Abstract
Description

4,6−ジクロロピリミジン−2−メチルスルホン(A):以下のようにして、Koppellら、JOC,26,1961,792で述べられた方法と実質的に類似の方法により、調製した。4,6−ジクロロ−2−(メチルチオ)ピリミジン(50g、0.26mol)のジクロロメタン(1L)攪拌溶液に、0℃で、20分間にわたって、メタ−クロロ過安息香酸(143.6g、0.64mol)を加えた。この溶液を室温まで温め、そして4時間攪拌した。その混合物をジクロロメタン(1.5L)で希釈し、次いで、50%Na2S2O3/NaHCO3溶液(2×200mL)、NaHCO3飽和溶液(4×300mL)およびブライン(200mL)で連続的に処理し、次いで、乾燥した(MgSO4)。真空中で溶媒を除去して、灰白色固形物を得、これを、EtOAc(1L)に再溶解し、そしてNaHCO3飽和溶液(3×300mL)およびブライン(100ml)で連続的に処理し、次いで、乾燥した(MgSO4)。真空中で溶媒を除去して、白色固形物(55.6g、収率96%)として、表題化合物(A)を得た。1H NMR CDCl3 δ3.40(3H,s,CH3),7.75(1H,s,ArH)。
シクロプロパンカルボン酸[4−(4,6−ジクロロ−ピリミジン−2−イルスルファニル)−フェニル]−アミド(C):化合物A(10g、44.04mmol)およびシクロプロパンカルボン酸(4−メルカプト−フェニル)−アミド(B、8.51g、44.04mmol)のt−ブタノール(300mL)懸濁液を真空排気により脱気し、次いで、窒素でフラッシュした。その混合物を、90℃で、窒素雰囲気下にて、1時間攪拌し、次いで、真空中で溶媒を除去した。その残留物を酢酸エチル(600mL)に溶解し、そして炭酸カリウムおよび塩化ナトリウムの水溶液で洗浄した。その有機抽出物を硫酸マグネシウムで乾燥し、低容量まで濃縮し、そして結晶化させた。無色結晶として、生成物Cを集めた(11.15g、74%)。1H−NMR DMSO−d6,δ0.82−0.89(4H,m),1.80−1.88(1H,m),7.55(2H,d),7.70−7.76(3H,m),10.49(1H,s);M+H,340。
シクロプロパンカルボン酸{4−[4−クロロ−6−(5−メチル−2H−ピラゾール−3−イルアミノ)−ピリミジン−2−イルスルファニル]−フェニル} アミド(D):ジメチルホルムアミド(6mL)中の化合物C(1.0g、2.94mmol)および3−アミノ−5−メチルピラゾール(314mg、3.23mmol)の混合物を、ジイソプロピルエチルアミン(0.614mL、3.53mmol)およびヨウ化メチル(530mg、3.53mmol)で処理した。この混合物を、窒素下にて、85℃で、4時間攪拌し、室温まで冷却し、そして酢酸エチルで希釈した。その溶液を水(×4)で洗浄し、硫酸マグネシウムで乾燥し、そして5mlまで濃縮して、結晶化し無色結晶を収集すると、表題化合物D(920mg、78%)が得られた。
シクロプロパンカルボン酸{4−[4−(4−メチル−ピペラジン−1−イル)−6−(5−メチル−2H−ピラゾール−3−イルアミノ)−ピリミジン−2−イルスルファニル]−フェニル}−アミド(V−1):化合物D(2.373g、5.92mmol)をN−メチルピペラジン(10mL)で処理し、その混合物を、110℃で、2時間攪拌した。真空中で過剰のN−メチルピペラジンを除去し、その残留物を酢酸エチルに溶解し、炭酸水素ナトリウム水溶液で洗浄し、硫酸マグネシウムで乾燥し、そして濃縮した。その残留物をメタノールから結晶化して、所望生成物V−1の無色結晶(1.82g、66%)を得た。
シクロプロパンカルボン酸{4−[4−(4−メチル−4−オキシ−ピペラジン−1−イル)−6−(5−メチル−2H−ピラゾール−3−イルアミノ)−ピリミジン−2−イルスルファニル]−フェニル}−アミド(V−19):化合物V−1(1g、2.1mmol)をジクロロメタン(20mL)に溶解し、0℃まで冷却し、そして10分間隔にて、10アリコートで、mCPBAのジクロロメタン溶液で処理した(各アリコートは、DCM(1mL)中の100mg、0.44mmolからなる)。アリコートを加えるたびに、その溶液は褐色になり、そしてmCPBAが消費されるたびに、徐々に、黄色に戻った。一旦、全ての出発物質が消費されると、真空中で溶媒を除去し、得られた橙色オイルを分取HPLCで精製して、灰白色固形物(69mg、7%)として、表題化合物を得た;
シクロプロパンカルボン酸{4−[4−(4−メチル−ピペラジン−1−イル)−6−(5−メチル−2H−ピラゾール−3−イルアミノ)−ピリミジン−2−イルスルファニル]−フェニル}−アミドメタンスルホネート(V−1ii):化合物V−1(515mg、1.1lmmol)をエタノール(80mL)に懸濁し、そして還流状態まで加熱した。この透明溶液に、メタンスルホン酸(106mg、1.11mmol)を加え、その反応混合物をさらに10分間還流した。その混合物を室温まで冷却し、そして沈殿物が形成し始めるまで、溶媒を蒸発させた。次いで、この混合物を0℃まで冷却し、そして得られた沈殿物を濾過により集めた後、真空中で乾燥して、白色固形物(290mg、47%)として、表題化合物を得た;
本発明の手順に従い、そして上記実施例1〜8で示した方法と実質的に類似の方法により、下記の表4で示した以下の化合物を調製した。これらの化合物の特性付けデータを以下の表4で要約するが、これは、1H NMR、融点(m.p.)および質量スペクトル(MS)データを含む。
本発明の化合物のキナーゼ阻害剤としての活性は、インビトロ、インビボまたは細胞株内でアッセイされ得る。インビトロアッセイとしては、活性化されたAurora酵素および/もしくはFLT−3酵素のキナーゼ活性またはATPase活性のどちらかの阻害を決定するアッセイが挙げられる。代替的なインビトロアッセイは、阻害剤のAuroraおよび/またはFLT−3に結合する能力を定量化し、そして、結合の前に阻害剤を放射標識する工程、阻害剤/Aurora複合体および/もしくは阻害剤/FLT−3複合体を単離し、放射標識の結合量を決定する工程、または、公知の放射性リガンドに結合するAuroraおよび/もしくはFLT−3とともに新規の化合物をインキュベートする競争実験を実施する工程のいずれかによって測定され得る。どのAurora型もしくはどのアイソフォームが阻害されるかに依存して、Auroraの任意の型もしくはアイソフォームを使用し得る。酵素アッセイに使用される条件の詳細は、以下の実施例に示す。
(Auroraの阻害に対するKiの決定)
標準結合酵素アッセイ(Foxら、(1998)、Protein Sci 7、2249)を使用して、化合物のAuroraを阻害する能力について、以下の様式で化合物をスクリーニングした。0.1M HEPES 7.5、10mM MgCl2、1mM DTT、25mM NaCl、2.5mM ホスホエノールピルビン酸、300mM NADH、30mg/ml ピルビン酸キナーゼ、10mg/ml 乳酸脱水素酵素、40mM ATPおよび800μM ペプチド(LRRASLG、American Peptide、Sunnyvale、CA)を含有するアッセイストック緩衝溶液に、本発明の化合物のDMSO溶液を添加し、最終濃度30μMにした。生じた混合物を、30℃で10分間インキュベートした。10μLのAuroraストック溶液を添加することにより反応を開始し、アッセイ中で70nMの最終濃度を得た。BioRad Ultramarkプレートリーダー(Hercules、CA)を用いて、30℃で5分間より長い読み取り時間にわたって、340nmで吸光度をモニタリングすることにより、反応速度を得た。Ki値を、インヒビター濃度の関数として、速度データから決定した。
(FLT−3の阻害に対するKiの決定)
放射測定フィルター結合アッセイを使用して、化合物のFLT−3活性を阻害する能力について、化合物をスクリーニングした。このアッセイは、33Pの基質ポリ(Glu、Tyr)4:1(pE4Y)への取り込みをモニタリングする。反応を、100mM HEPES(pH7.5)、10mM MgCl2、25mM NaCl、1mM DTT、0.01% BSAおよび2.5% DMSOを含有する溶液中で行なった。このアッセイの最終基質濃度は、90μM ATPおよび0.5mg/ml pE4Y(両方ともSigma Chemicals、St Louis、MOから入手した)であった。本発明の化合物の最終濃度は、一般的に、0.01μMと5μMとの間である。代表的に、試験化合物の10mM DMSOストックから連続希釈を調製することによって、12点滴定を行なった。反応は、室温で行った。
(Colo205細胞アッセイにおけるAuroraの阻害に対するIC50の決定)
化合物はまた、細胞増殖の阻害についてアッセイした。このアッセイにおいて、RPMI1640培地(Sigma)に10% ウシ胎仔血清、L−グルタミンおよびペニシリン/ストレプトマイシン溶液を添加することによって、完全培地を調製した。結腸癌細胞(COLO−205細胞株)を、1.25×104細胞/ウェル/150μLの播種濃度で、96ウェルプレートに添加した。試験化合物の溶液を、連続希釈によって、完全培地中に調製し、試験化合物溶液(50μL)を各ウェルに添加した。
(腫瘍細胞型および正常細胞型のパネルにおける細胞増殖の測定:3Hチミジン取り込みアッセイ)
細胞増殖決定のよく特徴付けられた方法として、3Hチミジン取り込みアッセイを選択した。正常組織および広範な種々の腫瘍型由来の細胞を、分析のために選択した。腫瘍細胞は、高レベルのAuroraタンパク質を発現し(例えば、MCF−7、PC3、A375、A549)(節5.3.5およびBischoffら、EMBO J.1998、17、3052〜3065を参照のこと)、そして/または、ヌードマウスおよびヌードラットにおいて腫瘍を形成し得る(例えば、HCT116、MCF−7およびMDA−MB−231)ので、多数の腫瘍細胞を選択した。

を投与する工程を包含する、方法。 - 患者における癌を治療する方法であって、該患者に、請求項40に記載の組成物を投与する工程を包含する、方法。
- 前記患者に、追加化学療法薬または抗増殖薬を投与する工程を包含する、請求項51に記載の方法。
- 前記癌が、黒色腫、リンパ腫、神経芽細胞腫、白血病、もしくは大腸癌、乳房癌、肺癌、腎臓癌、卵巣癌、膵臓癌、腎臓癌、CNS癌、頚部癌、前立腺癌から選択される癌、または消化器の癌から選択される、請求項51に記載の方法:。
- 前記癌が、急性骨髄性白血病(AML)、急性リンパ性白血病(ALL)、肥満細胞症または胃腸間質性腫瘍(GIST)から選択される、請求項51に記載の方法。
- 患者における癌を治療するかその重症度を減らす方法であって、Auroraタンパク質キナーゼを請求項34に記載の化合物で阻害することにより、該癌細胞の有糸分裂を中断させる工程を包含する、方法。
- 前記患者に、請求項40に記載の組成物を投与する工程を包含する、請求項55に記載の方法。
