Description
TGFβ1に対する5種のタイプの特異的な受容体が特徴づけられている(Cheifets S. など. (1988) J. Biol. Chem. 163: 17225-17228及びLopez Casillas F. (1991) Cell 67: 785-795)。それらの受容体は、異なったタイプのTGFβ1に対して異なった親和性を有する。タイプI, II及びIIIの受容体は、これまでの最良の理解である(Attisano L. など. (1994) Biochim. Biophys. Acta 1222: 71-80; Derynck R. (1994) Trends Biochem. Sci. 19: 548-553; Yingling など. (1995) Biochim. Biophys. Acta 1242: 115-136)。
生成される効果は、成長のモーメント及び細胞の型に依存して異なる。
・肝臓星状細胞(Ito細胞)、すなわちマトリックスタンパク質の主要源に対して作用する細胞外マトリックスの拡大(Mustoe TA など. (1987) Science 237: 1333-1336)。
・上皮細胞及び肝細胞の分化(Florini JRなど. (1986) J. Biol. Chem. 261: 16509-16513)。
・ラット胎児肝臓細胞の培養物において観察されるような上皮増殖因子(EGF)の受容体のエンドサイトーシスの阻害(Noda M. and Rodan GA (1987) J. Cell Physiol. 133: 426-437)。
TGFβ1は、細胞外マトリックスのタンパク質の生成において、肝臓星状細胞(脂肪細胞又はIto細胞)によりそれらの受容体の上昇を引き起こし、そしてマトリックスを分解するタンパク質分解酵素の合成を阻害する(Ignot RA and Massague J. (1986) J. Biol. Chem. 261: 4337-4345)、肝線維症の工程に関連することが見出された(Czaja MJ. など. (1989) J. Cell Biol. 108: 2477-2482; Annoni G. など. (1992) J. Hepatol. 14: 259-164)。肝臓においては、TGFβ1は、肝臓星状細胞におけるコラーゲン及びフィブロネクチンの合成を誘発する(Weiner FR (1990) Hepatology 11: 111-117)。そのmRNAの誘発によるそれ自体の合成を高めることによっての自己−調節が存在する。
慢性肝臓損傷を有する患者の調査は、TGFβ1の発現と、タイプIプロコラーゲンについてのmRNAの発現及びプロコラーゲンのタイプIIIペプチドの血清レベルとの間に相互関係が存在することを示した(Castilla A. など. (1991) N. Engl. J. Med. 324: 933-940)。
肝硬変を有する患者は、疾病、例えば門脈圧亢進症又は肝疾患の間に生じる合併症のために正常な生命の予測よりも短い生命の予測を有する。
細胞受容体とTGFβ1との相互作用は、次のものを引き起こす:
・プロコラーゲン、フィブロネクチン(Ignots RA など. (1987) J. Biol. Chem. 262: 6443-6446)、及び関連するタンパク質、例えば細胞外マトリックスの成分と相互作用することができる膜タンパク質(Carter WG (1982) J. Biol. Chem. 257: 13805: 12805-12815)の合成の活性化;
・マトリックスを分解することができるタンパク質分解酵素の合成の阻害(Fukamizu H. and Grinnell F. (1990) Exp. Cell Res. 190: 276-282);
・タンパク質分解酵素のインヒビターの合成の刺激(Fukamizu H. and Grinnell F. (1990) Exp. Cell Res. 190: 276-282)。
身体に存在する分子の類似体として、それらの機能の模倣のために、小さな分子、すなわち合成ペプチドを使用することの可能性が存在する。LeSateur などにより行われた研究は、受容体へのその結合を可能にする、β回転領域を模倣する神経成長因子(NGF)の環状化された類似体の使用の可能性を示す(LeSateur L. など. (1996) Nature Biotechnology 14: 1120-1122)。ペプチドにより介在されるブロッキングによりその受容体と相互作用する生来の因子を妨げる、ペプチド分子のアンタゴニストとしてそれらのペプチドを使用することも可能である(Lasarte JJ など. (1994) J. Acquired Immue Deficiency Syndromes 7: 129-134; Le Sateur など. (1995) J. Biol. Chem. 270: 6564-6569)。
上記理由のために、TGFβ1及びその受容体の両者に由来し、又はTGFβ1に結合する能力を有するタンパク質に由来するペプチドが、TGFβ1の作用のインヒビターであると思われる。従って、本発明者は、この可能性を調査することを決定した。
合成のためのペプチドを、それらがTGFβ1に由来するか又はその受容体に由来するかどうかに依存して、異なった手段で選択した。
TGFβ1の配列の場合、ペプチドは、TGFβ1の完全な配列を含む15個のアミノ酸から合成された。個々のペプチドは、その2種の中間隣接物と同じような10個のアミノ酸を有した。
ペプチドは、α−アミノ基の一時的保護基としてフルオレニルメチルオキシカルボニル(Fmoc)を用いて、固相法(Merrifield (1963) J. Am. Chem. Soc. 85: 2149-34)により合成された(Atherton など. (1989) Journal of Chemical Society Perkins Transactions 1: 538-546)。少量の多数のペプチドの合成のために、96個のペプチドの同時合成を可能にする多合成機が使用された(Barras-Cuestaなど. (1991) Biologicals 19: 187-190)。ペプチドは、使用まで、固体状態で−80℃で貯蔵された。
合成されたペプチドを、Waters 600E-900システム(Millpore Corp., Bedford, USA)を用いて、高性能液体クロマトグラフィー(HPLC)により分析し、そして精製した。
細胞系
ミンク肺上皮に由来する系統、すなわちMV−1−Luを用いた(CCL−64, American Type Cell Culture, Virginia, USA)。細胞を、集密性以下に達するまで、37℃及び5%CO2で、ストーブにおいて、162cm2の培養フラスコ(Costar Corporation, Cambridge, USA)において増殖した。次の完全培地を使用した:5%のウシ胎児血清(FCS, Biological Industries, Kibbuts Beit Haemek, Israel), 10mMのHEPES(1MのHEPES緩衝液、Bio−Whittaker, Verviers, Belgium)及び抗生物質(100U/mlのペニシリン及び100μg/mlのストレプトマイシン)により補充された、L−グルタミンを有するRPMI 1640 (GibcoBRL, Life Technologies Ltd., Paisley, Scotland)。
上記のようにして増殖されたMV−1−Lu細胞を、5mlのトリプシン−EDTA(Biological Industries, Kibbutz Baemek, Israel)を用いて、培養フラスコの底から除去し、完全培地に再懸濁し、そして1500回転/分で8分間、遠心分離した。遠心分離の後、細胞を完全培地において、50,000個の細胞/mlの濃度に再懸濁した。試験を行うために、10mlの細胞懸濁液を採取し、そして96−ウェルの平底培養プレート(Costar Corporation, Cambridge, USA)において、100μl/ウェルで分散し、そして37℃及び5%のCO2で一晩、インキュベートし、ウェルの底への細胞の付着を可能にした。
細胞受容体へのTGFβ1の結合の阻害の調査
細胞受容体の選択的ラベリング(親和性ラベリング)
MV−1−Lu細胞を、培養フラスコから除去し、それらを37℃で10分間、10mlの溶液1(128mMのNaCl、5mMのKCl、25mMの4−(2−ヒドロキシエチル)−1−ピペラジンエタンスルホネート、pH7.5, 5mMのグルコース及び1mMのEDTA) と共にインキュベートした。このようにして除去された細胞を、溶液2(128mMのNaCl, 5mMのKCl, 50mM の4−(2−ヒドロキシエチル)−1−ピペラジンエタンスルホネート、pH7.5, 1.2mMのCaCl2, 1.2mMのMgSO4及び5mg/mlのBSA)に再懸濁し、そして1000×gでの5分間の遠心分離により集めた。遠心分離の後、細胞を106個の細胞/mlの濃度で溶液2に再懸濁した。
界面活性剤において可溶性及び不溶性の画分を、7.5%及び220ボルトで5〜6時間のアクリルアミド/ビスアクリルアミドゲルにおける電気泳動による分析のために使用した。
タンパク質を、メタノール(50%)、酢酸(10%)及び蒸留水におけるクーマシーブリリアントブルー(商標)R250 (Serra Feinbiochemi ca gmbH, Heidelberg, Germany) の溶液により30分間、染色した。続く洗浄を、第1の洗浄においては、メタノール(50%)、酢酸(10%)及び蒸留水の溶液により15分間もたらし、そして続く洗浄においては、メタノール(2.5%)、酢酸(0.5%)及び蒸留水の溶液により、バックグラウンドの色が除去されるまで、もたらした。
細胞受容体への、ペプチドにより仲介されるTGFβ1の結合の阻害を、直接的な免疫蛍光方法により測定した。免疫蛍光キットをこのために使用した(Fluorokine rh TGFβ−ビオチン、R & D Systems Europe Ltd., Abingdon, UK)。子の試験は、シグナル強度が細胞受容体に結合されるTGFβ1の量に依存するよう、特定の態様での細胞受容体に結合するビオチニル化されたTGFβ1の能力、及びフルオレセインによりラベルされたアビジンとビオチンとの続く相互作用に基づかれている。
同腹子(生後5週±1.5週)からの雄白色ラット(albino Wistar 株)を、年齢及び初期体重において均等であるグループを得るために使用した。実験を通して、動物は、明及び暗の12時間サイクルを伴って、一定温度(22℃)の条件下に保持された。それらは、水及び食物を自由に得ることができた。
動物を、肝硬変の誘発工程の開始の前、4種のグループに分けた。
健康な対照(Co):線維症工程にゆだねられなかった動物。
処理された健康な対照(Co+P144):線維症工程にゆだねられておらず、そして最後の3週間、ペプチドp144を投与された動物(ラットTto2のグループの処理と同時に起こる)。
硬変対照1(Ci1):週2度、CCl4の吸入により硬変の誘発工程にゆだねられた動物。それらの動物は、5週目に達すると、次の2つのグループに分離された:
治療された硬変1(Tto 1 ):5週から11週まで、線維症の誘発工程の間、1日おきに、タイプIII受容体の配列に由来するペプチドP144を投与された動物。
硬度対照2(Ci2):ペプチドP144又は塩溶液血清を受けないで、線維症の誘発工程にゆだね続けた動物。このグループは、11週に達すると、さらに2つのグループに細分割された。
治療された硬変2(Tto 2 ):3週間(13週〜15週まで)、タイプIII受容体の配列に由来するペプチドにより処理された硬変動物。
・グループTto1:それらの動物は、線維症工程の間、処理を受けた。
ペプチドによる処理は5週目に開始し(5分間のCCl4への暴露の前)、そして硬変誘発工程の11週目の最後まで続けた。
グループTto2:それらの動物は、硬変の誘発工程の完結の後(11週目)、処理を受けた。処理は、CCl4の最後の吸入の1週間後、開始し、そして21日間続けた。
処理の開始の前、及びその完結に基づいて、ペプチドにより処理されたすべての動物から血液を採取した。ペプチドを、生理食塩水500μl中、70μgの用量(動物当たり)で、腹部領域における皮下注射により投与した。
ラットのモデル及びマウスのモデルの両者において、ペプチドによる動物の処理の完結に基づいて、毛細管を有する眼窩後方叢から採血した後、断頭により、動物を、殺害した。
すぐにこれに続いて、肝臓を切開し、そしてサンプルを採取した。
サンプルを切断し、そして後での組織学的試験のために、固定溶液としてのホルモルに配置した。他のフラグメントを、凍結管に配置し、これを液体窒素に含浸し、そして次に、−80℃で貯蔵した。
組織学的試験を、少なくとも24時間、前もってホルモルに固定された肝臓のフラグメントに対して行い、この後、それらをエタノール(70%)に配置した。
脱水の後、それらをパラフィンブロックに埋封した。3μmの厚さの連続的切片を、Leitz回転ミクロトーム及び鋼製羽を用いて、得られたブロックから調製した。染色の前、断片を、60℃でストーブにおいて15分間、それらを加熱した後、15分間、キシレンにおいてパラフィン除去し(AnalaR, BDH, Poole UK)、そしてそれらを、100%、96%、80%、及び70%の低下する濃度のアルコールに連続的に通すことにより、そして最終的に水において水和化した。次の染料を使用した:
ヘマトキシリン−エオシン;
Masson’s 三原色(Locquin M. and Langeron, (1985) Manual de Microscopia Ed. Labor S.A. Burcelona):コラーゲンタンパク質のための特定の染料の使用(緑色光);
Sirius Red: コラーゲンに対して特異的な染料。
得られるサンプルの像分析のために、個々の調製物の種々の視野の写真を取るビデオカメラ(Sony DXP-950P, Sony Co., Tokyo, Japan)に連結された光顕微鏡を使用した(Olympus BH-2, Tokyo, Japan)。6個の視野を、Sirius Red により染色された個々の調製物からランダムに採取した。獲得された種々の像を、線維症の領域及び調製物の合計領域を計算するコンピュータープログラム(Visilog 4.1.5, Noesis, Orsay, France)により分析した。
この技法のために使用される14μmの切片を、前記3μm切片と同じ手段で得た。それらの切片を、キシレンにおいて12時間、パラフィン除去の工程にゆだねた。パラフィンが除去されるとすぐに、サンプルを、それらを、96%、80%、50%の異なった濃度のアルコールに通し、その工程を蒸留水において完結することにより水和化した。
インビボ実験において得られたデータを、統計学的分析にゆだねた。定量変数の正規性は、Shapiro−Wilks試験により確かめられた。
データは正常な分布に調節されていないので、非パラメーター統計学的分析を採用した。グループ間の比較は、Kruskal-Wallis H, 続いてMann-Whitney Uの比較によりもたらされた。データは、四分位数間領域(ボックスの高さ)と共に、データのメジアン(個々のボックス内の濃い線)の表示を伴って、ボックスによりグラフ的に示され、そして個々のボックスの“ホイスカー(whiskers)”は、与えられる四分位数間領域内の最高及び最低の観察を表す。
0.05に等しいか又はそれ以下のPの値は、有意として見なされた。
すべての統計学的分析は、Window V6.1.3のためのプログラムSPSSを用いて行われた。
MV−1−Lu系の細胞増殖の阻害試験
TGFβ1は、MV−1−Lu細胞系のインビトロ増殖を阻害できるサイトカインであり(Grubeck−Loebenstein B. など. (1989) J. Clin. Invest. 83: 764-770; Brennan FNなど. (1990) Clin. Exp. Immunol. 81: 278-285)、従って、この系はTGFβ1に対するペプチドのブロッキング効果を試験するために使用された。培地、細胞及びチミジンの種々の組み合わせの後、本発明者は、試験のための最も適切な条件を決定するために、培養におけるNV−1−Lu細胞による[メチル−3H]
最適濃度のMV−1−Lu細胞(5000個の細胞/ウェル)及び約90%の阻害率を生成できる最小濃度のTGFβ1(200pg/ml;図18)の両者が決定されると、200μg/mlの濃度での合成ペプチドの阻害効果が試験された。
上記セクション:合成されるべきペプチドの選択に示されるようにして選択された、TGFβ1活性の実質的なインヒビターである合成ペプチド(TGFβ1に結合するタンパク質及びTGFβ1自体に由来するそれら)を、MV−1−Lu細胞系を用いて試験した。ペプチドを、ウシ胎児血清を有さない緩衝されたRPMI培地に溶解し、そして次の方法を使用した:
第1の段階においては、TGFβ1由来のオーバーラッピングペプチドを合成した。それらのペプチド(表2)を、それらのいくつかが細胞受容体に結合し、従ってそれらの受容体への天然のTGFβ1の結合を妨げることを期待して合成した。
表2.TGFβ1由来のペプチド。ペプチドの番号が、その完全な配列におけるその位置及びそのアミノ配列と共に示されている。合成を便利にするために、すべてのペプチドは、表に示されないC−末端で付加されたアラニンにより合成された。
図4から見出され得るように、TGFβ1の配列に由来するペプチドP12は、TGFβ1のより高い阻害活性を示すペプチドである。ペプチドP12の阻害効果のより詳細な研究のためには、下記に記載される、サイトカインの阻害に対するペプチドの濃度の効果についての研究が行われた。
TGFβ1の活性の阻害に対するペプチドP12の濃度の効果を調べた。このペプチドは試験培地において容易に溶解できないので、原溶液又は懸濁液は公称濃度のペプチド(ペプチドが完全に溶解している場合に達成されている)により調製され、そしてアリコートがそれらから取られ、そして濾過され、又はさらに、阻害試験のために直接的に使用された。
結果がペプチドP12により得られると、ペプチドを、N−末端及びC−末端方向に延長し、そしてその活性に対する効果を調べることが決定された。さらに、その溶解性を改良し、そしてTGFβ1の阻害活性に対するその配列における2個のシステインの重要性を研究するために、その配列の変更が行われた。合成されるペプチドが表3に示される。
ペプチドP29が活性的であることが図6から見出され得る。このペプチドは、前に試験されたペプチドP12を包含し、そしてN−末端の方に9個の特別なアミノ酸を有する(図4)。キメラ組換えタンパク質を用いて、Quian SWなど. (1992) Proc. Natl. Acad. Sci. 89: 6290-6294及びBurmester JK など. (1993) Proc. Natl. Acad. Sci. 90: 8628-8632) により行われた調査は、このサイトカインの活性のために必要であるTGFβ1の領域(成熟TGFβ1の配列におけるアミノ酸40〜82)を同定した。
TGFβ1の配列に由来するペプチドP29を、その細胞受容体へのTGFβ1の結合の阻害についてのその能力を確証するために親和性ラベリング試験に使用した(材料及び方法)。
使用される一連の125I− TGFβ1の異なった活性のために、試験において使用されるペプチドの濃度を、個々の場合に使用される一連の125I−TGFβ1の濃度に従って調節した。それらの試験の結果は、図7及び8に示される。
さらなる試験を、細胞受容体への125I−TGFβ1の結合を阻止するために必要とされる最少濃度を見出すために行った。
TGFβ1の活性のインヒビターである新規ペプチドを見出すために、ラットのタイプIII受容体に由来するペプチドを合成した。TGFβ1の配列のアミノ酸阻止に対する相補的であるものとして予測されるそれらの配列の領域に基づいて、いくつかのペプチドを選択した。それらのペプチドが遊離TGFβ1に結合することができ、それを隔離し、そして細胞受容体へのその結合を妨げることが所望される。
図9〜12に見出され得るように、MV−1−Lu細胞系の増殖を、高いか又は低い程度に阻害することができる種々のペプチドが存在するが、しかしペプチドP54のみがTGFβ1の活性をほとんど完全に阻害することができる。このペプチドのより十分な調査を行うために、試験を、200pg/mlの固定された濃度のTGFβ1に対して異なった濃度のペプチドを用いて行った。
TGFβ1の活性の阻害に対するペプチドP54の濃度を調べた。このペプチドの低い溶解性の観点から、呼称濃度のペプチドを有する原液を、ペプチドP12の場合に行われたようにして、調製し、そしてアリコートをそれらから取り、そして濾過し、又はさらに、阻害試験について直接的に使用した。
図13は、濾過の前及び後、呼称濃度のペプチドの阻害効果を試験する。ペプチドP54の濾液において測定できる阻害活性が存在しないことが見出され得る。

ペプチドP144によるTGFβ1の阻害の用量−応答試験
用量−応答試験を、ヒトタイプIII受容体の配列に由来するペプチドP144により、その活性が濃度に依存するかどうかを試験するために行った(図15)。ペプチドの活性が試験に使用されるペプチドの濃度の低下と共に低下することが見出され得る。
ヒトタイプIII受容体の配列に由来するペプチドP144を、その細胞受容体へのTGFβ1の結合の阻害についてのその能力を確証するために親和性ラベリング試験に使用した(材料及び方法)。
使用される一連の125I− TGFβ1の異なった活性のために、試験において使用されるペプチドの濃度を、個々の場合に使用される一連の125I−TGFβ1の濃度に従って調節した。それらの試験の結果は、図15に示される。
TGFβ1に結合する能力を有し、そしてTGFβ1に対して相補的であるとして予測される他のタンパク質に由来するペプチドによるTGFβ1の阻害:
表8及び9.TGFβ1に結合することができる種々のタンパク質(タイプII受容体P146、P149に対するフェチュインP147, P154に対するエンドグリンP150及びP179に対するα2−マクログロブリンP155)に由来するペプチド。ペプチドの番号が、完全な配列におけるその位置、そのアミノ酸配列及びその起源と共に示されている。合成を便利にするために、すべてのペプチドは、表に示されないC−末端で付加されるアラニンにより合成された。
図17及び18に見出され得るように、ペプチドP150のみが、50%以上の活性を示した。しかしながら、Demetriou M. など. (1996) J. Biol. Chem. 271: 12755-12761により活性的であるものとして記載されたペプチドP146及びP149は、この試験のために使用される条件下で活性的であることが見出されなかった。
TGFβ1のその細胞受容体への結合に対する合成ペプチドの阻害効果の流動細胞計測法による測定:
MV−1−Lu細胞系の増殖の阻害のバイオアッセイにおいて活性的であることがわかっているヒトタイプIII受容体の配列に由来するペプチドP144を、CCl4による試験的硬変の誘発におけるその阻害効果を研究するためにラットにおけるインビボ試験に使用した。
このモデルにおいては、肝硬変を、材料及び方法に記載のようにして、1週当たり2度、11週間、四塩化炭素の注入により誘発する(Lopez Novoa JM など. (1976) Patologia IX: 223-240: Camps J. など. (1987) Gastroenterology93: 498-505)。
1.プロトコール1:ペプチドを、硬変誘発工程(11週間)の間、腹腔内経路により1日おきに投与した。図20及び21。
2.プロトコール2:ペプチドを、硬変が確立されると、すなわち硬変の誘発の開始から12週目で、3週間、腹腔内経路により1日おきに投与した。図22及び23。
図36及び38は、Fast Green 及びDirect Redによる肝臓切片(動物当たり2個)の染色、色彩の溶出及び分光計による読み取りにより測定される全コラーゲン生成を示す(材料及び方法)(Lopez de Leon A. and Rojkind (1985) Histochem. Cytochem. 33: 737-743; Gaudio E. など。(1993)Int. J. Exp. Path. 74: 463-469)。
図20に見出され得るように、ペプチドP144により処理されたラットグループ(Tto1)と硬変ラットの対照グループ(Ci1)との間の、合計タンパク質に対するコラーゲンの割合の研究に基づく有意な差異が観察される(P<0.05)。図37においては、ペプチドP144により処理されたラットのグループ(Tto1)と硬変ラットの対照グループ(Ci1)との間の差異はまた、線維症の領域が調べられる場合、有意である(P<0.001)。
コラーゲンを測定するために使用される2種の技法を、個々の調製における調査のための視野の選択のランダム性、及び従って、像分析の有効性を確かめるために、線状回帰を用いて比較した(図24及び25)。
図26及び27は、硬変誘発工程の間に処理されたラット(Ci1及びTto1)の肝臓から得られた、Sirius Red により染色された肝臓調製物からの10倍の倍率での光顕微鏡により得られる像を示す。
図27は、それらの像が特定のソフトウェアーを用いて研究のために改良された後の像を示す。それらの改良は、像の品質を改良し、そしてそれらの自動試験を促進するために、2種のフィルター、すなわち偏光された光及び緑色光の2種のフィルターの適用から成る。
図26及び27は、硬変ラット(Ci1)から得られた像と、ペプチドP144により処理されたラット(Tto1)から得られた像との間に差異が存在することを示す。
硬変の誘発の工程の最後での処理されていない硬変ラットのグループ(Ci1)と、誘発の最後から4週目での処理されていない硬変ラット(Ci2)とを比較する場合、2種のグループ間に有意な差異(P=0.016)が存在(図28)することが見出され、これは、硬変剤が除去される場合、硬変の部分的緩衝が存在することを示し、この観察は種々の業者により公開されている(Szende-Zなど. (1992) in Viro 6: 355-361: Columbano A (1996) Carcinogenesis 17: 395-400)。

- 形質転換成長因子β1(TGF−β1)受容体へのTGF−β1の結合のアンタゴニストペプチドであって、そして下記のアミノ酸配列:
P152 Leu Asp Ser Leu Ser Phe Gln Leu Gly Leu Tyr Leu Ser Pro His
から成るポリペプチド。 - 請求項1に記載のアンタゴニストペプチドを含んでなる肝臓疾患治療剤。
- 前記肝臓疾患が肝線維症である請求項2に記載の治療剤。
- 前記肝臓疾患が肝硬変である請求項2に記載の治療剤。
- 肝臓疾患治療剤の製造のための請求項1に記載のアンタゴニストペプチドの使用。
- 前記肝臓疾患が肝線維症である請求項5に記載の使用。
- 前記肝臓疾患が肝硬変である請求項5に記載の使用。
