Description
- Die vorliegende Erfindung betrifft neue α-Aminosäure-Derivate, Verfahren zu ihrer Herstellung, sie enthaltende pharmazeutische Zubereitungen sowie ihre Verwendung als Inhibitoren der Dipeptidyl-petidase IV (DPP IV).
- Die Dipeptidyl-peptidase IV ist eine Membran-Serinprotease, welche in einer Vielzahl von menschlichen Geweben vorliegt und bei einer Reihe von Erkrankungen beteiligt ist.
- Insbesondere konnte gezeigt werden, daß DPP IV für die Inaktivierung von GLP-1 (des Glucagon-artigen Peptids 1) verantwortlich ist. GLP-1 ist ein wichtiger Stimulator der Sekretion von Insulin im Pankreas und entfaltet demzufolge eine direkte günstige Wirkung auf den Blut-Glucosespiegel.
- Die Inhibierung der DPP IV stellt demzufolge einen extrem interessanten Ansatz dar bei der Behandlung der Glucoseunverträglichkeit und von mit Hyperglykämie verknüpften Erkrankungen, wie beispielsweise dem nicht insulinabhängigen Diabetes (Diabetes Typ II) oder der Fettsucht.
- In der Literatur wurden bereits Inhibitoren der DPP IV beschrieben, insbesondere Amidderivate in der Patentanmeldung
EP 0 490 379 und in den Literaturstellen Current Med. Chem. 6 (4), (1999), 311-327, Arch. Biochem. Biophys. 323 (1), (1995), 148-154, Bioorg. Med. Chem. Lett. 6 (22), (1996), 2745-2748 und Adv. Exp. Med. Biol. 421, (1997), 157-160, und Carbamatderivate in der PatentanmeldungDE 198 26 972 . - Andererseits wurden in der Patentanmeldung WO 96/27593 zu Arginin analoge α-Aminosäure-Derivate beschrieben als Inhibitoren der NO-Synthase, welche daher nützlich sind zur Behandlung von Erkrankungen des zentralen und peripheren Nervensystems, pathologischen Dysfunktionen des gastrintestinalen und Harnsystems und Erkrankungen, die mit dem kardiovaskulären oder dem bronchopulmonalen System verknüpft sind.
- Die erfindungsgemäßen Verbindungen entfalten ihrerseits inhibierende Wirkungen auf die Dipeptidyl-peptidase IV, was sie besonders nützlich macht für die Behandlung der Glucoseunverträglichkeit und von mit der Hyperglykämie verknüpften Erkrankungen.

- – entweder eine Gruppe ausgewählt aus geradkettigem oder verzweigtem (C1-C6)-Alkyl, Nitro und Cyano bedeutet, für den Fall, da Y CH-NO2 darstellt,
- – oder eine Gruppe ausgewählt aus Nitro und Cyano bedeutet, wenn Y NR4 darstellt,
- Als pharmazeutisch annehmbare Säuren kann man in nicht einschränkender Weise nennen Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Trifluoressigsäure, Milchsäure, Brenztraubensäure, Malonsäure, Bernsteinsäure, Glutarsäure, Fumarsäure, Weinsäure, Maleinsäure, Citro nensäure, Ascorbinsäure, Methansulfonsäure, Camphersäure, Oxalsäure, etc.
- Unter einem stickstoffhaltigen Heterocyclus mit 5 Kettengliedern versteht man eine monocyclische gesättigte Gruppe mit 5 Kettengliedern, die eines, zwei oder drei Heteroatome enthält, wobei eines dieser Heteroatome das Stickstoffatom ist und das oder die gegebenenfalls vorhandenen weiteren Heteroatome aus Sauerstoff-, Stickstoff- und Schwefelatomen ausgewählt sind.
- Die bevorzugten stickstoffhaltigen Heterocyclen mit 5 Kettengliedern sind die Pyridinyl- und Thiazolidinyl-gruppen.

- Die bevorzugten Verbindungen der Formel (I) sind jene, bei denen die Amidfunktion in der α-Konfiguration (S) vorliegt.
- Ein vorteilhafter Gegenstand der Erfindung betrifft die Verbindungen der Formel (I), bei denen
Ak die Gruppe (CH2)4 darstellt. -
- Ein weiterer vorteilhafter Gegenstand der Erfindung betrifft die Verbindungen der Formel (I), bei denen X eine Einfachbindung bedeutet.
- Ein weiterer vorteilhafter Gegenstand der Erfindung betrifft die Verbindungen der Formel (I), bei denen Y eine Gruppe NR4 darstellt, worin R4 ein Wasserstoffatom oder eine geradkettige oder verzweigte (C1-C6)-Alkylgruppe bedeutet.
- Ein weiterer vorteilhafter Gegenstand der Erfindung betrifft die Verbindungen der Formel (I), bei denen R3 die Nitrogruppe bedeutet.
- Als bevorzugte Verbindungen der Erfindung kann man insbesondere nennen:
- – N-{(4S)-4-Amino-5-[(2S)-2-cyano-1-pyrrolidinyl]-5-oxopentyl}-N'-nitroguanidin, seine Tautomeren, seine optischen Isomeren sowie seine Additionssalze mit einer pharmazeutisch annehmbaren Säure,
- – N-{(5S)-5-Amino-6-[(2S)-2-cyano-1-pyrrolidinyl]-6-oxohexyl}-N'-nitroguanidin, seine Tautomeren, seine optischen Isomeren sowie seine Additionssalze mit einer pharmazeutisch annehmbaren Säure und
- – N-{(5S)-5-Amino-6-(1,3-thiazolidin-3-yl)-6-oxohexyl}-N'-nitroguanidin, seine Tautomeren, seine optischen Isomeren sowie seine Additionssalze mit einer pharmazeutisch annehmbaren Säure.
- Die Erfindung erstreckt sich weiterhin auf das Verfahren zur Herstellung der Verbindungen der Formel (I), welches dadurch gekennzeichnet ist, daß man eine Verbindung der Formel (II): in der Ak und X die bezüglich der Formel (I) angegebenen Bedeutungen besitzen, P1 eine Schutzgruppe für die Aminofunktion darstellt und P2 eine von P1 verschiedene Schutzgruppe für die Aminofunktion darstellt,
mit einer Verbindung der Formel (III): in der die bezüglich der Formel (I) angegebenen Bedeutungen besitzt,
bei den klassischen Bedingungen der Peptidkupplung umsetzt,
so daß man nach der Abspaltung der Schutzgruppe die Verbindung der Formel (IV) erhält: in der Pi, Ak und X die oben angegebenen Bedeutungen besitzen,
welche man anschließend mit Hilfe klassischer Reaktionen der organischen Chemie, gefolgt von einer Reaktion zur Abspaltung der Schutzgruppen, in die Verbindung der Formel (I) umwandelt,
welche man gegebenenfalls mit Hilfe einer klassischen Reinigungsmethode reinigt, die man gewünschtenfalls mit Hilfe einer klassischen Trennmethode in ihre optischen Isomeren auftrennt und welche man gewünschtenfalls in ihre Additionssalze mit einer pharmazeutisch annehmbaren Säure überführt. - Man kann die Verbindungen der Formel (Ie), einem Sonderfall der Verbindungen der Formel (I): in der R1, Ak und X die bezüglich der Formel (I) angegebenen Bedeutungen besitzen, ebenfalls erhalten ausgehend von der Verbindung der Formel (V): in der P1, Ak und X die oben angegebenen Bedeutungen besitzen,
welche man bei den klassischen Bedingungen der Peptidkupplung mit der Verbindung der Formel (III) umsetzt, so daß man nach der eventuellen Abspaltung der Schutzgruppe die Verbindung der Formel (Ie) erhält, welche man gegebenenfalls mit Hilfe einer klassischen Reinigungsmethode reinigt, gewünschtenfalls mit Hilfe einer klassischen Trennmethode in ihre optischen Isomeren auftrennt und gewünschtenfalls in ihre Additionssalze mit einer pharmazeutisch annehmbaren Säure über führt. - Die Verbindungen der vorliegenden Erfindung besitzen neben der Tatsache, daß sie neu sind, interessante pharmakologische Wirkungen. Sie entfalten inhibierende Wirkungen auf die Dipeptidylpeptidase IV, was sie nützlich macht bei der Behandlung der Glucoseunverträglichkeit und von mit einer Hyperglykämie ver knüpften Erkrankungen, wie Diabetes Typ II oder der Fettsucht.
- Die Erfindung erstreckt sich daher auch auf pharmazeutische Zubereitungen, die als Wirkstoff mindestens eine Verbindung der Formel (I) zusammen mit einem oder mehreren inerten, nichttoxischen und geeigneten Hilfsstoffen enthalten. Als erfindungsgemäße pharmazeutische Zubereitungen kann man insbesondere jene nennen, die für die Verabreichung auf oralem, parenteralem (intravenösem, intramuskulärem oder subkutanem) oder nasalem Wege geeignet sind, einfache oder dragierte Tabletten, Sublingualtabletten, Gelkapseln, Compretten, Suppositorien, injizierbare Präparate, trinkbare Suspensionen, etc.
- Die nützliche Dosierung hängt ab von der Art und der Schwere der Erkrankung, dem Verabreichungsweg sowie dem Alter und dem Gewicht des Patienten und eventuellen begleitenden Behandlungen. Diese Dosierung variiert von 0,5 mg bis 2 g im Verlaufe von 24 Stunden bei einer oder mehreren Gaben.
- Die folgenden Beispiele verdeutlichen die Erfindung, ohne sie in irgendeiner Weise einzuschränken.
- Die verwendeten Ausgangsprodukte sind bekannte Produkte oder werden mit Hilfe bekannter Vefahrensweisen hergestellt.
- Die Strukturen der in den Beispielen beschriebenen Verbindungen wurden mit Hilfe der üblichen spektrometrischen Methoden (Infrarotspektrum, NMR- Spektrum, Massenspektrum) bestimmt.
- Unter einer Verbindung mit der Konfiguration (2α) oder (2β) versteht man eine Verbindung ausgewählt aus den Verbindungen mit den absoluten Konfigurationen (2R) und (2S), wobei es sich versteht, daß, wenn die Verbindung (2α) die Verbindung mit der absoluten Konfiguration (2R) bedeutet, dann die Verbindung (2α) für die Verbindung (2S) steht.
- BEISPIEL 1: N-{(4S)-4-Amino-5-[(2S)-2-cyano-1-pyrrolidinyl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Stufe A: N-{(4S)-4-((tert.-Butyloxycarbonyl)-amino]-5-[(2S)-2-cyano-1-pyrrolidinyl]-5-oxopentyl}-N'-nitroguanidin
- Man gibt zu 10 mMol N2-(tert.-Butyloxycarbonyl)-N5-[(imino)-(nitroamino)-methyl]-ornithin in Lösung in Dimethylformamid 10 mMol (2S)-2-Cyano-pyrrolidin, 10 mMol 1-Hydroxybenzotriazol und 10 mMol Dicyclohexylcarbodiimid. Nach dem Rühren über Nacht bei Raumtemperatur filtriert man den gebildeten Dicyclohexylharnstoff ab und verdampft das Dimethylformamid. Man reinigt den in dieser Weise erhaltenen Rückstand chroamtographisch über Siliciumdioxid (Elutionsmittel: Dichlormethan/Ethanol 95/5) und erhält das erwartete Produkt.
- Stufe B: N-{(4S)-4-Amino-5-((2S)-2-cyano-1-pyrrolidinyl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man gibt zu 10 mMol der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Dioxan eine 4N Lösung von Chlorwasserstoffsäure in Dioxan. Nach dem Rühren während 24 Stunden bei Raumtemperatur verdampft man das Lösungsmittel, gibt Wasser zu und gefriertrocknet die Lösung unter Erhalt des erwarteten Produkts.
- BEISPIEL 2: N-[(45)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N'-cyanoguanidin, Sesquihydrochlorid
- Stufe A: 1-((S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man erhält das erwartete Produkt nach dem in der Stufe A des Beispiels 1 beschriebenen Verfahren ausgehend von (S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-onithin und Pyrrolidin.
- Stufe B: 1-[(S)-N2-(tert.-Butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man hydriert 10 mMol der in der vorhergehenden Stufe erhaltenen Ver bindung in Lösung in Ethanol in Gegenwart von 10 % Palladium-auf-Kohlenstoff bei Raumtemperatur und Umgebungsdruck während 6 Stunden. Anschließend filtriert man das Reaktionsmedium, dampft ein, setzt Wasser zu, gefriertrocknet und erhält das erwartete Produkt.
- Stufe C: N-{(4S)-4-((tert.-Butyloxycarbonyl)-amino]-5-(1-pyrrolidinyl)-5-oxopentyl}-N'-cyanoguanidin
- Man erhält das erwartete Produkt nach der in Synthesis (1975), 332 beschriebenen Verfahrensweise ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung und von NaN(CN)2.
- Stufe D: N-[(4S)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N'-cyanoguanidin, Sesqui hydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe B des Beispiels 1 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung.
- BEISPIEL 3: N-[(4S)-4-Amino-5-(1,3-thiazolidin-3-yl)-5-oxopentyl]-N'-cyano guanidin
- Stufe A: 3-{(S)-N5-(tert.-Butyloxycarbonyl)-N2-((9H-fluoren-9-yl-methoxy)-carbonyl]-ornithyl}-1,3-thiazolidin
- Man erhält das erwartete Produkt nach dem in der Stufe A des Beispiels 1 beschriebenen Verfahren ausgehend von (S)-N5-(tert.-Butyloxycarbonyl)-N2-(9H-fluo ren-9-yl-methoxy)-carbonyl]-ornithin und von 1,3-Thiazolidin.
- Stufe B: 3-{(S)-N2-((9H-Fluoren-9-yl-methoxy)-carbonyl]-ornithyl}-1,3-thiazolidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe B des Beispiels 1 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung.
- Stufe C: N-[(4S)-4-Amino-5-(1,3-thiazolidin-3-yl)-5-oxopentyl]-N'-cyanoguanidin
- Man erhält das erwartete Produkt nach dem in Synthesis (1975), 332 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung und von NaN(CN)2.
- BEISPIEL 4: N-[(5S)-5-Amino-6-(1-pyrrolidinyl)-6-oxohexyl]-N'-cyanoguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in Beispiel 2 beschriebenen Verfahren unter Ersatz von (S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)- ornithin durch (S)-N6-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-lysin.
- BEISPIEL 5: N1-[(4S)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N2-methyl-2-nitro-1,1-ethylendiamin, Dihydrochlorid
- Stufe A: 1-((S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man erhält das erwartete Produkt nach dem in der Stufe A des Beispiels 1 beschriebenen Verfahren ausgehend von (S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-ornithin und von Pyrrolidin.
- Stufe B: 1-((S)-N2-(tert.-Butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man hydriert 10 mMol der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Ethanol in Gegenwart von 10 % Palladium-auf-Kohlenstoff bei Umgebungstemperatur und Umgebungsdruck während 6 Stunden. Anschließend filtriert man das Reaktionsmedium, dampft ein und gefriertrocknet das Material nach der Zugabe von Wasser unter Erhalt des erwarteten Produkts.
- Stufe C: NI-{(4S)-4-((tert.-Butyloxycarbonyl)-amino]-5-(1-pyrrolidinyl)-5-oxopentyl}-N2-methyl-2-nitro-1,1-ethylendiamin
- Man erhält das erwartete Produkt nach der in Bioorg. Med. Chem. 7(23) (1997), 3045-3048 beschriebenen Verfahrensweise ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung und von N-Methyl-1-methylthio-2-nitroethylendiamin.
- Stufe D: N1-[(4S)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N2-methyl-2-nitro-1,1-ethylendiamin, Dihydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe B des Beispiels 1 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung.
- BEISPIEL 6: N-[(4S)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N''-cyano-N'-methylguanidin, Hydrochlorid
- Stufe A: 1-((S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man erhält das erwartete Produkt nach dem in der Stufe A des Beispiels 1 beschriebenen Verfahren ausgehend von (S)-N5-(Benzyloxycarbonyl)-N2-(tert.-butyloxycarbonyl)-ornithin und von Pyrrolidin.
- Stufe B: 1-((S)-N2-(tert.-Butyloxycarbonyl)-ornithyl]-pyrrolidin
- Man hydriert 10 mMol der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Ethanol in Gegenwart von 10 % Palladium-auf-Kohlenstoff bei Umgebungstemperatur und Umgebungsdruck während 6 Stunden. Anschließend filtriert man das Reaktionsmedium, dampft ein, gibt Wasser zu, gefriertrocknet und erhält das erwartete Produkt.
- Stufe C: N-{(4S)-4-[(tert.-Butyloxycarbonyl)-amino]-5-(1-pyrrolidinyl)-5-oxopentyl}-N''-cyano-N'-methylguanidin
- Man erhält das erwartete Produkt nach dem in Chem. Pharm. Bull. 45(1) (1997), 53-61 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung, von N-Cyanoimido-S,S-dimethyldithiocarbonat und Methylamin.
- Stufe D: N-((4S)-4-Amino-5-(1-pyrrolidinyl)-5-oxopentyl]-N''-cyano-N'-methylguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe B des Beispiels 1 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung.
Massenspektrum: (M + H]+ = 267; (M + Cl]+ = 301 ; (M – H]– = 265. - BEISPIEL 7: N-{(4S)-4-Amino-5-[(4R)-4-cyano-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in Beispiel 1 beschriebenen Verfahren unter Ersatz von (2S)-2-Cyano-pyrrolidin durch (4R)-4-Cyano-1,3-thiazolidin.
- BEISPIEL 8: N-{(4S)-4-Amino-5-[(2a)-2-cyano-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Stufe A: N-{(4S)-4-((tert.-Butyloxycarbonyl)-amino]-5-(2-carbamoyl-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe A des Beispiels 1 beschriebenen Verfahren unter Ersatz des (2S)-2-Cyano-pyrrolidins durch (±)-1,3-Thiazolidin-2-carboxamid.
- Stufe B: N-{(4S)-4-((tert.-Butyloxycarbonyl)-amino]-5-((2a)-2-carbamoyl-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man trennt die in der vorhergehenden Stufe A erhaltene Mischung der Diastereoisomeren durch Chromatographie über Siliciumdioxid (Elutionsmittel: Dichlormethan/Methanol/NH4OH 90/10/0,5).
- Das erwartete Produkt ist das erste in dieser Weise abgetrennt Diastereoisomere.
- Stufe C: N-{(4S)-4-[(tert.-Butyloxycarbonyl)-amino]-5-((2a)-2-cyano-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin
- Man gibt zu 10 mMol der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Pyridin 20 mMol Imidazol, kühlt die Reaktionsmischung auf –30 °C ab und gibt tropfenweise 40 mMol POCl3 zu. Anschließend bringt man die Temperatur im Verlaufe von 1 Stunde auf –20 °C, verdampft dann das Pyridin und löst den Rückstand in Ethylacetat. Man wäscht die Lösung, trocknet sie, dampft sie ein und reinigt den erhaltenen Rückstand chromatographisch über Siliciumdioxid (Elutionsmittel: Dichlormethan/Methanol/NH4OH 95/5/0,5) und erhält das erwartete Produkt.
- Stufe D: N-{(4S)-4-Amino-5-((2a)-2-cyano-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in der Stufe B des Beispiels 1 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe C erhaltenen Verbindung.
- BEISPIEL 9: N-{(4S)-4-Amino-5-((2β)-2-cyano-1,3-thiazolidin-3-yl]-5-oxopentyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in den Stufen C und D des Beispiels 8 beschriebenen Verfahren ausgehend von dem zweiten Diastereoisomeren, das man in der Stufe B des Beispiels 8 abgetrennt hat.

- Man trennt die in der obigen Stufe A erhaltene Mischung der Diastereoisomeren durch Chromatographie über Siliciumdioxid (Elutionsmittel: Dichlormethan/Methanol/NH4OH 90/10/0,5). Das erwartete Produkt ist das erste der in dieser Weise abgetrennten Diastereoisomeren.
- Stufe C: N-{(5S)-5-Amino-6-[(2a)-2-cyano-1,3-thiazolidin-3-yl]-6-oxohexyl}-N'-nitro guanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in den Stufen C und D des Beispiels 8 beschriebenen Verfahren ausgehend von der in der vorhergehenden Stufe erhaltenen Verbindung.
- BEISPIEL 15: N-{(5S)-5-Amino-6-[(2β)-2-cyano-l,3-thiazolidin-3-yl]-6-oxo hexyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in den Stufen C und D des Beispiels 8 beschriebenen Verfahren ausgehend von dem zweiten der in der Stufe B des Beispiels 14 getrennten Diastereoisomeren.

- BEISPIEL 30: N-[(6S)-6-Amino-7-(1,3-thiazolidin-3-yl)-7-oxoheptyl]-N'-nitroguanidin, Dihydrochlorid
- Man erhält das erwartete Produkt nach dem in Beispiel 29 beschriebenen Verfahren ausgehend von (2S)-2-[(tert.-Butyloxycarbonyl)-amino]-7-amino-heptansäure und von 1,3-Thiazolidin.
- BEISPIEL 31: N-[(6S)-6-Amino-7-[(2S)-2-cyano-1-pyrrolidinyl]-7-oxoheptyl]-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in Beispiel 29 beschriebenen Verfahren ausgehend von (2S)-2-[(tert.-Butyloxycarbonyl)-amino]-7-amino-heptansäure und von (2S)-2-Cyano-pyrrolidin.
- BEISPIEL 32: N-{4-[(2S)-2-Amino-3-oxo-3-[(2S)-2-cyano-1-pyrrolidinyl]-propyl]-phenyl}-N'-nitroguanidin, Hydrochlorid
- Man erhält das erwartete Produkt nach dem in Beispiel 29 beschriebenen Verfahren ausgehend von (2S)-2-[(tert.-Butyloxycarbonyl)-amino]-3-(4-aminophenyl) propansäure und von (2S)-2-Cyano-pyrrolidin.
- PHARMAKOLOGISCHE UNTERSUCHUNG DER ERFINDUNGSGEMÄSSEN VERBINDUNGEN
- BEISPIEL 33: Inhibierung der Dipeptidyl-peptidase IV in vitro
- Man bewertet die Wirkung der erfindungsgemäßen Verbindungen auf die enzymatische Wirkung der DPPIV in vitro wie folgt. Man bestimmt das Nierenenzym von Schweinen mit Hilfe eines chromogenen Substrats, nämlich 1,4 mM Glycyl-Propyl-p-Nitroanilid, das unter Freisetzung von p-Nitroanilin hydrolysiert wird, welches bei 405 nm absorbiert. Man bestimmt die Enzymaktivität durch Absorption in Gegenwart von variablen Konzentrationen der zu bewertenden Verbindung (meistens 10–4 bis 10–9 M). Die erhaltenen Zahlenwerte ermöglichen die Bestimmung der wirksamen Dosis zur Inhibierung der Kontrollaktivität um 50 % (IC50). Die erfindungsgemäßen Verbindungen besitzen einen IC50-Wert zwischen 1 nM und 10 μM.
mit Ausnahme der Verbindungen, bei denen gleichzeitig einen nichtsubstituierten stickstoffhaltigen Heterocyclus mit 5 Kettengliedern darstellt, Ak die Gruppe -(CH2)3- darstellt, X eine Einfachbindung darstellt, Y NH darstellt und R3 die Nitrogruppe darstellt.
Claims (15)
- Verbindung der Formel (I): in der: einen stickstoffhaltigen Heterocyclus mit 5 Kettengliedern bedeutet, der gegebenenfalls durch eine Cyanogruppe substituiert ist, – Ak eine geradkettige oder verzweigte (C1-C6)-Alkylenkette bedeutet, – X eine Einfachbindung oder eine Phenylengruppe bedeutet, – R1 und R2, die gleichartig oder verschieden sind, jeweils ein Wasserstoffatom oder eine geradkettige oder verzweigte (C1-C6)-Alkylgruppe bedeuten, – NR4 oder CH-NO2 bedeutet, worin • R4 ein Wasserstoffatom oder eine geradkettige oder verzweigte (C1-C6)-Alkylgruppe darstellt, – R3: – entweder eine Gruppe ausgewählt aus geradkettigem oder verzweigtem (C1-C6)-Alkyl, Nitro und Cyano bedeutet, für den Fall, da Y CH-NO2 darstellt, – oder eine Gruppe ausgewählt aus Nitro und Cyano bedeutet, wenn Y NR4 darstellt, ihre Tautomeren, falls sie existieren, ihre optischen Isomeren und ihre Additionssalze mit einer pharmazeutisch annehmbaren Säure, mit Ausnahme der Verbindungen, bei denen gleichzeitig einen nichtsubstituierten stickstoffhaltigen Heterocyclus mit 5 Kettengliedern darstellt, Ak die Gruppe -(CH2)3- darstellt, X eine Einfachbindung darstellt, Y NH darstellt und R3 die Nitrogruppe darstellt, wobei es sich versteht, daß man unter einem stickstoffhaltigen Heterocyclus mit 5 Kettengliedern eine monocyclische gesättigte Gruppe mit 5 Kettengliedern versteht, die eines, zwei oder drei Heteroatome enthält, wobei eines dieser Heteroatome das Stickstoffatom ist und das oder die gegebenenfalls vorhandenen weiteren Heteroatome aus Sauerstoff-, Stickstoff- und Schwefelatomen ausgewählt sind.
- Verbindung der Formel (I) nach einem der Ansprüche 1 oder 2, worin die Konfiguration in der α-Stellung zu der Amidfunktion die (S)-Konfiguration ist.
- Verbindung der Formel (I) nach einem der Ansprüche 1 bis 3, worin Ak die Gruppe (CH2)4 bedeutet.
- Verbindung der Formel (I) nach einem der Ansprüche 1 bis 5, worin X eine Einfachbindung bedeutet.
- Verbindung der Formel (I) nach einem der Ansprüche 1 bis 6, worin Y eine Gruppe NR4 bedeutet, worin R4 ein Wasserstoffatom oder eine geradkettige oder verzweigte (C1-C6)-Alkylgruppe darstellt.
- Verbindung der Formel (I) nach einem der Ansprüche 1 bis 7, worin R3 die Nitrogruppe bedeutet.
- Verbindung der Formel (I) nach Anspruch 1, nämlich N-{(4S)-4-Amino-5-[(2S)-2-cyano-1-pyrrolidinyl]-5-oxopentyl}-N'-nitroguanidin sowie dessen optische Isome re, dessen Tautomere und dessen Additionssalze mit einer pharmazeutisch annehmba-ren Säure.
- Verbindung der Formel (I) nach Anspruch 1, nämlich N-{(5S)-5-Amino-6-[(2S) 2-cycano-1-pyrrolidinyl]-6-oxohexyl}-N'-nitroguanidin und dessen optische Isomere, dessen Tautomere und dessen Additionssalze mit einer pharmazeutisch annehmbaren Säure.
- Verbindung der Formel (I) nach Anspruch 1, nämlich N-{(5S)-5-Amino-6-(1,3-thiazolidin-3-yl)-6-oxohexyl}-N'-nitroguanidin sowie dessen optische Isomere, dessen Tautomere und dessen Additionssalze mit einer pharmazeutisch annehmbaren Säure.
- Verfahren zur Synthese der Verbindungen der Formel (I) nach Anspruch 1, dadurch gekennzeichnet, daß man eine Verbindung der Formel (II): in der Ak und X die bezüglich der Formel (I) angegebenen Bedeutungen besitzen, P1 eine Schutzgruppe für die Aminofunktion darstellt und P2 eine von P1 verschiedene Schutzgruppe für die Aminofunktion darstellt, mit einer Verbindung der Formel (III): in der die bezüglich der Formel (I) angegebenen Bedeutungen besitzt, bei den klassischen Bedingungen der Peptidkupplung umsetzt, so daß man nach der Abspaltung der Schutzgruppe die Verbindung der Formel (IV) erhält: in der P1, Ak und X die oben angegebenen Bedeutungen besitzen, welche man anschließend mit Hilfe klassischer Reaktionen der organischen Chemie, gefolgt von einer Reaktion zur Abspaltung der Schutzgruppen, in die Verbindung der Formel (I) umwandelt, welche man gegebenenfalls mit Hilfe einer klassischen Reinigungsmethode reinigt, die man gewünschtenfalls mit Hilfe einer klassischen Trennmethode in ihre optischen Isomeren auftrennt und welche man gewünschtenfalls in ihre Additionssalze mit einer pharmazeutisch annehmbaren Säure überführt.
- Pharmazeutische Zubereitung enthaltend als Wirkstoff eine Verbindung nach einem der Ansprüche 1 bis 11 in Kombination mit einem oder mehreren iner-ten, nichttoxischen und pharmazeutisch annehmbaren Trägermaterialien.
- Pharmazeutische Zubereitung nach Anspruch 13, nützlich als Arzneimittel mit inhibierender Wirkung auf die Dipeptidyl-peptidase IV, nützlich bei der Behandlung der Glucoseunverträglichkeit, von Erkrankungen, die mit einer Hyperglykämie verknüpft sind, wie Diabetes Typ II oder Fettsucht.
- Pharmazeutische Zubereitung nach Anspruch 14, nützlich als antidiabetisches Arzneimittel.
