Abstract
本发明涉及一种生物移植用细胞片,其表面具有平均细胞密度为3.0×104细胞/cm2以下的间充质系干细胞。此外,本发明还涉及一种生物移植用细胞片的制造方法,包括在具有由纤维构成的三维结构的细胞培养载体上按3.0×105细胞/cm2以下的细胞数接种间充质系干细胞的工序、以及对间充质系干细胞进行培养、调制平均细胞密度为3.0×104细胞/cm2以下的细胞片的工序。
Description
技术领域
本发明涉及用于在生物移植中使用的细胞片及其制造方法。
背景技术
间充质系干细胞(Mesenchymal stem cell,以下也称为MSC)是具有多向分化能力和自身复制能力的干细胞,不仅能够分化为成骨细胞、软骨细胞、脂肪细胞、肌肉细胞等属于间充质系的细胞,而且具有超越胚层而分化为神经细胞、肝细胞等的能力。此外,已知MSC还具有因自身产生的液体因子导致的旁分泌效应和细胞粘附相互作用。MSC被认为基于这些作用发挥目标组织、细胞的修复/再生能力、以及抗炎症等免疫控制能力,结果表现出对各种疾病的治疗效果。
从分离培养容易且增殖能力旺盛、能够在短期内确保可移植的细胞数、能够实现没有免疫排斥的自身移植、伦理问题少、由于低免疫原性因而无需预处理即可实现同种移植等出发,MSC作为理想的细胞移植疗法的材料,正在进行在对多种疾病的治疗中的应用的研究。
期待应用使用了MSC的细胞移植疗法的疾病之一是肾病、特别是慢性肾病。慢性肾病是指以蛋白尿为代表的肾损伤和以肾小球过滤量为指标的肾功能低下中的任一方或两方持续3个月以上的状态,在日本,8个成年人中就有1个人患病。如果慢性肾病的症状推进达到终末期肾衰竭,则药物治疗不见效果,多数患者不得不进行人工透析。人工透析是对症疗法,因而患有慢性肾衰竭的患者有必要终生接受人工透析,给患者造成的身体和经济负担很大。进一步,透析医疗费用的增大也成了医疗经济上令人担忧的问题。
本发明人等在确立使用MSC的细胞移植疗法的过程中发现:患者例如糖尿病患者的MSC是异常的MSC,具体为失去前述那样的多种能力或这些能力与正常MSC相比降低,结果成为失去疾病治疗效果或治疗效果与正常MSC相比降低的MSC,来自哺乳动物胎儿附属物的提取物能够使前述异常MSC活化从而恢复治疗效果,以及可以进行使用该活化的MSC的自身移植治疗等;发明了含有来自哺乳动物胎儿附属物的提取物作为有效成分的对于异常MSC的活化剂,并申请了专利(专利文献1)。特别是在对处于有必要进行治疗阶段的患者也可以进行MSC自身移植一点上,该活化剂是具有重要意义的。
另一方面,细胞移植疗法中,为了减少使用的MSC的量,正在研究能够使MSC在必要的部位集中的可局部应用的制剂形态。作为对肾病使用MSC的细胞移植疗法,报道了通过将含有MSC的细胞片组合物应用于肾脏来治疗肾病的方法(专利文献2)。该细胞片组合物是通过在刺激应答性培养基材上接种MSC并进行培养后,进行剥离刺激从而将细胞剥离而调制的。

发明内容
发明所要解决的课题
本发明的目的在于,提供一种含有治疗效果好的MSC的新的生物移植用细胞片。
用于解决课题的方法
本发明人等发现,通过在特定的培养载体上以成为特定的低细胞密度的方式对MSC进行培养,能够制造具有好的疾病治疗效果的MSC的细胞片,完成了下述各发明。
(1)一种生物移植用细胞片,其表面具有平均细胞密度为3.0×104细胞/cm2以下的MSC。
(2)根据(1)所述的细胞片,进一步包含生物相容性的支撑体。
(3)根据(2)所述的细胞片,支撑体为具有由纤维构成的三维结构的细胞培养载体。
(4)根据(3)所述的细胞片,细胞培养载体具有开口部,所述开口部由具有纳米~微米单位的平均纤维直径的纤维在与细胞接触的面上形成。
(5)根据(4)所述的细胞片,开口部的平均直径为500nm~1000μm。
(6)根据(2)至(5)中任一项所述的细胞片,支撑体为含有由生物降解性聚合物构成的纳米纤维的细胞培养载体。
(7)根据(1)至(6)中任一项所述的细胞片,其表面具有用于在肾病的治疗中使用的、平均细胞密度为1.0×103细胞/cm2~3.0×104细胞/cm2的MSC。
(8)根据(7)所述的细胞片,其用于在肾脏的纤维被膜下应用。
(9)根据(1)至(6)中任一项所述的细胞片,其表面具有用于在脑损伤或神经变性疾病的治疗中使用的、平均密度为0.5×103细胞/cm2~1.5×104细胞/cm2的MSC。
(10)根据(9)所述的细胞片,其用于在脑的损伤部位、变性部位或它们附近应用。
(11)根据(1)至(10)中任一项所述的细胞片,MSC为骨髓或脂肪组织来源的MSC。
(12)根据(1)至(11)中任一项所述的细胞片,MSC为从有疾病的对象分离的MSC。
(13)一种生物移植用细胞片的制造方法,包括:
在具有由纤维构成的三维结构的细胞培养载体上按3.0×105细胞/cm2以下的细胞数接种MSC的工序,以及
对MSC进行培养,调制平均细胞密度为3.0×104细胞/cm2以下的细胞片的工序。
(14)根据(13)所述的制造方法,细胞培养载体具有开口部,所述开口部由具有纳米~微米单位的平均纤维直径的纤维在与细胞接触的面上形成。
(15)根据(14)所述的制造方法,开口部的平均直径为500nm~1000μm。
(16)根据(13)至(15)中任一项所述的制造方法,细胞培养载体为含有由生物降解性聚合物构成的纳米纤维的细胞培养载体。
(17)根据(13)至(16)中任一项所述的制造方法,MSC为骨髓或脂肪组织来源的MSC。
(18)根据(13)至(17)中任一项所述的制造方法,MSC为从有疾病的对象分离的MSC。
发明效果
根据本发明,能够制造治疗效果好的细胞片,而且能够使MSC的移植疗法中所必需的MSC的细胞数量与以往相比大幅减少。此外,还能够治疗以往没有除肾移植以外的根治疗法的、不得不终生接受人工透析的达到终末期肾衰竭的慢性肾病。

具体实施方式
生物移植用细胞片
本发明的第1方式涉及其表面具有平均细胞密度为3.0×104细胞/cm2以下的MSC的生物移植用细胞片(以下简单地表述为细胞片)。
细胞片在其表面具有平均细胞密度为3.0×104细胞/cm2以下的MSC(MSC集团)。MSC的平均细胞密度是细胞片每单位面积的MSC细胞数的算术平均值,可以通过下述方法算出:对用显微镜等观察细胞片的表面而确认到的MSC数量进行计数,用合计数除以细胞片整体的面积;或者用显微镜等观察细胞片的表面,对每单位面积的MSC数量进行计数。
本发明中,只要存在于细胞片表面上的MSC的平均细胞密度为3.0×104细胞/cm2以下即可,例如可以适当设定为0.1×103细胞/cm2~3.0×104细胞/cm2、0.1×103细胞/cm2~2.0×104细胞/cm2、0.1×103细胞/cm2~1.5×104细胞/cm2、0.1×103细胞/cm2~1.0×104细胞/cm2、0.1×103细胞/cm2~0.5×104细胞/cm2、0.3×103细胞/cm2~3.0×104细胞/cm2、0.5×103细胞/cm2~3.0×104细胞/cm2、1.0×103细胞/cm2~3.0×104细胞/cm2、3.0×103细胞/cm2~3.0×104细胞/cm2、6.0×103细胞/cm2~3.0×104细胞/cm2、1.0×104细胞/cm2~3.0×104细胞/cm2、0.3×103细胞/cm2~2.0×104细胞/cm2、0.5×103细胞/cm2~2.0×104细胞/cm2、1.0×103细胞/cm2~2.0×104细胞/cm2、3.0×103细胞/cm2~2.0×104细胞/cm2、6.0×103细胞/cm2~2.0×104细胞/cm2等。细胞片表面上的MSC的平均细胞密度优选为0.5×103~3.0×104细胞/cm2,更优选为0.5×103~2.0×104细胞/cm2,其中更优选为2.0×103细胞/cm2~2.0×104细胞/cm2,进一步优选为2.0×103细胞/cm2~1.8×104细胞/cm2,其中进一步优选为3.0×103细胞/cm2~1.6×104细胞/cm2,更加优选为3.0×103细胞/cm2~1.0×104细胞/cm2,特别优选为3.0×103细胞/cm2~8.0×103细胞/cm2。
平均细胞密度也可以用细胞片表面的MSC的占有率(confluency,汇合性)来表示。汇合(Confluent)或100%汇合是细胞片表面完全被MSC覆盖的状态,MSC处于密集地相互接触的状态。相对于这一状态的细胞密度为%汇合,可以通过用细胞片表面上MSC所占面积的合计值除以整个片的面积来算出。
人骨髓来源MSC的情况下,平均细胞密度2.0×104细胞/cm2相当于60%汇合。同样地,平均细胞密度1.0×103~2.0×104细胞/cm260%相当于10~60%汇合,1.0×103细胞/cm2~1.6×104细胞/cm2相当于10~50%汇合,1.0×103细胞/cm2~1.0×104细胞/cm2相当于10~40%汇合,3.0×103细胞/cm2~1.0×104细胞/cm2相当于20~40%汇合,3.0×103细胞/cm2~8.0×103细胞/cm2相当于20~35%汇合。
因此,在其表面具有平均细胞密度为2.0×104细胞/cm2以下的MSC的本发明的细胞片也可以表述为在其表面具有细胞密度为60%汇合以下的MSC的细胞片。优选的实施方式中,本发明的细胞片在其表面具有细胞密度为10~60%汇合、10~50%汇合、10~40%汇合、20~40%汇合、20~35%汇合的MSC。
细胞片只要在其表面具有平均细胞密度为3.0×104细胞/cm2以下的MSC,则也可以含有MSC以外的细胞。MSC以外的细胞可以是来源于作为MSC的分离源的组织的细胞,此外也可以是期待通过与MSC并用带来的优选效果而添加的细胞。后者的细胞例如为血管内皮细胞、成纤维细胞、上皮系细胞等,根据治疗对象的疾病、预定移植的部位适当选择。
本发明的细胞片的一个例子是MSC通过MSC自身产生的细胞外基质或培养时添加在培养基中的细胞外基质相互粘附而成的片。
本发明的细胞片的另一例子是进一步包含生物相容性的支撑体的细胞片。该例子中,细胞片中,可以将通过细胞外基质相互粘附的MSC和支撑体组合,或者也可以是MSC粘附在片状支撑体的表面、MSC细胞体的一部分或者全部埋入位于支撑体表面的凹陷或开口部等从而支撑体与MSC一体化。本发明中的“在其表面具有MSC的”细胞片也作为其一个方式而包括MSC细胞体的一部分或全部处于埋入位于支撑体表面的凹陷或开口部等的状态。以这种方式,本发明的细胞片处于MSC通过细胞外基质相互粘附的状态或与支撑体一体化的状态,与为了进行细胞培养而仅仅简单地在其上接种MSC的培养载体相区别。
本发明的细胞片所含的支撑体只要为由生物相容性材料构成的片状部件即可,只要在其后向生物体内移植时容易操作,则其形状、大小、厚度没有限制。作为生物相容性材料的例子,可以列举聚氟乙烯、聚苯乙烯等聚合化合物、二氧化硅等无机化合物、生物降解性聚合物等,优选生物降解性聚合物。作为生物降解性聚合物,例如合成高分子材料中可列举聚乙醇酸、聚乳酸、聚乙二醇、聚己内酯、聚二恶烷酮、其他乳酸-二醇酸共聚物等上述的共聚物,无机材料中可列举β-磷酸三钙、碳酸钙等,天然高分子材料中可列举胶原蛋白、明胶、海藻酸、透明质酸、琼脂糖、壳聚糖、纤维蛋白、丝素蛋白、几丁质、纤维素、蚕丝等。
本发明的细胞片所含的支撑体的优选例是具有由纤维构成的三维结构的片状细胞培养载体。特别优选为由具有纳米(nm)~微米(μm)单位的平均纤维直径的纤维构成的片状三维培养载体。细胞培养载体或培养载体是细胞培养时使用的、作为细胞粘附的基础的载体。此外,“平均纤维直径”是指,从细胞粘附面一侧、典型地从上方观察培养载体时,作为相对于纤维长度方向正交的方向上的长度而测得的纤维直径的算术平均值。其中,除非另有规定,否则,本说明书中的平均的意思均为数均。此外,作为由具有纳米(nm)~微米(μm)单位的平均纤维直径的纤维构成的三维培养载体的、具有由该纤维在与细胞接触的面上形成的开口部的支撑体也是本发明中的优选支撑体的例子。这里,“开口部”是指由上述纤维形成的、存在于载体的与细胞的接触面上的凹陷。
开口部的平均直径在纤维彼此接触的情况下是指从上方观察培养载体时观察到的、以纤维为轮廓的图形的直径的平均值。关于图形的直径,在图形为多边形的情况下相当于从各顶点出发的对角线长度的算术平均值,在图形为圆形的情况下相当于其直径,在图形为椭圆形或与之类似的形状的情况下相当于其长径。
此外,纤维彼此不接触的情况下,开口部的平均直径是指通过ASTM-F316中规定的方法得到的平均流量孔径,这例如可以使用孔度计(库尔特公司制等)通过平均流点法来测定。
构成三维培养载体的纤维的平均纤维直径可以在nm~μm单位的范围内,优选在10nm~500μm、更优选在10nm~300μm范围内。特定的实施方式中,平均纤维直径例如可以在10nm~1μm、100nm~1μm、500nm~1μm、1μm~10μm、1μm~100μm、1μm~300μm或1μm~500μm范围内,优选在10nm~1μm、1μm~10μm或10μm~300μm中的任一范围内。本发明中,也可以使用一般被称为纳米纤维的纤维。
在与细胞粘附的面上具有开口部的三维培养载体中,开口部的平均直径只要为500nm~1000μm即可,可以优选在700nm~600μm、更优选在900nm~400μm范围内。特定的实施方式中,开口部的平均直径例如可以在500nm~100μm、5μm~100μm、10μm~100μm、20μm~100μm、100μm~200μm、100μm~400μm、或100μm~600μm范围内、优选在500nm~100μm或100μm~400μm范围内。
特定的实施方式中,三维培养载体可以具有60%以上、优选具有70%以上、更优选具有75%以上、特别优选具有80%以上的空隙率。
另一实施方式中,三维培养载体的开口部的平均面积为0.1~100μm2、优选为0.2~60μm2、更优选为0.5~30μm2。开口部为孔的情况下,开口部面积相当于孔的面积。
三维培养载体是具有纤维三维堆积、即纤维在三维方向上堆积重叠的结构的载体,纤维的配置可以是规则的,也可以是不规则的;此外,纤维彼此可以是结合的,也可以不结合。
三维培养载体只要是在细胞粘附面具有由纤维构成的三维结构即可,也可以具有不与细胞粘附的部分,典型地,可以在由纤维构成的三维结构下方具有作为基础的构件。基础部件只要能够支撑前述三维结构即可,为何种结构体都可以,例如可以为无纺布、针织品、纺织品、多孔质基础材料等。此外,也作为支撑体使用的三维培养载体只要为由生物相容性材料构成的片状构件即可,只要在细胞培养时、其后向生物体内移植时容易操作,则其形状、大小、厚度没有限制,但不包括例如与细胞培养用容器一体成型的构件等无法在生物移植中使用的形态。
三维培养载体中的细胞粘附面只要其主要部分为具有由纤维构成的三维结构的部分即可,具体地,从细胞粘附面一侧、典型地从上方观察培养载体时,只要培养载体面积的50%以上被具有由纤维构成的三维结构的部分占据即可。因此,三维培养载体中,可以在细胞粘附面的一部分包含不是“由纤维构成的三维结构”的部分、例如不具有三维结构的平坦的膜状部分等,粘附面中该部分的面积在低于50%的范围,优选在低于40%的范围,进一步优选在低于30%的范围。
作为本发明中能够利用的三维培养载体的例子,可以列举Vessel株式会社的VECELL(注册商标)(聚四氟乙烯,平均纤维直径:<1μm,平均孔面积:1~20μm2,开口部的平均直径:20~100μm,空隙率:80~90%)、日本宝翎株式会社的Cellbed(注册商标)(高纯度二氧化硅纤维,平均纤维直径:1μm,平均流量孔径:7~8μm,空隙率:>95%)、3D Biotek的3D Insert-PS系列(聚苯乙烯纤维,平均纤维直径:PS-200为~150μm、PS-400为~300μm,开口部的平均直径:PS-200为200μm、PS-400为400μm)、国际公开WO2014/196549号小册子中记载的细胞培养基材、国际公开WO2016/068266号小册子和作为与之对应的美国申请的美国专利申请公开US2017/319747号公报中记载的细胞培养基材(生物降解性聚合物,平均纤维直径50nm~5μm)、Liu L、Kamei K等Biomaterials 124(2017)47-54中记载的细胞培养基材(聚乙醇酸,平均纤维直径:345±91nm,平均孔面积:0.68±0.02μm2,由平均孔面积算出的开口部的平均直径:0.93μm((0.68μm2/π)1/2)×2))、NEOVEIL和NEOVEIL nano(聚乙醇酸,GUNZE株式会社)等市售的组织增强材料等。上述各文献通过参照整体并入本说明书。

本说明书中使用的治疗和/或预防包括以疾病或症状的治愈、暂时缓解、预防等为目的的医学上允许的全部类型的治疗性和/或预防性介入。即,疾病或症状的治疗和/或预防包括各种目的的医学上允许的介入,包括疾病或症状推进的延缓或停止、病变的回缩或消失、发病的预防或复发的防止等。
本发明的细胞片以有效量局部给药至对象。这里,“有效量”的意思是对治疗和/或预防疾病有效果的量。该有效量根据疾病的种类、局部给药的脏器或组织、症状的严重程度、患者之外的医学因素适当调节。优选实施方式中,细胞片的有效量换算成片中所含MSC的数量,为每1kg给药个体的体重为102细胞~109细胞,优选为104细胞~106细胞。
本发明的细胞片的优选方式之一是用于在肾病的治疗中使用的、在其表面具有平均细胞密度为1.0×103细胞/cm2~3.0×104细胞/cm2、优选为2.0×103细胞/cm2~3.0×104细胞/cm2、更优选为2.0×103细胞/cm2~2.0×104细胞/cm2、进一步优选为2.0×103细胞/cm2~1.8×104细胞/cm2、更进一步优选为3.0×103细胞/cm2~1.6×104细胞/cm2、更加优选为3.0×103细胞/cm2~1.0×104细胞/cm2、特别优选为3.0×103细胞/cm2~8.0×103细胞/cm2的MSC的细胞片。本方式中,细胞片以粘贴于肾脏、优选为肾脏的纤维被膜下的方式移植,此时,优选使剥离的肾脏的杰氏筋膜和脂肪层尽量远离肾实质。通过将细胞片移植至肾病患者的肾脏,能够抑制或防止肾病和与之相伴的症状的推进,进一步,能够改善这种情况。例如,通过将细胞片移植至重度慢性肾病的、特别是达到终末期肾衰竭的肾脏的纤维被膜下,能够抑制肾损伤的推进、降低死亡率。
本发明的细胞片的另一优选方式是用于在脑损伤或神经变性疾病的治疗中使用的、在其表面具有平均细胞密度为0.5×103细胞/cm2~1.5×104细胞/cm2、优选为1.0×103细胞/cm2~1.5×104细胞/cm2、更优选为1.8×103细胞/cm2~0.5×104细胞/cm2的MSC的细胞片。本方式中,细胞片以粘贴于脑的损伤部位、变性部位或其附近的方式移植。通过将细胞片移植至脑损伤或神经变性疾病患者的脑中,能够抑制或防止脑损伤、神经变性疾病和与之相伴的症状的推进,进一步,能够改善这种情况。例如,通过将细胞片移植至神经变性疾病的变性部位,能够改善认知功能。
本发明的细胞片与以往的含有MSC的细胞片不同,MSC的平均细胞密度大幅降低。如后述实施例中所示,本发明人等发现,通过在具有由纤维构成的三维结构的细胞培养载体上以低细胞密度对MSC进行培养,各MSC的治疗效果显著提高。令人意外的是,含有低密度培养的MSC的细胞片尽管与以往的含有MSC的细胞片相比细胞数大幅减少,但整个片的治疗效果却比以往的细胞片好。
本发明中,作为另一方式,还包括对使用MSC的细胞移植疗法有效的疾病进行治疗和/或预防的方法,该方法包括将有效量的本发明的细胞片局部给药至有需要的对象。本方式中的各术语的意思如上所述。
细胞片的制造方法
本发明的细胞片可以通过包括下述工序的方法来制造:在具有由纤维构成的三维结构的细胞培养载体上按3.0×105细胞/cm2以下的细胞数接种MSC、例如从有疾病的对象分离的MSC的工序;以及对MSC进行培养,调制平均细胞密度为3.0×104细胞/cm2以下的细胞片的工序。该制造方法为本发明的另一方式。
术语“对象”和“从有疾病的对象分离的MSC”均解释为与作为专利文献1的国际公开WO2015/137419号小册子和作为与之对应的美国申请的美国专利申请公开US2017/0071984号公报中记载的各术语相同的意思。这些文献通过参照整体并入本说明书,将这些文献和本说明书中各术语的意思的概要示于下文。
“对象”的意思是具有MSC的任意的动物,优选为哺乳动物的个体、例如人、黑猩猩等灵长类、小鼠、大鼠、豚鼠、仓鼠等啮齿类、牛、山羊、绵羊、猪等偶蹄目、马等奇蹄目、兔子、狗、猫等的个体,进一步优选为人的个体。
本发明中细胞片的制造中使用的MSC可以是从健康对象分离的MSC,也可以是从有疾病的对象分离的MSC。此外,本发明中使用的MSC还可以是从人工多能干细胞(iPS细胞)、胚胎干细胞(ES细胞)、胚胎肿瘤细胞(EC细胞)、胚胎生殖干细胞(EG细胞)等多能干细胞分化诱导而得的细胞。
正如在专利文献1中也有记载的那样,已知具有某种疾病的对象和老化的对象的MSC与健康者的MSC相比治疗效果差,即使对这些对象直接移植自身MSC,也不能期待好的治疗效果。另一方面,本发明的制造方法中,通过在三维培养载体上的低密度培养,能够提高各MSC的治疗效果,因此即使以这样的治疗效果差的MSC为原料,也能够制造治疗效果好的细胞片。具有治疗效果差的MSC的对象患有的疾病为慢性疾病,其例子在专利文献1中作为MSC异常的疾病记载。
在考虑其后的细胞移植疗法中的安全性的情况下,MSC优选从与用细胞进行给药的个体为同种或近缘物种的个体采集。例如,对人的个体进行细胞移植的情况下,优选使用从作为同种的人采集的细胞、更优选使用从接受给药的同一人类个体采集的细胞、即自身MSC。
本发明中细胞片的制造中使用的MSC可以通过一般的方法从对象的骨髓液、脂肪组织、胎儿附属组织、牙髓等试样来采集。例如,使用骨髓液作为试样的情况下,可以通过密度梯度离心法、骨髓播散法等公知的方法分离MSC。本发明的优选实施方式之一中,MSC是骨髓或脂肪组织来源的MSC。
本发明的制造方法包括在具有由纤维构成的三维结构的细胞培养载体上按3.0×105细胞/cm2以下的细胞数接种MSC的工序。本工序可以通过下述方法来进行:将前述具有由纤维构成的三维结构的片状细胞培养载体置于通常的细胞培养用容器内,加入培养基浸泡细胞培养载体,在其上接种调节了细胞数的MSC。
细胞培养用容器的底面只要是能够以展开状态配置三维培养载体的程度的大小即可,优选在以展开状态配置三维培养载体时,不被三维培养载体覆盖的部分少。特定的实施方式中,细胞培养用容器的底面与三维培养载体为同一形状或者是与三维培养载体内接的形状。
接种的MSC的细胞数根据制造的细胞片的平均细胞密度适当调节。典型地,接种的MSC的细胞数只要在制造的细胞片平均细胞密度的10倍量至1/10倍量的范围内即可,优选在10倍量至1倍量的范围内。例如,调制平均细胞密度为3.0×104细胞/cm2的细胞片时,接种的MSC的细胞数为3.0×105细胞/cm2~3.0×103细胞/cm2。此外,调制平均细胞密度为2.0×104细胞/cm2的细胞片时,接种的MSC的细胞数为2.0×105细胞/cm2~2.0×103细胞/cm2;调制平均细胞密度为1.0×104细胞/cm2的细胞片时,接种的MSC的细胞数为1.0×105细胞/cm2~1.0×103细胞/cm2;调制平均细胞密度为5.0×103细胞/cm2的细胞片时,接种的MSC的细胞数为5.0×104细胞/cm2~5.0×102细胞/cm2;调制平均细胞密度为1.0×103细胞/cm2的细胞片时,接种的MSC的细胞数为1.0×104细胞/cm2~1.0×102细胞/cm2;调制平均细胞密度为0.5×103细胞/cm2的细胞片时,接种的MSC的细胞数为0.5×104细胞/cm2~0.5×102细胞/cm2。
其中,上述接种细胞数是三维培养载体的单位面积的数量值,实际的接种细胞数通过用细胞培养用容器的底面积除以三维培养载体的面积,将算出的值乘以上述接种细胞数而算出。
培养基可以利用MSC的培养中通常使用的培养基,例如α-MEM、DMEM等。这些培养基也可以含有MSC増殖所必需的各种成分、例如血清成分等。
本发明的制造方法包括在具有由纤维构成的三维结构的培养载体上对MSC进行培养,调制平均细胞密度为3.0×104细胞/cm2以下的细胞片的工序。
在三维培养载体上的MSC的培养进行24小时~144小时、优选进行24小时~96小时、更优选进行48~96小时。培养工序中的培养温度和气体浓度只要在MSC培养中通常使用的温度和气体浓度范围内即可,温度例如为25℃~37℃、优选为30℃~37℃、更优选为37℃,氧浓度例如为2%~30%、优选为2%~20%。通过将培养载体中接种的细胞数调节至上述范围,并且根据需要调节培养时间、温度等培养条件,能够调制具有目标平均细胞密度的细胞片。
通过上述方法制造的细胞片可以直接以包含三维培养载体作为支撑体的方式使用,在能够从三维培养载体剥离的情况下也可以剥离后使用,或者在从三维培养载体剥离后根据需要与其他支撑体组合使用。作为能够将细胞片剥离的培养载体,例如,除了聚(N-异丙基丙烯酰胺)以外,还可以列举用分子结构由于温度、pH、光等刺激而发生变化的高分子将表面覆盖的、具有由纤维构成的三维结构的细胞培养载体。
细胞片不含支撑体的情况下,即MSC相互粘附而成的细胞片的情况下,培养载体没有必要为片状。这种情况下,三维培养载体只要在培养时细胞能够与由纤维构成的三维结构接触,则形状没有限制。三维培养载体可以是设置在细胞培养容器内而使用的插入状的形状,或者也可以是由纤维构成的三维结构在细胞培养容器的内表面、例如孔的底面等一体成型的形状。
细胞片上的MSC可以维持未分化的状态,或者也可以使其分化成希望的细胞。MSC的未分化状态的维持可以通过使用适合于维持未分化状态的培养基、例如HyCloneAdvance STEM间充质干细胞表达试剂盒(赛默飞世尔科技)、MesenCult(商标)MSC基础培养基(STEMCELL Technology)、Stromal Cellutions(商标)介质(DV Biologics)、MSC专用培养基试剂盒(MSCGM BulletKit,Lonza)等对MSC进行培养来进行。此外,MSC的分化可以通过在加入了具有向希望的细胞进行分化诱导作用的因子的分化诱导培养基中的培养等一般的已知方法来进行。例如,在向成骨细胞的分化中,使用骨形成蛋白(Bone MorphogeneticProteins,BMP)4、BMP2等作为分化诱导因子;在向脂肪细胞的分化中,使用地塞米松、3-异丁基-1-甲基黄嘌呤、胰岛素等作为分化诱导因子。
制造方法中活化剂的利用
为了进一步提高调制的细胞片的治疗效果,本发明的制造方法中,优选使用含有以来自哺乳动物胎儿附属物的提取物为有效成分的活化剂的培养基进行MSC的培养。
本发明中能够利用的“来自哺乳动物胎儿附属物的提取物”的一个例子是,之外通过参考而整体并入本说明书的专利文献1的国际公开WO2015/137419号小册子和作为与之对应的美国申请的美国专利申请公开US2017/0071984号公报中记载的提取物。该提取物是如下调制的提取物:将在哺乳动物、优选为人的胎儿娩出后作为产后物从母体娩出或者通过剖腹产从母体摘出的胎儿附属物、优选为脐带组织、胎盘组织或卵膜直接或切断或破碎并在蒸馏水、生理盐水、磷酸缓冲生理盐水、细胞培养中通常使用的培养基等提取介质中浸泡等。提取物特别优选不含来自作为供体的哺乳动物的具有増殖能力的细胞。具体的提取操作和条件可以遵循专利文献1中记载的操作和条件。
此外,“来自哺乳动物胎儿附属物的提取物”的另一例子可以是通过对胎儿附属物实施从胎儿附属物、典型地从胎盘调制生理活性物质时本领域技术人员通常使用的处理、例如使用酸、酶水解等处理而调制的物质。该提取物的例子是由美思满制药株式会社出售的作为人胎盘酸水解物的胎盘制剂“美思满”、由株式会社日本生物制剂出售的人胎盘制剂“莱乃康”、其他市售的胎盘制剂和被称为胎盘提取物的各种市售品。
本发明中的“含有来自哺乳动物胎儿附属物的提取物作为有效成分的活化剂”是指含有前述提取物作为有效成分的、用于增大MSC的治疗效果的试剂,以下将其表述为“活化剂”。
培养中使用的培养基中的活化剂浓度以蛋白质换算的终浓度计为0.01μg/mL~500μg/mL则足够,优选为0.02μg/mL~300μg/mL,更优选为0.04μg/mL~100μg/mL,在特定的实施方式中可以为0.05μg/mL~10μg/mL。
通过以下的实施例进一步详细地对本发明进行说明,但本发明不受这些例子的限定。

根据专利文献1的记载,从人胎盘组织调制活化剂。简单来说,按对于湿重50g的切碎的人胎盘组织为100mL的比例加入无血清培养基(alpha-MEM),4℃振荡72小时。通过离心分离回收上清,得到作为胎盘组织提取物的活化剂。
2)MSC的培养
将在变形性髋关节病患者的人工关节置换术时采集的骨髓来源的MSC(OA-MSC)用不含活化剂的MSC培养基(含有15%FBS、1%青霉素、1%链霉素、4500mg/L葡萄糖和L-谷氨酰胺的DMEM)进行培养。回收细胞,分别在具有由纤维构成的三维结构的细胞培养载体即Preset VECELL 6孔(注册商标)(Vessel株式会社)上接种8×104细胞/孔、在Cellbed 24孔(注册商标)(日本宝翎株式会社)上接种2×104细胞/孔、在3D-insert PS-200 12孔和3D-insert PS-400 12孔(3DBiotek公司)上接种3.8×104细胞/孔,此外在作为比较例的培养载体的细胞培养基材A(平均纤维直径0.1~0.5μm,空隙率70%以下,平均孔面积0.2μm2,24孔)上接种2×104细胞/孔,使用不含活化剂的MSC培养基,37℃培养72小时。基材A是在其一部分具有由纤维构成的三维结构、但不具有三维结构的平坦的膜状部分占细胞粘附面大约50%的基材。将培养基除去后,分别将含有以蛋白质换算计为10μg/mL、1μg/mL或者0.1μg/mL的上述活化剂或不含活化剂的MSC培养基在Preset VECELL 6孔中加入2mL,在Cellbed24孔和基材A中加入0.6mL,在3D-insert PS-200 12孔和3D-insert PS-400 12孔中加入1mL,37℃进行4天培养。将该培养进行1~3次后,回收细胞,调制继代数1~3的OA-MSC。
进一步,在作为平板状二维细胞培养载体的Corning(注册商标)Costar(注册商标)细胞培养6孔板(Thermo Fisher Science公司)上接种用不含活化剂的MSC培养基培养的OA-MSC,6×104细胞/孔,在100μg/mL活化剂存在下进行培养。进一步将该培养重复1次,从而调制继代数2的对照OA-MSC。
实施例1细胞片的调制
用不含活化剂的MSC培养基(含有15%FBS、1%青霉素、1%链霉素、4500mg/L葡萄糖和L-谷氨酰胺的DMEM)对OA-MSC进行培养。回收细胞,在二维培养载体(Corning(注册商标)Costar(注册商标)细胞培养6孔板,Thermo Fisher Science公司)上放入2.5cm×2.5cm的细胞培养基材B,接种8×104细胞的MSC,加入2mL含有1μg/mL的活化剂的MSC培养基,37℃进行72小时培养,从而调制继代数1的OA-MSC的片。通过该方法调制的MSC片含有大约8,500细胞/cm2的MSC。其中,细胞培养基材B是通过国际公开WO2016/068266号小册子和Liu L、Kamei K等Biomaterials 124(2017)47-54记载的方法制造的、在由聚乙醇酸构成的基础部件上含有由聚乙醇酸构成的纳米纤维的三维培养载体。
实施例2MSC的基因表达分析
回收参考例1和实施例1中调制的MSC,使用Tri试剂(Molecular ResearchCenter,Inc)提取总RNA。通过逆转录反应合成克隆DNA,对于OCT4、Nanog、SOX2、DNMT1、TERT、IL-6、IDO、TSG-6、p16ink4a、p21、p53、α-SMA和18sRNA,使用由表1所示碱基序列构成的引物组实施实时PCR。

以18sRNA为持家基因,以在二维培养载体上在100μg/mL活化剂存在下培养的对照OA-MSC为基础算出ΔΔCT值,对基因表达谱进行分析、聚类。将在Preset VECELL上在0.1μg/mL活化剂存在下培养的OA-MSC(VECELL_0.1)、在Cellbed上在0.1、1或10μg/mL活化剂存在下培养的OA-MSC(分别为CellBed_0.1、CellBed_1、CellBed_10)、在3D-insert PS-200上在0.1μg/mL活化剂存在下培养的OA-MSC(3D.insert200_0.1)、在3D-insert PS-400上在10μg/mL活化剂存在下培养的OA-MSC(3D.insert400_10)、在基材A上在0.1μg/mL活化剂存在下培养的OA-MSC(A_0.1)、在基材A上在1μg/mL活化剂存在下培养的OA-MSC(A_1)、在基材B上在0.1μg/mL活化剂存在下培养的OA-MSC(B_0.1)以及在二维培养载体上在100μg/mL活化剂存在下培养的对照OA-MSC(2D_100)的结果示于图1。
确认到与2D_100相比,在三维培养载体上在活化剂存在下培养的OA-MSC中,被认为是与干细胞性相关的基因的DNMT1、Nanog、SOX2和OCT4、被认为是与免疫控制/抗炎症功能相关的基因的IDO、TSG6和IL-6、被认为是与端粒酶活性相关的基因的TERT的表达量均有增加,被认为是与细胞老化相关的基因的P53、被认为是与细胞骨架相关的基因的α-SMA的表达量减少。OCT4、SOX2、Nanog、DNMT1、IDO、TSG6、IL-6和TERT均为在治疗效果好的MSC中表达被促进的标记物,P53和α-SMA为在治疗效果好的MSC中表达被抑制的标记物。此外,p16ink4a是与本发明人等发现的OCT4、SOX2、Nanog、IDO和TSG6显示高相关性的治疗效果的正的标记物。根据这些结果,认为在任何三维培养载体上培养的OA-MSC治疗效果均比在二维培养载体上培养的OS-MSC好。另一方面,A_1和A_0.1显示出与2D_100类似的基因表达谱,推测在基材A上培养的OA-MSC的治疗效果与在通常的二维培养载体上培养的OA-MSC为同等程度。
实施例3细胞片的肾病治疗效果(糖尿病性肾病合并急性肾损伤)
对于糖尿病性肾病发病的14月龄雄性OLETF大鼠(星野试验动物),用利妥昔单抗(中外制药)按1天1次、5mg/只进行尾静脉给药4天。从利妥昔单抗初次给药开始10~14天后,在用利妥昔单抗给药的大鼠的肾脏中移植实施例1的MSC片(MSC片组,n=6)。将仅用利妥昔单抗给药作为空白(溶媒)(溶媒组,n=7)。在异氟烷吸入麻醉下,由侧卧位的大鼠的最下位肋骨下切开皮肤,切开肌肉层,将一个肾脏拉出体外。以不损伤副肾的方式小心地将杰氏筋膜、脂肪层和纤维被膜切开,从肾表面剥下,拉至肾门部。以包裹整个肾脏的方式粘贴1片2.5cm×2.5cm的MSC片和1片切成1/3的同样的MSC片。在另一肾脏上也同样地粘贴MSC片。在MSC片移植当天、移植后3周和6周时,从大鼠采集血液,使用酶法(SRL)测定血清肌酐。
将移植后11周存活的动物(溶媒组,MSC片组均为n=2)的血清肌酐的推移示于图2。溶媒组中确认到血清肌酐的经时性上升,而MSC片组中未确认到血清肌酐值的上升。这6周的肾功能维持相当于对于人使透析的开始推迟5年,表明MSC片的良好治疗效果。此外,将表示到移植后11周为止的存活率的Kaplan-Meier曲线示于图3。MSC片组与溶媒组相比显示高的存活率。
进一步,制成从移植后11周的动物摘出的肾脏的组织切片,进行PAS染色标本的光学显微镜观察和电子显微镜观察。溶媒组中几乎在全部肾小球中确认到硬化,观察到系膜细胞和足细胞的异常(图4左上、图5)。此外,在肾小管中确认到肾小管间质的炎症细胞浸润、肾小管上皮细胞的异常、肾小管上皮的脱落、基底膜的肥厚等(图4左下、图6、图7)。另一方面,在MSC片组中,残留有正常的肾小球(图4右上),系膜细胞和足细胞正常化(图8)。此外,确认到肾小管上皮细胞的再生图像(图4右下的箭头)、微绒毛明显的近端肾小管上皮、肾小管间质的炎症细胞的减少(图9)等表示治愈的组织学观察结果。进一步,在MSC片组中确认到在肾表面新的被膜状结构的形成、在其外侧大量毛细血管的再生(图10、图11)。
本试验中使用的大鼠是除了糖尿病性肾病以外还通过抗癌剂处理进一步诱导急性肾损伤从而使程度一致的严重慢性肾病发病的模型动物,表现相当于肾病第4期(肾衰竭期)的非常严重的末期肾损伤。确认本发明的细胞片即使对于这样严重的疾病也表现出优异的治疗效果。
实施例4细胞片的肾病治疗效果(糖尿病性肾病)
在糖尿病性肾病发病的14月龄雄性OLETF大鼠肾脏中,与实施例3同样地操作,将实施例1中调制的细胞片移植至肾纤维被膜下(n=2/组)。将实施仅将背部皮肤和筋膜切开的假手术的大鼠作为假模(Sham)组。在MSC片移植当天、移植后4周和11周时从大鼠采集血液,使用酶法(SRL)测定血清肌酐,使用UV法(SRL)测定尿素氮(BUN)。
将血清肌酐和BUN的推移示于图12。Sham组中确认到血清肌酐和BUN的经时性上升,而MSC片组中未确认到。确认本发明的片在不伴随急性肾损伤的糖尿病性肾病中也表现出优异的治疗效果。
实施例5细胞片的肾病治疗效果(糖尿病性肾病)
用不含活化剂的MSC培养基(含有10%FBS、1%青霉素、1%链霉素、4500mg/L葡萄糖和L-谷氨酰胺的DMEM)对OA-MSC进行培养。回收细胞,在二维培养载体(ThermoScientific(商标)Nunc(商标)Lab-TekTM,8孔,Thermo Fisher Science公司)上加入1×0.8cm的NEOVEIL nano(GUNZE株式会社),接种4×103细胞的MSC,加入200μL不含活化剂的MSC培养基,37℃进行24小时培养,从而调制继代数1的OA-MSC片。通过该方法调制的MSC片含有4781细胞/cm2(调制的各片的平均细胞密度3923~5884细胞/cm2的平均值)的MSC。
在糖尿病性肾病发病的16周龄雄性KK-Ay小鼠的两肾中,以不损伤副肾的方式小心地将杰氏筋膜、脂肪层和纤维皮膜切开,从肾表面剥下,以包裹整个肾脏的方式粘贴上述制作的MSC片,进行移植(n=12)。作为对照,准备未处理的糖尿病性肾病发病的同周龄KK-Ay(n=9)。在MSC片移植前和移植后4周时采集小鼠的尿,使用免疫比浊法(东方酵母工业株式会社)测定尿中白蛋白,使用酶法(东方酵母工业株式会社)测定尿中肌酐。
将MSC片移植前和MSC片移植后4周时尿中白蛋白/肌酐比之差示于图13。确认到尿中白蛋白/肌酐比在未处理组中増加,但在MSC片组中反而减少的倾向。确认到尽管本实施例中使用的细胞片的平均细胞密度比实施例1中调制的细胞片低,但也与实施例1中调制的细胞片同样地表现出对糖尿病性肾病的优异治疗效果。
实施例6细胞片的肾病治疗效果(缺血再灌流导致的急性肾损伤)
1)细胞片的调制
按照实施例1的方法,将表2所示细胞数的MSC接种于2.5cm×2.5cm的基材B,用不含活化剂的MSC培养基培养,从而调制具有不同平均细胞密度的MSC的细胞片。

将细胞片A~G上的MSC用DAPI(4’,6-二脒基-2-苯基吲哚)染色,进行显微镜观察(图14)。测定以将MSC在同一培养载体上进行培养至达到汇合时的细胞密度作为100%时各细胞片上的%汇合,结果,片A~C为90%~70%汇合、D~F为60%~10%汇合、G为低于10%汇合。
2)基因表达分析
与实施例2同样操作,通过实时PCR测定细胞片B、G和H的MSC中OCT4、Nanog、p16ink4a和TERT的表达量(图15)。平均细胞密度越低则全部基因的表达量越多,推测平均细胞密度低的细胞片上的各MSC的治疗效果比平均细胞密度高的细胞片上的各MSC好。
3)缺血再灌流导致的急性肾损伤模型大鼠中的移植试验
对麻醉下的5周龄雄性SD大鼠进行腹部正中切开。首先确定右肾,将右肾动脉用血管用夹钳阻断。阻断后立刻将上述1)中调制的细胞片A~G分别与实施例3同样地操作,移植于肾纤维被膜下。阻断60分钟后,打开血管用夹钳,解除阻断。对于左肾,也与上述同样地阻断左肾动脉,移植细胞片,阻断60分钟后解除阻断(n=1或2/组)。此外,准备不移植细胞片、将杰氏筋膜、脂肪层和纤维被膜从肾表面剥下且拉至肾门部的组(Sham-有肾被膜处理)、以及拉过以后再复原的组(Sham-无肾被膜处理)。
将到移植后10天为止各大鼠的存活天数与细胞片的平均细胞密度之间的关系示于图16,将表示各个平均细胞密度的组(高、中、低)的存活率的Kaplan-Meier曲线示于图17。移植了平均细胞密度为中等程度的片D~F的大鼠全部存活至移植后第10天,而移植了平均细胞密度高的片A~C的小鼠中的半数、和移植了平均细胞密度低的片G的小鼠在第10天均已死亡。推测MSC的绝对数量多的片A~C的上述结果是因为各MSC的治疗效果差。此外,推测各MSC中预测治疗效果最好的片G的上述结果是因为发挥治疗效果所必需的细胞的绝对数量不足。
此外,将表示到移植后10天为止的Sham-有肾被膜处理组和Sham-无肾被膜处理组的存活率的Kaplan-Meier曲线示于图18。Sham-有肾被膜处理组与Sham-无肾被膜处理组相比存活率的下降速度慢,因而确认到通过将杰氏筋膜、脂肪层和纤维被膜从肾表面剥下,远离肾实质,急性肾损伤的影响得到缓和。
实施例7细胞片的阿尔茨海默氏病治疗效果
1)细胞片的调制
按照实施例6中1)的方法,将表3所示细胞数的MSC接种于2.5cm×2.5cm的基材B,用不含活化剂的MSC培养基培养,从而调制具有不同平均细胞密度的MSC的细胞片。

2)阿尔茨海默氏病模型小鼠中的移植试验
将15月龄雄性APP/PS1小鼠(Charles River(查尔斯河实验室))在异氟烷吸入麻醉下,从前囟水平(bregma level)至人字点水平(lambda level)以能够形成8mm×5mm大小的开窗部的方式开颅。分别将1片切成8mm×5mm大小的细胞片从开窗部粘贴于脑表面,静置5分钟后,将颅骨放回,缝合。
在移植后第3周进行新物体识别试验。新物体识别试验的内容如下。第0天,将小鼠在没有放置任何东西的箱子中放置5分钟,熟悉环境。第1天,将小鼠在放入了2个相同物体的箱子中放置5分钟(Familiarization,熟悉),1小时后,将1个物体换成新物体,分别测定5分钟内探索新物体或旧物体的时间。算出新物体的探索时间相对于2个物体的总探索时间的比例(偏好指数,新/(新+旧物体探索时间)×100%),将50%以上设为认知功能良好。物体的位置在整个试验中置于相同位置。
将根据移植的片的平均细胞密度分为2组绘制偏好指数的结果示于图19。移植1.50×104细胞/cm2以下的片的小鼠中,偏好指数高,认知功能良好,因而确认到本发明的细胞片对于阿尔茨海默氏病也表现出治疗效果。
实施例8使用小鼠脂肪组织来源MSC的细胞片对肾病的效果
1)细胞片的调制
采集C57BL/6(雄性,10周龄)的精巢上体周围脂肪,切碎后,加入含有0.4PZ单位/mL的Liberase(商标)的PBS,每1g脂肪加入1mL,37℃静置2小时。加入5mL含有10%FBS的DMEM培养基并悬浮后,300g×5分钟离心分离,将上清除去。将从1g脂肪回收的细胞颗粒接种于1片15cm培养盘,用含有10%FBS、1%PS的DMEM培养基培养4天,替换培养基后,回收粘附细胞(小鼠脂肪组织来源MSC)。
将6×104/cm2、3×104/cm2、4×103/cm2的小鼠脂肪组织来源MSC分别接种于1×0.8cm的NEOVEIL nano(GUNZE株式会社),加入2mL不含活化剂的MSC培养基,37℃进行24小时培养,从而制成培养后的最终细胞密度(平均值±标准误差,DAPI染色细胞数)为45480±4953/cm2、27541±5475/cm2、3800±908/cm2的、继代数1的细胞片。将对各细胞片上的MSC进行DAPI染色的显微镜观察图像示于图20。
2)缺血再灌流导致的急性肾损伤模型小鼠中的移植试验
将麻醉下的10-11周龄雄性C57BL6小鼠的右侧后背部切开,使右肾露出。将右侧肾动静脉结扎,切除右肾脏动脉静脉,将右肾摘出。确认无出血,将肾脏放回体内原来的位置,将皮肤缝合。接下来,将左侧后背部切开,使左肾露出,将左肾动静脉用无创血管夹阻断血流。22分钟的阻断期间,以不损伤副肾的方式小心地将杰氏筋膜、脂肪层和纤维皮膜切开,从肾表面剥下,拉至肾门部。以包裹整个肾脏的方式粘贴45480细胞/cm2的细胞片(n=4)、27541细胞/cm2的细胞片(n=4)或3800细胞/cm2的细胞片(n=3),左肾的缺血开始22分钟后,取下血管夹,使血液再灌流。作为对照,准备不移植细胞片、将右肾摘出后使左肾缺血22分钟后再灌流的组(Sham组:n=3)和未处理的正常小鼠(n=4)。
从粘贴细胞片开始24小时后,从小鼠采集血液,使用UV法(SRL)测定尿素氮(BUN)。将结果示于图21。移植了高细胞密度(45480细胞/cm2)的细胞片的组的BUN与Sham组为同等程度,确认到急性肾损伤的推进,而移植了中细胞密度或低细胞密度(27541细胞/cm2、3800细胞/cm2)的细胞片的组的BUN与正常小鼠为同等程度。由以上结果确认到,使用了小鼠脂肪组织来源MSC的本发明的细胞片与使用了人骨髓来源MSC的本发明的细胞片同样地表现出抑制急性肾损伤的推进的效果。
Claims (18)
1.一种生物移植用细胞片,其表面具有平均细胞密度为3.0×104细胞/cm2以下的间充质系干细胞。
2.根据权利要求1所述的细胞片,进一步包含生物相容性的支撑体。
3.根据权利要求2所述的细胞片,支撑体为具有由纤维构成的三维结构的细胞培养载体。
4.根据权利要求3所述的细胞片,细胞培养载体具有开口部,所述开口部由具有纳米~微米单位的平均纤维直径的纤维在与细胞接触的面上形成。
5.根据权利要求4所述的细胞片,开口部的平均直径为500nm~1000μm。
6.根据权利要求2至5中任一项所述的细胞片,支撑体为含有由生物降解性聚合物构成的纳米纤维的细胞培养载体。
7.根据权利要求1至6中任一项所述的细胞片,其表面具有用于在肾病的治疗中使用的、平均细胞密度为1.0×103细胞/cm2~3.0×104细胞/cm2的间充质系干细胞。
8.根据权利要求7所述的细胞片,其用于在肾脏的纤维被膜下应用。
9.根据权利要求1至6中任一项所述的细胞片,其表面具有用于在脑损伤或神经变性疾病的治疗中使用的、平均密度为0.5×103细胞/cm2~1.5×104细胞/cm2的间充质系干细胞。
10.根据权利要求9所述的细胞片,其用于在脑的损伤部位、变性部位或它们附近应用。
11.根据权利要求1至10中任一项所述的细胞片,间充质系干细胞为骨髓或脂肪组织来源的间充质系干细胞。
12.根据权利要求1至11中任一项所述的细胞片,间充质系干细胞为从有疾病的对象分离的间充质系干细胞。
13.一种生物移植用细胞片的制造方法,包括
在具有由纤维构成的三维结构的细胞培养载体上按3.0×105细胞/cm2以下的细胞数接种间充质系干细胞的工序,以及
对间充质系干细胞进行培养、调制平均细胞密度为3.0×104细胞/cm2以下的细胞片的工序。
14.根据权利要求13所述的制造方法,细胞培养载体具有开口部,所述开口部由具有纳米~微米单位的平均纤维直径的纤维在与细胞接触的面上形成。
15.根据权利要求14所述的制造方法,开口部的平均直径为500nm~1000μm。
16.根据权利要求13至15中任一项所述的制造方法,细胞培养载体为含有由生物降解性聚合物构成的纳米纤维的细胞培养载体。
17.根据权利要求13至16中任一项所述的制造方法,间充质系干细胞为骨髓或脂肪组织来源的间充质系干细胞。
18.根据权利要求13至17中任一项所述的制造方法,间充质系干细胞为从有疾病的对象分离的间充质系干细胞。
