Abstract
Description
必ずしも必要ではないが、検出または治療目的のために、本発明の抗体または抗原結合性フラグメントは、検出可能部分(すなわち、検出もしくは診断目的に対応)または治療部分(治療目的に対応)とコンジュゲートしてもよい。
抗体(結合または非結合)の医薬組成物もまた、本発明に包含される。医薬組成物は、抗体または抗原結合性フラグメントを含んでもよく、また薬学的に許容可能な担体も含んでいてもよい。

相同性モデル化により、このタスクを達成した。マウス25D8(配列番号22および配列番号26)ならびに25E9(配列番号6および配列番号12)可変配列と最も類似した鋳型構造を、PDBに対するblast検索により同定した。マウス25D8可変領域の初期モデルを構築するために、以下の鋳型構造を用いた(PDBコード):軽鎖については3CFC、および重鎖については1NGQ。マウス25E9可変領域の初期モデルを構築するために、以下の鋳型構造を用いた:軽鎖については1AE6、および重鎖については1NMC。これらの鋳型構造上に、マウス25D8および25E9配列に従って突然変異を操作した:3CFC軽鎖(全てCDR内)において3つの突然変異、1NGQ重鎖において17の突然変異(フレームワーク内で3つ、CDR内で14)、1AE6軽鎖において7つの突然変異(フレームワーク内で4つ、CDR内で3つ)、および1NMC重鎖において34の突然変異(フレームワーク内で17、CDR内で17)。CDRループは、各抗体におけるCDR−H3ループ(1NGQから25D8に2残基欠失を、1NMCから25E9に1残基挿入を実施した)を除いて、CDRループは、長さの調節を一切必要としないと思われた。それぞれの鋳型構造の重鎖および軽鎖を重ね合せることにより、マウス25D8および25E9可変ドメインの重鎖および軽鎖に対応する突然変異構造を実質的に2本鎖抗体構造にアセンブルした。まず初めに、AMBER力場、ならびに最初に弛緩したCDRループから、最後の段階で初めて完全に弛緩したフレームワーク領域の骨格重原子まで変動する段階的な束縛開放を用いたエネルギー最小化により、アセンブルした25D8および25E9可変ドメインの得られる構造を精密化した。次に、各抗体可変ドメイン構造のCDR−H3ループをMonte−Carlo最小化(MCM)構造探索により精密化したが、ここで、CDR−H3領域中の二面角をMCM周期毎にサンプリングした後、CDR−H3ループの初期構造の周辺に10Å延びる、予め定めた領域のエネルギー最小化を行った。
このステップを実施して、ヒト度指数(humanness index)、抗原接触傾向指数を推定し、CDR、カノニカル残基(canonical residue)、分子鎖間パッキング(VH/VL界面残基)、可変/定常領域パッキング(VH/VLおよびVL/CL界面残基)、特異フレームワーク残基、潜在的なNおよびO−グリコシル化部位、埋没残基(buried residue)、バーニアゾーン(Vernier zone)残基、およびCDRとの隣接残基を画定した。インターネットで入手可能な試料およびローカルソフトウエアを用いて、これらの特性を評価した。
これは、ヒト生殖細胞系データベース(VBASE)の局所コピー、他の配列ライブラリー(GenbankおよびSwissProt)、ならびにヒトフレームワークコンセンサス配列のセットとの標準配列相同性比較により実施した。BLAST検索を実施して、フレームワーク領域内のみ(従って、CDRは除外する)で最も高い相同性を有する配列マッチを検索すると共に、CDRループの長さをマッチングした。重鎖および軽鎖について同定したヒトフレームワークは、25D8および25E9抗体のいずれも、それぞれ、k2およびh1クラスに対応する。突然変異の候補位置でのアミノ酸可変性を評価すると共に、ヒト化の際の親和性喪失のイベントにおけるバックアップとして好適なフレームワークのプールを提供するために、複数の高度に類似したヒトフレームワーク配列を保持した。
これは、特に注意して、対応するヒト配列に対し、突然変異を実施すべきアミノ酸残基をフラッグする重要なステップである。これらの残基は、親和性損失の場合に、マウス配列への復帰突然変異のための一次候補を示す。これは、とりわけ、抗体−抗原複合体の実験構造の非存在下で、設計によるヒト化の最も困難かつ予測不可能なステップである。これは、以下のカテゴリー:カノニカル(canonical)残基、CDR−H3残基、バーニアゾーン(Vernia zone)残基、特異残基、CDRとの近接残基(5Å以内)、分子鎖間パッキング残基、およびグリコシル化部位残基の1つまたは複数における残基の同定を利用する。このような残基は、抗原結合部位および親和性に影響を直接または間接的に与え得る。また、各位置でのヒト生殖細胞系データベースにおける抗原接触傾向指数およびアミノ酸発生も、特定の残基がマウス配列からヒト配列へと安全に突然変異することができるかどうかを決定する上で、極めて重要である。25D8および25E9軽鎖および重鎖可変配列の、提案されるヒト化配列をそれぞれ図2および3に示す。各ヒト化配列およびそのドナーマウス配列、ならびにいくつかのアラインメントした候補アクセプター配列の間のフレームワーク突然変異の数も記載する(括弧内にフレームワークの割合(%)として示す)。突然変異残基および復帰突然変異のための候補残基も図1、2および3に示す。図からわかるように、25D8および25E9抗体の軽鎖は、各々の提案されたヒト化フレームワークに対して、それぞれ9および11の突然変異を必要とすると考えられる。これは、軽鎖については100%フレームワークヒト化の試みを表す。各抗体の重鎖は、ヒト化のためにその軽鎖より実質的に多くの突然変異、すなわち、25D8の場合には18、25E9の場合には17を必要とすると考えられる。さらに、重鎖のヒト化配列は、ヒトフレームワーク配列と完全には(100%)対応しない。特に、25E9重鎖の場合には、提案されたフレームワークヒト化の最高レベルは、最初の試みで94%であり、これは、ヒト化配列が、最も近いヒトフレームワーク配列と5残基異なることを意味する。25E9マウス配列由来のこれら残基のうち4つを保持する決定を、入念な構造および比較配列解析に基づいて行ったところ、抗原結合CDRとの近接(図1bを参照)により、突然変異が、次の位置:GluH1、IleH2、ThrH93、およびSerH94に導入されれば、抗原結合親和性が改変される高い確率を示した。Gluは、ヒトフレームワーク配列においてH1位置にみいだされる共通の残基であることに留意しなければならない(図3参照)。ヒト化配列において直近のヒトフレームワークと異なる第5残基は、マウス配列におけるHisH43であり、これは、ヒト化配列においてGlnH43に突然変異した(Glnは、この位置で共通であるが、Hisは、希有である)。25D8可変重鎖フレームワークのヒト化の場合、これは、99%に達した。すなわち、直近のヒトフレームワークに出現するAspH81をマウス配列のGln81で置換したヒト化配列において、直近のヒトフレームワーク配列に対し、提案されたヒト化配列のフレームワークにおける1つの残基:GluH81が異なった。位置H81のAspをGluに突然変異させる決定は、ヒトフレームワークにおいて考えられるこれら2つの置換の相対的発生頻度に基づいて行った(図2を参照)。全体として、25D8抗体のヒト化は、25E9抗体のそれより容易であると結論付けることができる。
ヒト化配列を組換え発現に付す前の追加的構造解析は、シグナルペプチドの選択、アイソタイプの選択、可変/定常領域結合部での構造適合性の解析を含んだ。加えて、マウスおよびヒト化配列同士の鎖間パッキングおよび可変/定常領域パッキングの比較解析から、25D8および25E9ヒト化の場合に、ヒト化およびキメラ(マウス可変領域)鎖を組み合わせたハイブリッド抗体、すなわち軽鎖/重鎖ペアリングとしてのマウス/マウス(M/M)、マウス/ヒト化(M/H)、ヒト化/マウス(H/M)およびヒト化/ヒト化(H/H)抗体を作製することが実現可能となり得ることが判明した。25D8および25E9全長IgG2抗体についてアセンブリングしたヒト化およびキメラ配列を図4Aおよび図4Bにそれぞれ示す。25D8および25E9全長IgG1抗体についてアセンブリングしたヒト化およびキメラ配列を図12Aおよび12Bにそれぞれ示す。
一過性トランスフェクションにより、マウスおよび選択されたヒト化またはキメラ25D8 IgG2およびヒト化25E9(L1H1 IgG2、L1H1 IgG1、L1H2 IgG、L1H3およびL1H1 IgG1変異体)抗体の小さなロットを生成して、いくつかの比較解析を実施することができるように、これらを精製した。表面プラズモン共鳴(SPR)方法を用いて、組換えSiglec−15と様々な抗体の直接結合を測定した。ELISA方法と同様に、SPR実験で用いたSiglec−15を293−6E細胞においてFc−Siglec−15融合タンパク質として発現させた。Fcコンジュゲートとして、Fc領域でのホモ二量体相互作用により、タンパク質を二量体として発現させ得ることに留意すべきである。この事象は、結合中に結合活性効果を生み出すことができたが、これにより、親和性定数の直接決定を実施することができなかった。さらに、抗体およびSiglec−15タンパク質の両方にFc領域が存在すると、直接親和性決定を行うことができない。従って、各抗体サンプルの結合結果は、互いに対する関連でのみ表示する。
細胞培養
破骨細胞分化を誘導するために、10%胎仔ウシ血清(Gibco)および1mMピルビン酸ナトリウムを含有するDMEM中に増殖させたマウスRAW264.7細胞を擦り取り、PBS中に再懸濁させた。100ng/mlマウスRANKL(R&D Systems,Minneapolis,MN)を含む培地中で細胞を2×104細胞/cm2で平板培養した。細胞を3日間(免疫蛍光顕微鏡検査のために)または4日間(他の全ての実験のために)分化させた。CD14マイクロビーズおよびMSカラム(Miltenyi Biotec,Cologne,Germany)を用い、製造者の指示に従い、ヒト破骨細胞前駆細胞(CD14+末梢血液単核細胞(PBMC))を正常ヒトPBMC(AllCells,Emeryville,CA)から単離した。10%胎仔ウシ血清(HyClone)、1mMピルビン酸ナトリウム(HyClone)、25ng/mlのヒトMCSFおよび30ng/mlのヒトRANKL(R&D Systems)を含有するAlpha−MEM(Gibco)中で、細胞を3.1×105細胞/cm2で平板培養した。第4日に培地の半分を取り換え、細胞を7日間分化させた。
単一抗体による細胞刺激のために、様々な時点で細胞を溶解させる前に、表示した抗体濃度を含む新鮮な増殖培地(RANKLを含まない)と分化培地を取り換えた。一次および二次(架橋)抗体による刺激のために、分化培地を、10μg/mlで一次抗体を含有する低温増殖培地と取り換えた後、細胞を4℃で20分インキュベートした。次に、抗ヒトIgGポリクローナル抗体(Jackson Immunoresearch,West Grove,PA)を含む温かい増殖培地と取り換えた後、37℃で、表示した時間、溶解まで細胞をインキュベートした。
破骨細胞分化および機能に対する抗体の作用を試験するために、表示した濃度の抗体を含む培地において、前述のように細胞を誘導して分化させた。TRAP染色により、培養して4日後に破骨細胞を視覚化した。手短には、細胞を3.7%ホルムアルデヒドで固定し、0.2%Triton X−100/PBSで透過性にし、37℃で約30分TRAP染色バッファー(100mM酢酸ナトリウム、pH5.2、50mM酒石酸ナトリウム、0.01%ナフトールASMXおよび0.06%Fast Red Violet)中にインキュベートした。TRAP酵素は、破骨細胞中に赤い反応産物を生成する。破骨細胞の吸収活性を試験するために、リン酸カルシウム基質でコーティングしたウェル(Osteologic,BD BioSciencesまたはOsteoAssay,Corning)内に細胞を塗布した後、前述のように分化させた。7日後、ウェルを漂白剤で処理することにより、細胞を除去し、基質吸収の領域を光学顕微鏡検査により観察した。Siglec−15の活性(破骨細胞または破骨細胞前駆細胞において)を阻止することができる抗体は、例えば、より少ないTRAP陽性多核細胞を示すか、または改変形態のTRAP陽性多核細胞をもたらし得る。これを図9に示す。上方のパネルに示すように、1μg/mlのヒト化25E9抗体(L1H1 IgG1変異体)に暴露したヒト破骨細胞(上方の右側パネル)は、成熟した多核化破骨細胞を適正に形成することができなかった。対照的に、等量の対照抗体で処理したヒト破骨細胞は、正常に分化した(上方の中央パネル)。破骨細胞が、ミネラル化基質を活発に消化することから、Siglec−15の活性(破骨細胞または破骨細胞前駆細胞において)を阻止することができる抗体は、例えば、対照(例えば、Siglec−15に結合していない抗体、抗体の非存在など)と比較して、カルシウム基質が消化された領域(露出領域)が、より少ないと考えられる。ヒト破骨細胞を、骨様表面として作用するリン酸カルシウム基質上で分化させたところ(図9を参照)、対照抗体で処理した細胞(下方の中央パネル)は、広い面積の露出したリン酸カルシウムを生成したが、これは、破骨細胞が、骨吸収活性を発揮することを示している。対照的に、1μg/mlの25E9(L1H1 IgG1変異体)で処理した細胞(下方の右側パネル)は、基質を吸収することができなかったが、これは、非分化前駆細胞(下方の左側パネル)と同様であった。
細胞表面タンパク質をビオンチン標識するために、分化RAW264.7由来の破骨細胞を1mM CaCl2および1mM MgCl2(PBS/Ca/Mg,HyClone)含有の低温PBSで2回すすいだ後、PBS/Ca/Mgで1mg/mlに希釈したビオチン標識試薬:スルホ−NHS−SS−ビオチン(Pierce)と一緒に4℃で1時間インキュベートした。グリシン(PBS/Ca/Mg中100mM)で、未反応ビオチン標識試薬をクエンチングすることにより、反応を停止した。Siglec−15内在化を誘導するために、「細胞刺激」において前述したように、抗Siglec−15抗体で、もしくは対照ヒトIgG単独で、または二次架橋抗体と組み合わせて、細胞を処理した。抗体処理後、低温NTバッファー(20mM Tris/HCl、pH8.6、150mM NaCl、1mM EDTAおよび0.2%BSA)で細胞を2回すすいでから、低温NTバッファー中25mMで調製したナトリウム−2−メルカプトエタンスルホネート(MesNa)で2×25分インキュベートすることにより、スルホ−NHS−SS−ビオチンのジスルフィド結合を低減し、これによって、残留する細胞表面ビオチンを除去した。Siglec−15ビオチン標識の最大可能レベルを測定するために、1つの対照についてMesNa処理を省いた(これらの対照細胞は、NTバッファー単独と一緒2×25分インキュベートした)。次に、PBS/Ca/Mgで5mg/mlに希釈したヨードアセトアミドで、残留MesNaを15分間クエンチングした。
結合したリガンドおよび抗体のエンドサイトーシスを媒介する能力は、Siglecファミリーのいくつかのメンバーについて立証されたが、実際に、治療抗体の細胞取込みは、CD22およびCD33Siglecをターゲティングする抗体薬剤コンジュゲートの作用のメカニズムの重要な要素である(O’ReillyおよびPaulson,2009)。興味深いことに、Siglec−15はまた、その細胞質ドメイン中にYxxφ配列も含む(このチロシン、Y309はまた、推定ITIMモチーフの一部でもある);Yxxfモチーフは、クラスリンアダプターAP−2と相互作用して、受容体内在化を調節することができる(Angata et al.,2007;BonifacinoおよびTraub,2003)。従って、本発明者らは、破骨細胞におけるSiglec−15エンドサイトーシスに対する抗体結合の効果を調べた。
細胞溶解物の調製および免疫沈降
プロテアーゼおよびホスファターゼ阻害剤(50mM NaF、1mM NaVO4および1xRoche Complete EDTA非含有ホスファターゼ阻害剤)を含むmRIPAバッファー(50mM Tris/HCl、pH7.4、1%NP−40、0.25%デオキシコレート、150mM NaCl)を用いて、細胞溶解物を調製した。溶解物タンパク質濃度をBCAアッセイ(Pierce)により測定した。全細胞溶解物のウェスタンブロッティングのために、等量のタンパク質(10〜15ug)を、βメルカプトエタノール含有SDSサンプルバッファー中で熱変性させ、10または12%SDS−PAGEゲル上で分離してから、PVDFに移した後、表示した抗体でプロービングした。免疫沈降のために、2mgまたは1mgの全溶解物を、4μgの抗体および15μlのタンパク質G−Sepharoseビーズと一緒に、4℃で回転しながら、4時間インキュベートした。ビーズをmRIPAで4回洗浄した後、沈降物質の半分を前述したようにウェスタンブロッティングにより分析した。
近年の研究から、293T細胞におけるSiglec−15およびDAP12のエピトープタグ付加形態の共発現時に、複合体を検出することができ、これは、K273の存在に依存することが立証された(Angata et al.,2007)。本発明者らは、類似の過剰発現条件下で複合体を検出することもでき(データは示さず)、この複合体が、破骨細胞において内性発現レベルでも存在するかどうかを決定した。分化RAW264.7由来の破骨細胞、ならびに非分化対照細胞からタンパク質溶解物を調製し、Siglec−15およびDAP12抗体を用いて免疫沈降を実施した。DAP12は、抗Siglec−15を用いて沈降したタンパク質複合体中に容易に検出され、同様に、抗DAP12も多量のSiglec−15を沈降した。予測した通り、Siglec−15タンパク質発現レベルに基づいて、この複合体は高度に破骨細胞特異的であり、非分化細胞中には検出されなかった。特に、DAP12発現は、RAW264.7破骨細胞の分化中に著しく変化しなかった。
IgG1およびIgG2抗体変異体の比較
続いて、本発明者らは、ヒト化抗Siglec−15 IgG1抗体変異体と、対応するヒト化抗Siglec−15 IgG2抗体変異体(すなわち、これらの抗体は、同じ可変ドメインを有するが、重鎖のヒト定常領域が異なる)とを比較して、以下に説明するin vitro実験において、IgG1が、対応するIgG2よりはるかに活性であることをみいだした。
Siglec−15をターゲティングする抗体−薬剤コンジュゲート(ADC)
本出願人は、破骨細胞の表面上にSiglec−15との抗体の結合が効率的に内在化された後、分解されたことを明らかにした。また、別の研究から、内在化後にエンドソーム経路が続くため、Siglec−15/抗体複合体を、後期エンドソーム/リソソームのマーカであるLAMP2と共局在化することができることも判明した。実験を実施して、ADCによりSiglec−15発現細胞をターゲティングする実現可能性を調べた。ヒト化25E9 L1H1 IgG1変異体を、増殖細胞ならびに非増殖細胞、または完全に分化した細胞、とりわけ破骨細胞に有毒なペイロードとコンジュゲートした。ヒト化25E9 L1H1 IgG1コンジュゲート抗体を25E9−ADCと呼ぶ。コンジュゲート形成は、25E9が、ネイティブSiglec−15と相互作用する能力に影響を与え得ることから、フローサイトメトリーを実施して、25E9−ADCとSiglec−15発現細胞の結合を測定した。ヒトSiglec−15をコードするcDNAでトランスフェクトした293−6E細胞を用いて、前述のように実験を実施した。図15に示すように、非コンジュゲート25E9および25E9−ADCは、Siglec−15トランスフェクト細胞と、よく似た親和性で相互作用した(黒い曲線、h−Siglec−15を参照)。これは、コンジュゲート形成反応が、Siglec−15と結合する25E9−ADCの能力を変えないことを示した。対照として、Siglec−15cDNAを含まないプラスミドでトランスフェクトした細胞(図15のグレーの曲線、ベクターを参照)は、25E9と結合しなかったが、これは、抗体が、上記細胞と非特異的に結合しないことを示している。
in vivo機能アッセイ−マウス
in vivo効果の評価は、急速に成長する骨を有する非常に若いマウスを用いて、Schenk(Muhlbauer et al.,1991)により記載される方法から改変した。手短には、生後3〜4週の雄マウス(5匹/グループ)をPBS、対照マウスIgGまたはマウスSiglec−15と結合することができる抗Siglec−15抗体のいずれかで処理した。抗体は、26G針を用いて、4週間にわたり毎週2回腹腔内に投与した。マウスを死なせ、骨を切除し、24時間にわたり4%パラホルムアルデヒド中に固定した。PBSで洗浄した後、PIXImus Densitometer(GE Medical Systems)を用いて、骨をスキャンすることにより、大腿骨、脛骨および脊椎の骨ミネラル密度(BDM)を決定した。骨の3次元画像をSkyScan high resolution microCT(SkyScan Inc.,Kontich,Belgium)で作成した。
霊長類における骨バイオマーカに対するSiglec−15阻害の作用を調べるために、Siglec−15をターゲティングするヒト化モノクローナル抗体、25E9を、エストロゲン欠乏雌カニクイザルに投与した。

- Siglec−15との特異的結合が可能であり、以下:
a.配列番号6と少なくとも80%同一の軽鎖可変領域および/または配列番号12と少なくとも80%同一の重鎖可変領域を有する抗体またはその抗原結合性フラグメントであって、配列番号6または配列番号12と比較して少なくとも1つのアミノ酸置換を含み、前記アミノ酸置換が、相補性決定領域(CDR)の外側にある、抗体またはその抗原結合性フラグメント;ならびに
b.配列番号22と少なくとも80%同一の軽鎖可変領域および/または配列番号26と少なくとも80%同一の重鎖可変領域を有する抗体またはその抗原結合性フラグメントであって、配列番号22または配列番号26と比較して少なくとも1つのアミノ酸置換を含み、前記アミノ酸置換が、相補性決定領域(CDR)の外側にある、抗体またはその抗原結合性フラグメント
