発明の分野
本発明は眼科用溶液およびその用途の分野に関する。特に、本発明は、コンタクトレン
ズ洗浄用溶液、コンタクトレンズすすぎおよび保存溶液、眼に対して活性の医薬剤を送達
するための溶液、眼科用具を殺菌するための溶液等に関する。
背景
本発明は眼科用溶液の分野に関し、特に長期間使用後の防腐効力および心地よさの態様
に関する。これらの眼科用溶液は一定の期間にわたって使用され、一般薬（OTC製品）と
して入手できる。溶液は、例えば、眼に活性医薬剤を送達するような角膜組織に直接接触
するのに使用されるか、コンタクトレンズのような角膜組織と接触しうる器具の洗浄、コ
ンディショニングまたは保存のような間接的に使用され、これらの溶液が細菌またはその
他の微生物感染源を導入しないことを保証する必要性がある。したがって、多くの溶液が
購入され、開放され、使用され、封をされ、そして次に再使用されるので、溶液中の微生
物の生存性を減少させ、使用者による溶液の汚染の機会を少なくするために防腐剤を含有
させる。
従来技術の防腐剤には、ポリヘキサメチレンビグアニド(polyhexamethylene biguanide
: phmb)、polyquad（登録商標）、クロルヘキシジン、および塩化ベンザルコニウム等が
あるが、これらのすべてが一定の濃度で角膜組織を刺激し、使用者に不快感をもたらす。
したがって、一定量の防腐剤を使用するが、しかし防腐剤でない薬剤の添加によりより効
果的になる溶液が望まれる。
発明の概要
本発明は選択ビタミンＢ類、すなわち、ピリドキシンおよびその塩類、ならびにチアミ
ンおよびその塩類を使用する改良された眼科用溶液に関し、溶液をより効果的に防腐処理
をし、コンタクトレンズ上にカチオン性防腐剤が沈着するのを一定程度減少させることを
目的とする。ここで、眼科用溶液は、クリーナーのようなコンタクトレンズ処理溶液、浸
漬溶液、コンディショニング溶液ならびにレンズ保存液、さらに湿潤溶液および眼のコン
ディションの処置のための眼内溶液等を含むと理解される。
詳細な記述
本明細書中で具体的に記載される溶液は、０．００１〜約１％の選択ビタミンＢ類、す
なわち、ピリドキシンおよびその塩類、ならびにチアミンおよびその塩類と、張性剤、バ
ッファー、防腐剤、界面活性剤、および抗微生物剤のような眼科用溶液に有用なその他の
活性成分とを組合せて含有する。
ビタミンＢ族にはチアミン(B1)、リボフラビン(B2)、ナイアシン(B3)、パントテン酸(B
5)、ピリドキシン(B6)、およびコバラミン(B12)等がある。ビタミンＢの各形態は化学的
に区別されるが、それらはしばしば同じ栄養源中に見出され、それ故、一の欠乏がしばし
ばその他の形態の欠乏に関連する。代謝的に、それらは互いにボルスター代謝(bolster m
etabolism)に対して作用し、免疫および神経系機能を増強し、健康な皮膚および筋肉を正
常に維持し、そして細胞増殖および分化を促進する。それらは、ストレス、抑圧および心
臓血管系疾病も軽減する。一種のビタミンＢの欠乏はしばしばすべてのビタミンＢの摂取
が低いことを意味し、栄養源としてＢがマルチビタミンまたはＢ複合製剤で与えられる理
由である。
ナイアシンは非常に多くの体内の過程に貢献する。とりわけナイアシンは食物をエネル
ギーに変換し、赤血球を作り、ホルモン、脂肪酸およびステロイドを合成するのを助ける
。身体は炭化水素からエネルギーを放出する過程にナイアシンを使用する。ナイアシンは
炭化水素から脂肪を形成しアルコールを処理するのにも必要である。ナイアシンはコレス
テロールを調節するのを助ける。
ピリドキシンはセロトニン、メラトニン、およびドパミンを作るのに必要である。ビタ
ミンＢ−６は精神過程の調節における本質的な栄養素であり、雰囲気および多くのその他
の健康関心事の手助けする可能性がある。
コバラミンは正常神経細胞活性に必要である。ビタミンＢ−１２はＤＮＡ複製、および
ＳＡＭｅ(S-adenosyl-L-methionine)と呼ばれる雰囲気影響物質の産生にも必要である。
ビタミンＢ−１２は葉酸と作用してホモシスティンレベルを制御する。過剰のホモシステ
ィンはアミノ酸（蛋白質形成ブロック）であるが、心臓疾患、脳出血、ならびにおそらく
骨粗鬆症およびアルツハイマー病の危険性を増加させ得る。
葉酸や葉酸塩のようなその他の化合物はビタミンＢと組み合わせて活性であり、ＤＮＡ

に特に重要である。葉酸はＳＡＭｅを作り、血液中のホモシスティンレベルが上昇しない
ように維持するのにも必要である。葉酸(pteroylglutamic acid)はほ乳類器官中でそれ自
身活性でないが、むしろテトラヒドロ葉酸(tetrahydrofolic acid: THFA)である補酵素形
態に酵素により還元される。ビタミンＢ１２とビタミンＢ６にさらに関連する葉酸塩代謝
と相互関係が存在し、葉酸塩補酵素は大量の代謝反応に関与し、当該反応中に一炭素単位
の移動がある。
パントテン酸も時々補酵素Ａと呼ばれるが、パントテン酸の生理学的活性形態であり、
アセチル（二炭素）基の移動と関連する種々の酵素−異化反応の補酵素として代謝におけ
る生命機能を作用する。驚いたことに、パントテン酸は、病原細菌の多くの菌株を含む種
々の微生物の増殖に必須である。
コンタクトレンズ用すすぎ溶液の形態および／または医薬剤送達システムでは、溶液は
、レンズまたは医薬剤に加えて、チアミン(B1)、リボフラビン(B2)、ナイアシン(B3)、パ
ントテン酸(B5)、ピリドキシン(B6)、およびコバラミン(B12)、葉酸、カルニチンからな
る群から選択されるビタミンＢ形態またはビタミンＢの物質代謝酵素(co-metabolite)の
一種０．０００１〜約１．０重量％を含有しうる。
特定的に有用な防腐剤は、ポリヘキサメチレンビグアニド(polyhexamethylene biguani
de: phmb)、polyquad（登録商標）、クロルヘキシジン、および塩化ベンザルコニウムの
ようなカチオン性ポリマー防腐剤、ならびに同様に本発明に有用であることが証明され得
るその他のカチオン性防腐剤である。カチオン性防腐剤は防腐剤として有効量で使用され
るが、PHMBの例では０．０００１重量％から約０．０１重量％のより高いレベルまでであ
る。
選択ビタミンＢ類をカチオン性防腐剤と一緒に使用すると、予期せぬ防腐効果を示すこ
とが見出された。レンズの装着性を改良するためそして眼内で直接使用されるときに感染
に対する抵抗性を増加させるために、当業者に公知のレベルで溶液のその他の成分が使用
される。眼科用溶液に使用される選択ビタミンＢ類は一定の製剤中で防腐効果を向上させ
、一定の製剤中で角膜組織の感染に対する抵抗性を増加させ、そして一定の製剤中で涙の
質を改善させる。
製剤は、ホスフェート、ビカルボネート、シトレート、ボレート、ACES、BES、 BICINE
、 BIS-Tris、 BIS-Tris プロパン、 HEPES、 HEPPS、 イミダゾール、MES、 MOPS、 PIP
ES、 TAPS、 TES、およびトリシン(Tricine)のようなバッファーを含有することもできる
。
使用され得る界面活性剤はポリソルベート界面活性剤、ポリオキシエチレン界面活性剤
、ホスホネート、サポニンおよびポリエトキシル化ひまし油等があるが、好ましくはポリ
エトキシル化ひまし油である。これらの界面活性剤は商業的に入手できる。ポリエトキシ
ル化ひまし油はBASFより商標Cremaphorの下で販売されている。
本発明の溶液は、バッファー、張性剤(tonicity agents)、粘滑薬、湿潤剤、防腐剤、金
属イオン封鎖剤（キレート化剤）、界面活性剤、および酵素（これらに限定されない）を
含む他の添加剤をさらに含有できる。
その他の態様は、０．００１〜１重量％のキレート化剤（好ましくはEDTA二ナトリウム
）および／または追加の殺微生物剤、（好ましくは、０．００００１〜０．１または０．
００００１〜０．０１）重量％のポリヘキサメチレンビグアニド（PHMBO）、N-アルキル-
2-ピロリドン、クロルヘキシジン、ポリクォータニウム-1、ヘキセチジン、ブロノポール
、アレキシジン、低濃度過酸化水素、および眼科学的に許容できるその塩を溶液に加える
ことを含む。
本発明に有用な眼科学的に許容できるキレート化剤には、アミノカルボン酸化合物また
はその水溶性塩等があり、それはエチレンジアミン四酢酸、ニトリロ三酢酸、ジエチレン
トリアミン五酢酸、ヒドロキシエチルエチレンジアミン三酢酸、１，２−ジアミノシクロ
ヘキサン四酢酸、N,N,N',N' 四酢酸中のエチレングリコールビス（β−アミノエチルエー
テル）(EGTA)、アミノ二酢酸およびヒドロキシエチルアミノ二酢酸を含む。これらの酸を

シトレート(citrates)やポリホスフェートのようなその他のキレート化剤も本発明では
使用できる。本発明で使用できるシトレートにはクエン酸ならびにそのモノ−、ジ−およ
びトリ−アルカリ金属塩等がある。使用できるポリホスフェートにはピロホスフェート、
トリホスフェート、テトラホスフェート、トリメタホスフェート、テトラメタホスフェー
ト、ならびにより高度に濃縮された中性もしくは酸性アルカリ金属塩の形態のホスフェー
ト、例えば、ナトリウムおよびカリウム塩ならびにアンモニウム塩のようなものがある。
溶液のｐHは眼やコンタクトレンズと適合できるように調整すべきであり、６．０〜８
．０、好ましくは、６．８〜７．８または７．０〜７．６のようなｐHである。中性（ｐH
7.3）からの有意偏差は、一定のコンタクトレンズに物理的パラメーター（例えば、直径
）の変化をもたらす。低ｐH（５．５未満）は眼(eyes)のやけどおよび刺激をもたらす、
一方、非常に低いまたは非常に高いｐH（３．０未満または１０を超える）は眼(ocular)
の損傷をもたらす可能性がある。
本発明で使用される追加の防腐剤は、ポリヘキサメチレンビグアニド、N-アルキル-2-
ピロリドン、クロルヘキシジン、ポリヘキサメチレンビグアニド、アレキシジン、ポリク
ォータニウム-1、ヘキセチジン、ブロノポールおよび非常に低濃度の過酸化水素（例えば
、３０〜２００ppm）等のように公知である。
本発明の溶液は、保存、洗浄、湿潤、浸漬、すすぎおよび殺菌中、硬質のガス透過性お
よび親水性コンタクトレンズと相容性がある。
本発明の代表的な水性溶液は、レンズ洗浄または眼に潤滑を与えるのを補助する張性剤
、界面活性剤および粘性導入剤のような上述した活性成分の基本的で新規な特性に影響を
与えない追加の成分を含有できる。適切な張性剤には塩化ナトリウム、塩化カリウム、グ
リセリンまたはその混合物等がある。溶液の張性は典型的には溶液１kg当たりおおよそ２
４０〜３１０ミリオスモル(mOsm/kg)に調整され、溶液を眼組織とそして親水性コンタク
トレンズと相容性にする。一実施態様では、溶液は０．０１〜０．２重量％の塩化ナトリ
ウムを含有する。重要な因子は、約０．２モルパーセント以下の塩化物濃度を供給するよ
りも大きくない程度にこのような添加剤の濃度を維持することである。
適切な粘性導入剤にはレシチンや、ヒドロキシメチルセルロース、ヒドロキシプロピル
セルロースのようなセルロース誘導体等があり、上述界面活性剤の量と同程度の量で使用
する。
（実施例１）ピリドキシン
ピリドキシンＨＣｌ(Spectrum)およびチアミンＨＣｌ(Fisher)を含有する製剤を０．２
％ホスフェートバッファー中で調製した。溶液を塩化ナトリウムで等張とし、０．０００
１％のポリヘキサメチレンビグアニドで防腐処理をした。１N水酸化ナトリウムまたは１N
塩酸でｐHを７．２に調整した。溶液のインビトロ殺微生物活性を、室温で４時間１０ｍ
ｌの各溶液にC. albicansを露出することにより決定した。次いで、各溶液のアリコット
を寒天プレート上に順次希釈し、上昇させた温度で４８時間インキュベートした。インキ
ュベート時間の結果、コローニーの展開についてプレートを試験した。接種コントロール
に対する比較に基づいて対数減少を決定した。下記の表はインビトロ検討の結果を与える

 ピリドキシンＨＣｌおよびチアミンＨＣｌを含有する溶液はバッファーコントロールと
比較してC. albicansに対して活性において改良を示した。
（実施例２）拡張したチアミンおよびピリドキシン（ビタミンＢおよびビタミンＢ前駆
体）
実施例２ デクスパンテノールを含有する製剤
デクスパンテノールを含有する製剤を０．２％ホスフェートバッファー中で調製した。
溶液を塩化ナトリウムで等張とし、０．０００１％のポリヘキサメチレンビグアニドで防
腐処理をした。１N水酸化ナトリウムまたは１N塩酸でｐHを７．２に調整した。溶液のイ
ンビトロ殺微生物活性を、室温で４時間１０ｍｌの各溶液にC. albicansを露出すること
により決定した。次いで、各溶液のアリコットを寒天プレート上に順次希釈し、上昇させ
た温度で４８時間インキュベートした。インキュベート時間の結果、コローニーの展開に
ついてプレートを試験した。接種コントロールに対する比較に基づいて対数減少を決定し
た。下記の表はインビトロ検討の結果を与える。

 このデータはデクスパンテノールが防腐剤単独の溶液よりも防腐効果が改良されたこと
を示す。

Claims (10)

  1. ０．００１〜１０重量％の、リボフラビン(B2)である防腐剤増強剤、ならびに少なくとも０．０００１重量％のカチオン性防腐剤を含むコンタクトレンズ処理溶液であって、塩化物濃度が０．２重量％以下であり、ただし、前記コンタクトレンズ処理溶液は、下記式（１）化合物を含まない

     式中、Ｒ １ 、Ｒ ２ 及びＲ ３ は、各々、水素原子であるか、または置換されてもよいアルキル基である、
    前記コンタクトレンズ処理溶液。
  2. 前記カチオン性防腐剤の濃度が１〜１００ｐｐｍである、請求項１に記載のコンタクトレンズ処理溶液。
  3. 生理学的適合性バッファーをさらに含む請求項１に記載のコンタクトレンズ処理溶液。
  4. ０．０１％〜５．０％のグリセリンをさらに含む請求項１に記載のコンタクトレンズ処理溶液。
  5. ０．０１％〜２．０％のデカン二酸をさらに含む請求項１に記載のコンタクトレンズ処理溶液。
  6. ポリソルベート界面活性剤、ポリオキシエチレン界面活性剤、ホスホネート、サポニンおよびポリエトキシル化ひまし油からなる群から選択される湿潤剤をさらに含む請求項１に記載のコンタクトレンズ処理溶液。
  7. エチレンジアミン四酢酸、ホスホネート、シトレート、グルコネートおよびタートレートからなる群から選択される金属イオン封鎖剤をさらに含む請求項１に記載のコンタクトレンズ処理溶液。
  8. 前記カチオン性防腐剤はポリヘキサメチレンビグアニドである請求項１に記載のコンタクトレンズ処理溶液。
  9. コンタクトレンズと請求項１に記載のコンタクトレンズ処理溶液とを接触させる工程を含むコンタクトレンズ処理方法。
  10. ０．００１〜１０重量％の、リボフラビン(B2)である防腐剤増強剤、ならびに少なくとも０．０００１重量％のカチオン性防腐剤を含むコンタクトレンズ処理溶液であって、塩化物濃度が０．２重量％以下であり、０．０１％〜２．０％のデカン二酸をさらに含み、ただし、前記コンタクトレンズ処理溶液は、下記式（１）化合物を含まない
