Claims (15)
- バイオリアクターのための増殖媒体であって、
基板と、複数の多層マトリックスを形成するために積層にされた、2つまたはそれ以上の静電紡糸法により実質的に連続して合成された、ファインファイバーのネットワークと、を有し、
各ファインファイバーの層は、25nm〜2000nmの厚さと、70%以下の固体性と、非細胞傷害性のポリマーを含み、かつ10nm〜1000nmのファイバー直径を有するファイバーと、を有し、
前記ファインファイバーの層の2つまたはそれ以上のうちの1つの層は、ファインファイバーの層中に分散された生物活性な微粒子を含み、
前記ファインファイバーの層の2つまたはそれ以上のうちの1つの層は、前記ファインファイバーの層の前記固体性を低下させかつ前記ファインファイバーの層を通って移動する流体流れを増加させるために、前記ファインファイバーの層内に空隙空間を形成するように前記ファインファイバーの層内に分散されている活性微粒子、不活性微粒子またはそれらの混合物をさらに含み、
前記活性微粒子は、吸収性微粒子、吸着性微粒子、反応性微粒子またはそれらの混合物から選択され、前記吸収性微粒子は流体中の物質を吸収し、前記吸着性微粒子は前記流体中の物質を吸着し、前記反応性微粒子は前記流体中の物質と反応し、
前記不活性微粒子は、不吸収性微粒子、不吸着性微粒子、不反応性微粒子またはそれらの混合物から選択され、前記不吸収性微粒子は前記流体中の物質を吸収せず、前記不吸着性微粒子は前記流体中の物質を吸着せず、前記不反応性微粒子は前記流体中の物質と反応しないことを特徴とする増殖媒体。 - 前記ポリマーはポリエステルまたはポリアミドであることを特徴とする請求項1に記載の増殖媒体。
- 前記ポリエステルは、ポリε−カプロラクトン、ポリグリコレート、またはポリアセテートンであることを特徴とする請求項2に記載の増殖媒体。
- 前記生物活性な微粒子は、細胞、微生物、酵素、生物活性分子またはそれらの混合物であることを特徴とする請求項1乃至請求項3のいずれか1項に記載の増殖媒体。
- 前記生物活性な微粒子は、哺乳動物細胞、昆虫細胞、植物細胞、酵母、細菌、または真菌であることを特徴とする請求項4に記載の増殖媒体。
- 前記微生物は、クロストリジウム属種またはサッカロミセス属種であることを特徴とする請求項4または請求項5に記載の増殖媒体。
- 前記生物活性な微粒子は、セルロース結合領域(CBD)を含み、前記ファインファイバーの層はセルロースまたはセルロース誘導体を含むことを特徴とする請求項1乃至請求項6のいずれか1項に記載の増殖媒体。
- 前記層は、5%〜95%のファイバーと、前記活性微粒子、前記不活性微粒子またはそれらの混合物を含む95%〜5%の微粒子とを含むことを特徴とする請求項1に記載の増殖媒体。
- 前記ファインファイバーの層の2つまたはそれ以上のうちの1つの層は、30%〜75%のファイバーと、前記活性微粒子を含む70%〜25%の微粒子とを含むことを特徴とする請求項1に記載の増殖媒体。
- 少なくとも1つのファインファイバーの層は前記生物活性な微粒子が無く、少なくとも1つのファインファイバーの層は前記ファインファイバーの層全体にわたって分散された前記生物活性な微粒子を有することを特徴とする請求項1乃至請求項9のいずれか1項に記載の増殖媒体。
- 請求項1乃至請求項10のいずれか1項に記載の増殖媒体を含むことを特徴とするバイオリアクター。
- セルロースバイオマスをエタノールまたはブタノールに変換する方法であって、
(a)請求項1乃至請求項10のいずれか1項に記載の増殖媒体上でセルロース、ペクチン、デンプン、キシランまたはそれらの混合物を発酵することができる微生物を培養する工程であって、前記エタノールまたはブタノールが前記発酵の代謝生成物である、前記工程と、
(b)微生物による発酵を促進する条件下で、前記培養した増殖媒体をセルロースバイオマスと接触させる工程と、
を有することを特徴とする方法。 - セルロースバイオマスをエタノールに変換する方法であって、
(a)請求項1乃至請求項10のいずれか1項に記載の第1の増殖表面をセルロースバイオマスと接触させる工程であって、前記第1の増殖表面の生物活性な微粒子が、セルロース、ペクチン、デンプン、キシランまたはそれらの混合物をペントース糖またはヘキソース糖に変換することができるセルラーゼ、または、セルローサムを含む、前記工程と、
(b)微生物による発酵を促進する条件下で、請求項1乃至請求項10のいずれか1項に記載の第2の増殖表面を前記ペントース糖またはヘキソース糖と接触させる工程であって、前記第2の増殖表面の生物活性な微粒子が微生物を含み、エタノールが発酵の代謝生成物である、前記工程と、
を有することを特徴とする方法。 - 前記微生物は、クロストリジウム属種またはサッカロミセス属種を含むことを特徴とする請求項12または請求項13に記載の方法。
- 前記微生物は、クロストリジウム属種またはサッカロミセス属種であることを特徴とする請求項14に記載の方法。
