Description
- Die am häufigsten verwendeten mikrobiellen Pestizide sind von dem Bakterium Bacillus thuringiensis abgeleitet. Dieses bakterielle Agens wird dazu verwendet, um ein breites Spektrum von blattfressenden Raupen und Käfern sowie Moskitos zu kontrollieren. Bacillus thuringiensis erzeugt einen proteinhaltigen Parasporen-Körper oder Kristall, der nach Aufnahme durch einen empfänglichen Insektenwirt toxisch ist. Beispielsweise erzeugt B. thuringiensis subsp. kurstaki HD-1 einen Kristalleinschluß, bestehend aus einem Biotoxin mit der Bezeichnung Delta-Toxin, der für die Larven einer Anzahl von Lepidoptera-Insekten toxisch ist. Die Klonierung, Sequenzierung und Expression dieses B.t.-Kristallprotein-Gens in Escherichia coli wurde von Schnepf et al., [1981] Proc. Natl. Acad. Sci. USA 78:2893-2897 beschrieben. US-A-4,448,885 und US-A-4,467,036 offenbaren beide die Expression des B.t.-Kristallproteins in E. coli.
- Die vorliegende Erfindung betrifft ein neues Bacillus thuringiensis- Isolat mit der Bezeichnung PS81RR1, welches Aktivität gegen alle getesteten Lepidoptera-Schädlinge aufweist.
- Ebenfalls offenbart und beansprucht ist ein neues Toxin-Gen, welches ein für Lepidoptera-Insekten toxisches Toxin exprimiert. Dieses Toxin-Gen kann mit Hilfe eines Plasmidvektors in geeignete Wirte überführt werden.
- Speziell umfaßt die Erfindung ein neues B.t.-Isolat mit der Bezeichnung B.t. PS81RR1, Mutanten davon und ein von diesem B.t.- Isolat abgeleitetes neuartiges Delta-Endotoxin-Gen, welches für ein Protein kodiert, das gegen Lepidoptera-Schädlinge aktiv ist. Konkreter kodiert das Gen in B.t. PS81RR1 fur ein 133,367 Dalton Endotoxin.
- Das neue Toxin-Gen der vorliegenden Erfindung wurde aus einem neuen Lepidoptera-aktiven B. thuringiensis (B.t.)-Isolat mit der Bezeichnung PS81RR1 erhalten.
- Kolonien-Morphologie -- große Kolonie, matte Oberfläche, typisch für B.t.

- Plasmidpräparationen -- Die Agarose-Gelelektrophorese von Plasmidpräparationen unterscheidet PS81RR1 von B.t. HD-1 und anderen B.t.-Isolaten.
- Alkali-lösliche Proteine -- B.t. PS81RR1 erzeugt ein 133,367 Dalton-Protein.
- Einzigartiges Toxin -- Das 133,367 Dalton-Toxin unterscheidet sich von jedem bisher identifizierten.
- Aktivität -- B.t. PS81RR1 tötet alle getesteten Lepidoptera (Trichoplusia ni, Spodoptera exigua und Plutella xylostella).
- Bioassay-Verfahren:
- -- Von einem Sporen- und Kristallpellet werden Verdünnungen hergestellt, mit USDA-Insektennahrung (Technisches Bulletin 1528, U.S. Department of Agriculture) gemischt und in kleine Plastiktabletts gegossen. Frisch geschlüpfte Spodoptera exigua- Larven werden auf die Nahrungsmischung gesetzt und bei 25ºC gehalten. Die Sterblichkeitsziffern werden nach 6 Tagen aufgezeichnet.
- -- Verdünnungen und Nahrung werden auf dieselbe Weise wie für den Spodoptera exigua-Bioassay hergestellt. Es werden Larven in der vierten Erscheinungsform eingesetzt und die Sterblichkeitsziffern nach 8 Tagen aufgezeichnet.
- B. thurinqiensis PS81RR1, NRRL B-18458, und Mutanten davon können unter Verwendung bekannter Standardmedien und Fermentationstechniken in Kultur gezüchtet werden. Nach Vollendung des Fermentationszyklus können die Bakterien geerntet werden, indem man zuerst die B.t.- Sporen und -Kristalle von der Fermentationsbrühe durch auf dem Gebiet wohlbekannte Mittel abtrennt. Die gewonnenen B.t.-Sporen und -Kristalle können als benetzbares Pulver, flüssiges Konzentrat, Granulat oder andere Formulierungen durch die Zugabe von oberflächenaktiven Mitteln, Dispergiermitteln, inerten Trägersubstanzen und anderen Komponenten formuliert werden, um die Handhabung und die Anwendung für bestimmte Zielgruppen-Schädlinge zu erleichtern. Die Formulierungs- und Anwendungsverfahren sind auf dem Gebiet alle wohlbekannt und werden bei im Handel erhältlichen Stämmen von B. thuringiensis (HD-1) mit Aktivität gegen Lepidoptera, z.B. Raupen, eingesetzt. B.t. PS81RR1 und Mutanten davon können zur Kontrolle von Lepidoptera-Schädlingen eingesetzt werden.
- Eine Subkultur von B.t. PS81RR1 und der E. coli-Wirt, der das Toxin-Gen der Erfindung beherbergt, wurden in der permanenten Sammlung des Northern Research Laboratory, U.S. Department of Agriculture, Peoria, Illinois, USA hinterlegt. Die Hinterlegungsnummern und Hinterlegungsdaten sind wie folgt:
- Die Toxin-Gene der vorliegenden Erfindung können in eine große Vielfalt von mikrobiellen Wirten eingeführt werden. Die Expression des Toxin-Gens resultiert direkt oder indirekt in der intrazellulären Produktion und Aufrechterhaltung des Pestizids. Bei geeigneten Wirten, z.B. Pseudomonas, können die Mikroorganismen auf dem Aufenthaltsort von Lepidoptera-Insekten aufgebracht werden, wo sie sich vermehren und von den Insekten aufgenommen werden. Das Ergebnis ist eine Kontrolle der unerwünschten Insekten. Als Alternative kann der Mikroorganismus, der das Toxin-Gen beherbergt, unter Bedingungen behandelt werden, die die Aktivität des in der Zelle erzeugten Proteins verlängern. Die behandelte Zelle kann dann in der Umwelt eines Zielschädlings oder von Zielschädlingen ausgebracht werden. Das resultierende Produkt behält die Toxizität des B. t.-Toxins.
- Wenn das B.t.-Toxin-Gen mittels eines geeigneten Vektors in einen mikrobiellen Wirt eingeführt wird und der Wirt in der Umwelt in lebendem Zustand ausgebracht wird, ist es essentiell, daß bestimmte Wirtsmikroorganismen verwendet werden. Es werden Mikroorganismen- Wirte ausgewählt, von denen bekannt ist, daß sie die "Phytosphäre" (Phylboberfläche, Phyllosphäre, Rhizosphäre und/oder Rhizooberfläche) einer oder mehrerer interessierenden Erntepflanze(n) bewohnen. Diese Mikroorganismen werden so ausgewählt, daß sie imstande sind, in der besonderen Umwelt (Erntepflanze und anderen Insekten-Habitaten) erfolgreich mit den Wildtyp-Mikroorganismen im Wettbewerb zu stehen, für stabile Aufrechterhaltung und Expression des Gens sorgen, welches das Polypeptid-Pestizid exprimiert, und wünschenswerterweise für verbesserten Schutz des Pestizids vor umweltbedingtem Abbau und Inaktivierung zu sorgen.
- Von einer großen Anzahl von Mikroorganismen ist bekannt, daß sie die Phylboberfläche (die Oberfläche der Pflanzenblätter) und/ oder die Rhizosphäre (den Boden in der Umgebung von Pflanzenwurzeln) einer großen Vielfalt von wichtigen Erntepflanzen bewohnen. Diese Mikroorganismen schließen Bakterien, Algen und Pilze ein. Von besonderem Interesse sind Mikroorganismen wie Bakterien, z.B. der Gattungen Bacillus, Pseudomonas, Erwinia, Serratia, Klebsiella, Xanthomonas, Streptomyces, Rhizobium, Rhodopseudomonas, Methylophilius, Agrobacterium, Acetobacter, Lactobacillus, Arthrobacter, Azotobacter, Leuconostoc, und Alcaligenes; Pilze, insbesondere Hefe, z.B. der Gattungen Saccharomyces, Cryptococcus, Kluyveromyces, Sporobolomyces, Rhodotorula und Aureobasidium. Von besonderem Interesse sind solche Bakterienspezies der Phytosphäre wie Pseudomonas syringae, Pseudomonas fluorescens, Serratia marcescens, Acetobacter xylinum, Agrobacterium tumefaciens, Rhodopseudomonas spheroides, Xanthomonas campestris, Rhizobium melioti, Alcaligenes entrophus und Azotobacter vinlandii; und Hefespezies der Phytosphäre wie Rhodotorula rubra, R. glutinis, R. marina, R. aurantiaca, Cryptococcus albidus, C. diffluens, C. laurentii, Saccharomyces rosei, S. pretoriensis, S. cerevisiae, Sporobolomyces roseus, S. odorus, Kluvveromyces veronae und Aureobasidium pollulans. Von besonderem Interesse sind die pigmentierten Mikroorganismen.
- Eine große Vielfalt von Wegen sind zur Einführung eines B. t.- Gens, das ein Toxin exprimiert, in den Mikroorganismus-Wirt unter Bedingungen verfügbar, die die stabile Aufrechterhaltung und Expression des Gens erlauben. Es ist möglich DNA-Konstrukte bereitzustellen, welche einschließen die transkriptionalen und translationalen Regulationssignale zur Expression des Toxin-Gens, das Toxin-Gen unter deren regulatorischer Kontrolle und eine DNA-Sequenz, die mit einer Sequenz im Wirtsorganismus homolog ist, wodurch Integration eintreten wird, und/oder ein Replikationssystem, das in dem Wirt funktionell ist, wodurch Integration oder stabile Aufrechterhaltung eintreten wird.
- Die transkriptionalen Initiationssignale werden einen Promotor und eine transkriptionale Initiationsstartstelle einschließen. In einigen Fällen mag es wünschenswert sein, für eine regulative Expression des Toxins zu sorgen, wodurch die Expression des Toxins nur nach der Freisetzung in die Umwelt eintreten wird. Dies kann mit Operatoren erreicht werden oder einem Bereich, der an einen Aktivator oder an "Enhancer" bindet, die nach einer Änderung in der physikalischen oder chemischen Umwelt der Mikroorganismen zur Induktion imstande sind. Beispielsweise kann ein temperaturempfindlicher regulatorischer Bereich eingesetzt werden, wobei die Organismen im Labor ohne Expression eines Toxins gezüchtet werden können, aber die Expression nach Freisetzung in die Umwelt beginnen würde. Andere Techniken können ein spezielles Nährmedium im Labor verwenden, welches die Expression des Toxins verhindert, während das Nährmedium in der Umwelt die Expression des Toxins erlauben würde. Zur translationalen Initiation wird eine ribosomale Bindungsstelle und ein Initiationscodon vorhanden sein.
- Zur Erhöhung der Expression der "Messenger"-RNA können verschiedene Manipulationen durchgeführt werden, insbesondere die Verwendung eines aktiven Promotors ebenso wie der Einsatz von Sequenzen, welche die Stabilität der "Messenger"-RNA erhöhen. Der transkriptionale und translationale Terminationsbereich wird ein Stopcodon oder Stopcodons einschließen, einen Terminationsbereich und gegebenenfalls ein Polyadenylierungssignal. Eine hydrophobe "Leader"- Sequenz kann am Aminoterminus der translatierten Polypeptidsequenz eingesetzt werden, um die Sekretion des Proteins durch die innere Membran zu fördern.
- In Transkriptionsrichtung, nämlich in 5'- zu 3'-Richtung der kodierenden oder "Sense"-Sequenz, wird das Konstrukt den transkriptionalen regulatorischen Bereich, falls vorhanden, und den Promotor, wobei der regulatorische Bereich entweder 5' oder 3' vom Promotor gelegen sein kann, die ribosomale Bindungsstelle, das Initiationscodon, das Strukturgen mit einem offenen Leserahmen in Phase mit dem Initiationscodon, das oder die Stopcodons, die Polyadenylierungs-Signalsequenz, falls vorhanden, und den Terminationsbereich einschließen. Diese Sequenz kann als Doppelstrang für sich allein zur Transformation eines Mikroorganismen-Wirts verwendet werden, wird aber üblicherweise mit einer DNA-Sequenz, die einen Marker beinhaltet, eingeschlossen sein, wobei während der Einführung der DNA in den Wirt die zweite DNA-Sequenz mit dem Toxin-Expressionskonstrukt verknüpft sein kann.
- Durch einen Marker ist ein Strukturgen beabsichtigt, das die Selektion derjenigen Wirte, die modifiziert oder transformiert worden sind, ermöglicht. Der Marker wird normalerweise einen Selektionsvorteil bieten, beispielsweise Resistenz gegen ein Biozid, z.B. Resistenz gegenüber Antibiotika oder Schwermetallen; Komplementierung, z.B. um einem auxotrophen Wirt Prototrophie zu verleihen, oder dgl. Vorzugsweise wird Komplementierung eingesetzt, so daß der modifizierte Wirt nicht nur selektioniert werden kann, sondern auch im Freiland wettbewerbsfähig ist. Es können einer oder mehrere Marker bei der Entwicklung der Konstrukte ebenso wie zur Modifizierung des Wirts eingesetzt werden. Die Organismen können weiter modifiziert sein, indem für einen Wettbewerbsvorteil gegenüber anderen Wildtyp-Mikroorganismen im Freiland gesorgt wird. Beispielsweise können Gene, die Metall-chelierende Agentien, z.B. Siderophore, exprimieren, zusammen mit dem Strukturgen, welches das Toxin exprimiert, in den Wirt eingeführt werden. Auf diese Weise kann die erhöhte Expression eines Siderophors dem Toxin-erzeugenden Wirt einen Wettbewerbsvorteil bieten, so daß er effektiv mit den Wildtyp-Mikroorganismen in Wettbewerb stehen und stabil eine Nische in der Umwelt besetzen kann.
- Wenn kein funktionales Replikationssystem vorhanden ist, wird das Konstrukt auch eine Sequenz von mindestens 50 Basenpaaren (bp), vorzugsweise mindestens etwa 100 bp, und üblicherweise nicht mehr als etwa 1000 bp einer Sequenz, die mit einer Sequenz im Wirt homolog ist, einschließen. Auf diese Weise wird die Wahrscheinlichkeit legitimer Rekombination erhöht, so daß das Gen in den Wirt integriert und durch den Wirt stabil aufrechterhalten werden wird. Wünschenswerterweise wird sich das Toxin-Gen in enger Nachbarschaft zu dem Gen befinden, das für die Komplementierung sorgt, ebenso wie das Gen, das den Wettbewerbsvorteil bietet. Deshalb wird, falls das Toxin-Gen verlorengeht, der resultierende Organismus wahrscheinlich auch das komplementierende Gen und/oder das Gen, das den Wettbewerbsvorteil bietet, verlieren, so daß er nicht mehr in der Lage sein wird, in der Umwelt mit dem Gen, welches das intakte Kontrukt enthält, in Wettbewerb zu stehen.
- Eine große Anzahl von transkriptionalen regulatorischen Bereichen sind aus einer großen Vielfalt von Mikroorganismen-Wirten wie Bakterien, Bakteriophagen, Cyanobakterien, Algen, Pilzen und dgl. verfügbar. Verschiedene transkriptionale regulatorische Bereiche schließen die Bereiche ein, die mit dem trp-Gen lac-Gen, gal-Gen, den linken und rechten Lambda-Promotoren, dem Tac-Promotor, den in der Natur mit dem Toxin-Gen assoziierten Promotoren, wenn diese im Wirt funktionell sind, assoziiert sind. Siehe beispielsweise die US-Patente Nr. 4,332,898, 4,342,832 und 4,356,270. Der Terminationsbereich kann derjenige Terminationsbereich sein, der normalerweise mit dem transkriptionalen Initiationsbereich assoziiert ist, oder ein anderer transkriptionaler Initiationsbereich, solange die beiden Bereiche kompatibel und im Wirt funktionell sind.
- Wenn eine stabile episomale Aufrechterhaltung oder Integration gewünscht ist, wird ein Plasmid eingesetzt werden, welches ein im Wirt funktionelles Replikationssystem aufweist. Das Replikationssystem kann von dem Chromosom, einem episomalen Element, das normalerweise in diesem Wirt oder einem anderen Wirt vorhanden ist, oder einem Replikationssystem eines Virus, der im Wirt stabil ist, abgeleitet sein. Es sind eine große Anzahl von Plasmiden verfügbar, z.B. pBR322, pACYC184, RSF1010, pRO1614 und dgl. Siehe beispielsweise Olson et al. (1982) J. Bacteriol. 150:6069, und Bagdasarian et al. (1981) Gene 16:237, und die US-Patente Nr. 4,356,270, 4,362,817 und 4,371,625.
- Das B.t.-Gen kann zwischen dem transkriptionalen und translationalen Initiationsbereich und dem transkriptionalen und translationalen Terrninationsbereich eingeführt werden, um so unter der regulatorischen Kontrolle des Initiationsbereichs zu stehen. Dieses Konstrukt wird in einem Plasmid enthalten sein, das mindestens ein Replikationssystem beinhalten wird, aber mehr als eines beinhalten kann, wenn ein Replikationssystem zur Klonierung während der Entwicklung des Plasmids eingesetzt wird und das zweite Replikationssystem zur Funktion im endgültigen Wirt nötig ist. Zusätzlich können ein oder mehrere Marker vorhanden sein, die oben beschrieben worden sind. Wenn Integration gewünscht ist, wird das Plasmid wünschenswerterweise eine Sequenz einschließen, die mit dem Wirts- Genom homolog ist.
- Die Transformanten können nach bekannten Verfahren isoliert werden, üblicherweise unter Verwendung einer Selektionstechnik, die die Selektion des gewünschten Organismus gegenüber unmodifizierten Organismen oder übertragenden Organismen, falls diese anwesend sind, ermöglicht. Die Transformanten können dann auf Pestizid- Aktivität getestet werden.
- Geeignete Wirtszellen, wobei die Pestizid-enthaltenden Zellen behandelt werden, um die Aktivität des Toxins in der Zelle zu verlängern, wenn die dann behandelte Zelle in der Umwelt des Zielschädlings oder der Zielschädlinge ausgebracht wird, können Prokaryoten oder Eukaryoten einschließen und sind normalerweise auf solche Zellen beschränkt, die keine Substanzen erzeugen, die für höhere Organismen wie Säugetiere toxisch sind. Es könnten jedoch Organismen, die Substanzen erzeugen, welche für höhere Organismen toxisch sind, eingesetzt werden, bei denen das Toxin instabil ist oder das Niveau der Anwendung ausreichend niedrig, um jede Möglichkeit der Toxizität für einen Säugerwirt zu vermeiden. Als Wirte werden von besonderem Interesse die Prokaryoten und die niederen Eukaryoten, z.B. Pilze, sein. Beispielhafte Prokaryoten, sowohl Gram-negative als auch -positive, schließen Enterobacteriaceae, wie Escherichia, Erwinia, Shigella, Salmonella und Proteus; Bacillaceae; Rhizobiceae, wie Rhizobium; Spirillaceae, wie Photobaktenum, Zymomonas, Serratia, Aeromonas, Vibrio, Desulfovibrio, Spirillum; Lactobacillaceae; Pseudomonadaceae, wie Pseudomonas und Acetobacter; Azotobacteraceae, Actinomycetales und Nitrobacteraceae ein. Unter den Eukaryoten sind Pilze wie Phycomycetes und Ascomycetes, die Hefe, z.B. Saccharomyces und Schizosaccharomyces, einschließen; und Basidiomycetes-Hefe, wie Rhodotorula, Aureobasidium, Sporobolomyces und dgl.
- Eigenschaften, die für die Auswahl einer Wirtszelle für Produktionszwecke von besonderem Interesse sind, schließen die Leichtigkeit, mit der das B.t.-Gen in den Wirt eingeführt werden kann, die Verfügbarkeit von Expressionssystemen, die Effizienz der Expression, die Stabilität des Pestizids im Wirt und das Vorhandensein unterstützender genetischer Fähigkeiten ein. Eigenschaften, die für die Verwendung als Pestizid-Mikrokapsel von Interesse sind, schließen Schutzeigenschaften für das Pestizid wie dicke Zellwände, Pigmentierung und intrazelluläre Packung oder Bildung von Einschlußkörpern; Blattaffinität; mangelnde Toxizität gegenüber Säugern, Attraktivität für die Schädlinge zur Aufnahme; Leichtigkeit des Abtötens und Fixierens ohne Beschädigung des Toxins und dgl. ein. Andere Gesichtspunkte beinhalten die Leichtigkeit der Formulierung und der Handhabung, wirtschaftliche Gesichtspunkte, Lagerungsbeständigkeit und dgl.
- Wirtsorganismen von besonderem Interesse schließen Hefe, z.B. Rhodotorula sp., Aureobasidium sp., Saccharomyces sp. und Sporobolomyces sp.; phylloplane Organismen wie Pseudomonas sp., Erwinia sp. und Flavobacterium sp.; oder andere Organismen wie Escherichia, Lactobacillus sp., Bacillus sp., Stredtomyces sp. und dgl. ein. Spezielle Organismen schließen Pseudomonas aeruginosa, Pseudomonas fluorescens, Saccharomyces cerevisiae, Bacillus thuringiensis, Escherichia coli, Bacillus subtilis, Streptomyces lividans und dgl. ein.
- Die Zelle wird üblicherweise intakt sein und im wesentlichen in der Vermehrungsform vorliegen, wenn sie behandelt ist, eher als in der Sporenf orm, obwohl in einigen Fällen Sporen eingesetzt werden können.
- Die Behandlung der mikrobiellen Zelle, z.B. ein Mikroorganismus, der das B.t. Toxin-Gen enthält, kann auf chemische oder physikalische Weise oder einer Kombination von chemischen und/oder physikalischen Mitteln erfolgen, solange sich die Methode nicht schädlich auf die Eigenschaften des Toxins auswirkt oder die zelluläre Fähigkeit zum Schutz des Toxins nicht verringert. Beispiele chemischer Reagenzien sind Halogenierungsmittel, insbesondere Halogene der Atomzahl Nr. 17-80. Spezieller kann Iod unter milden Bedingungen und für eine ausreichende Zeit, um die gewünschten Resultate zu erreichen, eingesetzt werden. Andere geeignete Techniken beinhalten die Behandlung mit Aldehyden, z.B. Formaldehyd und Glutaraldehyd; Anti- Infektionsmitteln, z .B. Zephiranchlorid und Cetylpyridiniumchlorid; Alkoholen wie Isopropanol und Ethanol; verschiedenen histologischen Fixativen, z.B. Lugol-Iod, Bouin's Fixativ und Helly's Fixativ (siehe: Humason, Gretchen L., Animal Tissue Techniques, W.H. Freeman and Company, 1967); oder eine Kombination von physikalischen (Wärme) und chemischen Agenzien, die die Aktivität des in der Zelle erzeugten Toxins erhalten und verlängern, wenn die Zelle dem Wirtstier verabreicht wird. Beispiele physikalischer Mittel sind Strahlung kurzer Wellenlänge, z.B. Gamma-Strahlung und Röntgenstrahlung, Einfrieren, UV-Bestrahlung, Lyophilisierung und dgl.
- Die Zellen werden im allgemeinen erhöhte Strukturstabilität aufweisen, welche die Resistenz gegenüber Umweltbedingungen erhöht. Wenn das Pestizid in einer Proform vorliegt, sollte das Verfahren zur Inaktivierung so gewählt werden, daß das Processing der Proform zur reifen Form des Pestizids durch das Zielschädlingspathogen nicht behindert wird. Beispielsweise wird Formaldehyd Proteine vernetzen und könnte das Processing der Proform eines Polypeptid- Pestizids verhindern. Das Verfahren zur Inaktivierung oder Abtötung hält mindestens einen wesentlichen Teil der Bioverfügbarkeit oder Bioaktivität des Toxins aufrecht.
- Der zelluläre Wirt, der das insektizide B. t.-Gen enthält, kann in jedem geeigneten Nährmedium gezüchtet werden, worin das DNA- Konstrukt einen Selektionsvorteil bietet, vorausgesetzt, es ist ein selektives Medium, so daß im wesentlichen alle oder alle Zellen das B. t.-Gen enthalten. Diese Zellen können dann nach üblichen Verfahren geerntet werden. Alternativ können die Zellen vor der Ernte behandelt werden.
- Die B. t.-Zellen können auf vielfältige Weise formuliert werden. Sie können als benetzbare Pulver, Granulat oder Staub, durch Mischen mit verschiedenen inerten Materialien, z.B. anorganischen Mineralien (Phyllosilikaten, Carbonaten, Sulfaten, Phosphaten und dgl.) oder botanischen Materialien (gepulverten Maiskolben, Reishülsen, Walnußschalen und dgl.) eingesetzt werden. Die Formulierungen können Streu-Haft-Adjuvantien, Stabilisierungsmittel, andere pestizide Additive oder oberflächenaktive Mittel beinhalten. Flüssige Formulierungen können auf wäßriger oder nicht-wäßriger Basis sein und als Schäume, Gele, Suspensionen, emulgierbare Konzentrate oder dgl. verwendet werden. Die Bestandteile können rheologische Agenzien, oberflächenaktive Mittel, Emulgatoren, Dispergiermittel oder Polymere beinhalten.
- Die Pestizid-Konzentration wird in großem Umfang variieren, abhängig von der Natur der besonderen Formulierung, insbesondere davon, ob es ein Konzentrat ist oder zur direkten Anwendung bestimmt ist. Das Pestizid wird in mindestens 1 Gew.-% vorhanden sein und kann 100 Gew.-% betragen. Die trockenen Formulierungen werden 1-95 Gew.-% des Pestizids enthalten, während die flüssigen Formulierungen im allgemeinen etwa 1-60 Gew.-% der Feststoffe in der flüssigen Phase enthalten werden. Die Formulierungen werden im allgemeinen etwa 10² bis etwa 10&sup4; Zellen/mg enthalten. Diese Formulierungen werden bei etwa 50 mg (flüssig oder trocken) bis 1 kg oder mehr pro Hektar ausgebracht werden.
- Die Formulierungen können in der Umwelt des oder der Lepidoptera- Schädlinge(s), z.B. Pflanzen, Boden oder Wasser, durch Sprühen, Bestäuben, Besprengen oder dgl. ausgebracht werden.
- Mutanten von PS81RR1 können nach auf dem Gebiet wohlbekannten Verfahren hergestellt werden. Beispielsweise kann eine sporenlose Mutante durch Ethylmethansulfonat (EMS)-Mutagenese von PS81RR1 erhalten werden. Die Mutanten können unter Verwendung von UV-Licht und Nitrosoguanidin nach auf dem Gebiet wohlbekannten Verfahren hergestellt werden.
- Ein kleinerer Prozentsatz der sporenlosen Mutanten wird intakt bleiben und über längere Fermentationszeiträume nicht lysieren; diese Stämme werden als Lysis-minus bezeichnet (-). Lysis-minus- Stämme können identifiziert werden, indem sporenlose Mutanten in Schüttelkolbenmedien gescreent und diejenigen Mutanten ausgewählt werden, die bei Fermentationsende noch intakt sind und Toxinkristalle enthalten. Lysis-minus-Stämme sind für ein Zellfixierungsverf ahren geeignet, das ein geschütztes verkapseltes Toxinprotein ergeben wird.
- Zur Herstellung einer Phagen-resistenten Variante der genannten sporenlosen Mutante wird ein Aliquot des Phagenlysats auf Nähragar ausgebreitet und trocknengelassen. Ein Aliquot des Phagen-sensitiven Bakterienstammes wird dann direkt über dem getrockneten Lysat ausplattiert und trocknengelassen. Die Platten werden bei 30ºC inkubiert. Die Platten werden 2 Tage lang inkubiert und zu diesem Zeitpunkt ist das Wachstum von zahlreichen Kolonien auf dem Agar zu sehen. Einige dieser Kolonien werden gepickt und auf Nähragar- Platten subkultiviert. Diese anscheinend resistenten Kulturen werden auf Resistenz durch Überkreuz-Ausstreichen mit dem Phagenlysat getestet. Eine Linie des Phagenlysats wird auf der Platte ausgestrichen und trocknengelassen. Die mutmaßlich resistenten Kulturen werden dann quer über die Phagenlinie ausgestrichen. Resistente Bakterienkulturen zeigen nach einer Inkubation über Nacht bei 30º0 im Ausstrich quer über die Phagenlinie nirgends Lysis. Die Phagenresistenz wird dann bestätigt, indem ein Rasen der resistenten Kultur auf eine Nähragar-Platte ausplattiert wird. Der sensitive Stamm wird auf dieselbe Weise ebenfalls ausplattiert, um als positive Kontrolle zu dienen. Nach dem Trocknen wird ein Tropfen des Phagenlysats im Zentrum der Platte ausplattiert und trocknengelassen. Resistente Kulturen zeigten nach 24-stündiger Inkubation bei 30ºC keine Lysis in dem Gebiet, wo das Phagenlysat plaziert worden war.
- Es folgen Beispiele, welche Verfahren, einschließlich der besten Ausführungsform, zur Ausführung der Erfindung erläutern. Diese Beispiele sind nicht als beschränkend anzusehen. Alle Prozentangaben sind auf das Gewicht bezogen und alle Lösungsmittel-Mischungsverhältnisse sind auf das Volumen bezogen, wenn nicht anders angegeben.
- Eine Subkultur von PS81RR1 oder Mutanten davon kann dazu verwendet werden, um das folgende Medium, ein Medium mit Pepton, Glucose und Salzen, anzuimpfen.

- Die Salzlösung und die CaCl&sub2;-Lösung werden Filter-sterilisiert und zur autoklavierten und gekochten Brühe zum Animpfungszeitpunkt zugegeben. Die Kolben werden bei 30ºC auf einem Rotationsschüttler bei 200 Upm 64 h lang inkubiert.
- Das obige Verfahren kann ohne weiteres nach auf dem Gebiet wohlbekannten Verfahren auf große Fermentoren maßstäblich übertragen werden.
- Die B. t.-Sporen und/oder -Kristalle, die bei der obigen Fermentation erhalten werden, können nach auf dem Gebiet wohlbekannten Verfahren isoliert werden. Ein häufig verwendetes Verfahren besteht darin, die geerntete Fermentationsbrühe Trennungstechniken, z.B. Zentrifugation, zu unterwerfen.
- Gesamtzell-DNA wurde hergestellt aus B.t.-Zellen, die bis zu einer niedrigen optischen Dichte (OD&sub6;&sub0;&sub0; = 1,0) gezüchtet worden waren. Die Zellen wurden durch Zentrifugation gewonnen und in TES- Puffer (30 mM Tris-Cl, 10 mM Ethylendiamintetraessigsäure, 50 MM NaCl, pH = 8,0), der 20% Sucrose und 50 mg/ml Lysozym enthielt, in Protoplasten überführt. Die Protoplasten wurden durch Zugabe von Natriumdodecylsulfat (SDS) bis zu einer Endkonzentration von 4% lysiert. Das Zellmaterial wurde über Nacht bei 4ºC in 100 mM (Endkonzentration) neutralem Kaliumchlorid gefällt. Der Überstand wurde zweimal mit Phenol/Chloroform (1:1) extrahiert. Die DNA wurde mit Ethanol gefällt und durch isopyknische Bandenkonzentration auf einem Cäsiumchlorid-Gradienten gereinigt.
- Gesamtzell-DNA aus PS81RR1 und B.t.k. HD-1 wurde mit EcoRI verdaut und durch Elektrophorese auf einem TAE-gepufferten 0,8% Agarosegel aufgetrennt. Ein "Southern-Blot" des Gels wurde mit dem NsiI- bis NsiI-Fragment des im Plasmid pM3,130-7 von NRRL B-18332 enthaltenen Toxin-Gens und dem NsiI- bis KpnI-Fragment des "4,5 Kb-Klasse"-Toxin-Gens (Kronstad und Whitely [1986] Gene USA 43:29-40) sondiert. Diese beiden Fragmente wurden kombiniert und als Sonde verwendet. Die Ergebnisse zeigen, daß die hybridisierenden Fragmente von PS81RR1 von denjenigen aus HD-1 verschieden sind. Insbesondere wurde statt der hybridisierenden Banden von 3,8 Kb und 1,8 Kb, die bei HD-1 zu sehen waren, eine hybridisierende Bande von 2,3 Kb in PS81RR1 nachgewiesen.
- 200 µg PS81RR1-Gesamtzell-DNA wurden mit EcoRI verdaut und durch Elektrophorese auf einem präparativen 0,8% Agarose-TAE-Gel aufgetrennt. Der 2,2 Kb bis 2,4 Kb-Bereich des Gels wurde ausgeschnitten und die DNA daraus elektroeluiert und mittels einer ELUTIP -d- Ionenaustauschersäule (Schleicher und Schuell, Keene, NH) konzentriert. Die isolierten EcoRI-Fragmente wurden mit LAMBDA ZAP EcoRI-Armen ligiert (Stratagene Cloning Systems, La Jolla, CA) und unter Verwendung von Gigapak GOLD -Extrakten (Stratagene) verpackt. Die verpackten rekombinanten Phagen wurde mit dem E. coli-Stamm BB4 (Stratagene) ausplattiert, um eine hohe Plaque-Dichte zu ergeben.
- Die Plaques wurden nach Standard-Nukleinsäurehybridisierungsverfahren mit radioaktiv markierter Sonde gescreent. Die hybridisierenden Plaques wurden gereinigt und bei einer niedrigeren Plaque- Dichte erneut gescreent. Die resultierenden gereinigten Phagen wurden mit dem R408 M13-Helferphagen (Stratagene) gezüchtet und das rekombinante Bluescript -Plasmid (Stratagene) automatisch ausgeschnitten und verpackt. Mit dem "Phagemid" wurden XL1-Blue E. coli- Zellen (Stratagene) als Teil des automatischen Ausschneidungsverfahrens reinfiziert. Die infizierten XL1-Blue-Zellen wurden auf Arnpicillin-Resistenz gescreent und die resultierenden Kolonien durch ein Standard-Plasmid-Schnellanreicherungsverfahren analysiert, um das gewünschte Plasmid aufzufinden. Das Plasmid mit der Bezeichnung pM3,31-3 enthält ein etwa 2,3 Kb langes EcoRI-Insert und wurde unter Verwendung von Stratagene's T7- und T3-Primern plus einem Set von existierenden B. t.-Endotoxin-Oligonukleotidprimern sequenziert. Etwa 600 bp des Toxin-Gens wurden sequenziert und die Analyse der Daten zeigte im Vergleich von PS81RR1 zu anderen klonierten B.t.- Endotoxin-Genen, daß die PS81RR1-Sequenz einzigartig war. Ein synthetisches Oligonukleotid (CGTGGATATGGTGAATCTTATG) wurde für einen der Bereiche in der PS81RR1-Sequenz konstruiert, der bezüglich anderer existierender B.t.-Endotoxin-Gene die geringste Homologie aufwies.
- PS81RR1-Gesamtzell-DNA, partiell mit Sau3A verdaut und durch Elektrophorese auf einem 0,6% Agarose-TAE-Gel in eine Mischung von 9-23 Kb-Fragmenten fraktioniert, wurde in Lambda GEM -11 (PROMEGA) ligiert. Die verpackten Phagen wurden auf P2392 E. coli-Zellen (Stratagene) bei einem hohen Titer ausplattiert und unter Verwendung des (obigen) radioaktiv markierten synthetischen Oligonukleotids als Nukleinsäure-Hybridisierungssonde gescreent. Hybridisierende Plaques wurden bei einer niedrigeren Plaque-Dichte erneut gescreent. Ein einzelner gereinigter hybridisierender Plaque wurde dazu verwendet, um P2392 E. coli-Zellen in Flüssigkultur zur Herstellung von Phagen für die DNA-Isolierung zu infizieren. DNA wurde nach Standardverfahren isoliert. Präparative Mengen von rekombinanter Phagen-DNA wurden mit Sall verdaut, um die inserierte DNA von den Lambda- Armen zu befreien, und durch Elektrophorese auf einem 0,6% Agarose- TAE-Gel aufgetrennt. Die großen Fragmente, elektroeluiert und konzentriert wie oben beschrieben, wurden mit SalI-verdautern und dephosphoryliertem pUC19 (NEB) ligiert. Die Ligierungsmischung wurde mittels Transformation in kompetente E. coli-DH5(α)-Zellen (BRL) eingeführt und auf LB-Agar ausplattiert, der Ampicillin, Isopropyl- (β)-D-thiogalactosid (IPTG) und 5-Brom-4-Chlor-3-indolyl-(β)-D- galactosid (XGAL) enthielt. Weiße Kolonien (mit Insertionen im (β)- Galactosidase-Gen von pUC19) wurden Standard-Plasmid-Schnellanreicherungsverfahren unterworfen, um das Plasmid mit der Bezeichnung pM1,RR1-A zu isolieren. Das Toxin-Gen vollständiger Länge wurde unter Verwendung von Oligonukleotid-Primern, die für das "4,5 Kb Klasse"- Toxin-Gen hergestellt worden waren und durch "Walking" mit Primern, die für die Sequenz von PS81RR1 hergestellt worden waren, sequenziert.
- Das Plasmid pM1,RR1-A enthält etwa 13 Kb PS81RR1-DNA, einschließlich der 3,540 Kb, welche für das 133,367-Dalton-Endotoxin kodieren. Der ORF des PS81RR1-Toxin-Gens wurde aus pM1,RR1-A auf einem 3,8 Kb NdeI- Fragment isoliert und in den Bacillus-"Shuttle"-Vektor pBC1ac ligiert. E. coli NM522-Zellen wurden transformiert und die resultierenden Kolonien durch Standard-Schnellanreicherungsverfahren analysiert, um Plasmide zu isolieren, welche das korrekte Insert enthielten. Das gewünschte Plasmid pMYC390 enthält die kodierende Sequenz des PS81RR1-Toxin-Gens.
- Die obigen Klonierungsverfahren wurden, soweit nicht anders angegeben, unter Verwendung von Standardverf ahren durchgeführt.
- Die verschiedenen Methoden, die bei der Herstellung der Plasmide und der Transformation von Wirtsorganisrnen eingesetzt wurden, sind auf dem Gebiet wohlbekannt. Ebenso sind Verfahren für den Einsatz des Bakteriophagen Lambda als Klonierungs-Vehikel, d.h., die Präparation von Lambda-DNA, in vitro-Verpackung und Transfektion rekombinanter DNA, auf dem Gebiet wohlbekannt. Diese Verfahren sind alle in Maniatis, T., Fritsch, E.F., und Sambrook, J. (1982) Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Laboratory, New York, beschrieben. Deshalb liegt es im Rahmen der Fähigkeiten von Fachleuten auf dem Gebiet des "Genetic Engineering", DNA aus mikrobiellen Zellen zu extrahieren, Verdauungen mit Restriktionsenzymen durchzuführen, DNA-Fragmente zu elektrophoresieren, Plasmid und Insert-DNA mit Enden zu versehen und zu verschmelzen, DNA zu ligieren, Zellen zu transformieren, Plasmid-DNA herzustellen, Proteine zu elektrophoresieren und DNA zu sequenzieren.
- Die hier offenbarten Restriktionsenzyme können von Bethesda Research Laboratories, Gaithersburg, MD, New England Biolabs, Beverly, MA, oder Boehringer-Mannheim, Indianapolis, IN, bezogen werden. Die Enzyme werden nach den vom Hersteller gegebenen Anweisungen verwendet.
- Das Plasmid pMYC3B6, welches die B.t.-Toxin-Gene enthält, kann aus den transformierten Wirts-Mikroorganismen mittels bekannter Standardverfahren entfernt werden. Beispielsweise kann E. coli NRRL B- 18449 isopyknischen "cleared lysate" -Dichtegradientenverfahren und dgl. unterworfen werden, um pMYC3B6 zu gewinnen.
- Das für das neue insektizide Toxin kodierende neue Gen, wie hier offenbart, kann in Pflanzenzellen unter Verwendung des Ti-Plasmids aus Agrobacter tumefaciens eingeführt werden. Die Pflanzenzellen können dann dazu veranlaßt werden, zu Pflanzen zu regenerieren (Zambryski, P., Joos, H., Gentello, C., Leemans, J., Van Montague, M. und Schell, J. [1983] Cell 32:1033-1043). Ein besonders nützlicher Vektor in dieser Beziehung ist pEND4K (Klee, H.J., Yanofsky, M.F. und Nester, E.W. [1985] Bio/Technology 3:637-642). Dieses Plasmid kann sowohl in Pflanzenzellen als auch in Bakterien replizieren und weist mehrfache Klonierungsstellen für "Passenger-Gene" auf. Das Toxin-Gen kann beispielsweise in die BamHI-Stelle von pEND4K inseriert, in E. coli vermehrt und in geeignete Pflanzenzellen transformiert werden.
- Das neue erfindungsgemäße Gen kann in Baculoviren wie Autographa californica Kern-Polyhedrosis-Virus (AcNPV) kloniert werden. Es können Plasmide konstruiert werden, die das AcNPV-Genom in einen im Handel erhältlichen Klonierungsvektor wie pUC8 kloniert enthalten. Das AcNPV-Genom ist so modifiziert, daß der kodierende Bereich des Polyhedrin-Gens entfernt ist und eine nur einmal vorkommende Klonierungsstelle für ein "Passenger"-Gen direkt hinter dem Polyhedrin-Promotor plaziert ist. Beispiele solcher Vektoren sind pGP-B6874, beschrieben von Pennock et al. (Pennock, G.D., Shoemaker, C. und Miller, L.K. [1984] Mol. Cell. Biol. 4:399-406) und pAC380, beschrieben von Smith et al. (Smith, G.E., Summers, M.D. und Fraser, M.J. [1983] Mol Cell. Biol. 3:2156-2165). Das Gen, welches für das erfindungsgemäße neue Proteintoxin kodiert, kann mit BamHI-"Linkern" an geeigneten Bereichen sowohl stromaufwärts als auch stromabwärts des kodierenden Bereichs modifiziert werden und in die "Passenger"- Stelle eines der AcNPV-Vektoren eingebaut werden.
- Die Nukleotid-Sequenz, welche für das neue B.t.-Toxingen kodiert und die abgeleitete Aminosäuresequenz sind in SEQ ID Nr. 1 unten dargestellt.
- Aus dem Stand der Technik ist wohlbekannt, daß die Aminosäuresequenz eines Proteins durch die Nukleotidsequenz der DNA bestimmt wird. Aufgrund der Redundanz des genetischen Codes, d.h. mehr als ein kodierendes Nukleotidtriplett (Codon) kann für die meisten der zur Herstellung von Proteinen verwendeten Aminosäuren eingesetzt werden, können verschiedene Nukleotidsequenzen für eine spezielle Aminosäure kodieren.
- Die neuen B.t.-Toxine können mittels jeder Nukleotidsequenz (äquivalent zu der dargestellten) hergestellt werden, die für dieselbe Aminosäuresequenz kodiert; die vorliegende Erfindung schließt solche äquivalenten Nukleotidsequenzen ein.
- Es ist gezeigt worden, daß Proteine mit einer identifizierten Struktur und Funktion durch Änderung der Aminosäuresequenz konstruiert werden können, falls solche Änderungen die Proteinsekundärstruktur nicht verändern; siehe Kaiser, E.T. und Kezdy, F.J. (1984) Science 223:249-255. Die vorliegende Erfindung schließt Mutanten der hier dargestellten Aminosäuresequenzen ein, die eine unveränderte Proteinsekundärstruktur aufweisen oder, wenn die Struktur verändert ist, die Mutante die biologische Aktivität bis zu einem gewissen Grad beibehalten hat.

Claims (22)
1. Bacillus thuringiensis PS81RR1, wie erhältlich unter der
Hinterlegungsnummer NRRL B-18458.
2. DNA, die für ein Bacillus thuringiensis-Toxin mit der in
SEQ ID Nr.1 dargestellten Aminosäuresequenz kodiert.
3. DNA nach Anspruch 2, welche die in SEQ ID Nr.1
dargestellte Nukleotidsequenz aufweist.
4. Toxin, welches die in SEQ ID Nr.1 dargestellte
Aminosäuresequenz aufweist.
5. Rekombinanter DNA-Transfer-Vektor, der DNA nach Anspruch
2 oder Anspruch 3 umfaßt.
6. Prokaryotischer oder eukaryotischer Wirt, in den ein DNA-
Transfer-Vektor nach Anspruch 5 transferiert und
repliziert worden ist.
7. Mikroorganismus, der imstande ist, ein Bacillus
thuringiensis-Toxin mit der in SEQ ID Nr.1 dargestellten
Aminosäuresequenz zu exprimieren.
8. Mikroorganismus nach Anspruch 7, der eine Spezies ist von
Pseudomonas, Azotobacter, Erwinia, Serratia, Klebsiella,
Rhizobium, Rhodopseudomonas, Methylophilius,
Agrobactenum, Acetobacter oder Alcaligenes; ein aus
Enterobacteriaceae, Bacillaceae, Rhizobiaceae, Spirillaceae,
Lactobacillaceae, Pseudomonadaceae, Azotobacteraceae und
Nitrobacteraceae ausgewählter Prokaryot; oder ein aus
Phycomycetes, Ascomycetes und Basidiomycetes ausgewählter
niederer Eukaryot.
9. Mikroorganismus nach Anspruch 8, der Pseudomonas
fluorescens oder Escherichia coli ist.
10. Mikroorganismus nach Anspruch 9, der E. coli (NM522)
(pMYC 390) ist, wie erhältlich unter der
Hinterlegungsnummer NRRL B-18449.
11. Mikroorganismus nach Anspruch 7, der ein pigmentiertes
Bakterium, Hefe oder ein Pilz ist.
12. Mikroorganismus nach irgendeinem der Ansprüche 7 bis 11,
der pigmentiert ist und Phyllooberflächen-adhärent.
13. Im wesentlichen intakte Zellen eines einzelligen
Mikroorganismus nach irgendeinem der Ansprüche 1 und 6 bis 12,
der das Toxin enthält.
14. Zellen nach Anspruch 13, wie erhalten durch Behandlung
mit bd oder anderen chemischen oder physikalischen
Mitteln, um die insektizide Aktivität in der Umwelt zu
verlängern.
15. Zusammensetzung, umfassend einen Mikroorganismus nach
irgendeinem der Ansprüche 1 und 6 bis 12 in Verbindung mit
einem Insektizid-Träger oder mit
Formulierungsbestandteilen für die Anwendung als Samenüberzug.
16. Zusammensetzung nach Anspruch 15, worin der
Mikroorganismus in Form von Sporen oder Kristallen vorliegt.
17. Zusammensetzung nach Anspruch 15 oder Anspruch 16, worin
der Träger Phagostimulantien oder Lockstoffe für Insekten
umfaßt.
18. Verfahren zur Kontrolle eines Lepidoptera-Insekten-
Schädlings, welches umfaßt, daß der Schädling oder
dessen Umwelt mit einem Mikroorganismus nach irgendeinem
der Ansprüche 1 und 6 bis 12 in Kontakt gebracht wird.
19. Verfahren nach Anspruch 18, worin die Ausbringung in der
Rhizosphäre, der Phylboberfläche oder einem
Wasserkörper erfolgt.
20. Verfahren nach Anspruch 18, welches umfaßt, daß ein
Köderkörnchen, welches den Mikroorganismus, z.B. als Sporen
oder Kristalle, umfaßt, auf oder in dem Erdreich
ausgebracht wird, wenn Samen einer Pflanze, von der sich der
Schädling bekanntermaßen ernährt, gepflanzt wird.
21. Verfahren nach Anspruch 20, worin das Köderkörnchen
gleichzeitig mit dem Einpflanzen von Getreidesamen in
das Erdreich ausgebracht wird.
22. Plasmid pMYC 390, wie erhältlich in einem Wirt nach
Anspruch 10.
