Description
- Piperazinecarbonzuurderivaat, bereiding ervan en farmaceutische preparaten die het bevatten -
De Britse octrooiaanvrage 2.157.685 beschrijft op de 4-plaats gesubstitueerde 2-piperazinecarbonzuren die een werking op het centrale zenuwstelsel hebben.
Gevonden is, dat een verbinding van deze 5 groep, die tot nu toe niet is beschreven, een bijzonder sterke werking op het centrale zenuwstelsel heeft, goed wordt getolereerd en een langdurende werking heeft.
De uitvinding heeft betrekking op /R-(E).7-4-(3-fosfono-2-propenyl)-2-piperazinecarbonzuur met formule 1, 10 farmaceutisch aanvaardbare zouten ervan, een werkwijze voor de bereiding ervan, farmaceutische preparaten die het bevatten en de toepassing ervan als een farmacon.
De verbinding met formule 1 kan worden bereid door dealkylering van een verbinding met formule 2, waarin R een 15 alkylgroep met 1-4 koolstofatomen of een arylalkylgroep met 1-4 koolstof atomen in de alkylgroep is.
De werkwijze kan op bekende wijze worden uitgevoerd . De dealkylering kan bijvoorbeeld worden bewerkstelligd door silylering gevolgd door hydrolyse van de verkregen silylester.
20 De silylering kan worden uitgevoerd met bijvoorbeeld broomtrimethyl-silaan in een organisch oplosmiddel zoals dichloormethaan of chloroform bij kamertemperatuur. De daarop volgende hydrolyse kan onder milde omstandigheden worden bewerkstelligd.
Wanneer R een arylalkylgroep met 1-4 koolstofato-25 men in de alkylgroep is, is de arylrest geschikt een eventueel gesubstitueerde fenylgroep, waarbij geschikte substituenten onder meer lage alkyl- of lage alkoxygroepen zijn.
De verbinding met de formule 1,met inbegrip van de zouten ervan, kan ook in de vorm van hydraten, in het bijzonder het 30 monohydraat worden verkregen, De hydraten, in het bijzonder het mono-hydraat, vormen ook een deel van de uitvinding.

De verbinding met formule 1 kan kationische zouten en zuuradditiezouten vormen. Dergelijke zouten worden gemakkelijk met standaardmethoden bereid. Kationische zouten zijn 5 bijvoorbeeld ammonium-, natrium-, kalium-, calcium-, piperidinium-, morfolinium- of pyrrolidiniumzouten. Zuuradditiezouten zijn bijvoorbeeld de zouten die worden gevormd met zoutzuur, broomwater-stofzuur, zwavelzuur, methaansulfonzuur, benzeensulfonzuur, p-tolueensulfonzuur en trifluorazijnzuur.
^0 De hydraten, in het bijzonder het monohydraat, van de verbinding met formule 1 hebben bijzonder belangwekkende fysisch-chemische en andere eigenschappen, bijvoorbeeld wat betreft oplosbaarheid, zuiveringsgemak en stabiliteit. Het hydraat kan worden verkregen door kristallisatie van de verbinding met ^ formule 1 uit een waterig milieu, bijvoorbeeld zoals beschreven in het voorbeeld onder e), bijvoorbeeld uit water/methanol.
In het volgende voorbeeld zijn de temperaturen 20 en de -waarden niet gecorrigeerd.
20 Voorbeeld : /R-(E)j/-4-(3-fosfono-2-propenyl)- 2-piperazinecarbonzuur a) (R)-1,4-Bis(fenylmethyl)-2-piperazinecarbon- zure (1R,2S,5R)-5-methy1-2-(1-methylethyl)cyclohexyblester 25
Aan een oplossing ran 761 g 1,4-dibenzyl-2- piperazinecarbonzure ethylester (bereid volgens E. Jucker en E.Rissi,
Helv. Chim. Acta 45 (1962) 2383) en 458 g (-)-menthol wordt bij 45°C 15 g NaH (55-60% dispersie ) en 1 1 tolueen toegevoegd.
^ Ongeveer 700 ml van het oplosmiddel wordt vervolgens langzaam door destillatie verwijderd en continu vervangen door nieuwe porties tolueen ( de reactie wordt met TLC, ethylacetaat/hexaan 1:3 gevolgd).
Het mengsel wordt tot kamertemperatuur afgekoeld, met 1,25 1 2N waterig HC1 en 6 1 diethylether behandeld en 1 uur krachtig geroerd. Het kristallijne neerslag wordt afgefiltreerd en met di-35 ethylether en 0,1N waterig HC1 uitgewassen. Het ruwe produkt wordt herkristalliseerd uit ethanol/0,2N waterig HCl, waardoor het 20 hydrochloridehydraat van de titelverbinding, = +18,5® (c=1,2 in .8800412 * - 3 - - % CHC13) wordt verkregen, dat zonder verdere zuivering in de volgende stap wordt gebruikt.
b) (R)—2-Piperazineearbdnzure (1R,2S,5R)-5-methyl- * 2-(1-methylethyl)cyclohexylester 210 g Van het produktvan stap a) in 2 1 ethanol en 10,5 g Pd/C (10%) wordt 7 uur bij kamertemperatuur en normale druk gehydrogeneerd, daarna gefiltreerd en onder vacuum ingedampt. Het residu wordt behandeld met ethanolisch HC1 , waarna het 10 neerslag wordt gefiltreerd en uitgewassen met ethanol/diethylether (1:1). Het produkt wordt herkristalliseerd uit water/methanol/ethyl- acetaat onder verkrijging van het dihydrochloride van de titelver- binding , smpt. 225-226^0,^0(^ =-52,0° (c=1,34 in water).
_ Het dihydrochloride wordt behandeld met diethylether/ammonia, waarna 15 de organische fase wordt drooggedampt onder verkrijging van de titel- 20 verbinding, smpt. 50-52 °C, /T|%2-q ~ -56,7° (c= 1,05 in CHCl^).
c) ZR-(E)2~4-(3-Diethoxyfosfinyl-2-propenyl)-2-piperazinecarbonzure (1R,2S,5R)-5-methyl-2-(1-methylethyl)cyclohexyl-ester
Aan een oplossing van 11,3 g van de vrije base van stap b) en 5,9 ml triethylamine in 90 ml tetrahydrofuran wordt bij -30°C binnen 30 min 10,7 g 3-broom-propen-2-yl-fosfonzure 2^ diethylester (bereid volgens K. Hemmi, H.Takeno, M. Hashimoto en T.Kamiya, Chem. Pharm. Buil 30 (1982) ,111) in 45 ml tetrahydrofuran onder roeren toegevoegd. Het roeren wordt 20 uur bij -25°C voortgezet. Na filtratie van het neerslag wordt de oplossing bij verminderde druk ingedampt, waarna de verkregen siroop wordt ge-3q chromatografeerd op silicagel met dichloormethaan onder toevoeging van een toenemende concentratie van een 1:19 geconcentreerd ammonia/ ethanolmengsel tot 10% na 2 uur. De met dichloormethaan/geconcen-treerd ammonia/ethanol 200:1:19 geelueerde fraetie (Rf = 0,35) wordt gewonnen en onder vacuum ingedampt onder verkrijging van de titel- O Λ 35 verbinding als een olie, 0^0.^ = -56,0° (c= 1,3 in 2N HC1). Door behandeling van het produkt met ethanolisch HCl/ether wordt - -20 het dichlonde verkregen, smpt. 157-163°C>/(X/d = -48,2° (c= 1,4 in 2N HC1).
.8800412 4 - 4 - 4 d) /R-(E)J-4-(3-Dlethoxyfosfinyl-2-propeny1)-2- piperazinecarbonzuur
Aan een oplossing van 5,79 g van het produkt van stap c) in 58 ml absoluut CE^Cl^ wordt bij -30 °C binnen 30 min 5 22,8 ml van een oplossing van boriumtrichloride in 1,2-dichloor- ethaan ( ongeveer 2,2 M) onder roeren toegevoegd. Het reactiemengsel wordt 1 uur bij -25°C en 3,5 uur bij 0°C geroerd. Bij 0°C wordt 50 ml water toegevoegd , waarna het mengsel wordt geneutraliseerd door toevoeging van 2N natronloog en verdeeld tussen H^O en CH^Cl^· 10 De waterfase wordt onder vacuum drooggedampt, het residu wordt opgenomen in CHCl^, gefiltreerd, gedroogd over Na2S0^ en onder vacuum drooggedampt onder verkrijging van de titelverbinding, welke zonder verdere zuivering in de volgende stap wordt gebruikt.
Een analytisch monster wordt gezuiverd met 15 behulp van HPLC (Nucleosil RP-8, ^O/CH^OH 3:2) onder verkrijging van een schuim -18,0° (c = 1,1 in 0,5 N HC1 ).

De verbinding met formule 1 heeft significante I
remmende werking op de LH- en testosteronsecretie bij een dosis van I
3,2 mg/kg i.p.. (±)-4-(3-Fosfono-2-propenyl)-2-piperazinecarbonzuur I
(hierna CPP-een genoemd), het overeenkomstige racemaat van de I
verbinding met formule 1 heeft, bij 3,2 mg/kg i.p. , geen effect op I
20 de LH-secretie en remt de testosteronsecretie slechts zwak. I
In vrouwtjesratten remt de verbinding met formule 1 de LH-afhankelijke spontane ovulatie in de volgende proef I

Vrouwtjesratten van de Wistarstam (SIV, Kissleg, 22 West-Duitsland, 200-300 g) met regelmatige cycli van 4 dagen ontvangen de proefverbinding tijdens de pro-oestrus om 13.00 en 15.00 uur intraperitoneaal. De volgende dag om 9.00 uur ,vanneer de ratten in . 8 8 0 C ·'. 1 2 * ’ - 6 - de oestrus zijn, worden zij gedood, waarna de eileiders microscopisch worden onderzocht en de eicellen worden geteld. Geoordeeld wordt dat de ovulatie slechts is geremd als geen eicellen worden geteld.
De verbinding met formule 1 ( onderzocht als het monohydraat) remt 5 de spontane ovulatie in belangrijke mate, toegediend in doseringen van 2 x 3,2 mg/kg i.p. ( om 13.00 uur 3,2 mg/kg i.p. en om 15.00 uur 3,2 mg/kg i.p.).CPP-een remt de spontane ovulatie slechts zwak, toegediend in doseringen van 2 x 3,2 mg/kg i.p. onder dezelfde pro efoms t and i gheden.
10 De verbinding met formule 1 wordt dan ook geïndiceerd voor toepassing bij de behandeling van aandoeningen met een etiologie die verband houdt met of geregeld wordt door de LH-secretie of met een etiologie waarbij de fysiologische regulering van de LH-secretie betrokken is, bijvoorbeeld bij de behandeling van 15 prostaathypertrofie, bij de behandeling van menopauze-syndroom, alsmede bij de behandeling van mamma- en prostaatcarcinoom.Voor deze toepassing bedraagt een geïndiceerde dagelijkse dosering ongeveer 1 tot ongeveer 800 mg van de verbinding, doelmatig inverdeelde doses 2 tot 4 maal per dag in een eenheidsdoseringsvorm met bijvoorbeeld 20 ongeveer 0,25 tot ongeveer 400 mg van de verbinding of in een vorm met geleidelijke afgifte.
De verbinding met formule 1 vertoont spierontspannende activiteit bij het bij kennis zijnde konijn in doseringen van 0,01 tot 0,05 mg/kg i.v. Zii.J. Teschendorf et al., Arch. Exp.
25 Pharmacol.266, 467-468 (1970)7. Bii deze proef veroorzaakt het monohydraat van de verbinding met formule 1 een remming van 50% van de spierreflextonus na toediening van 0,02 mg/kg i.v., terwijl CPP-een de spierreflextonus met 47% remt na toediening van 0,5 mg/kg i.v.; de activiteit van de verbinding met formule 1 is derhalve 25 maal 30 zo groot als die van het overeenkomstige racemaat.
De verbinding met formule 1 wordt dan ook geïndiceerd voor toepassing bij de behandeling van verhoogde spier-tonus, bijvoorbeeld bij de behandeling van pijnlijke spierspasma's welke toegeschreven kunnen worden aan statische en functionele aan-35 doeningen van de lenden- of halswervels of na een chirurgische ingreep, of bij de behandeling van spasticiteit, bijvoorbeeld door multiple sclerose, aandoeningen van het ruggemerg, cerebrovasculaire accidenten, .8800412

De verbinding wordt dan ook geïndiceerd voor I
toepassing bij de profylaxe en therapie van aandoeningen die verband I
houden met hersenischemie , bijvoorbeeld beroerte. Voor deze I
toepassing bedraagt een geïndiceerde dagelijkse dosering ongeveer I
25 tot ongeveer 800 mg van de verbinding , doelmatig toegediend in I
30 2 tot 4 doses per dag in eenheidsdoseringsvorm welke bijvoorbeeld I
ongeveer 6 tot ongeveer 400 mg van de verbinding bevat of in een I
vorm voor geleidelijke afgifte. I
Voorts heeft de verbinding met formule 1 een I
krachtige, selectieve en competitieve antagonistische werking op I

NMDA (N-methyl-D-asparaginezuur ) -receptoren. Zo remt het monohydraat I
van de verbinding met formule 1 door NMDA opgewekte depolarisaties I
bij de bepaling aan geïsoleerde ruggemerg van de kikker /JP.L. I
Herrling, Neuroscience J_4 , 417-426 (1985)/ met een pA2_waarde van I
.8800412 - 8 - 6,8 , en CPP-een met een pA£ -waarde van 6,2 ; de verbinding met formule 1 heeft derhalve een ongeveer 4 maal zo krachtige werking als het overeenkomstige racemaat. CPP ^(±)-3-(2-carboxypiperazin-4-yl)propyl-1-fosfonzuur7, de verbinding volgens de Britse octrooi-5 aanvrage 2.157,685, heeft een pA2~waarde van 5,8.
Bij de door NMDA opgewekte natriumeffluxproef met schijfjes rattehersenen (Luini A., Goldberg 0. + Teichberg V.I., Proc. Natl. Acad. Sci. USA _78 , 3250-3254 (1981 )) heeft het monohydraat van de verbinding met formule 1 een pA2~waarde van ^ 6,5 en CPP-een een pA2 van 6,2 ; de verbinding met formule 1 heeft derhalve een ongeveer 2 maal zo krachtige werking als het overeenkomstige racemaat . CPP heeft een pA2~waarde van 6,0.
De selectiviteit van de werking als antagonist van NMDA blijkt uit het feit dat de verbinding met formule 1, even- 1 s . # als CPP-een en CPP , niet actief is bij de door quisqualaat en kainaat opgewekte natriumeffluxproef tot een concentratie van 1 mM.
De verbinding met formule 1 wordt ten gevolge van het NMDA receptorantagonisme geïndiceerd voor toepassing bij 20 .
de behandeling van angst , schizofrenie en depressie of van degene- ratie-aandoeningen van het centrale zenuwstelsel, zoals de ziekten van Huntington, Alzheimer of Parkinson . Voor deze toepassingen bedraagt een geïndiceerde dagelijkse dosering ongeveer 25 tot ongeveer 800 mg van de verbinding,doelmatig toegediend in 2 tot 4 J doses per dag in eenheidsdosermgsvorm,welke bijvoorbeeld ongeveer 6 tot ongeveer 400 mg van de verbinding bevat,of in een vorm voor geleidelijke afgifte.
De verbinding met de formule 1 wordt ten gevolge van het NMDA receptorantagonisme voorts geïndiceerd voor toe-J passing bij de behandeling van tinnitus. Voor deze toepassing bedraagt een geïndiceerde dagelijkse dosering ongeveer 25 tot ongeveer 800 mg van de verbinding, doelmatig toegediend in 2 tot 4 doses per dag in eenheidsdoseringsvorm,welke bijvoorbeeld ongeveer 6 tot ongeveer 400 mg van de verbinding bevat,of in een vorm voor gelei-delijke afgifte.
Bovendien vertoont de verbinding met formule 1 anticonvulsieve werking bij door elektrische schokken opgewekte con- .8800412

Bij deze proef ontvangen groepen van zes mannetjesmuizen (18-26 g, I
OF-t, Sandoz Basel) de proefverbinding intraperitoneaal. Na 1, 2 en I
5 4 uur wordt een 50 mA, 200 ms lange schok toegediend via cornea- I
electroden, ingesmeerd met elektrolytgel. Deze bovenmaximale schok I
geeft tonische extensorconvulsies van alle ledenmaten . Remming van I
het strekken van de achterpoot wordt opgevat als een beschermende I
werking. Na onderzoek van verscheidene dosisniveau’s wordt de I
10 drempelwaarde van de dosis vastgesteld. Na 1 uur bedraagt de drempel- I
waarde, de dosis die nodig is voor het remmen van het strekken I
van de achterpoot , minder dan 1 mg/kg i.p. voor de verbinding met I
formule 1 (monohydraat) en 3-10 mg/kg i.p. voor CPP-een . De I
literatuurwaarde voor de dosisdrempelwaarde van CPP is 10 mg/kg i.p. I

binding met formule 1 een sterkere anticonvulsieve werking dan het I
overeenkomstige racemaat bij onderzoek van deze werking tegen door I
.8800412 - 10 - elektrische schokken opgewekte convulsies bij muizen.
De verbinding met formule 1 remt voorts de door N-methyl-D-asparaginezuur ( NMDA) opgewekte convulsies bij muizen.
Bij deze proef werden groepen van 6 vrouwtjesmuizen (18-26 g, 5 OF- 1 , Sandoz Basel) intraperitoneaal voorbehandeld met de proef- verbinding. 30 Min later worden zij geprikkeld met 400 mg/kg s.c. NMDA in de nek en 30 min geobserveerd. De tijd die verloopt tot het verschijnen van de eerste tekenen van convulsies, tot de eerste tonische convulsies en tot het intreden van de dood worden geno-10 teerd. De significantie van eventuele verschillen wordt beoordeeld met de Mann-Whitney U-test /jS. Siegel, Non-parametric Statistics, McGraw-Hill, New York 1956/. dosis drempelwaarde is de kleinste dosis waarbij er een significante remming van convulsieve symptomen is. De dosis drempelwaarde van de verbinding met formule 1 (mono-15 hydraat) is ongeveer 5 mg/kg i.p. , en van CPP 10 mg/kg i.p.
De verbinding met de formule 1 wordt ten gevolge van de anticonvulsieve activiteit geïndiceerd voor toepassing bij de behandeling van epilepsie .Voor deze toepassing bedraagt een geïndiceerde dagelijkse dosering ongeveer 25 tot ongeveer 800 mg 20 van (feverbinding,doelmatig toegediend in 2 tot 4 doses per dag in eenheidsdoseringsvorm,welke bij voorbeeld ongeveer 6 tot ongeveer 400 mg van de verbinding bevat,of in een vorm voor geleidelijke afgifte.
De verbinding met formule 1 vertoont voorts 25 analgetische activiteit bij de geïsoleerde ruggemergstaart van de rat in vitro onder toepassing van capsaicine (0,8 1 jiM) als een chemische schadelijke stimulus welke depolariserende ventrale wortelresponsen opwekt ZM· Yanagisawa et al., European J.
Pharmacol. 106 , 231-239 (1984)7 . Het monohydraat van de verbinding 30 met formule 1 vermindert de chemische stimulus met 75-80% in een concentratie van 10 jjM.
De verbinding met formule 1 wordt dan ook geïndiceerd voor toepassing voor het verlichten van pijn. Voor deze toepassing bedraagt een geïndiceerde dagelijkse dosering ongeveer 35 25 tot ongeveer 800 mg van de verbinding, doelmatig toegediend in 2 tot 4 doses per dag in eenheidsdoseringsvorm,welke bijvoorbeeld ongeveer 6 tot ongeveer 400 mg van de verbinding bevat,of in een vorm voor .8800412 - 11 -
geleidelijke afgifte. I
Daarnaast hebben de verbinding met formule 1 als- I
mede het overeenkomstige racemaat geen effecten op bloeddruk en I
hartsnelheid in een geanesthetiseerde kat in een dosis tot 10 mg/kg I
5 i.v. Bij proefonderzoeken wat betreft de toxiciteit met honden I
gedurende 4 weken wordt 3 mg/kg/dag i.v. van de verbinding met formule I
1 goed getolereerd. I
Zoals men uit de bovenstaande proefresultaten I
kan zien, gaan de sterke remmende werking op de LH-secretie , de I
10 opmerkelijke (25-voudige) toeneming van de spierontspannende werking I
alsmede de twee- tot viervoudige antagonistische werking voor I
NMDA-receptor van de verbinding met formule 1 , vergeleken met het I
overeenkomstige racemaat, niet gepaard met eajsoortgelijke toeneming I
van de neveneffecten, bijvoorbeeld cardiovasculaire effecten. I

of als farmaceutisch aanvaardbare zouten ervan worden toegediend. I
Dergelijke zouten vertonen dezelfde mate van activiteit als de I
verbinding met formule 1 . I
De uitvinding verschaft voorts farmaceutische I
20 preparaten die de verbinding met formule 1 of een farmaceutisch aan- I
vaardbaar zout daarvan in combinatie, met een farmaceutisch aanvaard- I
bare drager of verdunningsmiddel omvatten. I
De verbinding met formule 1 kan volgens elke I

worden toegediend. De verbinding met formule 1 kan bijvoorbeeld I
voor orale toediening, bijvoorbeeld in de vorm van tabletten of I
capsules, worden gemengd met conventionele farmaceutisch aanvaardbare I
30 excipienten, bijvoorbeeld inerte verdunningsmiddelen, zoals I
lactose, mannitol, calciumsulfaat, microkristallijne cellulose; I
middelen die het uiteenvallen bevorderen, bijvoorbeeld zetmeel, I
natriumcarboxymethylcellulose, natriumcarboxymethylzetmeel, alginezuur, I
crospovidon ; bindmiddelen zoals cellulosederivaten (methyl-, I
35 hydroxymethyl- , hydroxypropylmethyl-), povidon , gelatine; glij- I
middelen zoals siliciumdioxyde , stearinezuur, magnesium- of
calciumstearaat; geharde oliën zoals ricinusolie , glycerolesters I
zoals palmitostearaat en/of smaakstoffen, kleurstoffen en zoetstoffen.

Op de tabletten kan eventueel een laag worden aangebracht met bekende methoden om het uiteenvallen en de absorptie in het maag-darmkanaal te vertragen, waardoor een geleidelijke werking over een langere periode wordt verschaft. Parenterale preparaten zijn bij 5 voorkeur steriele waterige oplossingen voor injectiedoeleinden.
Dergelijke waterige oplossingen moeten zonodig geschikt worden gebufferd en isotonisch gemaakt met voldoende zoutoplossing. Eventueel kan een verduurzamingsmiddel zoals benzylalkohol worden toegevoegd.
Een eenheidsdosering kan ongeveer 0,25 tot onge-10 veer 400 mg van de verbinding met formule 1 of een farmaceutisch aanvaardbaar zout daarvan bevatten.
De farmaceutische preparaten kunnen volgens conventionele methoden worden bereid.
Voor het vervaardigen van tabletten kan de 15 verbinding met formule 1 worden gemengd met lactose en gekorreld met water, 0,5 % natriumalginaat of 5% hydroxypropylmethylcellulose-oplossing. Het gedroogde granulaat wordt samengeperst tot tabletten in aanwezigheid van ongeveer 20% maïszetmeel en 1% magnesium-stearaat. Op deze wijze worden bijvoorbeeld tabletten met de volgende 20 samenstelling verkregen:

Deze tabletten , die van een kerf zijn voorzien, kunnen oraal in een dosering van een half tot 1 tablet 1 tot 4 maal per dag worden toegediend.
Capsules kunnen het werkzame bestanddeel alleen 35 of gemengd met een inert vast excipient, bijvoorbeeld zoals boven vermeld, bevatten.

De uitvinding verschaft bijgevolg een werkwijze voor de behandeling van aandoeningen welke verband houden met de LH-secretie, de behandeling van verhoogde spiertonus, de behandeling van aandoeningen welke verband houden met hersenischemie, de behandeling van angst, schizofrenie , depressie of degeneratieve aandoeningen van het centrale zenuwstelsel, de behandeling van tinnitus , de behandeling van epilepsie of de verlichting van pijn 10 , * ...
bij een.patient, bij welke werkwijze men een therapeutisch doeltreffende hoeveelheid van de verbinding met formule 1 of een farmaceutisch aanvaardbaar zout daarvan toedient aan een patient die een dergelijke behandeling nodig heeft.
De uitvinding verschaft voorts de verbinding ^ met formule 1 of een farmaceutisch aanvaardbaar zout daarvan voor toepassing bij de vervaardiging van een farmaceutisch preparaat voor gebruik bij de behandeling van aandoeningen welke verband houden met de LH-secretie, verhoogde spiertonus, aandoeningen welke verband houden met hersenischemie , angst, schizofrenie, depressie, degeneratieve aandoeningen van het centrale zenuwstelsel, tinnitus of epilepsie of bij het verlichten van pijn.

Claims (10)
1. Werkwijze voor de bereiding van /R-(E)J-4- I (3-fosfono-2-propenyl)-2-piperazinecarbonzuur met formule 1 of een I zout daarvan, waarbij men een verbinding met formule 2 , waarin I R een alkylgroep met 1-4 koolstofatomen c£ een arylalkyLgtroep met 1-4koolstaf- I 5 atomen in de alkylgroep is,dealkyleert, en eventueel eèn zout daarvan I vormt. I
2. Werkwijze voor de bereiding van de verbin- . I ding met formule 1 of een zout daarvan in hoofdzaak zoals beschreven I in de beschrijving en/of het voorbeeld. I

I

Werkwijze voor het behandelen van aandoeningen I 25 welke verband houden met de LH-secretie, verhoogde spiertonus , I aandoeningen welke verband houden met hersenischemie, angst, schizo- I frenie, depressie, degeneratieve aandoeningen van het centrale I zenuwstelsel, tinnitus of epilepsie of de verlichting van pijn, bij I welke werkwijze men een therapeutisch doeltreffende hoeveelheid van I 30 de verbinding met formule 1 of een farmaceutisch aanvaardbaar zout I daarvan toedient aan een patient die een dergelijke behandeling nodig heeft.

Toepassing van de verbinding met formule 1 .8800412 V ft - 16 - of een farmaceutisch aanvaardbaar zout daarvan voor de vervaardiging van een farmaceutisch preparaat voor toepassing bij de behandeling van aandoeningen welke verband houden met de LH-secretie, verhoogde spiertonus, aandoeningen welke verband houden met hersenischemie, 5 angst, schizofrenie, depressie, degeneratieve aandoeningen van het centrale zenuwstelsel, tinnitus, epilepsie of bij de verlichting van pijn.
10. Farmaceutisch preparaat dat een verbinding volgens conclusie 4 of 5 of een farmaceutisch aanvaardbaar zout daarvan 10 tezamen met een farmaceutisch aanvaardbaar verdunningsmiddel of drager daarvoor omvat. -o-o-o- .8800412
