Description
- a) bei peroralen Arzneiformen:
10 bis 800 mg, vorzugsweise 20 bis 600 mg, insbesonde re 20 bis 200 mg Dihydroliponsäure. Die Dosen können beispielsweise 1 bis 6, vorzugsweise 1 bis 4, insbe sondere 1- bis 3mal täglich verabreicht werden. Jedoch soll eine Gesamtdosis von 800 mg pro Tag nicht über schritten werden. Dasselbe gilt auch für die folgenden unter b) bis e) aufgeführten Arzneiformen. - b) bei parenteralen Arzneiformen (z. B. intraokulär oder
intravenös, intramuskulär oder intraartikulär):
0,01 bis 300 mg, vorzugsweise 0,15 bis 200 mg, insbe sondere 1 bis 100 mg Dihydroliponsäure. Die Dosen kön nen beispielsweise 1 bis 6, vorzugsweise 1 bis 4, ins besondere 1- bis 3mal täglich verabreicht werden. - c) bei Arzneiformen zur rektalen oder vaginalen Applika
tion:
10 bis 500 mg, vorzugsweise 20 bis 400, insbesondere bis 200 mg Dihydroliponsäure. Diese Dosen können beispielsweise 1 bis 6, vorzugsweise 1 bis 4, insbe sondere 1- bis 3mal täglich verabreicht werden. - d) bei Arzneiformen zur Applikation auf die Haut und
Schleimhäute, Konjunktivalsack oder intraokulär (z. B.
als Lösungen, Lotionen, Emulsionen, Salben, Pflaster
usw.):
0,01 bis 800 mg Dihydroliponsäure, vorzugsweise 0,1 bis 250 mg, insbesondere 0,2 bis 200 mg Dihydrolipon säure. Die Dosen können beispielsweise 1 bis 6, vor zugsweise 1 bis 4, insbesondere 1- bis 3mal täglich verabreicht werden.
- 1) Dihydroliponsäure oder ein pharmazeutisch verwendbares Salz der Dihydroliponsäure, zusammen mit üblichen Träger und-/oder Verdünnungs- bzw. Hilfsstoffen vermischt bzw. homogenisiert und gegebenenfalls die so erhaltene Mi schung in Hohlzellen entsprechender Größe ausgegossen oder in Kapseln entsprechender Größe abgefüllt oder gra nuliert und dann gegebenenfalls unter Zusatz von weiteren üblichen Hilfsstoffen zu Tabletten verpreßt oder in Kap seln abgefüllt, enthaltend in der Dosiereinheit 0,01 bis 800 mg Dihydroliponsäure oder ein pharmazeutisch ver wendbares Salz der Dihydroliponsäure. Die Herstellung dieser Formulierung erfolgt bei Temperaturen zwischen 0 und 120°C, vorzugsweise 20 bis 80°C.
- 2) Dihydroliponsäure oder ein pharmazeutisch verwendbares Salz der Dihydroliponsäure, gegebenenfalls mit einem An tioxydans sowie gegebenenfalls einem oder mehreren der folgenden Stoffe: Stärke, Cyclodextrin, Harnstoff, Cellu lose, Lactose, Formalin-Casein, modifizierte Stärke, Mag nesiumstearat, Calciumhydrogenphosphat, Kieselsäure ver mischt, die erhaltene Mischung gegebenenfalls mit einer wäßrigen Lösung, die als Bestandteil mindestens Gelati ne, Stärke, Polyvinylpyrrolidon, Vinylpyrrolidon-Vinyl acetat-Copolymerisat und/oder Polyoxyethylensorbitan monooleat enthält, granuliert, das Granulat gegebenen falls mit einem oder mehreren der oben genannten Hilfs stoffe homogenisiert, und diese Mischung zu Tabletten verpreßt oder in Kapseln abgefüllt, wobei die Tabletten oder Kapseln in der Dosierungseinheit jeweils 0,01 bis 800 mg Wirkstoff Dihydroliponsäure oder ein Salz hiervon enthalten. Die Herstellung dieser Formulierung erfolgt bei Temperaturen zwischen 0 und 120°C, vorzugsweise 20 bis 80°C.
- 1) Dihydroliponsäure oder ein pharmazeutisch verwendbares
Salz der Dihydroliponsäure, gegebenenfalls mit 0,001 bis
1 Gewichtsteilen (bezogen auf 1 Gewichtsteil Dihydroli
ponsäure) Antioxydans sowie gegebenenfalls unter Zusatz
eines oder mehrerer Emulgatoren und/oder Komplexbildnern
mit mindestens einem der folgenden Stoffe zu einer Mi
schung, die 0,5 bis 20 Gewichtsprozent Dihydroliponsäure
enthält, homogenisiert und gegebenenfalls emulgiert: Was
ser, Glycerin, Paraffin, Vaseline, aliphatischer Alkohol
mit 12 bis 25 C-Atomen, aliphatische Monocarbonsäure mit
15 bis 20 C-Atomen, Sorbitanmonopalmitat, Polyoxyethylen
polyolfettsäureester, ein- oder mehrwertiger niedrigmole
kularer aliphatischer Alkohol, Fettsäureglycerid, Wachs,
Silikon, Polyethylenglykol, Polyethylenoxid. Die Herstel
lung dieser Formulierung erfolgt bei Temperaturen zwi
schen 20 und 120°C.
Es können selbstverständlich Polyphenole wie Rutin, Quer cetin oder Morin oder ein pharmazeutisch verwendbares Salz zugesetzt werden. - 2) Dihydroliponsäure oder ein pharmazeutisch verwendbares Salz der Dihydroliponsäure gegebenenfalls mit 0,001 bis 1 Gewichtsteilen (bezogen auf 1 Gewichtsteil Dihydrolipon säure) Antioxydans sowie gegebenenfalls unter Zusatz ei nes Komplexbildners und/oder eines Emulgators in Wasser, physiologisch unbedenklichen Alkoholen, Dimethylsulfoxid, Polyethylenglykol oder Ölen oder Mischungen hiervon auf gelöst und gegebenenfalls die so erhaltenen Lösung mit soviel Wasser, Alkohol, Dimethylsulfoxid, Polyethylengly kol oder Öl aufgefüllt, daß die Endlösung, Endsuspension oder Endemulsion 0,5-20 Gewichtsprozent an Wirkstoff Dihydroliponsäure enthält. Die Herstellung dieser Formu lierung erfolgt bei Temperaturen zwischen 30 und 100°C. Es können selbstverständlich Polyphenole wie Rutin, Quer cetin oder Morin oder ein pharmazeutisch verwendbares Salz zugesetzt werden.
- 3) Creme mit 10% Dihydroliponsäure
50 g Polyoxyethylen-40-stearat (Handelsname: Myrj®52),
80 g Cetylstearylalkohol, 200 g weißes Vaselin,
50 g dickflüssiges Paraffin und 5 g Dimethicone werden in einer Homogenisierungsapparatur zusammengeschmolzen. In der Schmelze werden 1,26 g Methyl-4-hydroxybenzoat und 0,533 g Propyl-4-hydroxybenzoat gelöst.
In 511,207 g gereinigtem Wasser werden 1,4 g Methyl-4-hy droxybenzoat und 0,6 g Propyl-4-hydroxybenzoat bei 70°C gelöst. Die Lösung wird in die oben erhaltene Fettschmel ze einemulgiert. Die Emulsion wird homogenisiert und un ter Rühren auf Raumtemperatur abgekühlt. Dann werden 100 g Dihydroliponsäure in die Creme eingerührt und unter Va kuum die Creme nochmals homogenisiert. - 4) Augentropfen mit 0,01 bis 25 mmol Dihydroliponsäure
als Racemat
Zur Herstellung von Augentropfen ist es bevorzugt, dem Präparat Konservierungsmittel, Gleitmittel sowie gut be netzende Agentien zuzugeben.
Als Konservierungsmittel eignen sich z. B. Benzalkonium chloride, die eine antiseptische sowie oberflächenaktive Wirkung besitzen. Im weiteren ist es bevorzugt, den Au gentropfen Glycerin und physiologische Kochsalzlösung zuzugeben.Für 100 ml Augentropfen Dihydroliponsäure 0,01 bis 25 mmol H₂NaPO₄· H₂O 0,018 g HNa PO₄ · 12 H₂O 0,190 g Glycerin und/oder Dextran 0,100 g steriles Wasser bis auf 100 ml
Die topische Anwendung erfolgt tropfenweise direkt ins Auge. Die Häufigkeit der Anwendung variiert von ein- bis fünfmal täglich. Eine andere Möglichkeit der Anwendung besteht darin, die oben angegebene Formulierung über ei nen Träger dem Auge zuzuführen. Als Trager eignen sich Keratinscheiben oder weiche Kontaktlinsen, die nach einer Vorinkubation appliziert werden. Alternativ können den Augentropfen für die direkte oder die Applikation auf ei nem Träger auch Liposomenpräparate zugegeben werden.
