Abstract
Description
複合コラーゲン及びリン酸カルシウムゲル分散物は、6gのコラーゲンを量り取って調製された(5体積%コラーゲンゲル)。10mM塩酸は5体積%ゲルを調製するために容量計量された(246ml)。12gの滅菌されたリン酸二カルシウム[CaHPO4]パウダー(66.7重量%リン酸カルシウム)は加えられて、均一濃度に撹拌された。混合物は、適度な粘度(約1000Pから約1500P)をもった均一なコラーゲンゲル分散物が得られるまで、例えば反復せん断材料輸送(shear material transport)によって混合された。
上記組成物の粒子が入った容器を開ける前に、修復する骨空洞の体積が決定された。骨空洞に基づいて、ヒトでなない動物の血液の適切な量が得られ、血液又は骨髄穿刺液:骨空洞容積が0.75:1の割合で使用された。続いて説明されるように適切な量の液体が加えられることで、所望の粘度の生成物(例えばペースト)が得られた。一実施形態として、1ccの乾燥粒子量につき、粘性のあるパテを得るために0.75mlの全血が加えられるか、又はペーストを得るために0.85mlの全血が加えられた。他の実施形態において、1ccの乾燥粒子量につき、粘性のあるパテを得るために0.75mlの骨髄穿刺液が加えられるか、又はペーストを得るためには0.85mlの骨髄穿刺液が加えられた。
Claims (53)
- 骨成長組成物であって、
粒子状の線維状コラーゲン成分、及び
リン酸カルシウム成分を含む組成物。 - コラーゲン成分は不溶性コラーゲンを含む請求項1の組成物。
- コラーゲン成分は架橋コラーゲンを含む請求項2の組成物。
- コラーゲンは熱的脱水架橋コラーゲンを含む請求項3の組成物。
- コラーゲン成分は多孔質粒子を含む請求項2の組成物。
- 粒子内の全開孔容積は約50%から約90%の範囲である請求項5の組成物。
- 多孔質粒子の孔の直径は約125から約300μmの範囲である請求項5の組成物。
- 粒子のサイズは約100から約840μmの範囲である請求項5の組成物。
- コラーゲン成分は20乾燥重量%を超える請求項1の組成物。
- コラーゲン成分は架橋されていないコラーゲンを含む請求項1の組成物。
- コラーゲン成分はウシ皮膚コラーゲンを含む請求項1の組成物。
- コラーゲン成分は1型コラーゲンを含む請求項1の組成物。
- リン酸カルシウム成分は酸性リン酸カルシウムを含む請求項1の組成物。
- 酸性リン酸カルシウムは、リン酸一カルシウム[Ca(H2PO4)2]、リン酸水素カルシウム[CaHPO4]、ピロリン酸カルシウム[2CaO・P2O5]及びそれらの組み合わせから成るグループより選択される請求項13の組成物。
- 酸性リン酸カルシウムはリン酸水素カルシウム[CaHPO4]である請求項14の組成物。
- リン酸カルシウム成分は30乾燥重量%を超える請求項1の組成物。
- 骨誘導成分を含む請求項1の組成物。
- 骨誘導成分は、精製された骨成長因子、組換え骨成長因子、骨髄成分、血液成分、脱塩骨及び自家骨から成るグループより選択される物質を含む請求項17の組成物。
- 骨誘導成分は骨髄穿刺液を含む請求項17の組成物。
- 液体を含む請求項1の組成物。
- 液体は25体積%を超える請求項20の組成物。
- 組成物は約pH5から約pH7.4の間のpHを有する請求項20の組成物。
- 骨成長組成物を調製するための方法であって、
線維状コラーゲン成分をリン酸カルシウム成分と混合することで鉱化コラーゲン成分を調製すること、及び
鉱化コラーゲン成分の粒子を調製することを含む方法。 - 粒子の調製は、すりつぶすこと、製粉すること、切断すること又は成形することを含む請求項23の方法。
- 粒子の調製は製粉することを含む請求項23の方法。
- 鉱化コラーゲン成分は、粒子を調製する前のゲルの形態である請求項23の方法。
- 鉱化コラーゲン成分は、粒子を調製する前のスポンジの形態である請求項23の方法。
- スポンジは架橋されている請求項27の方法。
- 粒子をキットに加えることを含む請求項23の方法。

骨誘導成分を含む骨成長組成物。
