、培養物あるいは培養菌体内部より酵素を採取することができる。また、本発明発明のフラビン結合型GDH遺伝子または該GDH遺伝子を含む組換え体DNAを包含し、フラビン結合型GDH生産能を有する微生物を培養し、該培養物よりフラビン結合型グルコースデヒドロゲナーゼを採取することができる。
微生物の培養は、通常の固体培養法で培養してもよいが、可能なかぎり液体培養法を採用して培養するのが好ましい。培養に用いる培地は、炭素源、窒素源、無機物、その他の栄養素を適宜含有するものであればよく、また、合成培地、天然培地の何れでもよく、目的の酵素を効率よく製造することのできる培地であれば、如何なる培地でもよい。
その他にも、添加することにより本発明のフラビン結合型GDH製造量を向上させることができる栄養源や成分があれば、単独で、あるいは組み合わせて用いてもよい。
1.GDH生産菌のスクリーニング
自然界から分離した菌株および微生物保存機関((財)野田産業科学研究所)から分与された保存菌約500株から、GDH生産菌のスクリーニングを行った。マルツエキス培地(マルツエキス2.0%、D-グルコース2.0%、ポリペプトン0.1%、pH6.0)3mlに供試菌株をそれぞれ接種し、3~5日、30℃で振とう培養した。この培養液を800×g、10分間遠心分離して菌体を沈殿として得た。その後、この菌体を10mM 酢酸緩衝液(pH5.0)中に懸濁し、ビーズショッカー(安井器械(株)製)により菌体を破砕(2,000rpm、60秒、16回)し、4℃、20,000×g、10分間の遠心によって回収された上清を粗酵素液とした。
以下の手順に従って各溶液を混合し、吸光度を測定して、粗酵素液中のGDH活性を調べた。100mM リン酸緩衝液(pH7.0) 1.79mL、1.25M D-グルコース溶液 0.08mLおよび20mM DCIP溶液 0.01mLを混合し、37℃で5分間保温後、20mM PMS溶液 0.02mLおよび酵素サンプル溶液0.1mLを添加し、反応を開始した。反応開始時から酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量(ΔA600)を測定し、次式に従いGDH活性を算出した。この際、GDH活性は、37℃において濃度50mMのD-グルコ-ス存在下で1分間に1μmolのDCIPを還元する酵素量を1Uと定義した。
前培養用培地(イーストエキス 2.0 %、グルコース 4%、pH6.0)0.1Lを0.5L容坂口フラスコに入れ、予めプレート培地上で培養したMucor prainii NISL0103、Mucor javanicus NISL0111、Mucor circinelloides f. circinelloides NISL0117を、約1cm2分それぞれ接種し、30℃、130rpmで2日間回転振とう培養した。これを種培養として、30L容ジャーファメンターに入れた上記培地20Lに0.2Lずつ接種し(ジャーファメンター2基)、30℃、200rpm、0.5vvmで3日間培養した。培養終了後、培養液40Lをろ布でろ過し、菌体を回収した。次いで、得られた菌体を10mM 酢酸緩衝液(pH5.0)に懸濁した。
この上清を、緩衝液A(10mM 酢酸緩衝液、2M 硫酸アンモニウム、pH5.0)にて予め平衡化したブチルトヨパール650C(東ソー社製)カラム(26φ×28.5cm)にかけ、緩衝液Aから緩衝液B(10mM 酢酸緩衝液、pH5.0)のリニアグラジエントによって溶出させた。溶出された活性画分をセントリコンプラス-70(ミリポア社製)で濃縮後、緩衝液C(10mM 酢酸緩衝液、pH4.5)で透析し、予め緩衝液Cで平衡化したSPセファロースFastFlow(GEヘルスケア社製)カラム(26φ×28.5cm)にかけ、緩衝液Cから緩衝液D(10mM 酢酸緩衝液、200mM 塩化カリウム、pH4.5)のリニアグラジエントで溶出させた。溶出された活性画分を濃縮し、精製酵素を得た。
以降、各精製酵素について、Mucor prainii NISL0103由来のGDHをMpGDH、Mucor javanicus NISL0111由来のGDHをMjGDH、Mucor circinelloides f. circinelloides NISL0117由来のGDHをMcGDHと表記する。
実施例2により得られた各精製GDHの諸性質を調べた。
(a)吸収スペクトルの測定
MpGDH、MjGDHおよびMcGDHを10mM 酢酸緩衝液(pH5.0)で透析し、250-800nmにおける吸収スペクトルを分光光度計U-3010(日立ハイテクノロジーズ社製)により測定した。測定結果を図1に示す(図1(A)はMpGDHの吸収スペクトル、図1(B)はMjGDHの吸収スペクトル、図1(C)はMcGDHの吸収スペクトルを示す)。いずれのGDHについても、波長340~350nm付近および波長420~430nm付近に極大を示す2つのピークが確認され、このような吸収スペクトルの形状がフラビン酵素に特有の形状であることから、本発明のGDHがフラビン結合型タンパク質であることが強く示唆された。
実施例2により得られたMpGDH、MjGDHおよびMcGDHと、Aspergillus niger由来の市販のグルコースオキシダーゼ(GOD、biozyme laboratories社製)を用いて、GDH活性およびGOD活性を測定した。結果を表2に示す。
GDH活性は、実施例1の方法に準じ、GOD活性は、4-アミノアンチピリン(4-AA)、およびN-エチルーN-(2-ヒドロキシー3-スルホプロピル)-3-メチルアニリン(TOOS)を使用した以下の方法で測定した。100mM リン酸緩衝液(pH7.0) 30.0mL、833mM D-グルコース溶液 6.0mL、25mM 4-AA溶液 0.3mL、40mM TOOS溶液 0.3mLおよび500U/mL POD溶液 0.3mLを混合後、3.0mLを試験管に移し、37℃で5分間保温後、酵素サンプル溶液0.1mLを添加して反応を開始した。酵素反応の進行に伴う555nmにおける吸光度の1分間あたりの増加量(ΔA555)を測定し、次式に従ってGOD活性を算出した。この際、GOD活性は、37℃において濃度131mMのD-グルコース存在下で1分間に1μmolのH2O2を生成する酵素量を1Uと定義した。
上記の本発明のフラビン結合型GDHにおける至適pHを調べた。結果を図2に示す(図2(A)はMpGDH、(B)はMjGDH、(C)はMcGDHの結果を示す)。具体的には、100mM 酢酸カリウム緩衝液(pH5.0-5.5、図中△印でプロット)、100mM MES-NaOH緩衝液(pH5.5-6.5、図中斜め四角印でプロット)、100mM リン酸カリウム緩衝液(pH6.0-8.0、図中丸印でプロット)、100mM Tris-HCl緩衝液(pH7.5-9.0、図中×印でプロット)を用い、それぞれのpHにおいて、温度37℃にて酵素反応を行い、相対活性を比較した。
その結果、上記のフラビン結合型GDHはいずれも、pH6.5もしくはpH7.0において最も高い活性を示し、pH7.0付近に至適pHを有していた。個別にみると、MpGDHおよびMcGDHの相対活性が最も高かったのはpH7.0においてであり、その周辺域として、pH6.5-7.5の範囲で最大相対活性値の80%以上を示したことから、この範囲で好適に使用できると考えられた。また、MjGDHの相対活性が最も高かったのはpH6.5においてであり、その周辺域として、pH6.0-7.0の範囲で最大相対活性値の80%以上を示したことから、この範囲で好適に使用できると考えられた。
実施例2記載の活性測定法に準じ、種々の温度にて本酵素の活性測定を行った。具体的には、100mM リン酸緩衝液(pH7.0) 30.0mL、833mM D-グルコース溶液 6.0mL、25mM 4-AA溶液 0.3mL、40mM TOOS溶液 0.3mLおよび500U/mL POD溶液 0.3mLを混合後、3.0mLを試験管に移し、37℃で保温する代わりに各温度で5分間保温し、次いで、20mM PMS溶液0.02mLおよび酵素サンプル溶液0.1mLを添加し、各温度にて反応を開始した。反応開始時、および、2分後の吸光度を測定し、酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量を求めた。結果を図3に示す(図3(A)はMpGDH、(B)はMjGDH、(C)はMcGDHの結果を示す)。いずれも37℃付近で最大の活性を示し、最大活性に対して80%以上の活性を示す温度範囲は30~40℃であった。以上より、本発明のフラビン結合型GDHの至適温度範囲は、30~40℃、最も好ましい温度は37℃であると考えられた。
前記活性測定法において、基質であるD-グルコース濃度を変化させて活性測定を行い、ラインウェーバー・バークプロットから、ミカエリス定数(Km)を求めた。この結果、D-グルコースに対するKm値は、MpGDHで31.1mM、MjGDHで26.4mM、McGDHで33.2mMであった。
100mM 酢酸カリウム緩衝液(pH5.0)を用いて、本発明のフラビン結合型GDHを各温度で15分間処理した時の熱安定性の結果を図4に示す(図4(A)はMpGDH、(B)はMjGDH、(C)はMcGDHの結果を示す)。本発明のフラビン結合型GDHは、40℃、15分間の熱処理後に80%以上の残存活性を有しており、約40℃まで安定であることがわかった。
本発明のフラビン結合型GDHにおける安定pHを調べた。結果を図5に示す(図5(A)はMpGDH、(B)はMjGDH、(C)はMcGDHの結果を示す)。具体的には、100mM グリシン-HCl緩衝液(pH2.5-3.5、図中四角印でプロット)、100mM 酢酸カリウム緩衝液(pH3.5-5.5、図中△印でプロット)、100mM MES-NaOH緩衝液(pH5.5-6.5、図中斜め四角印でプロット)、100mM リン酸カリウム緩衝液(pH6.0-8.0、図中丸印でプロット)、100mM Tris-HCl緩衝液(pH7.5-9.0、図中×印でプロット)を用い、それぞれのpHにおいて25℃で16時間処理した後、本発明のフラビン結合型GDHの残存活性をそれぞれ測定した。その結果、いずれも、最大の残存活性を示したpH5.0付近での活性に対し80%以上の活性を示すpH範囲はpH3.5-7.0であった。以上から、本発明のフラビン結合型GDHの安定pH範囲はpH3.5-7.0であることが判った。
スーパーセップエース 10-20%(和光純薬工業社製)を用いたSDS-ポリアクリルアミド電気泳動によりMpGDH、MjGDHおよびMcGDHの分子量を求めた。また、脱糖鎖キット(Enzymatic Deglycosylation Kit、PZM製)を用いて、本発明の各フラビン結合型GDHを脱糖鎖処理し、同様に電気泳動に供した。結果を図6に示す。泳動サンプルは以下の通りである。

実施例1の酵素活性測定方法に準じ、基質としてはD-グルコース、マルトース、D-ガラクトース、D-キシロース、マンノース、スクロース、トレハロース、マルトトリオース、マルトテトラオースをそれぞれ用いて、各基質に対する本発明のフラビン結合型GDHの活性を測定した。基質濃度は50mMとした。結果を表3に示す。
本発明のフラビン結合型GDHの活性に対する1,10-フェナントロリンの阻害効果を、以下の方法で調べた。実施例1の酵素活性の測定方法に準じ、ただし、終濃度が1mM、5mM、10mM、25mMおよび50mMとなるように1,10-フェナントロリンを添加した場合の酵素活性を求め、1,10-フェナントロリン無添加の阻害率を0%として、その阻害率を計算した。結果を表4に示す。
本発明のフラビン結合型GDHを用いて、グルコースの測定を行った。具体的には、100mM リン酸緩衝液(pH7.0) 1.79mL、D-グルコース溶液(250、750、1,250、1,750、2,500、3,250、4,000、5,000mg/dL) 0.08mLおよび20mM DCIP溶液 0.01mLを混合し、37℃で5分間保温後、20mM PMS溶液 0.02mLおよび0.8U/mL GDH溶液0.1mLを添加し、反応を開始した。酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量(ΔA600)とグルコースの終濃度の関係を図7に示す(図7(A)はMpGDHを用いた測定結果、(B)はMjGDHを用いた測定結果、(C)はMcGDHを用いた測定結果を示す)。
100mM リン酸緩衝液(pH7.0) 1.77mL、D-グルコース溶液(10,000、16,000mg/dL) 0.02mLおよび20mM DCIP溶液 0.01mLを混合した。続いて、マルトース溶液(3,000、6,000、9,000、12,000、15,000mg/dL)もしくはD-ガラクトース溶液(1,500、3,000、4,500、6,000、7,500mg/dL)もしくはD-キシロース溶液(1,000、2,000、3,000、4,000、5,000mg/dL) 0.08mLを添加して37℃で5分間保温した後、20mM PMS溶液 0.02mLおよび2.0U/mL GDH溶液0.1mLを添加し、反応を開始した。酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量(ΔA600)、およびグルコースの終濃度の関係を表5~7に示す。
100mM リン酸緩衝液(pH7.0) 1.61mL、D-グルコース溶液(10,000、16,000mg/dL) 0.02mLおよび20mM DCIP溶液 0.01mLを混合した。続いて、マルトース溶液(3,000、6,000、9,000、12,000、15,000mg/dL)、D-ガラクトース溶液(1,500、3,000、4,500、6,000、7,500mg/dL)およびD-キシロース溶液(1,000、2,000、3,000、4,000、5,000mg/dL) をそれぞれ0.08mL添加して37℃で5分間保温した後、20mM PMS溶液 0.02mLおよび2.0U/mL の本発明のフラビン結合型GDH溶液0.1mLを添加し、反応を開始した。酵素反応の進行に伴う600nmにおける吸光度の1分間あたりの減少量(ΔA600)とグルコースの終濃度の関係を表8~9に示した。
(1)mRNAの調製
Mucor prainii NISL0103をマルツエキス培地(マルツエキス 2.0%、グルコース 4.0%、ポリペプトン 0.1%、pH6.0)3mLに接種し、2日、30℃で振とう培養した。この培養液を濾紙濾過し、菌糸体を回収した。得られた菌糸を液体窒素中で凍結させ、乳鉢を用いて菌糸を粉砕した。次いで、ISOGEN(ニッポンジーン社製)を用いて、本キットのプロトコールに従って、粉砕した菌体からmRNAを得た。
実施例2より得られたMpGDHをスーパーセップエース 10-20%(和光純薬工業社製)に供し、電気泳動を行った。電気泳動後のゲルをQuick-CBB(和光純薬工業社製)を用いて染色し、当該酵素の分子量に相当するバンド部分を切り出した。切り出したゲル断片を外部機関に委託し、その中に含まれるタンパク質の内部アミノ酸配列情報を取得した。その結果、得られたアミノ酸配列は、LVENFTPPTPAQIE(配列番号5)およびIRNSTDEWANYY(配列番号6)であった。
上記の部分アミノ酸配列情報に基づき、ミックス塩基を含有するディジェネレートプライマー(プライマーの一例を配列番号7(フォワードプライマー)、配列番号8(リバースプライマー)に示す)を作製した。なお、配列番号7および8に記載の1文字表記において、混合塩基はそれぞれ、h=a+c+t、r=a+g、y=c+t、d=a+g+tを表す。上記(1)にて調製したMucor prainii NISL0103のmRNAをテンプレートとし、PrimeScript RT-PCR Kit(タカラバイオ社製)を用いて、本キットのプロトコールに従ってRT-PCRを行った。逆転写反応には本キット付属のオリゴdTプライマーを、PCRによるcDNA増幅には配列番号7、8に示すディジェネレートプライマーを用いた。反応液をアガロースゲル電気泳動に供したところ、800bp程度の長さに相当するシングルバンドが確認された。このバンドに含まれる増幅DNA断片を精製し、Ligation Convenient Kit(ニッポンジーン社製)を用いて、pT7Blue(ノバジェン社製)に前記増幅DNA断片をライゲーションすることにより、組換えプラスミドpTMGD-1を構築した。
N末領域のプライマー(配列番号15)およびC末領域のプライマー(配列番号16)を作製し、これらのプライマーおよび上記(1)にて調製したMucor prainii NISL0103のmRNAを用いて、RT-PCRを行った。
反応液をアガロースゲル電気泳動に供したところ、約2kbp程度の長さに相当するシングルバンドが確認された。このバンドに含まれる増幅DNA断片を精製し、制限酵素SmaIで消化したプラスミドpUC19(タカラバイオ社製)とライゲーションを行って、組換えプラスミドpuc-MGDを構築した。
この培養液を氷上で冷却下、超音波破砕器(Ultrasonicgenerator、Nissei社製)を用いて20秒間、4回処理し、破砕した。破砕液をエッペンドルフチューブに入れ、微量遠心機を用い、12,000rpmで10分間遠心分離後、上清画分を別のエッペンドルフチューブに移しかえ、粗酵素液とした。前述の酵素活性測定法によりこの粗酵素液中のGDH活性を測定したところ、本発明のフラビン結合型GDH活性が確認された。
Double-joint PCR(Fungal Genetics and Biology,2004年,第41巻,p973-981)を行い、5’アーム領域~PyrG遺伝子(ウラシル栄養要求性マーカー)~TEF1プロモーター遺伝子~フラビン結合型GDH遺伝子~3’アーム領域から成るカセットを構築し、Aspergillus sojae KK1-2株を宿主に、プロトプラスト-PEG法により形質転換を行った。得られた菌株の中から目的の形質転換体を、PCRで確認して選抜した。
培養後のふすま麹2gに対し、5倍量の10mM 酢酸緩衝液(pH5.0)を加え、ポリトロンホモジナイザーPT3000(キネマチカ社製)で30秒×8回破砕を行った。破砕後、14,000rpm、30分間遠心分離を行い、得られた上清画分を粗酵素液とした。前述の酵素活性測定法により、この粗酵素液中のGDH活性を測定したところ、コントロール株を用いて得られた粗酵素液中のGDH活性は0.3U/mLであったのに対し、形質転換体を用いて得られた粗酵素液中のGDH活性は14.0U/mLであり、形質転換体において本発明のフラビン結合型GDHが発現されていることが確認された。

- 以下の(i)から(iii)の性質:
(i)作用:電子受容体存在下でグルコース脱水素酵素活性を示す、
(ii)分子量:タンパク質のポリペプチド鎖部分の分子量が約80kDaである、
(iii)基質特異性:D-グルコースに対する反応性に対して、マルトース、及びD-ガラクトース、及びD-キシロースに対する反応性が低い、
を備えるフラビン結合型グルコースデヒドロゲナーゼ。 - D-グルコースに対する反応性を100%とした場合に、マルトース、D-ガラクトース、およびD-キシロースに対する反応性がいずれも2%以下である請求項1記載のフラビン結合型グルコースデヒドロゲナーゼ。
- 以下の(a)から(c)の糖化合物:
(a)マルトース
(b)D-ガラクトース
(c)D-キシロース
の1以上が存在する場合のD-グルコースに対する反応性が、前記(a)~(c)が存在しない場合のD-グルコースに対する反応性を100%とした場合の96%~104%である、請求項1~2に記載のフラビン結合型グルコースデヒドロゲナーゼ。 - 至適pHがpH6.5~7.0であり、至適温度が37~40℃であり、安定pH範囲がpH3.5~7.0であり、40℃、15分間の熱処理後に80%以上の残存活性を有する、請求項1~3のいずれか一項に記載のフラビン結合型グルコースデヒドロゲナーゼ。

- Mucor属に分類される微生物に由来する請求項5記載のフラビン結合型グルコースデヒドロゲナーゼ。
- Mucor属に分類される微生物を培地に培養し、該微生物菌体からフラビン結合型グルコースデヒドロゲナーゼを採取することを特徴とする請求項1~6のいずれか一項に記載のフラビン結合型グルコースデヒドロゲナーゼの製造方法。
- 微生物がMucor prainii、Mucor javanicus、Mucor circinelloides f.circinelloidesから選択される1以上である請求項7記載のフラビン結合型グルコースデヒドロゲナーゼの製造方法。
- 配列番号1又は配列番号3で示されるアミノ酸配列、又は該アミノ酸配列と80%以上相同なアミノ酸配列、又は該アミノ酸配列において1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列からなるを有することを特徴とする、請求項1~6のいずれか一項に記載のフラビン結合型グルコースデヒドロゲナーゼ。
- 以下の(A)~(E)からなる群より選択されるいずれかのDNAからなるフラビン結合型グルコースデヒドロゲナーゼ遺伝子:
(A)配列番号1で示されるアミノ酸配列をコードするDNA;
(B)配列番号2で示される塩基配列からなるDNA;
(C)配列番号3で示されるアミノ酸配列をコードするDNA;
(D)配列番号4で示される塩基配列からなるDNA;
(E)配列番号2又は配列番号4で示される塩基配列と80%以上相同な塩基配列を有し、且つフラビン結合型グルコースデヒドロゲナーゼ酵素活性をもつタンパク質をコードするDNA。 - 請求項10記載のフラビン結合型グルコースデヒドロゲナーゼ遺伝子をベクターDNAに挿入したことを特徴とする組換え体DNA。
- 請求11記載の組換え体DNAが導入されている形質転換体。
- 請求項10記載のフラビン結合型グルコースデヒドロゲナーゼ遺伝子または請求11記載の組換え体DNAを含み、フラビン結合型グルコースデヒドロゲナーゼ生産能を有する微生物を培養し、該培養物よりフラビン結合型グルコースデヒドロゲナーゼを採取することを特徴とする、D-グルコースに対する反応性に対して、マルトース、D-ガラクトース、D-キシロースに対する反応性が低いフラビン結合型グルコースデヒドロゲナーゼの製造方法。
