即ち、図12は、2枚の膵島細胞シート移植を受けた、ストレプトゾトシン誘発糖尿病化または非糖尿病化SCIDマウスの腹腔内グルコース負荷試験(IPGTT,体重1kg当たり2gのグルコース)の結果を示す。IPGTTは、膵島細胞シート移植後60日目に実施した。IPGTTの結果は、グルコース投与前(0分)及びグルコース投与後15分、30分、60分、90分、120分、150分におけるグルコース濃度を示す。この試験において使用したマウスは、2枚の膵島細胞シートを有する糖尿病化SCIDマウス(n=7,三角)、擬似手術された糖尿病化SCIDマウス(n=5,四角)、及び膵島細胞シート移植を受けた非糖尿病化SCIDマウス(n=11,黒丸)である。データはmean ± SEMとして示した。
生後8週のラット(Lewins rat、雄、体重約250g)に対し、イソフルレン麻酔下に開腹し、膵島の摘出を行った。摘出した膵島はコラゲナーゼ溶液で32分間反応させ、その後Ficoll液を用いた濃度勾配法により膵島を分離した。そして、得られた膵島に対し、0.125%トリプシン-EDTAで5分間反応させることで膵島を個々の細胞にした。その際、細胞は10%ウシ胎仔血清を含むRPMI1640培地(Sigma-Aldrich社製)中に保管した。得られた膵島細胞の活性(viability)は90%以上であった。
細胞培養基材として、ポリスチレン製の35 mm 細胞培養皿(コーニング社製)に対し温度応答性ポリマーであるポリ-N-イソプロピルアクリルアミドを被覆したものを用いた。温度応答性ポリマーの被覆は特開平02-211865号公報に示された方法に従って行い、最終的に温度応答性ポリマーの被覆量が4.9μg/cm2である温度応答性細胞培養基材を得た。本実験では、その温度応答性細胞培養基材表面へさらにラミニン-5(CHEMICON社)を0.2μg/cm2となるようにコーティングし、その表面へ上述の膵島細胞を5.7×105個/cm2となるよう播種し、そのまま2日間培養した。その際、培地としては10%ウシ胎仔血清を含むRPMI1640培地(Sigma-Aldrich社製)を利用した。ポリ-N-イソプロピルアクリルアミドが被覆された基材表面は、32℃以上では疎水性となり、32℃以下では親水性となる。これにより、37℃の培養条件下では接着培養が可能で、37℃以下になると、トリプシン処理することなくシート状に剥離される(図2)。
培養2日目、培養皿上で膵島細胞は、ほぼ100%に近い密度となった(図3)。この培養皿上でコンフルエントとなった膵島細胞をインスリン染色にて観察したところ、7~8割の細胞でインスリン陽性が認められた(図4)。次に、別に用意した温度応答性培養基材上の膵島細胞に対し、グルコース濃度が3mM、20mM、3mMとなるように作製した培地を用いて、それぞれ1時間間隔で培地交換し、培地中のインスリン濃度を測定した。その結果、糖濃度に応じたインスリン分泌能が証明された(図5)。さらに、膵島細胞シートをPKH26(SIGMA)を用いて蛍光染色を行った。その結果、得られた細胞は膵島細胞であることが分かった。
最後に、得られた膵島細胞シートを、ドナーと同じ種類のラット(Lewis rat、生後8週、雄、体重約250g)へ移植した。背部にL字状皮膚切開し、皮下を剥離し、同部位の筋膜を露出させた。そこへ上述の支持膜を用いて採取した膵島細胞シートを静置し、細胞シートと筋膜面が付着したところで、支持膜に生理食塩水を添加し、支持膜をゆっくり除去させた。支持膜に水分を添加することで、膵島細胞シート間との吸着力が低下することから、膵島細胞シートが筋膜上にとどまり、シートを乱すことなく移植できた(図9)。移植4日後、失血死させた後、背部の移植した組織を採取した。採取した組織は凍結切片として、インスリン染色での評価と、蛍光顕微鏡下での評価を行った。その結果、膵島細胞にあらかじめ蛍光染色を施すことにより、移植4日後に摘出した組織中から膵島細胞シートの状態で存在することが確認された。また、インスリン染色により、シートを形成する細胞にはインスリン陽性細胞の存在が確認された(図10)。

- 請求項25、26のいずれか1項記載の方法により得られた膵島細胞移植動物に対し被検物質を投与し、当該被検物質の肝機能への影響を判定することを特徴とする膵機能評価システム。
