Description
(1)培養液温度を上限臨界溶解温度以上または下限臨界溶解温度以下とし、
(2)培養した培養細胞シートをキャリアに密着させ、
(3)そのままキャリアと共に剥離する
ことを特徴とする培養細胞シートの製造方法を提供する。このようにして得られた培養細胞シートは、軟骨組織、骨組織表面への付着性が良好であるという特徴を有しており、本発明においては、このような付着性が良好な培養細胞シートのことを、高生着性培養細胞シートと呼ぶ場合がある。
(1)キャリアと密着した細胞シートを細胞培養培養基材に付着させ、その後培地を加えることでキャリアを細胞シートからはがし、そして更に別のキャリアと密着した細胞シートを付着させることを繰り返すことで細胞シートを重層化させる方法;
(2)キャリアと密着した細胞シートを反転させ細胞培養培養基材上でキャリア側で固定させ、細胞シート側に別の細胞シートを付着させ、その後培地を加えることでキャリアを細胞シートからはがし、再び別の細胞シートを付着させる操作を繰り返すことで細胞シートを重層化させる方法;
(3)キャリアと密着した細胞シート同士を細胞シート側で密着させる方法;
(4)キャリアと密着した細胞シートを生体の患部に当て、細胞シートを生体組織に付着させた後、キャリアをはがし、再び別の細胞シートを重ねていく方法;
などの方法を用いて作成することができる。
市販の3.5cmφ培養皿(ベクトン・ディッキンソン・ラブウェア(Becton Dickinson Labware)社製ファルコン(FALCON)3001)上に、N−イソプロピルアクリルアミドモノマーを53%(実施例1)、54%(実施例2)になるようにイソプロピルアルコールに溶解させたものを0.07ml塗布した。0.25MGyの強度の電子線を照射し、培養皿表面にN−イソプロピルアクリルアミドポリマー(PIPAAm)を培養皿上に固定化した照射後、イオン交換水により培養皿を洗浄し、残存モノマーおよび培養皿に結合していないPIPAAmを取り除き、クリーンベンチ内で乾燥し、エチレンオキサイドガスで滅菌することで、温度応答性ポリマーで被覆した細胞陪養培養基材材料を得た。
上述した市販の3.5cmφ培養皿(FALCON3001)上に、N−イソプロピルアクリルアミドモノマーを55%になるようにイソプロピルアルコールに溶解させたものを0.07ml塗布した以外は、実施例2と同様な方法で細胞培養培養基材材料を得た。
軟骨細胞の形質発現程度を軟骨細胞特有のコラーゲンII産生量(図5中のCOLIIバンド強度)をRt−PCR法にて調べた。検討には、実施例2で得られた単層状培養細胞シート、実施例3で得られた積層化培養細胞シート、並びに温度応答性ポリマーが被覆されていない市販の細胞培養基材からトリプシン処理を行って得られた細胞を使用した。
ブタ膝関節軟骨面を40号紙ヤスリで約1mmスクラッチし、変形性関節症モデルを作成し、このものへ実施例3で得られた積層化培養細胞シートを移植することを試みた具体的には、実施例3の培養細胞シート下部を上述した軟骨露出面に被覆し、創傷面を保護することで移植を終えた。
ブタ膝関節軟骨面を40号紙ヤスリで約1mmスクラッチし、変形性関節症モデルを作成し、積層化培養細胞シートを移植は行わなかった。
Claims (3)
- 水に対する上限もしくは下限臨界溶解温度が0〜80℃である温度応答性ポリマーで表面を被覆した細胞培養基材上で、軟骨細胞、軟骨前駆細胞、滑膜由来細胞、滑膜幹細胞、骨芽細胞、間葉系幹細胞、脂肪由来細胞、脂肪由来幹細胞から選択される1種類またはそれ以上の種類の細胞を培養し、
(1)培養液温度を上限臨界溶解温度以上または下限臨界溶解温度以下とし、
(2)培養した培養細胞シートをキャリアとしての支持膜に密着させ、
(3)そのままキャリアと共に剥離する
ことを特徴とする、単層状培養細胞シートの製造方法であって、
前記細胞培養基材の表面の温度応答性ポリマーの被覆量は0.5〜5.0μg/cm2である、
前記製造方法。 - 前記細胞シートは、タンパク質分解酵素による処理を施されることなく細胞培養基材から剥離される、請求項1に記載の単層状培養細胞シートの製造方法。
- 前記キャリアはリング状のキャリアである、請求項1又は2に記載の単層状培養細胞シートの製造方法。
