Claims (13)

  1. Beschichtete Tablette, umfassend einen Tablettenkern und
    a) eine innere Versiegelungsüberzugsschicht, aufgetragen auf dem Tablettenkern, wobei die innere Versiegelungsüberzugsschicht eine Überzugspolymerformulierung umfasst, welche ein Polymer auf Polyvinylalkohol-(PVA-)Basis und einen Weichmacher umfasst,
    b) eine zweite Überzugsschicht, umfassend ein Medikament, aufgetragen auf der inneren Versiegelungsüberzugsschicht des Tablettenkerns, wobei die zweite Überzugsschicht Saxagliptin

    oder ein pharmazeutisch verträgliches Salz davon und eine Überzugspolymerformulierung, welche ein Polymer auf PVA-Basis und einen Weichmacher umfasst, umfasst, und
    c) eine äußere Schutzüberzugsschicht, aufgetragen auf der zweiten Überzugsschicht des Tablettenkerns, wobei die äußere Schutzüberzugsschicht eine Überzugsschichtformulierung umfasst, welche ein Polymer auf PVA-Basis und einen Weichmacher umfasst, und wobei die zweite Überzugsschicht 0,1 bis 70 Gew.-% Medikament und 30 bis 99,5 Gew.-% Überzugspolymer umfasst, basierend auf dem Gewicht der zweiten Überzugsschicht.
  2. Beschichtete Tablette nach Anspruch 1, wobei die Überzugspolymerformulierung in der zweiten Überzugsschicht mindestens etwa 2 mg bei einem 200-mg-Tablettenkern ausmacht und das Saxagliptin bzw. ein pharmazeutisch verträgliches Salz davon mindestens etwa 0,2 mg bei einer 200-mg-Tablette ausmacht.
  3. Beschichtete Tablette nach Anspruch 1, wobei das Polymer auf PVA-Basis in der zweiten Überzugsschicht das gleiche ist wie das Polymer auf PVA-Basis in den anderen Überzugsschichten.
  4. Beschichtete Tablette nach Anspruch 1, wobei das Polymer auf PVA-Basis aus Polyvinylalkohol besteht.
  5. Beschichtete Tablette nach Anspruch 1, wobei in der zweiten Überzugsschicht das Saxagliptin bzw. ein pharmazeutisch verträgliches Salz davon in einer Menge im Bereich von 0,2 bis 140 mg vorhanden ist und das Überzugspolymer in einer Menge im Bereich von 2 bis 140 mg vorhanden ist und sowohl in der inneren Versiegelungsüberzugsschicht als auch in der äußeren Schutzüberzugsschicht das Überzugspolymer in einer Menge im Bereich von etwa 1 bis etwa 100 mg vorhanden ist.
  6. Beschichtete Tablette nach Anspruch 1, wobei der Tablettenkern aus einem oder mehreren Füllstoffen, gegebenenfalls einem oder mehreren Bindemitteln, gegebenenfalls einem oder mehreren Sprengmitteln und gegebenenfalls einem oder mehreren Tablettierschmiermitteln besteht.
  7. Beschichtete Tablette nach Anspruch 6, wobei der Tablettenkern aus mikrokristalliner Cellulose, Lactose-monohydrat, Croscarmellose-Natrium und Magnesiumstearat besteht.
  8. Beschichtete Tablette nach Anspruch 1, wobei man anstelle des Tablettenkerns eine Ausgangstablette verwendet.
  9. Beschichtete Tablette nach Anspruch 8, wobei es sich bei der Ausgangstablette um eine andere Arzneimitteltablette handelt.
  10. Beschichtete Tablette nach Anspruch 1 mit der folgenden Zusammensetzung: Material Tablette Placebo Bereich Gew.-% für eine 200-mg-Placebotablette Lactose 20 bis 75%/40 bis 150 mg Mikrokristalline Cellulose 20 bis 75%/40 bis 150 mg Croscarmellose-Natrium 2 bis 10%/4 bis 20 mg Magnesiumstearat 0,2 bis 2%/0,4 bis 4 mg
    Erste innere Versiegelungsüberzugsschicht Gew.-% für eine 200-mg-Placebokerntablette Überzugspolymer und gegebenenfalls Weichmacher und gegebenenfalls Gleitmittel 1 bis 3%/2 bis 6 mg
    Zweite Überzugsschicht Gew.-% für eine 200-mg-Placebokerntablette Saxagliptin (freie Base oder HCl-Salz) 0,25 bis 70%/0,5 bis 140 mg Überzugspolymer und gegebenenfalls Weichmacher und gegebenenfalls Gleitmittel 1 bis 50%/2 bis 100 mg
    Dritte äußere Schutzüberzugsschicht Gew.-% für eine 200-mg-Placebokerntablette Überzugspolymer und gegebenenfalls Weichmacher, gegebenenfalls Gleitmittel und Farbstoff 1 bis 5%/2 bis 10 mg
  11. Beschichtete Tablette nach Anspruch 1, wobei, für eine Wirkungsstärke von 5 mg, die zweite Überzugsschicht aus 5 mg Saxagliptin oder einem pharmazeutisch verträglichen Salz davon und 5 mg Überzug auf Polymerbasis besteht und, für eine Wirkungsstärke von 2,5 mg, die Überzugsschicht aus 2,5 mg Saxagliptin oder einem pharmazeutisch verträglichen Salz davon und 20 mg Überzug auf Polymerbasis besteht.
  12. Verfahren zur Herstellung einer beschichteten Tablette nach Anspruch 1, bei dem man:
    a) einen Tablettenkern bereitstellt,
    b) die Tablette mit einer inneren Versiegelungsüberzugsschichtformulierung beschichtet, die mindestens ein Polymer auf PVA-Basis und einen Weichmacher umfasst,
    c) die beschichtete Tablette zur Bildung eines inneren Versiegelungsüberzugs darauf trocknet,
    d) die auf diese Weise beschichtete Tablette mit einer zweiten Überzugsschichtformulierung beschichtet, umfassend ein Medikament, bei dem es sich um Saxagliptin oder ein pharmazeutisch verträgliches Salz davon handelt, und mindestens ein Überzugspolymer auf PVA-Basis und einen Weichmacher,
    e) die auf diese Weise beschichtete Tablette unter Bildung einer zweiten Überzugsschicht darauf trocknet und
    f) die auf diese Weise beschichtete Tablette mit einer dritten äußeren Schutzüberzugsschichtformulierung beschichtet, die mindestens ein Überzugspolymer auf PVA-Basis und einen Weichmacher umfasst, und
    g) die auf diese Weise beschichtete Tablette unter Bildung der erfindungsgemäßen beschichteten Tablette trocknet.
  13. Verfahren nach Anspruch 12, bei dem man die Überzugsschichten als Suspension des Überzugspolymers auf PVA-Basis und eines Weichmachers aufträgt.
