また、POMC遺伝子産物が免疫応答や炎症反応に影響を与えられるということが明らかになっている。例えば、αMSHは種々の炎症性サイトカインの反応を抑制し、また一方で抗炎症性サイトカインIL−10の産生がαMSHによって促進されていると考えられている。これはαMSHが免疫応答や炎症反応の抑制に重要な役割を果たしていることを意味している。数々の研究により、C末端トリペプチドの投与がこれらの効果を引き起こすに十分であることから、αMSHの免疫調節および抗炎症作用はαMSHのC末端領域(アミノ酸11−13:Lys−Pro−Val)によって調節されることが示唆されている(非特許文献1、非特許文献2)。

前述の化合物は、あらゆるタイプの急性または慢性炎症の治療剤として使用できる。これらはなかんずく、例えば皮膚の急性または慢性炎症、乾癬、アトピー性皮膚炎、鼻炎から接触アレルギー、喘息、食物アレルギーに至るあらゆるアレルギー反応、自己免疫疾患、線維症や強皮症、移植拒否、また血管障害も含む。本化合物は、炎症状態の皮膚の治療に好適に用いられる。この場合において、軟膏やクリームの形状で局所的製剤として本化合物を塗布するのが好ましい。軟膏またはクリーム中の本化合物の濃度範囲は、通常1μMから1mM、好ましくは10μMから100μMである。さらに、軟膏またはクリームは、例えば Dermatologie und Venerologie、ブラウン・ファルコ (Braun−Falco)他、スプリンガーフェアラーク(Springer Verlag) ベルリン(1996)、または Dermatopharmakologie und Dermatotherapie、Merk/Bickers(1992)に記載されている慣用の成分を含んでいてもよい。

本発明における好ましい実施形態として、ペプチドもまた炎症性腸疾患に対し使用することができる。炎症性疾患の例としては、比較的軽い食中毒によって生じる腸の短期間の炎症の他に、クローン病や潰瘍性大腸炎のような慢性腸疾患が挙げられる。

本発明者らは、驚くべきことに、インビトロで樹状細胞をハプテンおよびαMSHで処理し、次に実験動物へのその細胞を注入することにより、ハプテン特異耐性の産生やCHS反応(接触過敏性反応)の抑制がもたらされることを更に見出した。従って、さらに本発明は、抗原提示細胞を提供し、αMSHまたはその生物学的に活性のある誘導体若しくはフラグメントを細胞に接触させ、かつ細胞を抗原と接触させることを含む、抗原に対する耐性を与えうる細胞のインビトロでの産生方法に関する。なお、最後の2つの工程は、どんな順序でも又同時にでも行うことができる。

抗原提示細胞には様々な型がある。本発明によれば、樹状細胞またはランゲルハンス細胞が好ましい。抗原提示細胞が存在するのが他の成分や細胞を含まない製剤中である必要はない。抗原提示細胞は、また他の細胞と混合して供給することもできる。好ましい例は、抗原提示細胞としてランゲルハンス細胞が存在する上皮細胞の供給である。骨髄から樹状細胞を単離するか、または例えば自体公知のインビトロ培養によりPBMCのような前駆細胞から樹状細胞を生成することも可能である。抗原提示細胞を供給する方法は、例えばジャーナル オブ イムノロジー(J.of Immunol)、162巻、1号、168〜175頁、Laubeur他(1999)に記載されている。

次に細胞を、αMSHまたはその生物学的に活性のある誘導体若しくはフラグメントとインビトロで接触させる。αMSHの生物学的に活性のある誘導体若しくはフラグメントとは、例えば、αMSHの化学的修飾型、Lsy、Lys−Pro、Lys−pro−ValまたはLys−Pro−Thrを含むαMSHのフラグメント、あるいは前記物質の1つを含有する化合物である。実質的にαMSHの生物学的活性(耐性を産生する能力)が保たれてさえいれば、多種多様な修飾が考えられる。細胞と接触させる際のαMSHまたは前記誘導体の通常の濃度は、10-6M〜10-16M、好ましくは10-8M〜10-12Mである。

細胞をαMSHまたはその生物学的に活性な誘導体若しくはフラグメントに接触させた後、あるいはその前または同時に、細胞をインビトロで、耐性を誘発させたい抗原と接触させる。この場合抗原は、アレルギー反応の対象となる危険性があるタンパクであってもよい。例えば、もし免疫応答の対象となる抗原のハプテンが判明していれば、細胞をその特定のハプテンとのみ接触させることができる。この場合に可能な実施例として、長さ7〜20のアミノ酸、好ましくは長さ7〜15のアミノ酸を持つペプチドが挙げられる。

本発明のさらなる特徴は、抗原に対する耐性を誘発させる医薬を製造するためのαMSHまたはその生物学的に活性な誘導体若しくはフラグメントの使用である。製造される医薬は、耐性を与えることができる細胞のインビトロ産生のための上記方法によって得られる細胞を含有しているのが好ましい。

ペプチドであるLys−Pro−Thrは、内皮細胞やケラチノサイトでのTNFα、IL−1またはLPSによる転写因子NF−κBの活性化を抑制する。その結果、細胞接着分子(内皮細胞)やケモカイン(ケラチノサイト)の発現が減弱する。また発明者らは、例えばペプチドKPTが接触アレルギー(接触過敏反応、CHS反応)の発生を予防し、抗原特異的で長期間継続する耐性を誘発することを示すことが出来た。CHS反応は2段階に分けられる。抗原との最初の接触(誘導期)で後のCHS反応の基盤を据え、抗原との更なる接触により、その反応(接触性皮膚炎、すなわち、腫れ、かゆみなど)を生じさせる。本発明で使用される化合物は両段階の前に用いることができ、最初の接触前に用いられる時(注射、局所適用)は、CHSの抑制と耐性の誘発があり、接触性皮膚炎の惹起時に用いられる時は、本化合物は皮膚炎の発生を予防する。これらすべての適用において、実質的に完全にアレルギー反応が抑制される。

また、Lys−Pro−Thrは、樹状細胞の共刺激分子の発現を抑制することが分かった。これは、恐らく、CHSの抑制や耐性誘発と関連するメカニズムの一部であると考えられる。同時に本化合物は単核白血球による抗炎症性IL−10の分泌を増加する。この効果は同様に、アレルギー性接触皮膚炎と関連するメカニズムの1部である。

一つの説に拘束されることは決して望まないが、本発明の化合物は、β−アドレナリンレセプターと結び付く可能性がある。そのうえ本発明で用いられるペプチドは、I型IL−1レセプターと結合可能であると想定できる。本発明のペプチドが、例えばκオピオイドレセプターのような他のレセプターとも結合するということを除外することも出来ない。この前提に基づくと、本発明のペプチドは、炎症事象において独自のリガンドにより活性化された後に炎症を促進するよう介在する複数のレセプターと結合できると推測される。これらレセプターに本発明のペプチドが結合することにより、これらレセプターと独自のリガンドとの結合が妨げられ、かくして炎症促進の誘発が抑制される。一方、本発明のペプチドがそれらの最初の基質(αMSH)のレセプターと結合すると、これらレセプターが活性され、その結果、全般的に抗炎症に働く作用機構の更なる構成要素を誘起する。

【実施例1】

マウス:
7〜10週齢の雌Balb/Cマウスをチャールズリバー(Charles River; Sulzfeld, ドイツ)から入手し、政府規制に従って飼育した。

αMSHまたはKPVまたはKPTまたはKPの投与:
αMSHとペプチド類は、使用するまで分取して−20℃で貯蔵した。注射前に、特定の化合物を0.1%マウス血清含有PBSに溶解し、マウスの尾静脈に静脈注射するまで氷中で保存した。マウス一匹当りαMSH5μgまたはペプチド1.5μg(KP:50μg)を、感作2時間前に静脈注射した。

CHSおよび耐性の決定:
実験未使用マウスの剃毛した腹に、0.5%DNFB含有アセトン/オリーブ油(4:1)75μlを塗布することによりマウスを感作した。マウスの一方の耳の両面に0.3%DNFBを10μl塗布してCHSを誘起した。もう一方のコントロール処理耳と比較したハプテン曝露耳の耳の腫脹の度合いにより、CHSを測定した。CHSは、ハプテン曝露24時間後にバネ付きコンパス(spring-loaded dividers)を用いて測定した。耳を事前感作無しにハプテン曝露させたマウスを、ネガティブコントロールとして用いた。ハプテン投与前のαMSHまたはペプチド注射が耐性を誘導するか否か判定するため、感作2時間前に前述のようにマウスにαMSHまたはペプチドを静脈注射(腹部)、または曝露(左耳)した。αMSHに誘導されたCHS抑制を確認するため、感作7日後にマウスの一方の耳をハプテンで曝露し、その24〜36時間後に耳の腫脹反応を測定した。14日後、同じマウスの剃毛した背中に再感作し(外因性αMSHがなくなっている状態)、その1週間後、右耳に2度目のハプテン曝露を行いCHS反応誘導能力を調べた。
いくつかの実験ではαMSHの局所製剤を使用した。これらの実験では、マウスへの塗布は、感作の直前、または3時間または24時間前に感作部位(腹部)に施された。

結果:
αMSHおよびKPVまたはKPTまたはKPの静脈注射により、7日後のDNFB曝露に対するCHS反応を誘導するマウスの能力が抑制される。これらのマウスは、このようにDNFB特異的感作を発現しなかった。KPTは、CHS反応を最も効果的に抑制した(図1および13b参照)。
一時的な免疫抑制と特異な免疫学的耐性とを区別するため、マウスは2度目の感作を受け、ハプテン曝露された。最初の感作前にαMSHまたはKPVまたはKPTを注射したマウスは、2度目の感作量のハプテン投与によっても感作不可能であった。このことは、これらマウスがDNFBに対する耐性を発現したことを示す。KPVは弱い効果を示し、一方αMSHおよびKPTおよびKPは耳腫脹反応を非常に大幅に抑制した(図2および13b参照)。
【実施例2】

物質と方法:
単核細胞(PBMC)は、ヒト軟膜からフィコール・ハイパック(Ficoll-Hypaque)密度勾配遠心分離法によって分離した。抗生物質および10%FCSを添加したRPMI 1640中で培養した細胞(1×106/ml)は、処理されなかったか、またはIL−1β(10U/ml)の存在または非存在下でαMSHまたはペプチドKPVあるいはKPTで刺激された。PBMC培養の上清を24または48時間培養後に集め、さらに使用するまで−20℃で貯蔵した。IL−10の検出は商業的に利用可能な酵素免疫測定法(ELISA)によって行った。

結果:
無処理またはさまざまな濃度のαMSHまたはペプチド類で処理されたヒトPBMCは、24時間培養後に低濃度のIL−10(5〜10pg/ml)のみを産生した。αMSH(10-11M)、KPV(10-8〜10-9M)およびKPT(10-8〜10-9M)は、IL−10産生を明らかに誘導した(図3参照)。
ヒトPBMCは48時間培養後、有意な量のIL−10を産生した。αMSH、KPVおよびKPTはヒトPBLCによるIL−10の産生を有意に増加させた。αMSHとペプチド類には実質的な相違はなかった(図4参照)。
これら結果は、ペプチドKPTがαMSHやKPVと同様、静脈注射後にCHS感作を抑制できること、およびハプテン特異耐性を誘導できることを立証している。KPTはまた、インビボおよびインビトロにおいてIL−10を誘導できる。またデータよりインビボにおけるαMSHの免疫抑制効果がIL−10誘導にのみ基づくものではないことが推定される。
【実施例3】

物質および方法:
すべての工程は0〜4℃で行った。単球細胞株THP−1は、PBSで1回、酸性グリシン緩衝液(50mMグリシン、100mM塩化ナトリウム、pH3)で1回、およびRPMIで3回洗浄した。細胞(2.5×106/ml)を100μlのRPMI/1%BSAに再分散し、96穴マイクロタイタープレートに移した。ビオチン標識αMSH(10− 10M)の添加後、細胞を4℃で1時間インキュベートし、PBSで1回洗浄し、100μlのPBS/1%BSAに再分散し、FITC標識ストレプトアビジン(40μg/ml)と共に4℃、30分間暗所でインキュベートした。最後の洗浄処理後、細胞をPBSで再分散した。結合したビオチン標識αMSH量をフローサイトメーターを用いて分析した。対照実験では、細胞はビオチン標識αMSHを添加せずにFITC−スレプトアビジンの存在下でインキュベートした。死滅細胞はFACS分析直前にヨウ化プロピジウム(propidium iodide)を添加することにより排除した。ビオチン標識MSHの結合特異性を、非標識αMSH(10-6〜10-12M)またはKPVあるいはKPT(10-6〜10-12M)を添加することにより判定した。

結果:
ビオチン標識αMSHを用いたFACS分析によると、刺激されていないTHP−1細胞は、FITC−ストレプトアビジンのみと共にインキュベートした対照混合物と比較して、有意な量のαMSHに特異的な結合部位を発現した。この実験で用いられたαMSHの濃度は、10-10Mであった(図5参照)。
THF−1細胞が公知のメラノコルチン(MC)受容体の一つを発現しているか否かを判定するために、MC−1、MC−2、MC−3およびMC−4特異プライマーを用いてRT−PCRが実施された。総RNAはTHP−1細胞から得た。予期された416bpの長さを有するMC−1特有のPCR産生物が検出された(ジャーナル オブ ルーコサイト バイオロジー(J. Leuk. Biol.)、59巻、248頁、Rajora他(1996))。MC−2、MC−3あるいはMC−4特有のPCR産生物は検出されなかった。この結果は、THP−1細胞が他のメラノコルチン受容体とは異なり、αMSHおよびACTHに特異的であるMC−1を発現することを示している。
THP−1上に発現した結合部位がαMSHに特異的であるかどうかを調べるために、αMSH、KPVまたはKPTを用いた競合実験(competition experiments)を実施した。THP−1細胞をビオチン標識αMSH(10-10M)およびさまざまな濃度の非標識αMSHまたはペプチドと共にインキュベートし、その特異結合を測定した。濃度10-8Mの非標識αMSHでは、αMSHの結合が有意に抑制された。αMSHが10-6M、10-10M、または10-12Mの濃度で用いられた時には、有意な抑制は認められなかった(図6a〜6d)。
非標識KPVが用いられた時には、濃度が10-6Mの場合のみに有意な抑制が認められた(図7a〜7d参照)。
ペプチドKPTの場合には、テストした各濃度でαMSH結合の有意な抑制が認められた(10-6〜10-12M、図8a〜8d参照)。
これらの結果は、ペプチドKPTが、αMSHに特異的であるTHP−1細胞上のメラノコルチン受容体と結合することを示している。そしてそのことは、αMSHとKPTが共通の結合部位を有することを示している。しかし、KPTが非常に低濃度でも受容体への競合を示すため、実際にはこのペプチドはMSHよりも高いMC−1受容体への親和性をもつと推定できる。
【実施例4】

物質および方法:
ヒト皮膚微小血管細胞(HDMEC)および細胞株HMEC−1(ヒト微小血管内皮細胞株1)を、ペプチドのうち一つの存在または非存在下、TNFαまたはLPSのどちらかで処理した。細胞を、処理3時間後または6時間後にRNA単離のため、また処理3、6、16または24時間後に接着分子の酵素免疫測定(EIA)またはFACS分析のために採取した。RNAは逆転写を受け、半定量的な測定を行うため、Eセレクチン、ICAM−1、VCAMまたはハウスキーピング遺伝子としてのβ−アクチンを目的としてサンプルのPCRを行った。リンパ球接着能アッセイ用に、内皮細胞を培養皿に播種し、51Cr−標識リンパ球と共にインキュベートした。洗浄処理後、EC層と結合した残存リンパ球量をサンプル中の放射能を測定することにより決定した。

結果:
αMSHまたはKPTで内皮細胞の処理を行うと、LPSまたはTNFαに誘導された接着分子の発現が抑制された。この作用は、10-6〜10-12Mの濃度範囲のαMSHまたはペプチドに認められた。ペプチドKPTは接着分子mRNAの発現について最も強い作用を示した。
LPSまたはTNFαに誘導された接着分子の表面発現は、全てのアゴニストによってわずかに減少した。これらのデータは、全細胞を用いたEIAより、また特異抗体を用いたFACSより得られた(EIAデータを示す図9a参照)。
αMSHは、LPSまたはTNF−αMSHで処理されたEC層とのTおよびB細胞の結合を有意に抑制する(図9b参照)。総合すれば、これらの結果は、αMSHがリンパ球のECへの接着に対し影響を及ぼすことを示し、従って組織炎症状態におけるリンパ球の溢出をも減少させることを示している。これは、局部脈管炎に関するインビボのデータでサポートされている。
【実施例5】

上皮細胞(ECs)または正常ヒトケラチノサイト(HNK)を、ペプチドの存在または不存在下で、IL−1、LPSまたはTNFαで処理した。15または30分後、核タンパクを得、NF−κB特異結合シークエンスを有する放射性同位体でラベルしたオリゴヌクレオチドを用い、電気泳動度シフトアッセイ(EMSA)に付した。非標識オリゴヌクレオチドを競合物質として用いた。一部の実験において、NF−κBのp65やp50鎖に対する抗体を、検出されるバンドがp50ホモダイマーかp50/p65ヘテロダイマーのどちらであるかを確認するために使用した。

TNFα−またはLPS処理ECs、およびIL−1処理HNKsにペプチドを添加すると、転写調節因子NF−κBの活性が減少する(図10、13a参照)。これにより、数多くの炎症促進メディエーター(サイトカイン、ケモカイン、接着分子など)の遺伝子の転写が減少する。EMSAで観察されたバンドがNF−κBヘテロダイマーであることを、抗p65抗体を使って確認した。
【実施例6】

物質および方法:
マウスの一方の耳にLPSの皮下注射処理を行った。この事前の注射は、LPS注射部位での長持続的なEセレクチン発現の発生を誘導する。24時間後、第2回目のLPS投薬を腹腔内注射により行った(チャレンジ)。この2回目のLPSの注射は、急激な血管壊死症と、その大きさと数により容易に測定できる点状出血性病変の形成をもたらす。αMSH(25μg)は、LPSの事前注射時に投与した。

結果:
LPSの事前投与時にαMSHを注射すると、耳での局所Eセレクチン発現の誘導が抑制され(図11a参照)、LPSのチャレンジ注射後に形成された点状出血性病変の数と大きさが有意に減少する。
【実施例7】

物質および方法:
骨髄樹状細胞(BMDC)をマウスの大腿骨から単離し、IL−4およびGM−CSFで、6日または9日間処理した。6日または9日目に、細胞を、同じ遺伝的バックグランドを持つ実験未使用マウスへ再注射する3時間および2.5時間前に、αMSH(2×10-11M)またはペプチドKP(2×10-6M)で処理した。再注射2時間前に、細胞をハプテン(1mM DNBS、DNFBの水溶形状)で処理した。再注射直前に、細胞をPBSで2回洗浄した。5×10-5の細胞を動物各個体に静脈内注射した。対照細胞は、DNBS単独かαMSH単独で処理されるか、または未処理で放置した。注射5日後、動物の耳にDNFBを接触させ、次の日に耳の厚さを測定した。2週間後、動物をDNFBで再感作し、再度5日後に耳に再接触させた。最後に耳の腫脹を測定した。

未処理細胞、またはαMSH処理細胞を注射されたレシピエント動物は、予想されたとおり最初のチャレンジ後に免疫応答を示さなかった。DNBS処理したBMDCを注射されたレシピエント動物は、最初のチャレンジ時に妥当なCHS反応を示した。この反応は、インビトロでTNBSとαMSH、あるいはTNBSとKPで前もって処理された細胞を注射された動物において抑制された。このようにDCsをαMSHまたはペプチドと接触させると、CHSの抑制が十分に誘発される(図12参照)。
2回目のチャレンジ時、および対応する再感作時に、DNBS処理細胞を再び注射された動物は免疫反応を示さず、またDNBS/αMSH処理細胞を注射された動物は免疫反応を示さなかった。このことは、αMSHに誘導された耐性についてもDCが介在していることを示している(図12参照)。
【実施例8】

αMSHまたはαMSH誘導体処理樹状細胞あるいはT細胞を用いた免疫療法
樹状細胞(DC)は血液、骨髄または組織から単離した。しかし、DCを含む細胞混合物(例えば、上皮細胞の混合物)も使用できる。そしてGM−CSFやIL−4(好ましくは、各物質あたり250〜1000μ/ml)の存在下で培養した。
成熟期間(好ましくは、6〜9日)後、細胞は抗原(濃度は特定の抗原ごとに異なる。期間も同様)負荷し、αMSHまたはその誘導体で処理した。誘導体は、少なくともαMSHの12および13番目のアミノ酸(Lys−Pro)が一致し、Lys−Pro−Valを含有する誘導体が好ましい。アミノ酸のDおよびLの立体配置が可能であり、同様に保守的なアミノ酸置換も可能である。これは、なかんずく、IL−1β由来のLys−Pro−Thr、およびそのN末の伸展を有する誘導体を使う可能性を導く。C末伸展を有する誘導体もまた用いることができる。ペプチドの添加は、抗原の添加前、同時、添加後に、1回または1回以上(好ましい用量は特定のペプチドごとに異なり、例えばαMSHでは10-8〜10-14Mである。)行うことができる。
このように処理された細胞を、次にレシピエントの生物に静脈注射する(腹腔内、または皮下も可能);マウス:下限2×105細胞程度。抗原によって、1回の注射で十分であるか、または複数回注射を行うことが必要である。また、長期間の後に注射を繰り返す必要があることも可能である(これについてのデータはまだ得られていない)。
別の可能性としては、体外でDCをT細胞と接触させ、次にその混合物またはT細胞を注射する方法がある。この場合、DCの抗原負荷は、T細胞との接触前またはその最中に行うことが出来る。T細胞はさらに特定の抗原で既に感作された個々のものに由来していてもよい。マウスにおける下限はT細胞約100万であり、更に多くの細胞がより好ましい(図14と15参照)。

このような使用形態の利点は、抗原特異的であり抗原特異リンパ球(B細胞またはT細胞)が病原となっているあらゆるタイプの好ましからざる免疫反応を防止することである。これらはとりわけ、アレルギー、自己免疫疾患、慢性炎症や移植を包含する。十分に多くの数の細胞が用いられるのであれば、先在する病気の治療も可能である。
一つの説に拘束されることは望まないが、前記の驚くべき結果は、αMSHが効果の高い免疫調節剤で、多くの抗炎症的特性を有するという事実を示していると考えられる。これらは、なかんずく、DCにおける共刺激分子の発現を減少させる特性を包含する。同様の特性が、C末端のトリペプチドおよびジペプチドであるLys−Proを含む本発明のαMSH誘導体によっても示される。異なるアミノ酸組成(保守的アミノ酸置換)をもつ誘導体もまた類似の性質を持ち、これらは特にIL−1βに由来するLys−Pro−Thrを包含する(ゆえに、IL−1βと共直線性のシークエンスを持つ、N末端側(αMSHと相似)に伸びたペプチドは同じ作用を有すると考えられる。)。
αMSHおよびその誘導体もまた、インビボにおいてハプテン特異耐性を誘導できる。DCは、専門的には多様なタイプの免疫反応を誘導でき、またそのような反応の方向を決定する抗原提示細胞である。これらの免疫反応は、特にT細胞介在免疫反応を包含する。
インビトロにてαMSHまたは誘導体の存在下でDCまたはDC/T細胞混合物を抗原で処理すると、生物体内に注射後、その細胞が同様にハプテン特異耐性を誘発するということを今では示すことができる。
この場合、αMSHまたは誘導体によりサプレッサーT細胞が産生されることでDCによる抗原提示が調節されるというメカニズムであると思われる。このように、適切な混合物中のT細胞は高いCTLA−4の発現を示すということを明らかにすることができた(図16参照)。これはサプレッサーT細胞の特性を示す表面分子の一つである。
このようにして生成されたDCまたはT細胞が存在する時、自己免疫疾患、慢性炎症やアレルギーを予防的に妨ぐことが可能である。十分に多くの量の細胞を使用すれば、先在する病的な状態の治療もまた考えられる。
本発明の化合物は、樹状細胞を発生部位(in situ)で活性化することによって、腫瘍の治療に使用できる。この方法は本発明のペプチドの存在下で寛容化(tolerization)に使用することも可能である。

Claims (18)
- 式(I)の化合物のN末端がアシル化、および/またはC末端がアミド化またはエステル化されていることを特徴とする請求項1記載の使用。
- 式(I)の化合物がジペプチドであるリシン−プロリンまたはトリペプチドであるリシン−プロリン−トレオニンであることを特徴とする請求項1記載の使用。
- 炎症が乾癬、アトピー性皮膚炎、鼻炎、接触アレルギー、喘息、食物アレルギーからなる群より選択されることを特徴とする上記請求項のいずれかに記載の使用。
- 炎症が皮膚炎症であることを特徴とする上記請求項のいずれかに記載の使用。
- 式(I)の化合物が軟膏またはクリームの形態で投与されることを特徴とする上記請求項のいずれかに記載の使用。
- 式(I)の化合物が1μM〜1mMの濃度範囲で軟膏またはクリーム中に存在することを特徴とする請求項6に記載の使用。
- 式(I)の化合物が腹腔内、静脈内または経口的に投与されることを特徴とする請求項1〜5のいずれかに記載の使用。
- 式(I)の化合物の投与量が体重1kg当り20μg〜10mgであることを特徴とする請求項8に記載の使用。
- 式(I)の少なくとも2種の異なる化合物を使用することを特徴とする上記請求項のいずれかに記載の使用。
- 食品添加剤としての式(I)の化合物の使用。
- 化粧品組成物を製造するための式(I)の化合物の使用。
- (a)抗原提示細胞を供給する工程、
(b)細胞を、αMSHまたはその生物学的に活性のある誘導体若しくはフラグメントと接触させる工程、および
(c)細胞を抗原に接触させる工程
を含み、工程(b)および(c)はいかなる順序でも、または同時でも実施可能である工程を含むことを特徴とする、抗原に対する耐性を与えることができる細胞のインビトロでの産生方法。 - 抗原提示細胞が実質的に樹状細胞であることを特徴とする請求項13に記載の方法。
- 抗原提示細胞が実質的に上皮細胞であることを特徴とする請求項13に記載の方法。
- 工程(b)において、細胞を、αMSH、Lys−Pro−Val、Lys−Pro−Thr、Lys−ProまたはLysを含む化合物と接触させることを特徴とする請求項13〜15のいずれかに記載の方法。
- 抗原に対する耐性を誘発させる医薬を製造するための、αMSHまたはその生物学的に活性のある誘導体若しくはフラグメントの使用。
- 医薬が請求項13〜16のいずれかに記載の方法によって得られうる細胞を含有することを特徴とする請求項17に記載の使用。
