Yh ·»* hvor R^ er hydrogen eller hydroxy, og R2 er hydrogen eller fluor, 5 farmaceutiske præparater, der omfatter en forbindelse med formlen I, og anvendelse af forbindelserne til fremstilling af sådanne præparater, der er nyttige til behandling af sygdomstilstande, der er karakteriseret ved metabolisk calciummangel, især osteoporosis og renal osteodystrophi, eller til behandling af neoplastiske sygdomme såsom 10 leukæmi.
Eksempler på de i det følgende nævnte C^.g-alkylgrupper er methyl, ethyl, propyl, isopropyl, t-butyl, hexyl, heptyl og octyl. Eksempler på de i det følgende nævnte arylgrupper er phenyl og phenyl, der er substitueret med Cj^. g-alkyl, fluor, chlor, brom, iod, nitro, cyano og 15 trifluormethyl.

Forbindelserne med formlen V er kendte eller kan fremstilles i overensstemmelse med kendte metoder, fx som beskrevet i J. Org. Chem. 51, 1986, 3098. Reaktionen udføres i nærværelse af en stærk base, fx en 10 alkyllithiumforbindelse eller et dialkyl- eller alkylsubstitueret di-silylamid, i et konventionelt etheropløsningsmiddel under en inert atmosfære o§ ved en temperatur i området fra ca. -80eC. til -50°C.

Alternativt kan afbeskyttelsen udføres ved behandling af en forbindelse med formlen III eller IV med en C^.g-alkanol eller med blandinger af vand og et 20 blandbart organisk opløsningsmiddel i nærværelse af en syre, fx en mineralsyre, en lavere alkansyre eller sulfonsyre, fortrinsvis hydrogenformen af en kationisk ionbytterharpiks.
En forbindelse med formlen Ilb (dvs. formlen II, hvor R2 er hydrogen) kan fremstilles ved at tosylere forbindelsen med formlen VI (Tetra-25 hedron 40, 1984, 2283) til forbindelsen med formlen VII, fx med p-toluensulfonylchlorid, i et basisk opløsningsmiddel såsom collidin eller pyridin ved ca. fra -10eC til 10°C, fortrinsvis 0°C, under en inert atmosfære såsom nitrogen.

Forbindelsen med formlen VII omdannes til forbindelsen med formlen VIII ved omsætning med ethylvinylether i et aprot opløsningsmiddel og i nærværelse af en syre, fx benzoesyre eller p-toluensulfonsyre, ved ca. fra -90°C til -60"C, fortrinsvis -70°C, under en inert atmosfære 5 såsom nitrogen.

Forbindelsen med formlen IX omdannes til forbindelsen med formlen X ved omsætning med en syre, fx p-toluensulfonsyre, i en lavere alka-nol, fx methanol, indledningsvis ved ca. fra -10°C til 10°C, for-15 trinsvis 0°C, og derefter ved ca. stuetemperatur.
Forbindelsen med formlen X oxideres til forbindelsen med formlen Ila ved omsætning med et oxidationsmiddel, fx pyridiniumchlorchromat, i et q _ g -alkylhalogenid-opløsningsmiddel, fx chloroform, carbontetra-chlorid eller dichlormethan, ved ca. fra -10°C til 30°C, fortrinsvis 20 stuetemperatur.
Ketonen med formlen Ila kan omdannes til ketonen med formlen Ilb ved behandling med et silyleringsmiddel såsom en tri(R5)-substitueret silylimidazol, hvor R5 er som defineret ovenfor, fx med trimethylsi-lylimidazol, i et inert organisk opløsningsmiddel såsom en ether 25 eller et halogeneret carbonhydrid såsom dichlormethan og under en inert atmosfære såsom argon.
En forbindelse med formlen Ild (dvs. formlen II, hvor R2 er F) kan fremstilles ved, at forbindelsen med formlen XI tosyleres til forbindelsen med formlen XII på en måde, der er analog med det ovenfor 30 beskrevne til omdannelse af forbindelsen VI til tosylatet VII.
Forbindelsen med formlen XIII fremstilles ved at omsætte n-butyl- DK 171966 B1 7 lithiumderivatet af (trimethylsilyl)acetylen med forbindelsen med formlen XII og opvarme den resulterende blanding ved tilbagesvaling.
Forbindelsen med formlen XIII omdannes til forbindelsen med formlen XIV ved omsætning med sølvnitrat efterfulgt af kaliumcyanid i et 5 vandigt alkanolisk opløsningsmiddel, fx ethanol og vand.
Forbindelsen med formlen XIV omdannes til forbindelsen med formlen XV ved omsætning med hexafluoracetonegas i nærværelse af et alkyllit-hium, fx butyllithium, i et etheropløsningsmiddel, fx THF, ved ca. fra -90eC til -70eC, fortrinsvis -75°C.
10 Forbindelsen med formlen XV omdannes til forbindelsen med formlen XVI ved omsætning med flussyre i acetonitril og THF.
Forbindelsen med formlen XVI oxideres til forbindelsen med formlen Ile ved omsætning med et oxidationsmiddel i et C^.g-alkylhalogenid-opløsningsmiddel (som beskrevet ovenfor til omdannelse af alkoholen X 15 til ketonen Ila) i nærværelse af vandfrit natriumacetat.
Forbindelsen med formlen Ile kan omdannes til forbindelsen med formlen Ild ved behandling med et silyleringsmiddel som beskrevet ovenfor til omdannelse af Ila til Ilb.
Stimuleringen af den intestinale calciumabsorption (ICA) og af knog-20 lecalciummobiliseringen (BCM) i rotte blev målt såvel som den kom-petitive binding (CB) til la,25-dihydroxycholecalciferol(i det følgende kaldt forbindelse X eller 1,25-(ΟΗ)£θ3)-tarmreceptorer i rotte, kylling og kalv. Dataene i tabel I indikerer, at sammenlignet med 1,25-'(OH)2D3 udviser analogerne A til C, der bærer C-23 tredobbeltbindingen, i almindelighed større intestinal calciumabsorption end knoglecalciummobilise-ringsaktiviteter, og at disse analoger binder til intestinale 1,25-20 (OH)2D3-receptorer.
Calcium- og phosphationkoncentrationerne og creatininkoncentrationen i serum samt legemsvægten og knoglemassen blev evalueret i rotter med vitamin D3-mangel, hvilke rotter blev behandlet med 1,25-(0H)2D3 eller med forbindelse C, samt i rotter, som blev holdt på en vitamin 25 D3-fyldt diæt (D+ kontrol) eller vitamin D3-utilstrækkelig diæt (D-kontrol). Resultaterne er vist i tabel II:

De antiproliferative (AP) og differentieringsinducerende (DI) virk-5 ninger af forbindelserne A til D på humane promyelocytiske HL-60 tumorceller blev evalueret. I tabel III er AP-virkningen angivet som procentvise reduktion af antal celler og som den koncentration ID50 af forbindelserne, hvilken koncentration reducerede celleantallet med 50%. Disse data viser, at forbindelserne med formlen I, særlig forbindelserne A og C, inhiberer celleproliferation og inducerer celledifferentiering og følgelig er nyttige som midler til behandling af neo-plastiske sygdomme såsom leukæmi.
25 Fra dansk patentansøgning 4571/76 og fra US 4521410 kendes beslægtede vitamin D3- derivater, som alene anvendes til de for vitamin D2 og D3-derivater kendte indikationer såsom osteoporosis og homeostase. Ingen anti-proliferativ og differentieringsinducerende virkninger er imidlertid tilskrevet de førnævnte kendte forbindelser. I modsætning 30 hertil kan forbindelserne ifølge opfindelsen ud over de hidtil kendte indikationer anvendes til behandling af neoplastiske sygdomme som fx leukæmi.
Endvidere har forbindelserne ifølge den foreliggende opfindelse sammenlignet med de kendte forbindelser en karakterisk tredobbelt-35 binding i 23,24-positionen.
Forbindelserne med formlen I kan administreres i doser i området fra ca. 0,1 eller 0,25 til 2 mg pr. dag til varmblodede dyr, som har be- DK 171966 B1 11 hov derfor, til behandling af sygdomstilstande, der er karakteriseret ved metabolisk calciummangel såsom renal osteodystrophi og, især, osteoporosis, eller til behandling af neoplastiske sygdomme såsom leukæmi. De kan formuleres i præparater såsom tabletter, kapsler 5 eller eliksirer til oral administration eller i sterile opløsninger eller suspensioner til parenteral, fx subkutan, intramuskulær, intravenøs eller intraperitoneal administration eller i topiske formuleringer. Ca. 0,1 eller 0,25 til ca. 2 mg af en forbindelse med formlen I kan sammensættes med en farmaceutisk acceptabel vehikel, bærer, 10 excipiens, bindemiddel, konserveringsmiddel, stabilisator eller smagsstof i en enhedsdosisform.
Eksempler på adjuvanser, som kan inkorporeres i kapsler, er bindemidler såsom tragantgummi eller gelatine; excipienser såsom dicalcium-phosphat; sprængmidler såsom majsstivelse; glittemidler såsom magne-15 siumstearat; sødemidler såsom sucrose eller saccharin; smagsgivende midler såsom pebermynte. Tabletter kan overtrækkes med schellak, sukker eller begge dele. En sirup eller eliksir kan indeholde et sødemiddel, methyl- og propylparabener som konserveringsmidler, et farvestof og et smagsgivende middel.
20 Sterile kompositioner til injektion kan formuleres ved at opløse eller suspendere den aktive substans i en vehikel såsom vand, en naturligt forekommende vegetabilsk olie såsom sesamolie eller en syntetisk fedtvehikel såsom ethyloleat. Buffere, konserveringsmidler og anti-oxidanter kan også inkorporeres.
25 EKSEMPEL 1 a) En blanding af 2,12 g (0,010 mol) [IR-[la,3a/3,4a,7aa] ]-octahydro- 4-hydroxy-/J, 7a-dimethyl-lH-inden-l-ethanol (J. Org. Chem. , 48, 1983, 1414), 2,10 g p-toluensulfonylchlorid og 9 ml tør pyridin omrøres ved 0eC i 3 timer under nitrogenatmosfære. Reaktionsblandingen hældes ud 30 i isvand og ekstraheres med methylenchlorid. Den organiske fase vaskes successivt med vand, 1 N svovlsyre og mættet vandigt natrium-hydrogencarbonat. Opløsningen tørres, filtreres og inddampes til tørhed, hvilket giver 3,70 g [IR-[la,3a/?,4a,7aa]]-octahydro-4-hy- DK 171966 B1 12 droxy-/3,7a-dimethyl-lH-inden-l-ethanol a-(4-methylbenzensulfonat), smp. 97-98°C, efter omkrystallisation fra methanol.
b) En blanding af 3,68 g (0,010 mol) af produktet fra a), 100 ml ethylvinylether og 0,04 g p-toluensulfonsyremonohydrat omrøres ved 5 -70°C i 1 time under nitrogenatmosfære og lades varme op til 0°C i 0,5 time. Blandingen behandles med 2 ml triethylamin og inddampes til tørhed. Remanensen opløses i methylenchlorid og vaskes med mættet vandigt natriumhydrogencarbonat. Den organiske fase tørres, filtreres og inddampes til tørhed, hvilket giver 4,60 g [IR- [ la, 3a/9,4a, 7aa] ] -4- 10 (1-ethoxyethoxy)octahydro-β,7a-dimethyl-IH-inden-1-ethanol 4-methyl- benzensulfonat, [ajjp + 31°C (c 1,2, CHCI3).
c) En blanding af 1,26 g af tetrahydropyranyletheren af 3-methyl-l-butyn-3-ol og 5,0 ml 1,5 M butyllithium i hexan og 30 ml dioxan omrøres ved 5°C i 0,5 time og ved stuetemperatur i 1 time under argon- 15 atmosfære. Derefter tilsættes 1,32 g (0,0030 mol) af produktet fra b), og blandingen opvarmes ved tilbagesvaling i 36 timer og afkøles. Blandingen hældes ud i vand og ekstraheres med ethylacetat. Den organiske fase vaskes med vand og saltvand og tørres over magnesiumsulfat. Blandingen filtreres og inddampes til tørhed. Remanensen op- 20 renses på silicagel med 4:1 hexan/ethylacetat, hvilket giver 1,43 g [IR-[la,3a/?,4a, 7aa]]-2-[[5-[4-(1-ethoxyethoxy)-octahydro-7a-methyl-1H-inden-1-yl]-1,1,5-trimethyl-2-pentynyl]oxy]-tetrahydro-2H-pyran, [a]g5 +36°C (c 1,0, CHCI3).
d) En blanding af 3,50 g (0,0073 mol) af produktet fra c), 50 ml 25 methanol og 0,10 g p-toluensulfonsyremonohydrat omrøres ved 0°C i 0,5 timer og ved 23eC i 18 timer under nitrogenatmosfære. Blandingen koncentreres derefter til 10 ml. Blandingen fortyndes med methylenchlorid og vaskes med mættet vandigt natriumhydrogencarbonat. De samlede vandige faser tilbageekstraheres derefter med methylenchlorid.
30 De forenede organiske faser tørres, filtreres og inddampes. Produktet oprenses på silicagel med 5:1 hexan/ethylacetat, hvilket giver 1,84 g [IR-[la, 3a/9,4a,7aa]]-octahydro-1-(5-hydroxy-1,5,5-trimethyl-3-penty-nyl)-7a-methyl-lH-inden-4-ol, smp. 62-63°C efter omkrystallisation fra ether/hexan.
DK 171966 B1 13 e) Til en suspension af 2,40 g pyridiniumchlorchromat i 50 ml methy-lenchlorid ved 0°C tilsættes 0,60 g (0,0022 mol) af produktet fra d) i 10 ml methylenchlorid. Blandingen omrøres ved 0°C i 0,5 time og ved 23®C i 1 time under nitrogenatmosfære. Blandingen fortyndes med ether 5 og omrøres i 10 minutter. Blandingen filtreres, filtret vaskes med ether, og de samlede filtrater inddampes. Den resulterende olie suspenderes i ether og filtreres, filtret vaskes med ether, og de samlede filtrater inddampes. Produktet oprenses på silicagel med 6:1 hexan/ethylacetat, hvilket giver 0,21 g [1R-[la,3ayØ,7aa]]-octahydro-1-[1,5,5-trimethyl-5-[(trimethylsilyl)oxy]-3-20 pentynyl]-7a-methyl-4H-inden-4-on.
g) Til en blanding af 0,32 g [3S-(1Z,3a,50)]-[2-[3,5-bis[[(1,1-dimet-hylethyl)dimethylsilyl)oxy]-2-methylencyclohexyliden]ethyl]diphenyl-phosphinoxid (J.A.C.S. 104, 1982, 2945) og 8 ml THF, der er afkølet til -78°C under argonatmosfære, tilsættes 0,32 ml 1,6 M n-butyllit- 25 hium i hexan. Blandingen opvarmes til 30 25eC og fortyndes med 30 ml af opløsningen af kaliumnatriumtartrat og kaliumhydrogencarbonat. Opløsningen ekstraheres med ethylacetat. De samlede organiske faser vaskes med vand og derefter med saltvand. Den organiske fase tørres, filtreres og inddampes. Produktet oprenses på silicagel med 19:1 hexan/ethylacetat, hvilket giver 0,13 g 35 (la,3/?,5Z,7E)-1,3-bis[[(1,1-dimethylethyl)dimethylsilyl]oxy]-25- DK 171966 B1 14 [(trimethylsilyl)oxy]-9,10-secocholesta-5,7,10(19)-trien-23-yn, [a]fp +37,8eC (c 0,52, CHC13).
h) En blanding af 0,12 g (0,00017 mol) af produktet fra g) og 8 ml 1% tetrabutylammoniumfluorid i THF omrøres i 18 timer under argonatmos-5 fære. Blandingen fortyndes med vand og ekstraheres med ethylacetat.
De samlede organiske faser vaskes med vand og med saltvand. Den organiske fase tørres, filtreres og inddampes. Produktet oprenses på silicagel med 2:1 hexan/ethylacetat, hvilket giver 0,048 g (lo,3β,5Z,7E)-9,10-secocholesta-5,7,10(19)-trien-23-yn-l,3,25-triol, 10 [a]g2 +22,8°C (c 0,21, CHCI3).
EKSEMPEL 2 a) En blanding af 2,00 g (6,12 mmol) af [IR-[lo(s*),3a£,4a,7aa]]- β,7a-dimethyl-4-[ [ (1,l-dimethyl)dimethylsilyl]oxy]octahydro-lH-inden-1-ethanol, 2,92 g p-toluensulfonylchlorid og 50 ml pyridin omrøres 15 ved 0°C i 19 timer under argonatmosfære. Efter tilsætning af knust is og fortynding med vand ekstraheres blandingen med methylenchlorid.
Den organiske fase vaskes med 1 N H2SO4, vand og mættet vandig NaHC03. Opløsningen tørres og inddampes til tørhed. Remanensen kroma-tograferes på silicagel med 1:8 ethylacetat/hexan, hvilket giver 2,81 20 g (96%) [IR-[ la(S*) , 3a/9,4a, 7aa] ]-/3,7a-dimethyl-4-[ [ (1,1-dimethylet- hyl)dimethylsilyl]oxy]octahydro-IH-inden-1- ethanol 4-methylbenzensul-fonat, [a]^ +34,1°C (c 0,92, CHCI3).
b) Til en opløsning af 4,96 ml (trimethylsilyl)acetylen i 34 ml dioxan ved +5°C tilsættes dråbevis 22,0 ml 1,6 M butyllithium i 25 hexan. Efter omrøring i 30 minutter ved +4°C og derefter ved 25°C i 1,5 time tilsættes dråbevis en opløsning af 2,81 g af produktet fra a) i 44 ml dioxan. Blandingen opvarmes til tilbagesvaling i 20 timer. Saltvand ved 0°C tilsættes, og blandingen ekstraheres med ether. Den organiske fase vaskes med saltvand, tørres og inddampes. Remanensen 30 kromatograferes på silicagel med hexan, hvilket giver 2,09 g (88%) [IR- [lo(R*),3a/9,4a,7aa]]-4-[[4-(1,1-dimethylethyl)-dimethylsilyl]-oxy]octahydro-7a-methyl-IH-inden-1-yl]-1-pentynyl]trimethylsilan, [a](55 +46,9°C (c 0,95, CHCI3) .
DK 171966 B1 15 c) Til en opløsning af 2,09 g af produktet fra b) i 11 ml ethanol tilsættes en opløsning på 2,31 g sølvnitrat i 20 ml 3:1 ethanol/vand. Blandingen omrøres ved 50*C i 30 minutter og afkøles derefter til 25eC. Derefter tilsættes en opløsning af 4,28 g kaliumcyanid i 11 ml 5 vand, og blandingen omrøres ved 25“C i 2 timer. Blandingen fortyndes med vand og ekstraheres med ether. Den organiske fase vaskes med vand, tørres og inddampes. Remanensen kromatograferes på silicagel med hexan, hvilket giver 1,63 g (95%) [IR-[la(R*) , 3a/3,4a, 7aa] ] - [ [oc-tahydro-7a-methyl-1-(1-methyl-3-butynyl)-IH-inden-4-yl]oxy]-(1,Ι-ΙΟ dimethylethyl)dimethylsilan, [a]fp +53,8°C (c 0,64, CHCI3).
d) Til en opløsning af 1,20 g af produktet fra c) i 40 ml THF ved -75“C tilsættes dråbevis 3,70 ml 1,6 M butyllithium i hexan. Efter omrøring i 30 minutter ved -75eC, ledes hexafluoracetonegas ind i reaktionsblandingen i 10 minutter. Blandingen omrøres ved -75°C i 25 15 minutter. Derefter tilsættes en 1:1 blanding af 1 M vandig kaliumtar-trat og 2 M vandig KHCO3 ved 0eC. Blandingen omrøres ved 25°C i 1 time og ekstraheres derefter med methylenchlorid. Den organiske fase vaskes med den samme saltblanding, tørres og inddampes. Remanensen kromatograferes på silicagel med 5% ethylacetat/hexan, hvilket giver 20 1,78 g (99%) [IR- [ la(R*) , 3a/9,4a, 7aa] ] - [ [ 1,1-bis (trif luorme thyl)-5- [[4-(1,l-dimethylethyl)dimethylsilyl]oxy]octahydro]-7a-methyl-lH-inden-l-yl]-2-pentyn-l-ol, [a]^5 +34,4eC (c 0,42, CHCI3).
e) Til en opløsning af 1,51 g af produktet fra d) i 17 ml acetonitril og 15 ml THF tilsættes 13,4 ml 48% HF. Blandingen omrøres ved 25eC i 25 1,5 time og fortyndes med vand. Blandingen ekstraheres med methylen chlorid. Den organiske fase vaskes med mættet vandig NaHC03, tørres og inddampes. Remanensen kromatograferes på silicagel med 1:3 ethylacetat/hexan, hvilket giver 1,16 g (99%) [IR- [la(R*) ,3a/J,4a,7aa] ]-oc-tahydro-1-[5-hydroxy-6 -trifluor-5 -(trifluormethyl)-1-methyl-3-hexy-30 nyl]-7a-methyl-lH-inden-4-ol, [a]^5 +29,0eC (c 0,57, CHCI3).
f) Til en opløsning af 0,200 g (0,518 mmol) af produktet fra e) i 8 ml tør methylenchlorid tilsættes 0,304 g natriumacetat og 0,610 g 2',2'-dipyridiniumchlorchromat. Blandingen omrøres ved 25eC i 2 timer. Derefter tilsættes 0,305 g 2',2'-dipyridiniumchlorchromat, og DK 171966 B1 16 blandingen omrøres i 110 minutter. Efter tilsætning af 1,1 ml 2-propanol fortyndes opløsningen med vand og ekstraheres med 1:1 ethyl-acetat/ether. Den organiske fase vaskes med vand, saltvand, tørres og inddampes til tørhed. Remanensen kromatograferes på silicagel med 5 1:1 ethylacetat/hexan, hvilket giver [IR-[la(R*),3a/9,7aa]]-octahydro- 1-[5-hydroxy-6-trifluor-5-(trifluormethyl)-1-methyl-3-hexynyl]-7a-methyl-4H-inden-4-on, [a]^3 +2,3°C (c 0,48, CHCI3).
g) Til en opløsning af 181 mg [3S-(lZ,3a,5/9)]-[2-[3,5-bis[[(l,l-dimethylethyl)dimethylsilyl]oxy]- 2-methylencyclohexyliden]ethyl]-10 diphenylphosphinoxid i 3,5 ml THF ved -75°C tilsættes 0,164 ml 1,6 M butyllithium i hexan. Efter omrøring tilsættes dråbevis en opløsning af 40 mg af produktet fra f) i 2,5 ml THF. Blandingen omrøres ved -75°C i 110 minutter. Efter tilsætning af 1:1 blanding af 1 M vandig kaliumnatriumtartrat og 2 M vandig KHCO3 ekstraheres blandingen med 15 ethylacetat. Den organiske fase vaskes med saltvand, tørres og inddampes. Remanensen kromatograferes på silicagel med 1:5 ethylacetat/-hexan, hvilket giver 65 mg (87%) (la, 3/?, 5Z, 7E)-1,3-bis[ [ (1,1-dimet-hylethyl)dimethylsilyl]oxy]-26,26,26,27,27,27-hexafluor-9,10-secocho-lesta-5,7,10(19)-trien-23-yn-25-ol, [a]^3 +38,8°C (c 0,17, CHCI3).
20 h) Til en opløsning af 60 mg af produktet fra g) i 3 ml THF tilsættes 0,58 ml 1 M tetrabutylammoniumfluorid i THF. Blandingen omrøres ved 25°C i 21 timer. Efter tilsætning af 2 ml af halvmættet vandig NaHC03 omrøres blandingen ved 25°C i 15 minutter og ekstraheres derefter med ethylacetat. Den organiske fase vaskes med halvmættet vandig NaHC03 25 og saltvand og tørres derefter.
