Description
- Verfahren zur Darstellung von Furancarbonsäureamiden Es wurde die überraschende Beobachtung gemacht, daB den bisher nicht bekannten Furandicarbonsäureamiden bzw. Furandicarbonsäureesteramiden bemerkenswerte phärmakologische Eigenschaften zukommen, die sie therapeutisch wertvoll machen. Diese neuen Verbindungen werden erhalten, indem man Furandicarbonsäuren bzw. Abkömmlinge dieser Verbindungen, wie z. B. ihre Metallsalze, Halogenide, Ester oder Anhydride, auf sekundäre Amine bzw. deren Salze oder auf Carbaminsäurehalogenide in An-- oder Abwesenheit von Lösungsmitteln, von Kondensationsmitteln sowie von säurebindenden Mittelneinwirken läßt. Als Kondensationsmittel verwendet man vorteilhaft P'hosphorhalogenide, Phosphorpentoxyd, Thionylchlorid sowie Phosgen usw.
- Die neuen Verbindungen zeichnen sich besonders durch ihre vorzüglichen analeptischen Wirkungen aus. Die bisher unbekannten Furandicarbonsäureamide bzw. Furandicarbonsäureesteramide sind zum Teil sehr gut in Wasser löslich, zum Teil löslich in Alkohol, Äther, Chloroform, Glykoläther oder Propylenglykol.
- Bleispiel i 44 Gewichtsteile z - Methylfuran - 5 - @essigsäure-3-carbonsä'uredichlorid (Kp.l,4 i3o°), dargestellt aus der entsprechenden Säure mit Thionylchlorid, löst man in Soo Volumteilen absolutem Äther und tropft unter Rühren und Kühlen bei o bis io- eine Lösung von 36 Gewichtsteilen Diäthyl.amin in 4o0 Voluniteilen absolutem Äther dazu. Ist alles Amin eingetropft, so läßt man die Temperatur auf Raumtemperatur steigen und erwärmt noch 2 Stunden auf 25@. Dann saugt man vom abgeschiedenen Diäthylaminchlorhydrat ab, um hierauf die ätherische Lösung mit gesättigter Pottaschelösung zu behandeln. Anschließend trocknet man über Natriumsulfat. Nach Verjagen des Äthers destilliert das 2-Methylfuran-5-essig-3-carl)onsäurebis-diäthylamid als schwach gelb gefärbtes Öl bei o,5mm und i85'. Es ist in jedem Verhältnis mit Wasser mischbar. Ausbeute 95% der Theorie.
- In gleicher Weise kann man darstellen 2-1lethylfuran-5-essig-3-carbonsäure-bis-dimethylamid Kp0,1 i78" F. 80° 2-Methylfuran-5-essig-3-carbonsäure-bis-dipiperidid F. 97° 2-Methylfuran-5-essig-3-carbonsäure-bis-N-methylanilid F. 124' 2-Methylfuran-5-essig-3-carbonsäure-bis-diallylamid Kplo-4 1400 2-Methylfuran-ä-essig-3-carbonsäure-bis-dipropylamid Kplo-4 14o bis 145' a-Methylfuran-5-essig-3-carbonsäure-bis-morpholid Kpo,o; i4o bis i5o' F. 158 bis 16o° 2-1lethylfuran-j-essig-3-c arbonsäure-bis-äthylallylamid 2-Nlethy lfuran-5-essig-3-carbonsäure-bis-aa-dimethylpiperidid 2-Methylfuran-ä-essig-3-carbonsäure-bis-a-methylpipe. idid 2-Methylfuran-5-essig-3-carbonsäure-bis-N-methylbenzylamid. Beispiel 2 22 Teile 2-Methylfuran-5-essig-3-carbonsäuredichlorid erhitzt man mit 2o Teilen; Diäthylaminchlorhydrat während 2 Stunden auf etwa 16o°. Die erhaltene Schmelze wird in Wasser gelöst, mit Alkalilauge versetzt und mit Äther ausgeschüttelt. Die ätherische Lösung wird getrocknet, eingedampft und durch Destillation gereinigt. Man erhält das gleiche Bis-Diäthylamid wie in Beispiel i. Ausbeute 8o% der Theorie.
- An Stelle des Hydrochlorids kann eben,sagut ein anderes Salz verwendet werden. Beispiel 3 15 Teile des Dinatriumsal.zes der 4.-Methylfuran - 2 - essig - 3 - carbonsäure werden mit q.o Teilen Diäthylcarbaminsäurechlorid auf etwa i 5o` erwärmt. Unter Kohlensäureentwicklung setzt stürmische Umsetzung zum Bis-Diäthylamid der 4-Methylfuran-2-essig-3-carbonsäure ein. Dieses wird in Äther aufgenommen, und mit Wasser gewaschen; nach dem Trocknen und Verdampfen des Lösungsmittels erhält man das Bis-Diäthylamid als eine fast farblose; bei 158 bis i 6o" (o,65 mm) siedende Flüssigkeit. Ausbeute 850;ö der Theorie.
- In gleicher Weise wie die q:-Methylfuran-2-essig-3-carbonsäure können auch die Furan-2 :essig-3-carbonsäure, die 4.-Äthy1furan-2-essig-3-carbonsäure oder die ¢, 5-Dimethylfuran-2 iessig-3-carbonsäure in die Bis-Diäthylamide übergeführt werden. Die Umsetzung verläuft in gleicher Weise, wenn man an Stelle des Natriumsalzes andere Salze, z. B. Calciumsalze, verwendet. Beispiel q.
- 2o Gewichtsteile 2 - Methylfuran - 5 - essigsäureäthylester-3-carbonsäurechl.orid (Kpl2 1,15 bis 1q.7°), dargestellt aus 2-1fIethylfuran-5-essigsäureäthylester-3-carbonsäure und Thionylchlorid, werden in Äther gelöst, mit der berechneten Menge Diäthylamin versetzt und während 2 Stunden auf 25° gehalten. Nach dem Filtrieren von ausgeschiedenem Diäthylaminchlorhydrat wird die ätherische Lösung mit Wasser gewaschen, hierauf getrocknet und dann der Äther verjagt. Der Rückstand, das 2 - Methylfuran - 5 - @essigsäureäthyl.ester-3-carbonsäurediäthylamid, bildet ein helles Öl vom Kpi 16o bis 162°. Ausbeute 9 i o o der Theorie.
- Beispiel 5 18 Teile 2, 5-Dimethylfuran-3, 4.-dicarbonsäurechlorid (Kp" 135 bis 137'), dargestellt aus 2, 5-Dimethylfuran-3, 4-dicarbonsäure und Thionylchlorid, werden in absolutem Äther gelöst und mit 24 Teilen Diäthylamin versetzt. Sogleich scheidet sich Diäthylaminchlorhydrat aus. Nach 2stündigem Stehen wird dieses abgesaugt, der Äther mit gesättigter Pottaschelösung gewaschen, dann mit Kaliumhydroxyd getrocknet und verdampft. Der Rückstand, das 2, 5-Dimethylfuran-3, .1-dicarbonsäure-bis-diäthylamid, siedet bei 15,1 bis 156° und o,¢ mm. Aus Petroläther bildet es große, durchsichtige Kristalle vom F.62°. Es ist klar wasserlöslich. Ausbeute 8q.oio der Theorie.
- An Stelle des Chlorids kann auch das Anhydrid verwendet werden. Beispiel 6 8 Teile 2, ¢-Furandicarbonsäurechlorid, dargestellt aus 2, q.-Furandicarbonsäure und Thionylchlorid, werden in absolutem Benzol gelöst und mit der berechneten Menge Diäthylamin versetzt. Man :erwärmt während einiger Stunden auf q.0° und filtriert nach einigem Stehen vom ausgeschiedenen Chlorhydrat ab. Die Benzollösung wird mit konzentrierter Alkalilauge behandelt und hierauf mit festem Kaliumhydroxyd getrocknet. Nach dem Verdampfen des Benzols verbleibt das 2, ¢-Furandicarbonsäure-bis-diäthylamid als wasserlösliche Substanz. Ausbeute 86% der Theorie.
- Auf gleiche Weise läßt sich das 2, 5-Furandicarbonsäure-bis-diäthylamid gewinnen. Beispiel 7 2o Teile 2-Methylfuran-5-,essig-3-ca.rbonsäurediphenylester, dargestellt aus 2-Methy1-furan-5-essig-3-carbonsäuredichlorid und Phenol durch Erwärmen in benzolischer Lösung, werden mit 5o Teilen Diäthylamin einige Stunden im Rohr auf i6o° erhitzt. Aus dem Umsetzungsgemisch gewinnt man Methronsäure-bis-diäthylamid in einer Ausbeute von 76% der Theorie wie in Beispiel i.
- Beispiel 8 2o Teile 2 - Methyl- 5 -,essig- 3 - carbonsäure werden in das Diäthylaminsal.z umgewandelt und unter Zusatz von 25 Teilen Phosphoroxychlorid auf etwa i 5o' erwärmt. Nach beendigter Umsetzung arbeitet man wie in Beispiel i beschrieben auf und erhält so Methronsäure-bis-diäthylamid. Ausbeute 72% der Theorie. An Stelle von Phosphoroxychl.orid verwendet man zweckmäßig auch . Phosphorpentoxyd.

- PATENTANSPRUCH: Verfahren zur Darstellung von Furandicarbonsäureamiden bzw. Furandicarbonsäureesteramiden, dadurch gekennzeichnet, daß man Furandicarbonsäuren bzw. Furandica.rbonsäureester oder ihre Substitutionsprodukte bzw. Abkömmlinge auf sekundäre Amine, ihre Salze oder Carbamin.säurehalogenide in An- oder Abwesenheit von Lösungsmitteln, von Kondensationsmitteln oder säurebindenden Mitteln einwirken läßt.
