Description
驚くべきことに、糖アルコールを含む固形組成物はS1P受容体アゴニストの経口投与に特によく適合した製剤を提供する。当該組成物はS1P受容体アゴニストの全身投与に都合のよい手段を提供し、注射剤または経口用途用の液剤製剤の不便さを回避し、かつ、物理化学的にも、貯蔵性においてもよい性質を有している。特に、本発明組成物は高度な安定性を示すとともにS1P受容体アゴニストが組成物全体を通して高いレベルで均一な分布を示す。本発明組成物は高速な自動機器によって製造され、そのことにより手詰めのカプセル化は必要でない。
本発明によれば、以下の態様が提供される。
[1] (a)S1P受容体アゴニストおよび(b)糖アルコール類を含有する経口投与に適した固形医薬組成物。
[2] S1P受容体アゴニストが2−アミノ−2−[2−(4−オクチルフェニル)エチル]プロパン−1,3−ジオールもしくは2−アミノ−2−{2−[4−(1−オキソ−5−フェニルペンチル)フェニル]エチル}プロパン−1,3−ジオールまたはそれらの医薬上許容される塩である[1]記載の組成物。
[3] 糖アルコール類がマンニトールである[1]または[2]記載の組成物。
[4] さらに滑沢剤を含有する[1]記載の組成物。
[5] 滑沢剤がステアリン酸マグネシウムである[4]記載の組成物。
[6] 0.5から5重量%のS1P受容体アゴニストを含有する[1]記載の組成物。
[7] 90から99.5重量%の糖アルコール類を含有する[1]記載の組成物。
[8] 1.5から2.5重量%の滑沢剤を含有する[4]から[7]のいずれかに記載の組成物。
[9] 錠剤の形態である上記いずれかに記載の組成物。
[10] カプセル剤の形態である[1]から[9]に記載の組成物。
R1はアルキル部が直鎖または分枝鎖状の炭素数6〜20の炭素鎖であるフェニルアルキル;またはアルキル部が直鎖または分枝鎖状の炭素数1〜30の炭素鎖であり、フェニルアルキル部が、ハロゲンによって置換されていてもよい直鎖または分枝鎖状の炭素数6〜20の炭素鎖、ハロゲンによって置換されていてもよい直鎖または分枝鎖状の炭素数6〜20個のアルコキシ鎖、直鎖または分枝鎖状の炭素数6〜20のアルケニルオキシ、フェニルアルコキシ、ハロフェニルアルコキシ、フェニルアルコキシアルキル、フェノキシアルコキシ、フェノキシアルキルにより置換されたフェニルアルキル;アルキル部が炭素数6〜20の直鎖または分枝鎖状の炭素鎖であるシクロアルキルアルキル;アルキル部が炭素数6〜20の直鎖または分枝鎖状の炭素鎖であり、炭素数6〜20のアルキルによって置換されたシクロアルキルアルキル;アルキル部が直鎖または分枝鎖状の炭素数6〜20の炭素鎖であるヘテロアリールアルキル;アルキル部が炭素数6〜20の直鎖または分枝鎖状の炭素鎖であり、炭素数6〜20のアルキルによって置換されたヘテロアリールアルキル;ヘテロサイクリック−炭素数6〜20のアルキル;または炭素数6〜20のアルキルによって置換されたヘテロサイクリック−炭素数6〜20のアルキルであり、当該アルキル部は炭素鎖中に二重結合、三重結合、酸素原子、硫黄原子、スルフィニル、スルホニルまたはNR6(ここで、R6は上記定義通りである)から選ばれる結合またはヘテロ原子を有していてもよく、また置換基としてアルコキシ、アルケニルオキシ、アルキニルオキシ、アラルキルオキシ、アシル、アルキルアミノ、アルキルチオ、アシルアミノ、アルコキシカルボニル、アルコキシカルボニルアミノ、アシルオキシ、アルキルカルバモイル、ニトロ、ハロゲン、アミノ、水酸基またはカルボキシを有していてもよい。

Pharmacol.,2000;57;753u)を用いてHEK293細胞の単層へ移入するために用いた。すなわち、DNA25μgおよび0.25M塩化カルシウムを含むDNA混合物をHEPESで緩衝化された2mMリン酸水素2ナトリウムに加えた。HEK293の集密的細胞単層の下層(Subconfluent monolayers)を25mMクロロキンで汚染させ、ついでDNA沈降物を当該細胞に注入した。4時間後、単層をリン酸塩で緩衝化された生理食塩水および栄養(refed)培地(90%1:1ダルベッコの改変必須培地(DMEM):F−12 + 10%牛胎児血清)にて洗浄した。DNAを10%ショ糖含有HME緩衝液(20mMHEPES、5mM塩化マグネシウム、1mMEDTA;pH7.4)中で氷で冷却しながら断片化して添加し、48〜72時間後に細胞を取得し、ダウンス(Dounce)ホモジナイザーを用いて破砕した。800×gで遠心分離後、上清をショ糖を含有しないHMEで希釈し、100,000×gにて1時間遠心分離した。得られたペレットを再度ホモジナイザーに付し、100,000×gにてさらに1時間遠心分離した。この粗膜ペレットをショ糖を添加したHME中に再懸濁し、等分割し、液体窒素中に漬けて素早く凍結させた。膜を70℃で保存した。蛋白質濃度はブラッドフォード(Bradford)蛋白質アッセイ法により分光学的に測定した。
S1P受容体/HEK293膜調製物を用いたGTP γ S結合評価法
GTPγS結合試験は、DS.Im et al.,Mol. Pharmacol.,2000;57;753に記載されたように行う。G蛋白質へのリガンド介在GTPγS結合は一過的に移入されたHEK293細胞からの膜調製物25μgを用いてGTP結合緩衝液(50mM HEPES、100mM塩化ナトリウム、10mM塩化マグネシウム;pH7.5)中にて測定する。リガンドを10μMGDPおよび0.1nM[35S]GTPγS(1200Ci/mmol)の存在下に膜に添加し、30℃にて30分培養する。結合したGTPγSをブランデル(Brandel)ハーベスター(ガイタースバーグ、メリーランド)を用いて非結合物から分離し、液体シンチレーション計数管にて計数する。
本発明は、さらに医薬組成物の製造方法に関しており、それは、(a)S1P受容体アゴニストを糖アルコール類と混合し、(b)(a)で得られた混合物を粉砕および/または造粒し、(c)(b)で得られた粉砕および/または造粒混合物と滑沢剤とを混合することからなる。
a)臓器または組織の移植における拒絶反応の治療および予防、たとえば、心臓、肺、心臓と肺の組合せ、肝臓、腎臓、膵臓、皮膚または角膜移植を受けた患者の治療、および骨髄移植後にしばしば生じる移植組織対宿主病(graft−versus−host disease)の予防;特に、急性または慢性の同種または異種移植における拒絶反応の治療または膵臓小島細胞(pancreatic islet cell)等のインスリン産生細胞の移植等。
b)自己免疫疾患または炎症症状の治療および予防、たとえば、多発性硬化症、関節炎(たとえば、関節リウマチ)、炎症性腸疾患、肝炎等。
c)ウイルス性心筋炎およびウイルス性心筋炎により惹起される肝炎やAIDS等のウイルス性疾患の治療および予防に有用である。
1.上記に定義したような疾患または症状を治療または予防するのに有用な上記に定義した組成物。
を提供する。
実施例1
微粉化した化合物A、すなわち2−アミノ−2−[2−(4−オクチルフェニル)エチル]プロパン−1,3−ジオール塩酸塩(FTY720)を篩過し、篩過した化合物116.7gをマンニトール(エー・メルク社製Parteck M200)9693.3gと混合する。ついで、混合物を30メッシュ篩を用いてFrewitt MGI装置(米国キー・インターナショナル社製)にて粉砕する。ステアリン酸マグネシウムを20メッシュのスクリーンを用いて篩過し、篩過したステアリン酸マグネシウム200gをFTY720およびマンニトール混合物と混和して組成物を調製する。
ステアリン酸マグネシウムの代わりにCutina(登録商標:硬化ヒマシ油)を用いる以外は実施例1の工程を行う。
化合物A、すなわち、FTY720およびマンニトール(エー・メルク社製Parteck M200)をそれぞれ別に18メッシュスクリーンを用いて篩過する。篩過したFTY720 1.9gと篩過したマンニトール40gとを32rpmで混和機中にて120回転させて混合する。ついで、FTY720およびマンニトールの混合物を35メッシュスクリーンを通して篩過する。
ステアリン酸マグネシウムの代わりにCutina(登録商標:硬化ヒマシ油)を用いる以外は実施例3の工程を行う。
ヒドロキシプロピルセルロースの代わりにヒドロキシメチルプロピルセルロースを用いる以外は実施例3の工程を行う。
微粉化された化合物A、すなわちFTY720を400μm(40メッシュ)スクリーンを用いて篩過する。篩過した化合物58.35gをマンニトール(エー・メルク社製Parteck M200)4841.65gと25Lボーレ(Bohre)容器混合機にて240回転させて混合する。ついで、混合物を400μmのスクリーンを用いたFrewitt MGI装置にて粉砕し、粉砕された混合物をもう一度混和する。ステアリン酸マグネシウムを篩過し、篩過した化合物100gをFTY720およびマンニトールの混合物に混和することにより、混和物内でS1P受容体アゴニストがマンニトール中に実質的に均一に分散している目的とする組成物を得る。
別の実施例では、実施例6aに記載した成分および量を用いて、ただし、FTY720は最初(篩過前)にマンニトール14mgと混合してカプセルが製造される。ついで、この混合物を上記のとおり篩過する。篩過した混合物は残りのマンニトールと混和し、ステアリン酸マグネシウムを添加し、その後、追加の混和を行い、カプセルに充填する。
実施例6に記載されたようにカプセル剤を製造する。ただし、各カプセル剤は以下の量での各成分を含有している。
実施例6から8に記載されたようにカプセル剤を製造する。ただし、各実施例でのステアリン酸マグネシウムの代わりにCutina(登録商標:硬化ヒマシ油)を用いる。
実施例1から11に記載されたようにカプセル剤または錠剤が調製される。ただし、各実施例でのFTY720の代わりに2−アミノ−2−{2−[4−(1−オキソ−5−フェニルペンチル)フェニル]エチル}プロパン−1,3−ジオール塩酸塩を用いる。
各成分を秤量し、乳鉢で混合し、ついでカプセルに充填することによって以下の成分を含有するカプセル剤が得られる。
以下の成分を含有する医薬組成物が調製される。
以下の成分(含量はmg単位)を含有する錠剤が調製される。
仕込み量 1170g
給気風量 50m3/分
給気温度 75℃
噴霧速度 15mL/分
噴霧空気圧 1.5N/cm2
噴霧空気量 30L/分
結合剤溶液の量 351mL
以下の成分(含量はmg単位)を含有する錠剤が調製される。
Claims (8)
- (a)S1P受容体アゴニストをマンニトールである糖アルコール類と混合し、
(b)(a)で得られた混合物を粉砕および/または造粒し、
(c)(b)で得られた粉砕および/または造粒混合物と滑沢剤とを混合することを含む、経口投与に適した固形医薬組成物を製造する方法であって、
S1P受容体アゴニストが2−アミノ−2−[2−(4−オクチルフェニル)エチル]プロパン−1,3−ジオールの遊離体またはその医薬上許容される塩の形態であり、滑沢剤が0.01から5重量%の量で添加される方法。 - 0.01から20重量%のS1P受容体アゴニストを添加することを含む請求項1記載の方法。
- 75から99.99重量%の糖アルコール類を使用する請求項1または2記載の方法。
- 糖アルコール類が1m2/gから7m2/gの単点表面積を有する請求項1から3のいずれか1項に記載の方法。
- 糖アルコール類の平均粒子径が100から300μmである請求項1から4のいずれか1項に記載の方法。
- 糖アルコール類の嵩比重が0.4から0.6g/mLである請求項1から5のいずれか1項に記載の方法。
- 医薬組成物が、臓器または組織の移植における拒絶反応、移植組織対宿主病、自己免疫疾患、炎症症状、ウイルス性心筋炎またはウイルス性心筋炎により惹起されるウイルス性疾患の予防または治療用薬剤である請求項1から6のいずれか1項に記載の方法。
- 医薬組成物が、多発性硬化症の予防または治療用薬剤である請求項7記載の方法。
