A、およびTFASTA、Wisconsin Genetics Software Package Release 7.0中(Genetics Computer Group、575 Science Dr、Madison、Wis)、GENEWORKS(商標)、またはMACVECTOR(登録商標)ソフトウェアパッケージ)のコンピュータによる実施によって、あるいは調査によって行うことができ、様々な方法によって生じた、最良のアラインメント(すなわち、比較ウインドウで最高の割合の相同性をもたらす)を選択する。
基本的な抗体の構造単位は、テトラマーを含むことが知られている。それぞれのテトラマーは、ポリペプチド鎖の2つの同一の対から構成され、それぞれの対は、1本の「軽鎖」(約25kDa)および1本の「重鎖」(約50〜70kDa)を有する。それぞれの鎖のアミノ末端部分は、抗原認識を主に担う約100〜110以上のアミノ酸の可変領域を含む。それぞれの鎖のカルボキシ末端部分は、エフェクター機能を主に担う定常領域を規定する。ヒト軽鎖は、kappaおよびλ軽鎖として分類される。重鎖はmu、デルタ、ガンマ、アルファ、またはイプシロンとして分類され、それぞれIgM、IgD、IgA、およびIgEとして、抗体のイソ型を定義する。軽鎖および重鎖内では、可変領域と定常領域が、約12以上のアミノ酸の「J」領域によって結びつき、重鎖は約10以上のアミノ酸の「D」領域も含む。Fundamental Immunology Ch.7(Paul,W.編、第2編、Raven Press、N.Y.(1989))を一般的に参照のこと。それぞれの軽鎖/重鎖対の可変領域は、抗体結合部位を形成する。
ヒト抗体は、ネズミまたはラット可変領域および/または定常領域を有する抗体に関するいくつかの問題を回避する。このようなネズミまたはラット由来のタンパク質が存在することによって、抗体の迅速な除去がもたらされる可能性があり、あるいは患者による抗体に対する免疫応答の発生がもたらされる可能性がある。ネズミまたはラット由来の抗体の使用を回避するために、げっ歯類が完全ヒト抗体を生成するように、ヒト抗体の機能をげっ歯類に導入することによって、完全ヒト抗体を作製することができる。
本明細書で論じるように、TNFa抗体の機能は、その操作形式の少なくとも一部分に対して重要であるようである。機能とは、例えばTNFaと共に働くTNFa抗体の活性を意味する。したがって、いくつかの観点で、TNFaに対する治療候補としての抗体の作製に関して、抗体が補体を固定し、CDCに関与することができることが望ましい可能性がある。以下のネズミIgM、ネズミIgG2a、ネズミIgG2b、ネズミIgG3、ヒトIgM、ヒトIgG1、およびヒトIgG3を非制限的に含めた、同じことができるいくつかのイソ型の抗体が存在する。作製される抗体は、このようなイソ型を最初に有している必要はなく、作製される抗体は、任意のイソ型を有することができ、当分野でよく知られている従来の技法を使用して、抗体のイソ型を後に変えることができることは理解されよう。このような技法には、特に、直接的な組換え技法の使用(例えば、米国特許第4,816,397号を参照)、細胞-細胞融合技法(例えば、米国特許第5,916,771号および米国特許第6,207,418号を参照)がある。
本発明に従い、TNFaに対する本明細書で生成し特徴付けした抗体の活性に基づいて、抗体要素を超える他の治療的物理療法の設計が容易になる。このような物理療法には、非制限的に、二重特異性抗体などの最新の抗体療法、免疫毒素、および放射標識療法、ペプチド療法剤、遺伝子療法剤、特に細胞内発現抗体、アンチセンス療法剤、および小分子の作製がある。
本明細書に記載する抗体は、以下に記載するようにXENOMOUSE(登録商標)技術を使用することによって調製した。このようなマウスは、したがって、ヒト免疫グロブリン分子および抗体を産生することができ、ネズミ免疫グロブリン分子および抗体の産生において欠陥がある。同じことを達成するために使用する技法は、本明細書の背景技術の項中に開示する特許、出願、および参照文献中に開示する。しかしながら、特に、トランスジェニックマウスおよびそれに由来する抗体の産生の好ましい実施形態は、1996年12月3日に出願された米国特許出願第08/759,620号、および1998年6月11日に公開された国際特許出願WO98/24893号、および2000年12月21日に公開されたWO00/76310中に開示されている。Mendez他、Nature Genetics 15:146〜156(1997)も参照のこと。
本明細書に記載する生物学的に活性がある抗TNFa抗体を、滅菌済み薬剤調製物または配合物中で使用して、血清TNFaのレベルを低下させ、これによって、例えば血清TNFaが異常に増大している病的状態を有効に治療することができる。抗TNFa抗体は、標的治療範囲内にTNFaを強く抑制する、適度な親和性を有することが好ましく、低頻度の投与を可能にする充分な作用時間を有することが好ましい。長い作用時間によって、皮下または筋肉内注射などの他の非経口経路より、低頻度でさらに好都合な投与スケジュールが可能になるであろう。
抗原の調製
XENOMOUSE(登録商標)動物の免疫処置用の、TNFa-KLH抗原の調製
組換えヒトTNFaを、R&D Systems(Minneapolis、MNカタログ番号210-TA/CF)から得た。XENOMOUSE(登録商標)動物の免疫処置用に使用した、TNFa-KLH抗原は、以下のように調製した:ヒトTNF-a(200μg)(R&D)を、50μgのキーホールリンペットヘモシアニン(KLH;Pierce、Rockford、IL)と混合させて、蒸留水を使用して165μlの最終体積にした。250μlの結合バッファー(0.1MのMES、0.9MのNaCl、pH4.7)を加え、TNFaとKLHを、1-エチル-3- [3-ジメチルアミノプロピル] カルボジイミド塩酸塩(EDC、Pierce、Rockford、IL)の10mg/mLのストック溶液25μlを加えることによって架橋させた。結合体を室温で2時間インキュベートし、未反応EDCを、1kDaのフィルタ(Centrifugalフィルタ;Millipore、Bedford、MA)を介して、PBS、pH7.4を使用して遠心分離によって除去した。
ヒトTNFaを、XENOMOUSE(登録商標)動物の免疫処置用に、共通のT細胞エピトープ(TCE)(J.Immunol 1992 148(5):1499)とフレームが一致する融合タンパク質として、組換えによって作製した。
抗体の作製
免疫処置
ヒトTNFaに対するヒトモノクローナル抗体を、XENOMOUSE(登録商標)マウス(XENOMOUSE(登録商標)XMG2L3または3B-3L3Abgenix,Inc.Fremont、CA)を連続的に免疫処置することによって開発した。
抗hTNFa抗体の力価を、ELISAによって測定した。hTNFaを、4℃において一晩、Costar Labcoat Universal Binding Polystyrene96ウェルプレート(Corning、Acton、MA)上にコーティングした。非結合TNFaを含む溶液を除去し、プレートをUV光(365nm)で4分間(4000マイクロジュール)処理した。プレートはdH2Oで5回洗浄した。TNFaで免疫処置した動物、または非投薬のXENOMOUSE(登録商標)動物由来のXENOMOUSE(登録商標)の血清を、まったく同様に1:2に希釈した2%ミルク/PBS中において、1:100の最初の希釈液から滴定した。最後のウェルは空の状態にした。プレートはdH2Oで5回洗浄した。抗体と結合した、ヤギ抗ヒトIgGFc特異的ホースラディッシュペルオキシダーゼ(HRP、Pierce、Rockford、IL)を、室温で1時間、1μg/mLの最終濃度で加えた。プレートはdH2Oで5回洗浄した。プレートは、30分間TMB色原体基質(Gaithersburg、MD)を加えることによって顕色させ、ELISAは、1Mのリン酸を加えることによって停止させた。個々のXENOMOUSE(登録商標)動物の特異的力価を、450nmでの光学濃度から測定し、表2〜8に示す。力価は相互的な血清の希釈を表し、したがって、その数が大きくなるほど、hTNFaに対する体液性免疫応答は高くなる。
抗ヒトTNFα抗体の作製
ハイブリドーマによる抗hTNFα抗体の作製
リンパ細胞の回収、B細胞単離、ハイブリドーマの融合および作製
免疫処置したマウスを頸部脱臼によって殺傷し、リンパ節をそれぞれのコホートから採取し集めた。リンパ細胞をDMEM中で粉砕することによって解離させて、組織から細胞を切り離し、細胞をDMEM中に懸濁させた。細胞を計数し、リンパ球1億個当たり0.9mLのDMEMを細胞ペレットに加えて、完全ではないが軽く細胞を再懸濁させた。細胞1億個当たり100μLのCD90+磁気ビーズを使用して、15分間4℃で磁気ビーズと細胞をインキュベートすることによって、細胞を標識した。108個までの陽性細胞(または2×109個までの合計細胞)を含む、磁気標識した細胞の懸濁液をLS+カラムに充填し、DMEMでカラムを洗浄した。すべての溶出液を、CD90-陰性分画(これらの細胞の大部分はB細胞である)として回収した。
培養の14日後、ハイブリドーマ上清を、TNFa特異的モノクローナル抗体に関してスクリーニングした。ELISAプレート(Fisher、カタログ番号12-565-136)を、コーティングバッファー(0.1Mの炭酸バッファー、pH9.6、NaHCO3、8.4g/L)に溶かしたTNFa(2μg/mL)50μL/ウェルでコーティングし、次いで一晩4℃でインキュベートした。インキュベーションの後、プレートは洗浄バッファー(PBSに溶かした0.05%Tween20)で3回洗浄した。200μL/ウェルのブロッキングバッファー(1×PBSに溶かした0.5%BSA、0.1%Tween20、0.01%Thimerosal)を加え、室温で1時間プレートをインキュベートした。インキュベーションの後、プレートは洗浄バッファーで3回洗浄した。50μL/ウェルのハイブリドーマ上清、ならびに陽性および陰性対照を加え、室温で2時間プレートをインキュベートした。
Lnminexプラットホームは、多数のアッセイを一度に行うのを可能にする、蛍光ビーズ系技術である。Lnminex読み取り装置は、異なるコード付けされたミクロスフェアの陽性のシグナル事象を確認することができる。これによって、それぞれのビーズを別々にコーティングすることが可能であり、したがって差別的にコーティングされたミクロスフェアを一緒に混合することが可能であり、したがって1ステップのアッセイで、それぞれの異なるミクロスフェアとの抗体の結合が可能である。抗体をイソ型で分類するために、それぞれのビーズが、特定の重鎖または軽鎖イソ型と特異的に結合することができるような方法で、ミクロスフェアをコーティングした。次いでミクロスフェアを一緒に混合し、それぞれの抗体のハイブリドーマ上清を加えた。20分のインキュベーションの後、ミクロスフェアを洗浄し、結合した抗体は、蛍光標識した二次抗体を使用して検出した。Lnminex読み取り装置を使用して、次いでミクロスフェアを読み取った。表10は、異なる融合体群に関して見られた、それぞれのイソ型の数を示す。
47の抗TNFaハイブリドーマ抗体を、ヒトWM266.4細胞に対するTNFa誘導型アポトーシスの生物学的影響を中和する、それらの能力に関してアッセイした。Swell-Gel proteinA(Pierce)での精製によって、それぞれのハイブリドーマ上清からIgGを最初に増大させ、次いで溶出させ、中和し、正確に測定した。20,000個のWM266.6細胞を、96ウェルプレート中、完全培地(RPMI1640/10%FBS/Gln/P/S)中に平板培養し、37℃/10%CO2で一晩インキュベートした。培地を除去し、50μLの試験抗体およびTNFa(室温で30分プレインキュベートしたもの)を、無血清培地(RPMI1640/Gln/P/S)に加えた。50μLのシクロヘキサミドのプレートを、以下の最終的なアッセイ条件下で、前と同様に一晩インキュベートした。V=100μl、シクロヘキサミド=6μg/mL、トリマーとしてTNFa=600μg/mL=11.4pM、試験抗体濃度は、記載したように変わる。100μLのカスパーゼバッファーおよび0.3μLのカスパーゼ基質(APO-ONE、Promega)を、それぞれのウェルに加えた。
47の抗TNFaハイブリドーマ抗体の上清を、ヒトMCF-7細胞に対するTNFa誘導型アポトーシスの生物学的影響を中和する、それらの能力に関してさらにアッセイした。96ウェルプレートに、5000細胞/ウェル、200μl/ウェル、およびフェノールレッドを含まないDMEM+10%FCSで接種した。37℃+5%CO2で、細胞を一晩インキュベートした。それぞれのプレート上で、(実施例2に記載したのと同様に捕捉ELISAによって正確に測定し、標準曲線の対照抗体と比較した)ハイブリドーマ抗体の滴定値を、100pg/mL(トリマーとして1.9pM)TNFaの一定濃度において、三連で、アポトーシス培地(2.5%FCS、5μg/mLのCHXをフェノールレッドを含まないDMEMに溶かしたもの)中において、10μg/mL〜最終濃度0.005ng/mL(1:5に滴定)のウサギ014対照抗体と比べてアッセイした。TNFaのみを含む6つのウェルプレート、およびアポトーシス培地のみを含む6つのウェルも、インキュベートした。TNFa+/-中和抗体は、37℃+5%CO2で1時間プレインキュベートした。200μLの抗体を次いで細胞に移し、37℃+5%CO2で一晩インキュベートした。
ハイブリドーマ4.17および3.2から、mRNAを抽出した。逆転写酵素PCRを行って、cDNAを作製した。可変部位重鎖および軽鎖をコードするcDNAを、PCRを使用して特異的に増幅させた。可変部位重鎖領域は、IgG1発現ベクターにクローニングした。このベクターは、ヒトIgG1の定常ドメインを、pcDNA3.1+/Hygro(Invitrogen、Burlington、ON)の多重クローニング部位にクローニングすることによって作製した。可変部位軽鎖領域は、IgK発現ベクターまたはIgλにクローニングした。これらのベクターは、ヒトIgKまたはIgλの定常ドメインを、pcDNA3.1+/Neo(Invitrogen、Burlington、ON)の多重クローニング部位にクローニングすることによって作製した。次いで重鎖および軽鎖の発現ベクターを、70%の集合状態の60mm皿中に同時にリポフェクションし、ヒト胚腎臓293細胞およびトランスフェクトした細胞は、24〜72時間、元のプラズマ細胞と同一の特異性を有する組換え抗体を分泌させた。上清(3mL)をHEK293細胞から採取し、完全抗体の分泌をサンドウィッチELISAによって実証して、ヒトIgGを特異的に検出した。ELISAを使用して組換え抗体とTNFaの結合により、特異性を評価した。
B細胞の培養および選択
動物由来のB細胞を、採取し培養した。TNFa特異的抗体を分泌する細胞は、Babcook他、Proc.Natl.Acad.Sci.USA、93:7843〜7848(1996)中に記載されたのと同様に単離した。ELISAを使用して、主要なTNFa特異的ウェルを同定した。XENOMOUSE(登録商標)動物由来の約1800万個のB細胞を、500または150細胞/ウェルで48096ウェルプレート中において培養し、TNFaに関してスクリーニングして、抗原特異的なウェルを同定した。3,825ウェルが基底を大幅に超えるODを示し、その代表的なサンプルは表11に示す。ウサギB細胞も、抗hTNFa抗体を分泌するそれらの能力に関してスクリーニングして、陽性のものは以下に記載したようにさらにアッセイした。
ELISA法を使用して、抗原特異的な抗体の濃度の上清を正規化した。平行して滴定した知られている濃度の抗標的(TNFa)抗体を使用して、標準曲線を作製することができ、上清中の抗原特異的な抗体の量は、標準およびその測定した濃度と比較することができる、以下の表12を参照のこと。
限定的な抗原分析は、すべての他の抗原特異的な抗体に対して、B細胞培養物上清中で調製された抗原特異的な抗体を、親和性によってランク付けする方法である。非常にわずかな抗原コーティングの存在下では、最高親和性の抗体のみが、平衡状態で任意の検出可能なレベルに、結合することができるはずである(例えば、2003年6月12日に公開された、「IDENTIFICATION OF HIGH AFFINITY MOLECULES BY LIMITED DILUTION SCREENING」という表題のPCT公開WO/03048730A2を参照のこと)。
10ng/mL〜1000ng/mLの範囲の抗原特異的な抗体の濃度を有する、B細胞培養物上清を調製した。限定的な抗原分析から生じた結果は、4.17ハイブリドーマ由来抗体の滴定値と比較した。このアッセイでは、抗体の多くは検出可能な結合を与えることができなかったが、しかしながら、O.D.により測定して、すべての濃度で他の培養物上清および組換え抗体より明らかに優れていた、401A7および433F4を含めたいくつかのウェルが存在した(表13)。残りのクローンは、特異的な抗体の濃度を測定する充分な抗原データ(詳細に関しては前を参照)と、限定的な抗原の産生量を組み合わせることによって、さらに分析した。このようにして、B細胞培養物上清中の抗体と対照抗体のそれを、図2に示す濃度範囲で比較することができた。図2は、B細胞培養物上清中の抗体間の抗TNFa限定抗原結合と、対照抗体(4.17IgG2)の結合を、ある濃度範囲で比較する、点グラフである。三角形はB細胞培養物上清中のクローンを表し、ブロックは棒線の抗体(4.17IgG2)を表す。棒線の抗体曲線より上の位置の、B細胞培養物上清中のクローンは、潜在的に高い親和性を有するものとしてランク付けする。
フットパッド免疫処置したマウス由来のB細胞培養物の、ウェルの上清から同定した、全1455の抗TNFa抗体を、ヒトMCF-7細胞に対するTNFa誘導型アポトーシスの生物学的影響を中和する、それらの能力に関してさらにアッセイした。さらに、BIP免疫処置動物から同定した全2,370抗hTNFaの限定的な抗原分析の後、最高の動態ランキングを有する145の抗体を、TNFa活性の中和に関してさらに分析した。96ウェルプレートに、5000細胞MCF-7/ウェル、200μl/ウェル、およびフェノールレッドを含まないDMEM+10%FCSで接種した。37℃+5%CO2で、プレートを一晩インキュベートした。それぞれのプレート上で、B細胞培養物抗体上清を、100pg/mL(トリマーとして1.9pM)TNFaの一定濃度において、アポトーシス培地(2.5%FCS、5μg/mLのCHXをフェノールレッドを含まないDMEMに溶かしたもの)中において、最も強力な中和抗TNFaハイブリドーマ抗体、4.17および3.2および/またはウサギ014対照と比べてアッセイした。アポトーシス培地中にTNFaを含むまったく同様のウェル、およびアポトーシス培地のみを含むウェルを、対照としてインキュベートした。TNFa+/-試験サンプルは、37℃+/5%CO2で1時間プレインキュベートした。200μLのTNFa+/-を細胞に移し、37℃+/5%CO2で一晩インキュベートした。
ポリクローナルB細胞培養物上清中の抗原特異的な抗体の補外濃度を使用して、MCF-7細胞における、TNFa誘導型アポトーシスの中和の外見上の有効度を計算した。標準抗標的試薬と共にアッセイを行うことによって、この場合のハイブリドーマ由来の抗体3.2IgG2では、有効度線を設定して、標準より高い可能性がある有効度を有する、抗体を探すことが可能であった。
ウサギ抗TNFa中和抗体を、B細胞培養物上清由来の抗体が、そのp55レセプターとのTNFa結合を阻害することができたかどうか、調べることによって発見した。以下の手順に従った。96ウェルマイクロタイタープレートは、TNFaで一晩コーティングした。翌日、プレートを洗浄し、抗TNFa抗体+/-で1時間インキュベートした。次いでビオチン-p55をプレートに1時間加え、水で洗浄し、結合したp55は、ストレプトアビジン-HRPを使用して検出した。次いでプレートを洗浄し、前に記載した他のELISAで行ったのと同様に顕色させた。p55の結合を阻害した抗体は中和抗体と名付けた、表14を参照のこと。
いくつかの特殊な試薬を使用して、このアッセイを行った。これらの試薬は、以下のように調製した。
SRBCを、25%ストックとしてRPMI培地中に保存した。250μLのSRBC含有細胞ペレットは、1.0mLのSRBCを新たなエッペンドルフチューブに等分試料分けすることによって得た。マイクロフュージ中において8000rpm(6800rcf)でパルススピンによって、SRBCをペレット状にし、上清を除去し、ペレットを1.0mLのPBS、pH8.6に再懸濁させ、遠心分離を繰り返した。洗浄サイクルは2回繰り返し、次いでSRBCペレットを15-mLのファルコンチューブに移し、PBS、pH8.6を用いて5mLにした。別の50mLのファルコンチューブ中において、2.5mgのSulfo-NHSビオチンを、45mLのPBS、pH8.6に加えた。ひとたびビオチンが完全に溶けた後、5mLのSRBCを加え、室温で1時間チューブを回転させた。SRBCは3000rpmで5分間遠心分離にかけ、上清は除去した。ビオチン化したSRBCはエッペンドルフチューブに移し、前と同様に、ただしPBS、pH7.4で3回洗浄し、次いで15mLのファルコンチューブ(5%B-SRBCストック)中において、免疫細胞培地(RPMI1640)を用いて5mLにした。必要となるまで、ストックは4℃で保存した。
1mLの5%B-SRBCストックを、新たなエッペンドルフチューブに移した。B-SRBC細胞は前と同様に3回洗浄し、1.0mLのPBS、pH7.4に再懸濁させて、5%(v/v)の最終濃度を得た。10μLの10mg/mLストレプトアビジン(CalBiochem、San Diego、CA)ストック溶液を加え、チューブを混合し、室温で20分間回転させた。洗浄ステップを繰り返し、SA-SRBCを1mLのPBS、pH7.4(5%(v/v))に再懸濁させた。
SA-SRBCを、10μg/mLでビオチン化TNFaを用いてコーティングし、混合し、室温で20分間回転させた。SRBCは前と同様に1.0mLのPBS、pH7.4で2回洗浄した。TNFaコーティングSRBCは、RPMI(+10%FCS)に再懸濁させて、5%(v/v)の最終濃度にした。
10μLの5%SA-SRBCおよび10μLの5%TNFaコーティングSRBCを、40μLのPBSを含む別々の新たな1.5mLのエッペンドルフチューブにそれぞれ加えた。対照ヒト抗TNFa抗体を、45μg/mLでSRBCのそれぞれのサンプルに加えた。チューブを室温で25分間回転させ、次いで細胞を100μLのPBSで3回洗浄した。細胞を50μLのPBSに再懸濁させ、Alexa488と結合したGt抗ヒトIgGFc抗体(Molecular Probes、Eugene、OR)40μg/mLと共にインキュベートした。チューブを室温で25分間回転させ、次いで100μLのPBSで洗浄し、細胞は10μLのPBSに再懸濁させた。10μLの染色細胞を、透明なガラス製の顕微鏡スライド上にスポットし、ガラス製カバーガラスで覆い、蛍光下で観察し、0〜4の任意単位で記録した。
様々なアッセイによって、当該の免疫グロブリンを分泌するB細胞クローンを含むとして前に同定した、1つのミクロ培養ウェルの中身を採取した。100〜1000μLのピペットマンを使用して、37℃のRPMI(10%FCS)を加えることによって、ウェルの中身を回収した。ピペット処理することによって細胞を再懸濁させ、次いで新たな1.5mLのエッペンドルフチューブに移した(最終体積約500〜700μL)。室温で1分間、マイクロフュージ中において2500rpm(660rcf)で、細胞を遠心分離にかけ、次いでチューブを180℃で回転させ、2500rpmで1分間再度回転させた。凍結培地を除去し、免疫細胞を100μLのRPMI(10%FCS)に再懸濁させ、次いで遠心分離にかけた。RPMI(10%FCS)を用いたこの洗浄を繰り返し、細胞を60μLのRPMI(10%FCS)に再懸濁させ、使用する用意ができるまで氷上で保存した。
シリコーン端部を有するガラススライド(2×3インチ)を事前に調製し、室温で一晩硬化させた。使用前に、スライドを約5μLのSigmaCoat(Sigma、Oakville、ON)で処理し、ガラス表面を均一に拭き取り、乾燥させ、次いで強く拭き取った。細胞の60μLサンプルに、60μLのそれぞれのTNFaコーティングSRBC(5%v/vストック)、4×モルモット補体(Sigma、Oakville、ON)ストック、RPMI(10%FCS)中で調製したもの、および4倍増大させた血清ストック(1:150、RPMI(10%FCS)中)を加えた。調製したスライド上に、混合物-)をスポットし(10〜15μL)、スポットは非希釈パラフィン油で覆った。37℃で少なくとも45分間、スライドをインキュベートした。
TNFaコーティングされたヒツジ赤血球細胞を使用して、ウェルから抗原特異的なプラズマ細胞を同定した(表15参照)。
1種のプラズマ細胞を単離した後、mRNAを抽出し、逆転写酵素PCRを行って、可変部位重鎖および軽鎖をコードするcDNAを作製した。ヒト可変部位重鎖領域をクローニングして、IgG1発現ベクターにイソ型を変更した。このベクターは、ヒトIgG1の定常ドメインを、pcDNA3.1+/Hygro(Invitrogen、Burlington、ON)の多重クローニング部位にクローニングすることによって作製した。ヒト可変部位軽鎖領域は、IgK発現ベクターにクローニングした。これらのベクターは、ヒトIgKの定常ドメインを、pcDNA3.1+/Neo(Invitrogen、Burlington、ON)の多重クローニング部位にクローニングすることによって作製した。次いで重鎖および軽鎖の発現ベクターを、70%の集合状態の60mm皿中に同時にリポフェクションし、ヒト胚腎臓293細胞およびトランスフェクトした細胞は、24〜72時間、元のプラズマ細胞と同一の特異性を有する組換え抗体を分泌させた。上清(3mL)をHEK293細胞から採取し、完全抗体の分泌をサンドウィッチELISAによって実証して、ヒトIgGを特異的に検出した(表16)。ELISAを使用して組換え抗体とTNFaの結合により、特異性を評価した。
大規模生成用に、重鎖および軽鎖発現ベクター(2.5μgのそれぞれの鎖/皿)を、HEK293細胞を含み70%の集密状態であった100mm皿10枚にリポフェクションした。トランスフェクトした細胞は、37℃で4日間インキュベートし、上清(6mL)を採取し、6mLの新たな培地と交換した。第7日に、上清を除去し、最初の採取物と共に集めた(10プレートから合計120mL)。Protein-A Sepharose(Amersham Biosciences、Piscataway、NJ)親和性クロマトグラフィーを使用して、それぞれの抗体を上清から精製した(1mL)。500mcLの0.1Mグリシン、pH2.5を用いてプロテイン-Aカラムから抗体を溶出させた。溶出物はPBS、pH7.4中で透析し、濾過滅菌した。抗体は非還元SDS-PAGEにより分析して、純度および収率を評価した。OD250でのUV分析によって、濃度も測定した。
抗TNFa抗体と膜貫通TNFaの結合
可溶性および膜結合TNFaのいずれもTNFaレセプターと相互作用し、TNFaの炎症誘発性作用に寄与する可能性がある。したがって、299v2および263が、可溶型の分子以外に、膜結合TNFaとも有効に結合することができるかどうかを、確立することが重要であった。この目的のために、TNFaトランスフェクトCHO細胞および活性化T細胞を使用した。
エピトープ領域分けアッセイ
抗TNFa抗体のエピトープマッピング
以下の事項は、抗TNFa抗体のエピトープをマッピングするために使用する方法を記載する。キメラTNFaタンパク質を、ヒトおよびマウスTNFaを使用して構築し、発現させた。ヒトおよびマウスTNFaの配列を表17に与える。
抗マカークザルTNFa結合の交差反応性
ヒトおよびサル可溶性組換えTNFaとの結合
抗TNFa抗体を、可溶性組換えTNFaと結合する、それらの能力に関しても試験した。ヒトおよびサル(カニクイマカークザル)TNFaを、GSTとの融合タンパク質として大腸菌中で発現させた。結合はELISAによって評価した。299v2、263、エタネルセプト、インフリキシマブおよびアダリムマブ(「抗TNFa抗体」)を、0.5μg/mlのヒトGST-TNFa、2μg/mlのサルGST-TNFa、および10μg/mlのGSTを一晩コーティングした96ウェルプレート中でインキュベートした。HRP結合ヤギ抗ヒトIgG抗体を使用して、結合した抗体を検出した。抗TNFa抗体はいずれも、類似した用量応答でヒトTNFaと結合することが、結果によって示された(図5)。抗TNFa抗体は、サルTNFaとは異なる結合をする。299v2、エタネルセプト、およびアダリムマブは、類似した方式でカニクイマカークザルTNFaと結合するが、263およびインフリキシマブは、カニクイマカークザルTNFaとは結合しないようである(図6)。
動態分析
KinExA(登録商標)およびBIACORE(登録商標)技術を使用して、抗TNFa抗体の動態測定値を評価した。KinExA(登録商標)法は、平衡状態における正式な親和性の測定値の溶液系の測定を含む。それぞれのヒト抗TNFa抗体の結合動態を測定するために、3つのうちのまったく同様の2つの実験を行った。両方の実験において、知られている濃度の抗原を滴定し、異なる抗体濃度をそれぞれの抗原滴定物に加え、結合平衡状態に到達させた。ヒトTNFaにおけるKd測定値を決定するために、1つのトリマー(52.5kDa)、表22参照、3つのモノマー(17.5kDa)、表23参照の、TNFa結合部位のモル濃度を使用して、Kdを計算した。これらの結果は、二重曲線分析によって分析した。ウサギR014抗体の動態測定は前とほぼ同様に行ったが、しかしながら、知られている抗体濃度を使用して、未知の抗原濃度による方法を行い、Kdを計算した。さらに、親和性の影響の可能性を無効にするために、パパインによる切断によってFab断片を作製し、動態分析を繰り返した(表24参照)。
IN VITROでの抗HTNFa抗体の特徴付け。
ヒトMCF-7細胞におけるTNFa誘導型アポトーシスの阻害
IgG2kappaおよびλハイブリドーマを、前に記載したのと同様に、大量に培養し、精製し正確に測定した。イソ型変更したハイブリドーマ、およびXENOMAX(登録商標)由来IgG1組換え抗体を、前に記載したのと同様に、発現させ、精製し正確に測定した。ヒトMCF-7細胞に対するTNFa誘導型アポトーシスの生物学的影響を中和する、それらの能力に関して、抗体をさらにアッセイした。96ウェルプレートに、5000細胞MCF-7/ウェル、200μl/ウェル、およびフェノールレッドを含まないDMEM+10%FCSで接種した。プレートは37℃+5%CO2で一晩インキュベートした。それぞれのプレート上で、0.005ng/ml〜10μg/mlの最終濃度で、それぞれの抗体の滴定値をアッセイした。抗TNFa試薬を、100pg/mL(トリマーとして1.9pM)TNFaの一定濃度において、三連または六連までで、アポトーシス培地(2.5%FCS、5μg/mLのCHXをフェノールレッドを含まないDMEMに溶かしたもの)に希釈した。アポトーシス培地中にTNFaのみを含む6つのウェル、およびアポトーシス培地のみを含む6つのウェルも、インキュベートした。TNFa+/-中和抗体は、37℃+5%CO2で1時間または18時間プレインキュベートした。200μLのTNFa+/-中和抗体を細胞に移し、37℃+5%CO2で一晩インキュベートした。
IgG2kappaおよびλハイブリドーマを、前に記載したのと同様に、大量に培養し、精製し正確に測定した。イソ型変更したハイブリドーマ、およびXENOMAX(登録商標)由来IgG1組換え抗体を、前に記載したのと同様に、発現させ、精製し正確に測定した。ヒトWM266.4細胞に対するTNFa誘導型アポトーシスの生物学的影響を中和する、それらの能力に関して、抗体をさらにアッセイした。20,000個のWM266.6細胞を、96ウェルプレート中、完全培地(RPMI1640/10%FBS/Gln/P/S)中に平板培養し、一晩37℃/10%CO2でインキュベートした。培地を除去し、50μLの試験抗体およびTNFa(室温で30分プレインキュベートしたもの)を、無血清培地(RPMI1640/Gln/P/S)に加えた。50μLのシクロヘキサミドのプレートを、以下の最終的なアッセイ条件下で、前と同様に一晩インキュベートした。V=100μL、シクロヘキサミド=6μg/mL、トリマーとしてTNFa=600μg/mL=11.4pM、試験抗体濃度は、記載したように変わる。100μLのカスパーゼバッファーおよび0.3μLのカスパーゼ基質(APO-ONE、Promega)を、ウェル毎に加えた。励起波長@485nmおよび発光波長@530nmで、Victor Wallacにおいて、カスパーゼ活性を測定した。アポトーシスを中和する抗体の能力の一例は、図10に示す。図10は、抗TNFaの中和に関する平均IC50値を示す、棒グラフである。中和はヒトWM266細胞に行い、カスパーゼ活性は、TNFa誘導型アポトーシスの指標として測定した。抗体のIC50の計算は、図7の簡単な説明中に記載したのと同様に行った。
ヒト全血の培養によって、細胞培養物または実験動物中には存在する可能性がない、本来存在する臨床関連の状態が再生される。全血培養を使用して、TNFa誘導型IL-8産生を中和するための、抗TNFa抗体の有効性を評価した。静脈穿刺によって正常なドナーから全血を得て、EDTAチューブに回収し、96ウェルプレートに平板培養した。抗TNFa抗体をRPMI培地に希釈し、全血と混合させた。無関係なヒトIgG1抗体を、対照として使用した。これに、TNFaの添加を続けた(最終濃度100pg/ml、トリマーとしてTNFaを考えると1.9pMに対応)。次いでプレートを、37℃で6時間インキュベートした。インキュベーションの後、TritonX-100を0.5%v/vの最終濃度で培養物に加えて、細胞溶解を引き起こした。IL-8産生はELISAによって測定した。結果を表すために、IgG1対照の存在下でTNFaによって誘導されたIL-8を、100%として設定した。表28は、阻害曲線(図11)を使用して計算した、抗TNFa抗体のIC50を報告する。299v2およびエタネルセプト代用物は、最も低いIC50および最高の有効度を示す。
抗TNFa抗体をアッセイして、PBMC、主にNK細胞によって仲介される、TNFaトランスフェクトCHO細胞の殺傷を支持する、それらの能力を測定した。簡潔には、ヒトPBMCを正常なドナーから得て、エフェクター細胞に加えて、1:100エフェクター/標的細胞の比率を生み出すように測定した濃度で再懸濁した。同時に、膜結合TNFaを安定して発現する、TNFaトランスフェクトCHO細胞を、膜染色液PKH-26で標識した。次いでCHO細胞を、5μg/ml抗体有りまたは無しで、三連で96ウェル皿に接種した。30分のインキュベーションの後、エフェクター細胞を加えて、ADCC反応を37℃で一晩進行させた。この時点で、三連のサンプルを集め、製造者の教示書に従い染色液TOPO-3で染色し、FACSによって分析した。PKH-26およびTOPO-3二重陽性細胞(死んだ標的細胞)とPKH-26単独陽性細胞(生きた標的細胞)の数の比率を計算し、これを使用して結果をパーセンテージとして表した。これらの結果は、モノクローナル抗体が、エタネルセプト代用物として使用した、p75-hFcとは著しい違いで、ADCCを支持する能力を有することを示す(表29)。
抗TNFa抗体を、補体を固定し、それによってTNFaトランスフェクトCHO細胞の殺傷を仲介する能力に関してもアッセイした。簡潔には、CHO細胞を125000/ウェルで96ウェルプレートに接種し、5μg/mlで抗体をまったく同様に加えた。氷上での3時間のインキュベーションの後、ウサギ補体を10%の最終濃度まで加え、CDC反応を室温で30分進行させた。この時点で、細胞を0.5pg/mlのPIおよび2.5μg/mlのヘキスト33342で1時間染色し、オートスコープを使用して計数した。実験は三連で行った。TNFa誘導型アポトーシスアッセイに関して前に記載したのと同様に、結果を計算し表した。ADCCの場合と同様に、これらの結果は、モノクローナル抗体が、エタネルセプト代用物として使用した、p75-hFcとは違い、CDCを刺激する能力を有することを示す(表29)。
IN VIVOでの抗HTNFa抗体の特徴付け。
マウスにおけるTNFa誘導型の肝臓損傷の阻害
抗ヒトTNFa抗体が、ヒトTNFaをin vivoで中和するかどうかを試験するために、マウスにおけるヒトTNFaおよびD-ガラクトサミン(D-Ga1N)投与によって誘導される肝臓損傷に対して保護する、抗ヒトTNFa抗体の能力を試験した(Lehmann V他、J.Exp.Med.、1987 165(3):657〜63)。TNFaおよびD-Ga1Nを投与することによって、ショックおよび死を最終的にもたらす、肝細胞の広範囲のアポトーシス死によって特徴付けられる、LPSおよびD-Ga1Nによって誘導される肝臓損傷と似た、劇症の肝臓損傷が誘導される。D-Ga1N処理によって、マウスは、リポ多糖(LPS)およびネズミTNFaの致死的影響に対して、100〜1000倍を超えて敏感になる(Lehmann V他、J.Exp.Med.、1987 165(3):657〜63)。LPSおよびD-Ga1Nによって誘導されるアポトーシス性肝臓損傷は、内因的に生成されるTNFaに依存することが示されてきている(Leist M他、Am.J Pathol.、1995、146(5):1220〜34)。この肝臓損傷がp55レセプターを介して分泌されるTNFaシグナルのみに依存することも実証されてきており(Nowak M他、Am.J.Physiol.2000、278(5):R1202〜9)、D-Ga1Nは、マウス中ではp55TNFaレセプターにのみ結合する、ヒトTNFaの致死的影響にも敏感であることが示唆される。hTNFaおよびD-Ga1Nによって誘導される肝臓損傷は、アラニンアミノトランスフェラーゼ(ALT)の血清酵素活性を測定することによって評価した。
in vivoでTNFaを阻害する抗TNFa抗体の能力を試験するための他の手法として、抗TNFa抗体を使用して、ヒトによってマウス中で誘導されるIL-6の産生を阻害した。TNFaは、IL-6の誘導を含めた、多くの急性の生物学的作用を引き起こす(Benigni他、J.Immunol.157:5563、1996)。1グループ当たり8〜10匹のマウスを使用した。時間行程の実験で最初に確定したように、ヒトTNFaをマウスに注射することによって、注射後2時間でピークに達する、血清IL-6レベルの急速な上昇が引き起こされる。TNFaの投与の用量および経路を定義することを目的とした、他の予備実験の結果に基づいて、1μg/マウスのヒトTNFをマウスに静脈内注射した。IL-6レベルは、市販のELISAキット(R&D System)を使用してTNFa投与後2時間で測定した。用量応答性の実験を、TNF(1μg/マウス、静脈内)の90分前に、抗TNF抗体(1〜10静脈内μg/マウス)を注射することによって行った。対照マウスには、TNFの前に生理食塩水を与えた。データは対照の割合として表し、中和曲線を作製した(図13)。IC50は4つのパラメータ適合曲線を使用して計算した。表30は、異なる実験から平均した、異なる抗TNF抗体に関するIC50を示す。
抗TNFa抗体の構造分析
前の表1に示した抗体の、可変部位重鎖および可変部位軽鎖を塩基配列決定して、それらのDNA配列を決定した。ヌクレオチドおよびアミノ酸配列を含めて、すべての抗TNFa抗体に関する完全な配列情報を、ここに提示した配列表中に示す。
カノニカルクラスの抗体の決定
Chothia他は、それぞれの免疫グロブリン鎖の超可変領域に関する「カノニカルクラス」という観点で、抗体構造を記載している(J Mol Biol.1987 Aug 20;196(4):901〜17)。様々な免疫グロブリンのFabおよびVL断片の原子構造を分析して、それらのアミノ酸配列とそれらの抗原結合部位の三次元構造の間の関係を決定した。Chothia他は、そのパッキング、水素結合中には、比較的少数の残基が存在し、あるいは異常なphi、psiまたはomegaの立体配座をとる能力は、超可変領域の主鎖の立体配座に主に原因があったことを見出した。これらの残基は、超可変領域内の部位、および保存型βシート骨格中に存在することが見出された。未知の構造を有する免疫グロブリンの配列を調べることによって、Chothia他は、多くの免疫グロブリンが、知られている構造の1つの大きさと大きさが類似しており、観察された立体配座の原因である部位に同一の残基をさらに含んでいた、超可変領域を有することを示す。
TNF-aエピトープに対する発現および結合アッセイによる、抗TNF-a抗体のドメイン分析
配列/領域分けの結果
可変(V)領域の免疫グロブリン鎖は、多数の生殖細胞系DNAセグメントによってコードされており、これらはB細胞個体発生中、機能的可変領域(VHDJHまたはVKJK)に接合している。TNF-aに対する抗体応答の、分子的および遺伝的多様性は、詳細に研究された。これらのアッセイによって、抗TNF-aに特異的ないくつかの点が明らかになった。TNF-aに特異的な65の個別の抗体の分析によって、13の生殖細胞系VH遺伝子が与えられ、そのうちの54個がVH3ファミリー由来であり、そのうちの34個はVH3-33遺伝子セグメントを使用した。最も多かった遺伝子、VH3-33生殖細胞系遺伝子は、分析した65個の抗体のうち34個中で発現され、結合(KappaA30とL2またはλ)と関係がある軽鎖の型と明らかな関係がある、2つの異なる領域に限られた。機能的抗体の選択および領域分けによって、特定の領域中の抗体は同じIgVHを発現したことが示され、いくつかの場合、同じVHDJHの再編成が示された。さらに、H鎖とL鎖の対は領域内で保存されたことも発見された。これらの発見によって、任意の所与のエピトープに関しては、生殖細胞系レパートリーのわずか数個のメンバーを使用して、対応するパラトープが形成され、それぞれの抗原エピトープに関しては、限られた数のL鎖およびH鎖遺伝子が対になって、特異的なパラトープを形成することができることが示唆される。
関節炎を治療するための、抗TNF-a抗体および抗体結合体の使用
関節炎を有するヒト患者における抗TNF-a抗体治療の、in vivoでの効果を測定するために、このようなヒト患者に、有効量の抗TNF-a抗体を一定量の時間注射する。治療中の定期的な時間で、ヒト患者を観察して、患者の関節炎が治療されているかどうか測定する。
診断用物質としての抗TNF-a抗体の使用
サンプル中のTNFa抗原の検出
サンプル中のTNFa抗原を検出するための、酵素結合免疫吸着アッセイ(ELISA)を開発することができる。このアッセイでは、マイクロタイタープレート、96ウェルマイクロタイタープレートまたは384ウェルマイクロタイタープレートなどのウェルに、この抗原を対照とする第1の完全ヒトモノクローナル抗体を数時間吸着させる。固定された抗体は、試験サンプル中に存在する可能性がある任意の抗原用の、捕捉抗体として働く。ウェルを洗浄し、乳タンパク質またはアルブミンなどのブロッキング剤で処理して、検体の非特異的吸着を防ぐ。
サンドウィッチELISAを開発して、ヒト血清中のTNFaレベルを正確に測定する。このサンドウィッチELISAからの、2つの完全ヒトモノクローナル抗TNFa抗体は、TNFa分子上の異なるエピトープを認識する。ELISAは以下のように行う:50μLの捕捉抗TNFa抗体を、コーティングバッファー(0.1MのNaHCO3、pH9.6)に2μg/mLの濃度で溶かしたものを、ELISAプレート(Fisher)にコーティングする。4℃で一晩のインキュベーションの後、200μLのブロッキングバッファー(0.5% BSA、0.1% Tween20、0.01% Thimerosal、PBS中)で、25℃において1時間プレートを処理する。0.05% Tween20をPBSに溶かしたもの(洗浄バッファー、WB)を使用して、プレートを3回洗浄する。正常または患者血清(Clinomics、Bioreclaimation)を、50%ヒト血清を含むブロッキングバッファーに希釈する。プレートは血清サンプルと共に4℃で一晩インキュベートし、WBで洗浄し、次いで100μL/ウェルのビオチン化検出抗TNFa抗体と共に25℃で1時間インキュベートする。洗浄後、プレートをHRP-ストレプトアビジンと共に15分間インキュベートし、前と同様に洗浄し、次いで色を生成させるために、100μL/ウェルのo-フェニレンジアミンをH2O2に溶かしたもの(Sigma現像液)で処理する。反応は50μL/ウェルのH2SO4(2M)を用いて停止させ、492nmにおいてELISAプレートリーダーを使用して分析した。血清サンプル中のTNFa抗原の濃度は、4パラメータ曲線適合プログラムを使用して、精製したTNFa抗原の希釈液との比較により計算する。
前に記載した本明細書は、当業者が本発明を実施するのを可能にするのに充分なものであると考えられる。前の記載事項および実施例は、本発明のいくつかの好ましい実施形態を詳細に述べ、本発明者によって企図される最良の形態を記載する。しかしながら、詳細な前述の記載事項が書物中にどのように表れようとも、本発明は多くの方法で実施することができ、添付の特許請求の範囲およびその任意の均等物に従って、本発明が解釈されるべきであることは理解されよう。

腫瘍壊死因子α(TNFa)と特異的に結合する、請求項1または請求項23に記載の完全ヒトモノクローナル抗体を治療上有効な用量、前記動物に投与することを含む方法。 - 前記腫瘍性疾患が乳癌、卵巣癌、膀胱癌、肺癌、神経膠芽細胞腫、胃癌、子宮内膜癌、腎臓癌、結腸癌、膵臓癌、および前立腺癌からなる群から選択される、請求項47に記載の方法。
- 免疫介在炎症性疾患を有効に治療する方法であって、
炎症状態の治療を必要とする動物を選択すること、および
腫瘍壊死因子α(TNFa)と特異的に結合する、請求項1または請求項23に記載の完全ヒトモノクローナル抗体を治療上有効な用量、前記動物に投与することを含む方法。 - 前記免疫介在炎症性疾患が、慢性関節リウマチ、糸球体腎炎、アテローム性動脈硬化症、乾癬、再狭窄、自己免疫疾患、クローン病、移植片対宿主反応、敗血症性ショック、悪液質、食欲不振、強直性脊椎炎および多発性硬化症からなる群から選択される、請求項49に記載の方法。
- 動物での腫瘍壊死因子α(TNFa)誘導型アポトーシスを阻害する方法であって、
TNFa誘導型アポトーシスの治療を必要とする動物を選択すること、および
TNFaと特異的に結合する、請求項1または請求項23に記載の完全ヒトモノクローナル抗体を治療上有効な用量、前記動物に投与することを含む方法。 - 腫瘍壊死因子(TNFa)と特異的に結合する請求項1または請求項23に記載の抗体の、動物の腫瘍性疾患を治療するための薬剤の調製における使用。
- 前記腫瘍性疾患が、乳癌、卵巣癌、膀胱癌、肺癌、神経膠芽細胞腫、胃癌、子宮内膜癌、腎臓癌、結腸癌、膵臓癌、および前立腺癌からなる群から選択される、請求項52に記載の使用。
- 腫瘍壊死因子(TNFa)と特異的に結合する請求項1または請求項23に記載の抗体の、動物の免疫介在炎症性疾患を有効に治療するための薬剤の調製における使用。
- 前記免疫介在炎症性疾患が、慢性関節リウマチ、糸球体腎炎、アテローム性動脈硬化症、乾癬、再狭窄、自己免疫疾患、クローン病、移植片対宿主反応、敗血症性ショック、悪液質、食欲不振および多発性硬化症からなる群から選択される、請求項54に記載の使用。
- 腫瘍壊死因子(TNFa)と特異的に結合する請求項1または請求項23に記載の抗体の、動物での腫瘍壊死因子誘導型アポトーシスを有効に治療するための薬剤の調製における使用。
