Neue Piperidylindole
Die Erfindung betrifft neue Piperidylindole, ins besondere 3-TPiperidyl-(4)j-indole der allgemeinen
Formel I
in der
R ein Wasserstoffatom oder eine Methoxygruppe und
R1 und R„ zugleich oder voneinander verschieden ein
Wasserstoffatom oder eine Methylgruppe bedeuten,
wobei R, R1 und R2 nicht zugleich Wasserstoffatome darstellen.
Die erfindungsgemHßen Piperidylindole sind insbesondere
als Ausgangsprodukte zur Synthese der in der DT-OS 2 338 ?83 beschriebenen 4-j~Indolyl-(3)j-piperidinoalkyl-arylketone

indol
Man gibt unter Ausschluß von Feuchtigkeit 7t5 g Natriumhydrid
(zu 50 i> in öl) in 75 ml Dimethylformamid und fügt
dann tropfenweise 43 g 3-[i'-Benzyl-1·,2*,3*,6'-tetrahydropyrldyl-(4')]-indol, gelöst In 225 ml Dimethylformamid hinzu.

Die Heaktionsmischung wird 30 Minuten lang auf 500C erwärmt.
Man kühlt dann ab und gibt zwisohen 0 und 50C 10,4 ml
Methyljodid hinzu. Dann wird 3 Stunden lang auf 400C erwärmt,
abgekühlt und hintereinander mit 200 ml Wasser versetzt. Der
gebildete Niederschlag wird abgesaugt bzw. abgeschleudert,
im Vakuum getrocknet und aus Aceton umkristallisiert. Man
erhält so 34,5 g 1-Methyl-3-[i '-benzyl-1·,2',3',e'-tetrehydropyridyl-(4')[-indol! P. % 103 bis 1040C

b) t-Methyl-3-fPiperidvl-( 4)1 -indol
Man gibt 30,2 g 1-Methyl-3- [if-benzyl-1·,2»,5'»61-tetrahydro-pyridyl-U1
j]-indol in 300 ml Essigsäure und 3 g Palladium auf Kohle (10.£lg). Bann wird die Mischung in Wasseretoffatmosphäre
bei Umgebungsdruck und 5O°G gerührt bzw. bewegt.
Kach Absorption der theoretischen Menge Wasserstoff wird der
Katalysator abfiltriert, die Essigsäure im Vakuum abgedampft und der Rückstand mit Wasser aufgenommen. Die Lösung wird
mit Katronaluge alkalisch gemacht und mit Methylenchlorid
extrahiert. Man trocknet über Natriumsulfat und dampft das
Lösungsmittel im Vakuum ab.
Haeh Umkristallisieren erhält man 14*5 g i-MethyX-3-[piperidyl-(4)}-indol;

Herstellung von 6-Methoxy->- fpiperidyl-(%>j -indol;
a) S-Methoxy-3- fpyridyl-C 4)1 -Indol
tOO g 6-Methoxyindol werden in 500 ml Pyridin gegeben.
Man kühlt auf -100C ab und fügt tropfenweise 158 ml Beneoylchlorid
hin2Eu. Die Mischung wird 3 Tage lang bei - Raumtemperatur
unter Liehtabaehlaß gertfchrt bsw. frewegt. Man dampft
dann das Pyrtdin unter vermindertem Brück ab, wäscht den Bttckatand
mit Äther und nimmt mit 1 1 Methanol wieder auf. Die Lösung wird mit Natronlauge alkalisch gemacht. Man dampft den
Methylalkohol unter vermindertem; Brück ab, säuert mit Salzsäure
an, wäscht mit Methylenchlorid, macht mit Natriumhydroxid alkalisch und extrahiert mit Chloroform. Die Organische Phase
wird über Natriumsulfat getrocknet und unter vermindertem Druck
eingedampft.

ratur über 30 bis 35°C hinausgeht, mit 0*95 S Matriumborkydrid
versetzt·

Die Mischung wird 2 Stunden lang bei Raumtemperatur gerührt
bzw. bewegt, mit Wasser versetzt, der gebildete Niederschlag abgesaugt bzw. abgeschleudert und mit Wasser gewaschen.
Nach Waschen mit Methanol und Trocknen erhält man 3,2 g 3- [1 ■ -Benzyl-1 ·, 2 ·, 3', 6»-tetrahydro-pyridyl-( 4')] -6-methoxyrindoli

d) 6-Methoxy-3-[piperidyl-(4)]-indol

Wasserstoff eingeleitet. Die theoretische Menge wird in 36 Stunden absorbiert. Der Katalysator wird abfiltriert und das
Äthanol abgedampft. Man erhält so ein öl, das man aus Äthylacetat
auskristalliBieren läßt. Nach Umkristallisieren aus
Aceton erhält man 8,1 g 6-Methoxy-3-jjpiperidyl-(4)] -indol.
Das Produkt ist ohne Reinigung für die Herstellung von

Herstellung von 5-Methoxy-3-jpiperidyl-(4η-Indolt
a) 5~Methoxy~3- fpyridyl-C 4)1 -indol
Das 5-Methoxy-3-[pyridyl-(4)]-indol wird nach den in Bei
spiel 1 der PR-PS 1 587 692 angegebenen Verfahren hergestellt.

d) 5-Methox.y-3-[piperidyl-U)]-indol
12,5 g 3-[1'-BeIiZyI-I1,2f ,3' ,6'-tetrahydro-pyridyl-(4· )]-5-methoxy-indol
und 3 g Palladium auf Kohle (10 %ig) werden in 300 ml Äthanol gegeben. Man erwärmt auf 5Q0C und läßt von
der Mischung 1770 cm^ Wasserstoff in 7 Stunden absorbieren. Nach
Beendigung der Reaktion wird der Katalysator abfiltriert und der Äthanol unter vermindertem Druck zur Trockne abgedampft.
Man erhält 8,3 g 5-Methoxy-3-[piperidyl-(4-)]-indolj F.» 1700C.

Herstellung von 2-Methyl-6-methoxy-3-[piperidyl-(4)~j-indol;
a) 2-Methyl-6-methoxy-3-[pyrrdyl-(4)1 -indol
50 g 2-ilethyl-6-methoxy-indol werden in 230 ml Pyridln
gegeben. Man kühlt auf -4-O0C ab und gibt tropfenweise 87 g
Benzoylchlorid hinzu. Die Mischung wird 3 Tage lang bei Zimmertemperatur unter Llchtabachluß gerührt bzw. bewegt. Man verdampft
dann daa Pyridln unter vermindertem Druck, wäscht mit
Natronlauge und verdampft daa Wasser. Das erhaltene Produkt wird in 500 ml eiedendem Methanol gelöst, dann unter Rühren
mit 200 ml Natriumhydroxid und schließlich zweimal mit 200 ml Wasser versetzt. Mau rührt 2 Stunden lang, laut eine Nacht
lang ruhen, verdampft das Methanol unter vermindertem Druck und extrahiert die wässrige Phase mit Chloroform. Die organische
Phase wird über Natriumsulfat getrocknet und unter verminertem Druck eingedampft. Man erhält 100 g Produkt, das durch

Chromatographie an Aluminiumoxid unter Elution mit Benaol gereinigt
wird; nach Abdampfen des Elutionsmittele erhält man
34 g Produkt, das aus Acetonitril umkristallisiert wird. Man erhält 19,5 g 2-Methyl-6-methoxy-3-[pyridyl-(4)] -indolj F. t
bis 1980G.

b) 1-Benzyl-4-[2t-Tnethyl-6,'-methoxy--lndolyl-(3')1-pyridiniumbromid
3,4 g 2-Methyl-6-methoxy-3-[pyridyl-(4 )]-indol und
2,65 g Benzylbromid werden in 30 ml Äthylacetat gegeben. Man erhitzt 4 Stunden lang unter Rückfluß zum Sieden. Man kühlt ab,
saugt bzw. schleudert die gebildeten Kristalle ab und wäscht diese mit Äthylacetat, Nach dem Trocknen erhält man 5,7 g
1 -Benzyl-4- [2' -methyl-6f -methoxy- indolyl-( 3') J-pyridiniumbromidf
F.* 244 bis 2460C.

c) 3- [l'-Benzyl-1',2' ,3 ',6'-tetrahydro-pyrldyl- (4')]-2-methyl-6-methoxy-indol
20,1 g l-Benzyl-4-{2'-methyl-6f-methoxy-indolyl-(5' )J pyridiniumbromid
werden bei 35 bis 40 0C in I65 ml Methanol
und 70 ml Wasser gegeben. Man kühlt auf 25 0C ab und gibt in
kleinen Portionen, ohne daß die Temperatur 30 bis 35 0C übersteigt,
4 g Natriumborhydrid hinzu.
Die Mischung wird 2 Stunden lang bei Raumtemperatur

gerührt bzw. bewegt und dann mit Wasser versetzt und der gebildete Niederschlag abgesaugt bzw. abgeschleudert und
mit Wasser gewaschen. Nach Trocknen unter vermindertem Druck erhält man 15,4 g 3-[ll-Benzyl-lT i2t,3' ,6'-tetrahydro-pyridyl-(4'-)]
^-methyl-ö-methoxy-indol; P.s 142
bis 14J 0C.

d) 2-Methyl-.6~meth03£y-3~ [piperldyl- (4)1 -Indol
24 g 3- [l1 -Benzyl-1 ',2',3!# β '-tetrahydro- pyridyl- (* ')]-2-methyl-6-methoxy-indol
und 4 g Palladium auf Kohle (10 #Lg) werden in 300 ml absolutes Äthanol gegeben. Man
erwärmt auf 50 0C und leitet Wasserstoff ein. In 12 Stunden
werden 2450 cnr Wasserstoff absorbiert. Der Katalysator
wird abfiltriert und erneut 3,0 g Palladium auf Kohle (10 #ig) hinzugegeben. Die Behandlung mit Wasserstoff wird
etwa 3 1/2 Stunden lang fortgesetzt s wonach 3250 cnr Wasserstoff
absorbiert sind. Man filtriert den Katalysator ab, verdampft das Äthanol unter vermindertem Druck und erhält
16,6 g 2-Methyl-6-mtthoxy-3-[piperidyl-(4)J-indolj F.: 166 0C.

Claims (2)
- ~12"Patentansprüche/. 3- jPiperidyl- (4)J -indole der allgemeinen Formel IN-Hmit R=H oder Methoxy und R1, R2 = H oder Methyl^wobei R, R. und R2 nicht zugleich H bedeuten.
- 2. Verfahren zur Herstellung der 3-[Piperidyl-(4)] -indole der Formel I nach Anspruch 1,gekennzeichnet durch folgende Schritte: (a) Umsetzung einer Verbindung der Formel IVuv)IImit R und R2 wie in Anspruoh mit Benzylbromid zu einer Verbindung der Formel Vmit R und R2 wie in Anspruch 1,6098S6/11U("b) Reduktion der Verbindung V mit Natriumborhydrid zu einer Verbindung der Formel VImit R und R2 wie in Anspruch 1(Yl)(el) Hydrierung der Verbindung VT mit Wasserstoff in Gegenwart eines Pd-Katalysators, wobei R. = H ist und R und R2 die obige Bedeutung besitzen, oder(c2) Umsetzung der Verbindung VI mit einem Alkylhalogenid HaI-R1 mit Hal = Halogen und R1 = Methyl in Gegenwart von Natriumhydrid zu einer Verbindung der Formel VII(VII)= Methyl und R und R2 wie in Anspruch 1raitund anschließende Hydrierung der Verbindung VEI mit'Wasserstoff in Gegenwart eines Pd-Katalysators, wobei R1 = Methyl ist und R und R2 wie oben definiert sind.609886/·^- l4 -j5. ¥erwenditing der 25-|_PiperIdyl-(4)j-±ndole der Formel I naeEii itaspraen 1 zur Herstellung von 4-|_Indolyl- (3)j-piperidlnoalkyl-arylice tonen.
