- 【特許請求の範囲】 1.L-トレオニンを産生するためのプロセスであって、以下の工程: (a)培地中でE.coliの株を培養する工程;および (b)該E.coliによって産生されるL-トレオニンを回収する工程; を包含し、 ここで、該E.coliの株が、 (i)少なくとも1つの非天然プロモーターと作動可能に連結される少なくと も1つのトレオニン(thr)オペロンを染色体上に含み;そして (ii)トレオニンを産生するために1つ以上のトレオニン生合成酵素をコード する1つ以上の遺伝子を含む任意の組換えプラスミドを必要としない、プロセス 。 2.前記E.coliが、約30時間で少なくとも約50g/LのL-トレオニンを産生し得 る、請求項1に記載のプロセス。 3.前記非天然プロモーターが、tacプロモーター、Iacプロモーター、trpプ ロモーター、Ippプロモーター、PLプロモーター、およびPRプロモーターからな る群より選択される、請求項1に記載のプロセス。 4.前記トレオニンオペロンが、フィードバック耐性アスパラギン酸キナーゼ −ホモセリンデヒドロゲナーゼをコードする遺伝子を含む、請求項1に記載のプ ロセス。 5.前記E.coliがkat-13株である、請求項1に記載のプロセス。 6.前記非天然プロモーターがtacプロモーターである、請求項3に記載のプ ロセス。 7.前記E.coliが、染色体上に欠損トレオニンデヒドロゲナーゼ遺伝子を含 む、請求項1に記載のプロセス。 8.前記トレオニンオペロンが、ATCC 21277から得られる、請求項1に記載の プロセス。 9.前記E.coliがボレリジン耐性である、請求項1に記載のプロセス。 10.1つ以上のアミノ酸の生合成酵素をコードする組換えプラスミドを有さ ないE.coliのアミノ酸産生株を産生する方法であって、以下の工程: (a)E.coli株の染色体にアミノ酸産生微生物からの遺伝物質を導入する工程; (b)その発現を制御するために、アミノ酸生合成遺伝子の染色体配置の前にそ して該遺伝子と作動可能に連結して、該染色体に非天然プロモーターを挿入する 工程;および (c)必要に応じて、該染色体から、アミノ酸生合成に対する調節妨害物および /またはアミノ酸生合成のための栄養必要物を除去する工程、 を包含する、方法。 11.以下の特徴: (i)その染色体が、少なくとも1つの非天然プロモーターと作動可能に連結さ れる少なくとも1つのトレオニン(thr)オペロンを含み;そして (ii)トレオニンを産生するために、1つ以上のトレオニン生合成酵素をコード する1つ以上の遺伝子を含む任意の組換えプラスミドを必要としない、 を有する微生物E.coliの株。 12.前記トレオニンオペロンが、フィードバック耐性アスパラギン酸キナー ゼI−ホモセリンデヒドロゲナーゼI遺伝子(thrA)、ホモセリンキナーゼ(th rB)遺伝子、トレオニンシンターゼ遺伝子(thrC)からなる、請求項11に記載 の株。 13.前記E.coliがkat-13株である、請求項11に記載の株。 14.前記非天然プロモーターがtacプロモーターである、請求項11に記載 の株。 15.前記E.coliが、染色体上に欠損トレオニンデヒドロゲナーゼ遺伝子を 含む、請求項11に記載の株。 16.前記トレオニンオペロンが、ATCC 21277から得られる、請求項11に記 載の株。 17.前記E.coliがボレリジン耐性である、請求項11に記載の株。 18.前記E.coliがNRRL B-21593株の特徴を有する、請求項11に記載の株 。 19.L-トレオニンを産生するためのプロセスであって、以下の工程: (a)培地中でボレリジン耐性E.coliの株を培養する工程;および (b)該E.coliによって産生されるL-トレオニンを回収する工程、 を包含する、プロセス。 20.前記E.coliが、約30時間で少なくとも約50g/LのL-トレオニンを産生し 得る、請求項19に記載のプロセス。 21.前記E.coliが、少なくとも1つの非天然プロモーターと作動可能に連 結される少なくとも1つのトレオニン(thr)オペロンを含む、請求項19に記 載のプロセス。 22.前記トレオニンオペロンが、フィードバック耐性アスパラギン酸キナー ゼ−ホモセリンデヒドロゲナーゼをコードする遺伝子を含む、請求項21に記載 のプロセス。 23.前記E.coliがkat-13株である、請求項19に記載のプロセス。 24.前記非天然プロモーターがtacプロモーターである、請求項21に記載 のプロセス。
