Claims (18)
- 곡류로부터 β-아밀라제를 추출하는 방법으로서,상기 방법은 β-아밀라제를 함유하는 추출물을 얻기 위하여 수성 매질 중에서 셀룰라제의 존재하에 곡류를 추출한 후, 상기 매질로부터 β-아밀라제를 회수하는 것을 특징으로 하고,상기 셀룰라제는 셀룰라제, 헤미셀룰라제 및 β-글루카나제로부터 선택되는 활성을 포함하는 것인 방법.
- 제1항에 있어서, β-아밀라제는 맥류, 소맥류 또는 호밀로부터 추출되는 것을 특징으로 하는 방법.
- 제1항에 있어서, β-아밀라제는 거피된 맥류로부터 추출되는 것을 특징으로 하는 방법.
- 제1항에 있어서, β-아밀라제는 거피, 제분, 분쇄, 연마 및 이들의 조합으로부터 선택된 공정에 의하여 처리된 곡류의 그레인으로부터 추출되는 것을 특징으로 하는 방법.
- 제1항 내지 제4항 중 어느 하나의 항에 있어서, 추출은 환원 조건하에서 수행되는 것을 특징으로 하는 방법.
- 제5항에 있어서, 상기 환원 조건은 그레인의 구조 단백질에 결합된 β-아밀라제를 방출할 수 있는 환원 활성을 제공하도록 변형되는 것을 특징으로 하는 방법.
- 제5항에 있어서, 상기 환원 조건은 메타중아황산나트륨 및 아황산나트륨으로 구성된 군으로부터 선택되는 화합물을 첨가함으로써 얻어지는 SO2 함유 물에 의하여 제공되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 거피된 맥류는 메타중아황산나트륨 및 아황산나트륨으로 구성된 군으로부터 선택되는 화합물을 첨가함으로써 얻어지는 SO2 함유 물을 사용하여 5:8∼2:3 (w/v) (거피된 맥류: SO2 함유 물) 비로 추출하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 추출은 25℃∼33℃의 온도에서 수행하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 추출 시간은 48∼66 시간인 것을 특징으로 하는 방법.
- 삭제
- 제1항에 있어서, 셀룰라제는 곰팡이의 셀룰라제를 포함하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 트리코더마(Trichoderma) 곰팡이의 셀룰라제를 사용하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 셀룰라제는 후미콜라(Humicola), 푸사륨(Fusarium), 마이셀리옵토라(Myceliopthora), 아스페르질루스(Aspergillus), 페니실륨(Penicillium), 트리코더마(Trichoderma) 및 이들의 조합으로부터 선택된 속의 셀룰라제를 포함하는 것을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, β-아밀라제는 제분된 그레인으로부터 추출되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 회수된 β-아밀라제는 정제, 농축 및 이의 조합으로부터 선택된 처리에 의하여 처리되는 것을 특징으로 하는 방법.
- 제9항에 있어서, 추출은 29℃∼31℃의 온도에서 수행하는 것을 특징으로 하는 방법.
- 제10항에 있어서, 추출 시간은 55∼62 시간인 것을 특징으로 하는 방법.
