Claims (1)
- 【特許請求の範囲】 1. 形質転換された糸状菌の生成方法であって、 (1)糸状菌に導入されるべき少なくとも1つの発現可能な遺伝子DNAフラ グメントを、まずAgrobacterium tumefaciensのベクター中に、そのベ クター中に存在するT−DNAボーダー間にクローニングし; (2)T−DNAボーダー間に上記DNAを含むベクターを、DNA中にvi r領域を含むAgrobacterium tumefaciens株に導入し; (3)vir誘導化合物の添加によって上記Agrobacterium tumefaciensから 上記DNAフラグメントを含むT−DNAを放出させ、形質転換される べき糸状菌とともにこのAgrobacterium tumefaciens株をインキュベー トし;そして (4)形質転換された糸状菌を、導入されたDNA又はその発現生成物の特徴 に依存して非形質転換糸状菌から選択し、そして場合によっては形質転 換された糸状菌を培養すること を特徴とする方法。 2. 糸状菌が、Eumycotaの群に属する、請求項1記載の方法。 3. 糸状菌が、Ascomycotina、Basidiomycotina、Deuteromycotina、Mastigomyc otina、及びZygomycotina菌類亜門からなる群から選択される、請求項1記 載の方法。 4. DNAフラグメントが、所望の遺伝子の複数のコピーを含む、請求項1記載 の方法。 5. DNAフラグメントが、糸状菌ゲノムの選択された遺伝子座に組み込まれる ,請求項1記載の方法。 6. DNAフラグメントが、糸状菌ゲノムのpyrG遺伝子座(A.nigerについ てはpyrA遺伝子座及びNeurospora crassaについてはpyr4遺伝子座 として知られる)に組み込まれる、請求項5記載の方法。 7. 形質転換された糸状菌が、T−DNAボーダーを含め、望ましくない細菌D NA配列を含まない、請求項5記載の方法。 8. T−DNAボーダーを含め、望ましくない細菌DNA配列を含まない、請求 項7記載のアグロバクテリウム媒介性形質転換によって得られる形質転換さ れた糸状菌。 9. 場合によっては所望のタンパクの生産を誘導することを含む、請求項8記載 の形質転換された糸状菌の培養方法。 10.DNAフラグメントが、糸状菌ゲノム中にランダムに組み込まれる、請求項 1記載の方法。 11.T−DNAボーダー配列の1又は2以上の部分を含む、請求項10記載のア グロバクテリウム媒介性形質転換によって得られる形質転換された糸状菌。 12.場合によっては所望のタンパクの生産を誘導することを含む、請求項11記 載の形質転換された糸状菌の培養方法。
