本発明での使用に適切な細菌抗原には、細菌から単離、精製、または由来することができるタンパク質、多糖類、リポ多糖類、外膜小胞が含まれる。さらに、細菌抗原には、細菌溶解物および不活化細菌処方物が含まれ得る。細菌抗原を、組換え発現によって産生することができる。細菌抗原には、好ましくは、その生活環の少なくとも1段階に細菌表面に曝露されるエピトープが含まれる。細菌抗原は、好ましくは、複数の血清型で保存される。細菌抗原には、下記の1つまたは複数の細菌由来の抗原および以下で同定した特定の抗原例が含まれる。

本発明で用いる真菌抗原は、1つまたは複数の下記の真菌に由来し得る。
本発明の組成物は、1つまたは複数の性感染症(STD)由来の抗原を含むことができる。かかる抗原は、STD(クラミジア、陰部ヘルペス、肝炎(HCVなど)、陰部疣贅、淋病、梅毒、および/または軟性下疳など)を予防または治療することができる(WO00/15255号を参照のこと)。抗原は、1つまたは複数のウイルスまたは細菌のSTDに由来し得る。本発明で用いるウイルスSTD抗原は、例えば、HIV、単純ヘルペスウイルス(HSV−1およびHSV−2)、ヒトパピローマウイルス(HPV)、および肝炎(HCV)に由来し得る。本発明で用いる細菌STD抗原は、例えば、淋菌、トラコーマクラミジア、梅毒トレポネーマ、軟性下疳菌、大腸菌、およびストレプトコッカス・アガラクチアに由来し得る。これらの病原体由来の特異的抗原の例は、上に記載している。
本発明の組成物は、呼吸器疾患を引き起こす病原体由来の1つまたは複数の抗原を含むことができる。例えば、呼吸器抗原は、呼吸器ウイルス(オルソミクソウイルス(インフルエンザ)、ニューモウイルス(RSV)、パラミクソウイルス(PIV)、モルビリウイルス(麻疹)、トガウイルス(風疹)、VZV、およびコロナウイルス(SARS)など)に由来し得る。呼吸器抗原は、呼吸器疾患を引き起こす細菌(肺炎連鎖球菌、緑膿菌、百日咳菌、結核菌、肺炎マイコプラズマ、クラミジア・ニューモニエ、炭疽菌、およびカタル球菌など)に由来し得る。これらの病原体由来の特異的抗原の例は、上に記載している。

本発明の組成物は、青年期被験体での使用に適切な1つまたは複数の抗原を含むことができる。青年期は、以前に投与した小児ワクチンの追加免疫が必要であり得る。青年期での使用に適切であり得る小児抗原は、上に記載されている。さらに、性行為の開始前に防御免疫または治療的免疫を確実にするために、STD病原体由来の抗原を投与するために青年期を標的化することができる。青年期での使用に適切であり得るSTD抗原は、上に記載されている。
本発明の他の態様では、吸着抗原を有する微粒子の生成方法を提供する。本方法は、(a)(i)水、(ii)界面活性剤、(iii)有機溶媒、および(iv)ポリ(α−ヒドロキシ酸)、ポリヒドロキシ酪酸、ポリカプロラクトン、ポリオルソエステル、ポリ酸無水物、およびポリシアノアクリラートからなる群より選択される生分解性ポリマーを含む混合物の分散によって乳濁液を準備する工程を含む。ポリマーは、典型的には、有機溶媒と比較して約1%〜約3%の濃度の混合物で存在する一方で、界面活性剤は、典型的には、約0.00001:1〜約0.1:1(より典型的には、約0.0001:1〜約0.1:1、約0.001:1〜約0.1:1、または約0.005:1〜約0.1:1)の水:水、界面活性剤:ポリマー比で混合物中に存在する、準備する工程、(b)乳濁液から有機溶媒を除去する工程、および(c)微粒子表面上に抗原を吸着させる工程を含む。一定の実施形態では、生分解性ポリマーは、有機溶媒と比較して約3%〜約10%の濃度で存在する。

アジュバント
本発明のワクチンを、他の免疫調節薬と併せて投与することができる。特に、組成物は、通常、アジュバントを含むであろう。本発明と共に使用するためのアジュバントには、1つまたは複数の下記のアジュバントが含まれるが、これらに限定されない。
本発明でのアジュバントとしての使用に適切なミネラル含有組成物は、ミネラル塩(アルミニウム塩およびカルシウム塩など)を含む。本発明は、水酸化物(例えば、オキシヒドロキシド)、リン酸塩(例えば、ヒドロキシホスフェート、オルトホスフェート)、硫酸塩などのミネラル塩(例えば、chapters8 & 9 of Vaccine Design…(1995)eds.Powell & Newman.ISBN:030644867X.Plenum.)または異なるミネラル化合物の混合物(例えば、リン酸塩アジュバントと水酸化物アジュバントとの混合物(任意選択的にリン酸塩が過剰))を含み、この化合物は任意の適切な形態(例えば、ゲル、血漿、無定形など)をとり、塩に吸着していることが好ましい。ミネラル含有組成物を、金属塩の粒子として処方することもできる(WO00/23105号)。
本発明でのアジュバントとしての使用に適切な油性乳濁液組成物には、スクアレン−水乳濁液(MF59(ミクロフルイダイザー(microfluidizer)を使用して、5%スクアレン、0.5%TWEEN(商標)80、および0.5%Span85をサブミクロン粒子に処方したもの)など)が含まれる。WO90/14837号を参照のこと。Podda,Vaccine(2001)19:2673−2680;Freyら、Vaccine(2003)21:4234−4237も参照のこと。MF59を、FLUAD(商標)インフルエンザウイルス3価サブユニットワクチンにおけるアジュバントとして使用する。
サポニン処方物を、本発明でアジュバントとして使用することもできる。サポニンは、ステロールグリコシドおよびトリテルペノイドグリコシドの異種群であり、広範な植物種の樹皮、葉、幹、根、さらに花弁中で見出される。キラヤサポナリア モリナ(Quillalaia saponaria Molina)の木の樹皮から単離したサポニンは、アジュバントとして広く研究されている。スミラックス・オルナタ(Smilax ornata)(サルサパリラ(sarsaprilla))、シュッコンカスミソウ(Gypsophilla paniculata)(ブライドベール(brides veil))、およびサボンソウ(ソープルート(soap root))由来のサポニンを購入することもできる。サポニンアジュバント処方物には、精製処方物(QS21など)および脂質処方物(ISCOMなど)が含まれる。

本発明での使用に適切なアジュバントには、以下などの細菌または微生物誘導体が含まれる。
かかる誘導体には、モノホスホリル脂質A(MPL)および3−O−脱アシル化MPL(3dMPL)が含まれる。3dMPLは、3−O−脱アシル化モノホスホリル脂質Aと4、5、または6アシル化鎖との混合物である。3−O−脱アシル化モノホスホリル脂質Aの好ましい「小粒子」形態は、EP0689454号に開示されている。かかる3dMPLの「小粒子」は、0.22ミクロン膜で濾過滅菌するのに十分に小さい(EP0689454号)。他の非毒性LPS誘導体には、モノホスホリル脂質A模倣物(アミノアルキルグルコサミニドホスフェート誘導体(例えば、RC529)など)が含まれる。Johnsonら(1999)Bioorg Med Chem Lett 9:2273−2278を参照のこと。
脂質A誘導体には、大腸菌(OM−174)由来の脂質A誘導体が含まれる。OM−174は、例えば、Meraldiら、Vaccine(2003)21:2485−2491およびPajak,ら、Vaccine(2003)21:836−842に記載されている。
本発明でのアジュバントとしての使用に適切な免疫刺激性オリゴヌクレオチドには、CpGモチーフ(非メチル化シトシンの後にグアノシンを含み、リン酸結合によって結合した配列)を含むヌクレオチド配列が含まれる。回分配列またはポリ(dG)配列を含む細菌二本鎖RNAまたはオリゴヌクレオチドも免疫刺激性を示すことが示されている。

生体接着剤および粘膜接着剤を、本発明でアジュバントとして使用することもできる。適切な生体接着剤には、エステル化ヒアルロン酸ミクロスフィア(Singhら(2001)J.Cont.Rele.70:267−276)または粘膜接着剤(ポリアクリル酸の架橋誘導体、ポリビニルアルコール、ポリビニルピロリドン、多糖類、およびカルボキシメチルセルロースなど)が含まれる。キトサンおよびその誘導体を、本発明でアジュバントとして使用することもできる。WO99/27960を参照のこと。
微粒子を、本発明でアジュバントとして使用することもできる。微粒子(すなわち、直径約100nm〜約150μm、より好ましくは直径約200nm〜約30μm、最も好ましくは直径約500nm〜約10μmの粒子)を、生分解性且つ非毒性の材料(例えば、ポリ(α−ヒドロキシ酸)、ポリヒドロキシ酪酸、ポリオルソエステル、ポリ酸無水物、ポリカプロラクトンなど)(ポリ(ラクチドコグリコリド)が好ましい)から形成し、任意選択的に、負電荷の表面(例えば、SDS)または正電荷の表面(例えば、CTABなどの陽イオン性界面活性剤)を有するように処理する。
アジュバントとしての使用に適切なリポソーム処方物の例は、米国特許第6,090,406号、米国特許第5,916,588号、および欧州特許第0626169号に記載されている。
本発明での使用に適切なアジュバントには、ポリオキシエチレンエーテルおよびポリオキシエチレンエステルが含まれる。WO99/52549号。かかる処方物には、さらに、オクトキシノール(WO01/21207)と組み合わせたポリオキシエチレンソルビタンエステル界面活性剤および少なくとも1つのさらなる非イオン性界面活性剤(オクトキシノールなど)と組み合わせたポリオキシエチレンアルキルエーテルまたはエステル界面活性剤(WO01/21152号)が含まれる。

本発明でのアジュバントとしての使用に適切なムラミルペプチドの例には、N−アセチル−ムラミル−L−トレオニル−D−イソグルタミン(thr−MDP)、N−アセチル−ノルムラミル−l−アラニル−d−イソグルタミン(nor−MDP)、およびNアセチルムラミル−l−アラニル−d−イソグルタミニル−l−アラニン−2−(1’−2’−ジパルミトイル−sn−グリセロ−3−ヒドロキシホスホリルオキシ)−エチルアミンMTP−PE)が含まれる。
本発明でのアジュバントとしての使用に適切なイミダゾキノリン化合物の例には、イミキモドおよびそのアナログが含まれ、Stanley,Clin Exp Dermatol(2002)27(7):571−577;Jones,Curr Opin Investig Drugs(2003)4(2):214−218;米国特許第4,689,338号、同第5,389,640号、同第5,268,376号、同第4,929,624号、同第5,266,575号、同第5,352,784号、同第5,494,916号、同第5,482,936号、同第5,346,905号、同第5,395,937号、同第5,238,944号、および同第5,525,612号にさらに記載されている。
チオセミカルバゾン化合物、ならびに本発明でのアジュバントとしての使用に適切な全ての化合物の処方、製造、およびスクリーニング方法の例には、WO04/60308号に記載のものが含まれる。チオセミカルバゾンは、TNF−αなどのサイトカインの産生のためのヒト末梢血単核細胞の刺激で特に有効である。
トリプタントリン化合物、ならびに本発明でのアジュバントとしての使用に適切な全ての化合物の処方、製造、およびスクリーニング方法の例には、WO04/64759号に記載のものが含まれる。トリプタントリン化合物は、TNF−αなどのサイトカインの産生のためのヒト末梢血単核細胞の刺激で特に有効である。

本発明でアジュバントとしての使用に適切なヒト免疫調節薬には、サイトカイン(インターロイキン(例えば、IL−1、IL−2、IL−4、IL−5、IL−6、IL−7、IL−12など)、インターフェロン(例えば、インターフェロン−γ)、マクロファージコロニー刺激因子、および腫瘍壊死因子など)が含まれる。
本発明は、治療において使用するための、上記の組成物を提供する。本発明は、化膿連鎖球菌に対する免疫応答を誘導または増大させるための、上記の組成物を提供する。本発明は、上記組成物を使用して化膿連鎖球菌に対する免疫応答を誘導または増大させる方法を提供する。免疫応答は、好ましくは防御的であり、抗体および/または細胞−媒介性免疫(全身免疫および粘膜免疫が含まれる)が含まれ得る。免疫応答には、ブースター応答が含まれる。
治療上の処置の有効性を評価する1つの方法は、本発明の組成物の投与後にGAS感染をモニタリングすることを含む。予防的治療の有効性を評価する1つの方法は、組成物の投与後に本発明の組成物における変異体SLOタンパク質に対する免疫応答をモニタリングすることを含む。
キット
本発明はまた、本発明の1つまたは複数の容器を含むキットを提供する。組成物は、個別の抗原と同様に、液体形態であり得るか凍結乾燥させることができる。組成物に適切な容器には、例えば、ボトル、バイアル、シリンジ、および試験管が含まれる。容器を、種々の材料(ガラスまたはプラスチックが含まれる)から形成することができる。容器は、滅菌アクセスポートを有することができる(例えば、容器は、静脈内注射用溶液のバッグまたは皮下注射針によって突き刺すことができるストッパを有するバイアルであり得る)。
野生型および変異体SLOタンパク質のクローニング
野生型および変異体SLOタンパク質をコードする遺伝子を、表1に示されたSF370ゲノム由来のプライマーを用いるPCRによって増幅した。
Hisタグ付きタンパク質の精製
E.coliペレットを溶解緩衝液に懸濁し、室温で30〜40分間、混合した。溶解物を、30〜40000×gで、20〜25分間、遠心分離し、上清を、洗浄緩衝液Aで平衡化したカラム(1mlのNi活性化キレート化セファロースファストフロー樹脂を含むPoly−Prep)に添加した。添加された樹脂を、洗浄緩衝液Aで3回、洗浄緩衝液Bで3回、洗浄した。最終2mMのDTTを含むEppendorfチューブにおいて溶出緩衝液でタンパク質を溶出した。全溶出タンパク質を、Bradford試薬で定量し、その後、SDS−ポリアクリルアミドゲル電気泳動によって分析した(図8および9)。

溶解物調製
約80〜110gの細菌培養ペレットを、6錠のCOMPLETE(登録商標)プロテアーゼ阻害剤、10mlの0.2M EDTA、pH7.5(5mMの最終濃度)、10mlの100mg/mlリゾチーム溶液、8mlの10000Kユニット/mlデオキシリボヌクレアーゼI溶液、および1mlの50mM MgCl2溶液を追加した200〜280mlのB−PER(商標)試薬(Pierce)に懸濁した。細菌懸濁液を60分間、均一な懸濁液が得られるまで振盪することによって、細菌溶解を達成した。

溶血アッセイ
定量的溶血アッセイについてのプロトコール
毒素の段階希釈物を、PBS+0.5%BSAを用いて、U字形底の96ウェルプレートに調製した。1mlのヒツジ血液を、PBS中で3回、洗浄し(3000×gでの遠心分離で)、血球を5mlのPBSに懸濁した。等容積の懸濁液を、50μlの各毒素希釈物に加え、37℃で30分間、インキュベートした。水中のTriton(2%)を用いて、100%溶血を得、PBS+0.5%BSAをネガティブコントロールとして用いた。その後、プレートを1,000×gで5分間、遠心分離し、上清を、96ウェル平底プレートへ慎重に移した。吸光度を540nmで読み取った。
SLO P427Lをコードする遺伝子を、SF370 M1ゲノムからPCRを用いて増幅し、Hisタグ付きタンパク質のE.coli BL21DE3における発現を可能にするベクターpET21b+へクローニングした。類似した量の野生型および変異したストレプトリシンOタンパク質を発現するE.coliの可溶性抽出物(図5参照)を用いて、溶血アッセイを行って、その2つの抗原の細胞溶解性を比較した。アッセイの結果は図2に示されており、変異したタンパク質は、毒性が野生型の多くとも100分の1であることを実証している。
SLO P427L変異体を、Hisタグ付き組換えタンパク質についての精製標準手順に従って精製した(図3)。様々な濃度の精製された野生型(wt)および変異したタンパク質を用いて、溶血アッセイを繰り返し、細胞溶解活性の減少を確認した(図4)。
本発明者らは、Hisタグなしの野生型組換えSLO(rSLO)(BL21 DE3、Novagen No.71382−pET24)およびHisタグなしのP427L変異体rSLO(BL21 DE3、Novagen No.71382−pET24)で形質転換されたE.coli溶解物の溶血活性を比較した。挿入なしのpET24で形質転換されたE.coli BL21 DE3(Novagen、No.71382)をネガティブコントロールとして用いた。ポジティブコントロールは、水中Triton2%を含む低張溶液であった。ネガティブコントロールは、タンパク質希釈緩衝液(0.5%BSAを含むPBS、pH7.4)であった。
野生型SLOの溶血活性を、いくつかの異なるSLO変異体の溶血活性と比較した。結果は、図13および下記の表5に示されている。1溶血単位(HU)は、血球を2%Tritonで処理して得られた最大溶解の50%を得るのに必要とされる毒素量として定義される。
30℃での細胞増殖、および25℃での1mMのIPTGでの誘導、およびOD600nm約0.4〜0.6の後に得られた、E.coli溶解物または200mg/mlのコレステロールを含むE.coli溶解物のPBS−BSA0.5%における2〜5倍の段階希釈物を、それらの溶血活性についてアッセイした。誘導の3時間後、溶解緩衝液(B−PER溶液(PIERCE)、1mMのMgCl2、100Kユニット/mlのデオキシリボヌクレアーゼ(Sigma)、およびリゾチーム(Sigma))で30〜40分間、細菌を溶解することによって得られた等容積のタンパク質調製物で、PBS中2%ヒツジ赤血球溶液の50マイクロリットルを処理した。その後、不溶性画分を遠心分離し(15分間、21000×g、4℃)、上清(E.coli溶解物)を、5mMの最終濃度でDTTを含む新しいEppendorfチューブへ移した。
溶血の阻害
プロトコール
(アジュバントを含まない、またはアジュバントとしてミョウバンもしくはMF59(商標)を含む)野生型または変異体SLOタンパク質で免疫化したマウス由来の血清の2倍段階希釈物を、U字形底の96ウェルプレートに、PBS+0.5%BSAを用いて調製した。必要に応じて、PBSで、またはアジュバントのみで免疫化したマウスの血清をネガティブコントロールとして用いた。PBS+0.5%BSA中の50〜100ng/ml(3.5〜7HU)の毒素溶液の等容積を加え、プレートを撹拌(800rpm)しながら室温で20分間、インキュベートした。インキュベーション後、この溶液の50mlを新しい96ウェルプレートに移し、(PBS中で3回洗浄した)等容積のヒツジ赤血球懸濁液を加え、37℃で30分間、インキュベートした。その後、プレートを1,000×gで1分間、遠心分離し、上清を96ウェル平底プレートへ慎重に移し、吸光度を540nmで読み取った。下記の結果において、阻害力価は、Triton誘導溶血を50%低下させた血清希釈度として表される。
抗野生型SLO抗血清によるSLO溶血の阻害は、図14、図15、図16、および表7〜9に示されている。抗SLO血清力価は、1/7,000〜1/14,000に含まれる(相加平均、1/12,167±2,714)。ネガティブコントロール血清(フロイントアジュバント)力価は、1/375〜1/4,000に含まれる(相加平均、1/1,854±1,384)。
野生型SLO、化学的に解毒された野生型SLO、およびSLO変異体(P427L;P427L+W535F)の溶血活性の滴定は、図17〜19および表10に示されている。
変異体SLOタンパク質に対する抗血清によるSLO溶血の阻害は、図20〜22および表11〜13に示されている。50ng/ml(3.5HU)の毒素を用いて、SLO変異体W535−P427LについてのSLO溶血活性の50%低下を得るのに必要とされる血清希釈度は、ミョウバンアジュバントを用いて1/17,860であり、MF59(商標)アジュバントを用いて1/7991である。ネガティブコントロール(アジュバントのみ)力価は、1/1,000(ミョウバン)および1/125(MF59(商標))である。
インビボ防御実験
精製されたSLO P427Lタンパク質をフロイントアジュバントと共に、40匹のマウスへ腹腔内に投与した。その後、3348 M1 GAS菌株でマウスを鼻腔内で攻撃誘発した。表14は、3つの別々の実験で得られたデータを報告し、100%防御が全ての実験において一貫して達成されたことを示している。
30匹のマウスを、SLO変異体P427L−W535Fで、アジュバントとしてミョウバンかまたはMF59のいずれかと共に、腹腔内で免疫化し、GAS M1菌株で鼻腔内攻撃誘発した。結果は図26に示されている。SLO変異体P427L−W535Fおよびミョウバンで免疫化したマウスの77パーセントが、(アジュバントのみで免疫化した)ネガティブコントロールマウスの3%と比較して、GAS M1菌株での鼻腔内攻撃誘発に対して防御された。SLO変異体P427L−W535FおよびMF59で免疫化したマウスの90パーセントが、(アジュバントのみで免疫化した)ネガティブコントロールマウスの10%と比較して、GAS M1菌株での鼻腔内攻撃誘発に対して防御された。これらの防御レベルは、野生型SLOでマウスを免疫化することによって得られたものに匹敵する。
プロトコール
SLOの静脈内注射。野生型SLOかまたは変異体SLOのいずれかのPBS中の溶液を、PBS+2mMのDTTの溶液中に希釈し、その後、マウスの尾静脈へ100μlを注射する。マウスを2〜3日間、観察する。野生型SLOの注射は、典型的には、数分以内で死をもたらす。
SLO P427L−W535Fでの免疫化は、野生型SLOの静脈内注射に対してマウスを防御する
5週齡のマウスを、野生型SLOかまたはSLO変異体P427L−W535Fかのいずれかで、アジュバントとしてミョウバンを用いて(2mg/ml水酸化アルミニウム中20μgのタンパク質)、腹腔内に3回(0日目、21日目、および35日目)免疫化した。アジュバントのみで免疫化したマウスをネガティブコントロールとして用いた。55日目、マウスに、PBS、2mMのDTT中の野生型SLOの様々な濃度の溶液を静脈内注射し、少なくとも72時間、モニターした。結果は表21に示されている。
Claims (23)
- アミノ酸P427、W535、C530、A248、およびD482からなる群より選択される1つまたは複数のアミノ酸位置にアミノ酸変化を含む精製された変異体ストレプトリシンO(SLO)タンパク質であって、該アミノ酸位置が配列番号1に従って番号付けられ、該変異体SLOタンパク質の溶血活性が野生型SLOに対して少なくとも50%低下している、精製された変異体SLOタンパク質。
- 配列番号20、配列番号21、配列番号22、配列番号23、配列番号24、配列番号25、配列番号26、および配列番号27からなる群より選択されるアミノ酸配列を含む、請求項1に記載の精製された変異体SLOタンパク質。
- 担体タンパク質にカップリングされている、請求項1に記載の精製された変異体SLOタンパク質。
- 前記担体タンパク質が、細菌毒素、細菌トキソイド、N.meningitidis外膜タンパク質、熱ショックタンパク質、百日咳タンパク質、H.influenzaeタンパク質D、サイトカイン、リンホカイン、ホルモン、成長因子、C.difficile毒素A、C.difficile毒素B、および鉄取り込みタンパク質からなる群より選択される、請求項3に記載の精製された変異体SLOタンパク質。
- 請求項1に記載の変異体SLOタンパク質をコードする核酸分子。
- 前記変異体SLOタンパク質が配列番号20、配列番号21、配列番号22、配列番号23、配列番号24、配列番号25、配列番号26、および配列番号27からなる群より選択されるアミノ酸配列を含む、請求項5に記載の核酸分子。
- 配列番号29、配列番号30、配列番号31、配列番号32、配列番号33、配列番号34、配列番号57、配列番号58、および配列番号59に示されたコード配列の群から選択されるヌクレオチド配列を含む、請求項5に記載の核酸分子。
- (a)以下:
(i)アミノ酸P427、W535、C530、A248、およびD482からなる群より選択される1つまたは複数のアミノ酸位置にアミノ酸変化を含む精製された変異体ストレプトリシンO(SLO)タンパク質であって、該アミノ酸位置が配列番号1に従って番号付けられ、該変異体SLOタンパク質の溶血活性が野生型SLOに対して少なくとも50%低下している、精製された変異体ストレプトリシンO(SLO)タンパク質、ならびに
(ii)該精製された変異体ストレプトリシンO(SLO)タンパク質をコードする核酸分子
から選択される活性剤、ならびに
(b)薬学的に許容可能な担体
を含むワクチン組成物。 - (iii)GAS抗原、および
(iv)GAS抗原をコードする核酸分子
からなる群より選択される1つまたは複数の活性剤をさらに含む、請求項8に記載のワクチン組成物。 - 小児用ワクチンで有用な活性剤をさらに含む、請求項8に記載のワクチン組成物。
- 前記活性剤が
(a)N.meningitidis、S.pneumoniae、Bordetella pertussis、Moraxella catarrhalis、Clostridium tetani、Chorinebacterim diphteriae、RSウイルス、ポリオウイルス、麻疹ウイルス、ムンプスウイルス、風疹ウイルス、およびロタウイルスのポリペプチド抗原からなる群より選択される病原体のポリペプチド抗原、ならびに
(b)該ポリペプチド抗原をコードする核酸分子
からなる群より選択される、請求項10に記載のワクチン組成物。 - 高齢者または免疫無防備状態の個体のためのワクチンで有用な第2の活性剤をさらに含む、請求項8に記載のワクチン組成物。
- 前記第2の活性剤が
(a)Enterococcus faecalis、Staphylococcus aureaus、Staphylococcus epidermis、Pseudomonas aeruginosa、Legionella pneumophila、Listeria monocytogenes、インフルエンザウイルス、およびパラインフルエンザウイルスのポリペプチド抗原からなる群より選択される病原体のポリペプチド抗原、ならびに
(b)該ポリペプチド抗原をコードする核酸分子
からなる群より選択される、請求項12に記載のワクチン組成物。 - Streptococcus pyogenesによる感染を治療または予防する方法であって、
(a)以下:
(i)アミノ酸P427、W535、C530、A248、およびD482からなる群より選択される1つまたは複数のアミノ酸位置にアミノ酸変化を含む精製された変異体ストレプトリシンO(SLO)タンパク質であって、該アミノ酸位置が配列番号1に従って番号付けられ、該変異体SLOタンパク質の溶血活性が野生型SLOに対して少なくとも50%低下している、精製された変異体ストレプトリシンO(SLO)タンパク質、および
(ii)該精製された変異体ストレプトリシンO(SLO)タンパク質をコードする核酸分子
から選択される活性剤、ならびに
(b)薬学的に許容可能な担体
を含むワクチン組成物を、それを必要としている個体に有効量投与する工程を含む、方法。 - 前記ワクチン組成物が、
(iii)GAS抗原、および
(iv)GAS抗原をコードする核酸分子
からなる群より選択される1つまたは複数の活性剤をさらに含む、請求項14に記載の方法。 - (a)請求項8から13のいずれか一項に記載のワクチン組成物を含む容器、および
(b)該組成物を用いてStreptococcus pyogenesによる感染を治療または予防するための使用説明書
を含むキット。 - Streptococcus pyogenesによる感染の予防または治療のためのワクチンを作製する方法であって、
(a)以下:
(i)アミノ酸P427、W535、C530、A248、およびD482からなる群より選択される1つまたは複数のアミノ酸位置にアミノ酸変化を含む精製された変異体ストレプトリシンO(SLO)タンパク質であって、該アミノ酸位置が配列番号1に従って番号付けられ、該変異体SLOタンパク質の溶血活性が野生型SLOに対して少なくとも50%低下している、精製された変異体ストレプトリシンO(SLO)タンパク質、ならびに
(ii)該精製された変異体ストレプトリシンO(SLO)タンパク質をコードする核酸分子
から選択される活性剤と、
(b)薬学的に許容可能な担体と、
を組み合わせる工程を含む、方法。 - 前記活性剤が前記精製された変異体SLOタンパク質であり、該変異体SLOタンパク質を:
(a)該変異体SLOタンパク質をコードする発現構築物を含む宿主細胞を培養する工程、および
(b)該変異体SLOタンパク質を回収する工程
を含む方法によって作製する、請求項17に記載の方法。 - (iii)GAS抗原、および
(iv)GAS抗原をコードする核酸分子
からなる群より選択される1つまたは複数の活性剤を薬学的に許容可能な担体と組み合わせる工程をさらに含む、請求項17に記載の方法。 - Streptococcus pyogenesによる感染を治療または予防するための薬物の製造における活性剤の使用であって、該活性剤が、
(i)アミノ酸P427、W535、C530、A248、およびD482からなる群より選択される1つまたは複数のアミノ酸位置にアミノ酸変化を含む精製された変異体ストレプトリシンO(SLO)タンパク質であって、該アミノ酸位置が配列番号1に従って番号付けられ、該変異体SLOタンパク質の溶血活性が野生型SLOに対して少なくとも50%低下している、精製された変異体ストレプトリシンO(SLO)タンパク質、ならびに
(ii)該精製された変異体ストレプトリシンO(SLO)タンパク質をコードする核酸分子
からなる群より選択される、使用。 - ワクチンとして使用する、請求項1から4のいずれか一項に記載の精製された変異体SLOタンパク質、または請求項5から7のいずれか一項に記載の核酸分子。
- 治療で使用する、請求項1から4のいずれか一項に記載の精製された変異体SLOタンパク質、請求項5から7のいずれか一項に記載の核酸分子、または請求項8から13のいずれか一項に記載のワクチン組成物。
- Streptococcus pyogenesによる感染を治療または予防するための、請求項1から4のいずれか一項に記載の精製された変異体SLOタンパク質、請求項5から7のいずれか一項に記載の核酸分子、または請求項8から13のいずれか一項に記載のワクチン組成物。
