Die erfindungsgemäßen Piperidylindole sind insbesondere als Ausgangsprodukte zur Synthese der in der

(a) durch Hydrierung der Verbindung Vl mit Wasserstoff in Gegenwart eines Pd-Katalysators, wobei

(b) durch Umsetzung der Verbindung Vl mit einem Methylhalogenid in Gegenwart von Natriumhydrid
zu einer Verbindung der allgemeinen Formel Vl I

Die Herstellung der erfindungsgemäßen Verbindungen wird im folgenden durch Beispiele näher erläutert.
Herstellung von l-Methyl-3-[piperidyl-(4)]-indol
a) l-Methyl-3-[l '-benzyl-1 \2',3',6'-tetrahydropyridyl-(4')]-indoi
Man gibt unter Ausschluß von Feuchtigkeit 7,5 g Natriumhydrid (zu 50% in öl) in 75 ml Dimethylformamid
und fügt dann tropfenweise 43 g 3-[l'-Benzyll',2'3',6'-tetralnydro-pyridyI-(4')]-indol,
gelöst in 225 ml Dimethylformamid hinzu.
Die Reaktionsmischung wird 30 Minuten lang auf 500C erwärmt. Man kühlt dann ab und gibt zwischen 0
und 5°C 10,4 ml Methyljodid hinzu. Dann wird 3
r> Stunden lang auf 40°C erwärmt, abgekühlt und hintereinander mit 200 ml Wasser versetzt. Der
gebildete Niederschlag wird abgesaugt bzw. abgeschleudert, im Vakuum getrocknet und aus Aceton
umkristallisiert. Man erhält so 34,5g l-Methyl-3-[l'-benzyl-l\2'3',6'-tetrahydro
pyridyl-(4')]-indol; F.: 103 bis 104" C.
b) 1 -Methy!-3-[piperidyl-(4)]-indol
Man gibt 30,2 g l-Methyl-3-[l'-benzyi l',2',3',6'-tetra-
2ί hydro-pyridyl-(4')]-indol in 300 ml Essigsäure und 3 g
Palladium auf Kohle (IO%ig). Dann wird die Mischung in Wasserstoffatmosphäre bei Umgebungsdruck und
500C gerührt bzw. bewegt. Nach Absorption der theoretischen Menge Wasserstoff wird der Katalysator
jo abfiltriert, die Essigsäure im Vakuum abgedampft und der Rückstand mit Wasser aufgenommen. Die Lösung
wird mit Natronlauge alkalisch gemacht und mit Methylenchlorid extrahiert. Man trocknet über Natriumsulfat
und dampft das Lösungsmittel im Vakuum ab.
j-, Nach Umkristallisieren erhält man 14,5 g l-Methyl-3-[piperidyl-(4)]-indol; F.: 78 bis 79"C.

I lerstellung vrm 6-Methoxy-3-Lpiperidyl-(4)]-indol
a) 6-Mcthoxy-3-[pyridyl-(4)]-indol
a) 6-Mcthoxy-3-[pyridyl-(4)]-indol
100 g 6-Methoxyindo! werden in 500 ml Pyridin gegeben. Man kühlt auf — 10°C ab und fügt tropfenweise
158 ml Benzoylchlorid hinzu. Die Mischung wird 3 Tage lang bei Raumtemperatur unter Lichlabschluß
gerührt bzw. bewegt. Man dampft dann das Pyridin unter vermindertem Druck ab, wäscht den Rückstand
mit Äther und nimmt mit 1 I Methanol wieder auf. Die Lösung wird mit Natronlauge alkalisch gemacht. Man
dampft den Methylalkohol unter vermindertem Druck ab, säuert mit Salzsäure an, wäscht mit Methylenehlorid.
macht mit Natriumhydroxid alkalisch und extrahiert mit Chloroform. Die organische Phase wird über Natriumsulfat
getrocknet und unter vermindertem Druck eingedampft.
Der Rückstand wird aus Acetonitril umkristallisiert
und man erhält so 23,3 g 6-Methoxy-3-[pyridyl-(4)]-indol;F.:187bisl88"C.

Berechnet: C 74.99, H 5,38, N 12,49%;

b) 1 -Ben/yl-4-[b'-incthoxy-indolyl-( J)J-pyridiniumbromid
3.1 g Methoxy-3-[pyridy1-(4)]-ind(>l und 2.5b μ licn/vlbmmid
werden in 30 ml Äthylacetat pegeben. Die
Mischung wird 4 Stunden lang unter Rückfluß zum Sieden erhitzt Es wird abgekühlt und der gebildete
Niederschlag abgesaugt bzw. abgeschleudert Man wäscht mit Äthylacetat, trocknet und erhält 5,1 g
l-Benzyl-4-[6'-methoxy-indolyl-(3')]-pyridii!iumbromid;
F.: 2020C.
Summenformel: C2, HmBrN2O; M =395,3
Berechnet: C 63,80, H 4,84, N 7.09%;
gefunden: C 63,57, H 4,91, N 6,93%.

c) 3-[r-Benzyi-l \2',3',6'-tetrahydro-
pyridyl-(4')]-6-methoxy-indol
4,6 g 1-Benzyl-4{6'-methoxy-indolyl-(3')]-pyridiniumbromid
werden bei 400C in 45 ml Methanol und 26 ml Wasser gegeben, die Mischung auf 25° C abgekühlt und,
ohne daß die Temperatur über 30 bis 35° C hinausgehl,
mit 0,95 g Natriumborhydrid versetzt.
Die Mischung wird 2 Stunden lang bei Raumtemperatur gerührt bzw. bewegt, mit Wasser versetzt, der
gebildete Niederschlag abgesaugt bzw. abgeschleudert und mit Wasser' gewaschen. Nach Waschen mit
Methanol und Trocknen erhält man 3,2 g 3-[1'-Benzyl-1 ',2',3'.6'-tetrahydro-pyridyl-(4')]-6-methoxy-indol; F.:

d) 6-Methoxy-3-[piperidyl-(4)]-indol
21 g 3-[1'-Bcnzyl-r.2',3'.6'-tetrahydro-pyridyl-(4')]-6-methoxy-indol
und 2 g Palladium auf Kohle (10%ig) werden in 300 ml Äthanol gegeben. Dann wird unter
Erwärmung auf 55°C Wasserstoff eingeleitet. Die π theoretische Menge wird in 36 Stunden absorbiert. Der
Katalysator wird abfiltriert und das Äthanol abgedampft.
Man erhält so ein Öl, das man aus Äthylacetat auskristallisieren läßt. Nach Umkristallisieren aus
Aceton erhält man 8,1 g 6-Methoxy-3-[piperidyl-(4)]-indol.
Das Produkt ist ohne Reinigung für die Herstellung von p-Fluor-ti)-|4-[6-methoxy-indolyl-(3)]-piperidino|-
butyrophenon-hydrochlorid brauchbar.
Herstellung von 5-Methoxy-3-[piperidyl-(4)]-indol -,n
a) 5-Melhoxy-3-[pyridyl-(4)]-indol
Das 5-Methoxy-3-[pyridyl-(4)]-indol wird nach dem in Beispiel 1 der FR-PS 15 87 692 angegebenen Verfahren
hergestellt.
b) 1 -Benzyl-4-[5'-methoxy-indolyl-(3')]-pyridiniumbromid
Eine Mischung aus 11 g 5-Methoxy-3-[pyridyl-(4)]-indol,
9,5 g Benzylbromid und 120 ml Äthylacetat wird 4 bo
Stunden lang unter Rückfluß zum Sieden erhitzt. Die Mischung wird abgekühlt, der erhaltene Niederschlag
abfiltriert, mit Äthylacetat gewaschen und im Vakuum getrocknet.
Man erhält 17.5g l-Benzyl-4-[5'-methoxy-indolyl- Μ
(3')]-pyridiniumbromid; F.: 254°C.
Das Produkt wird ohne weitere Reinigung für die folgende Stufe verwendet:
.·) 3-[1 '-Benzyl-1 '.2'3',6'-tetrahydropyridyl-(4')]-5-methoxy-indo!
17,5 g 1 -BenzyM^S'-Methoxy-indolyl-p'^-pyridiniumbromid
werden bei 400C in 150 ml Methanol und 65 ml Wasser gegeben und in kleinen Portionen mit
3,5 g Natriumborhydrid versetzt. Die Mischung wird 24 Stunden lang bei Raumtemperatur gerührt bzw. bewegt
und dann mil 150 ml Wasser versetzt.
Der gebildete Niederschlag wird abgesaugt bzw. abgeschleudert, mit Wasser gewaschen, getrocknet und
aus Methanol umkristallisiert.
Man erhält 13 g 3-[r-Benzyl-1\2\3',6'-tetrahydro-pyridyl-(4')]-5-methoxy-indol;
F.: 168=C.
Summenformel: C2,H22N2O; M = 318,30
Berechnet: C 79.21. H 6,96, N 8,80%:
gefunden: C 78,9. H 6,9, N 8,7%.

d) 5-Methoxy-3-[piperidyl-(4)]-indol
12,5 g 3-[1 '-Benzyl-1 \2',3',6'-tetrahydro-pyridyl-(4')]-5-methoxy-indol
und 3 g Palladium auf Kohle (10%ig) werden in 300 ml Äthanol gegeben. Man erwärmt auf
50°C und läßt von der Mischung 1770 cmJ Wasserstoff
in 7 Stunden absorbieren. Nach Beendigung der Reaktion wird der Katalysator abfiltriert und der
Äthanol un'er vermindertem Druck zur Trockne abgedampft. Man erhält 8.3 g 5-Methoxy-3-[piperidyl-(4)]-indol;F.:170°C.

Berechnet: C 73,01. H 7,88, N 12.17%:
gefunden: C 73,0. H 7,6, N 12.1%.

Herstellung von 2-Methyl-6-methoxy-3-[piperidyl-(4)]-indol
a) 2-Methyl-6-methoxy-3-[pyridyl-(4)]-indol
50 g 2-Methyl-6-methoxy-indol werden in 230 ml Pyridin gegeben. Man kühlt auf —40°C ab und gibt
tropfenweise 87 g Benzoylchlorid hinzu. Die Mischung wird 3 Tage lang bei Zimmertemperatur unter
Lichtabschluß gerührt bzw. bewegt. Man verdampft dann das Pyridin unter vermindertem Druck, wäscht mit
Natronlauge und verdampft das Wasser. Das erhaltene Produkt wird in 500 ml siedendem Methanol gelöst,
dann unter Rühren mit 200 ml Natriumhydroxid und schließlich zweimal mit 200 ml Wasser versetzt. Man
rührt 2 Stunden lang, läßt eine Nacht lang ruhen, verdampft das Methanol unter vermindertem Druck
und extrahiert die wäßrige Phase mit Chloroform. Die organische Phase wird über Natriumsulfat getrocknet
und unter vermindertem Druck eingedampft. Man erhält 100 g Produkt, das durch Chromatographie an
Aluminiumoxid unter Elution mit Benzol gereinigt wird; nach Abdampfen des Elutionsmittels erhält man 34 g
Produkt, das aus Acetonitril umkristallisiert wird. Man erhält 19,5 g 2-Methyl-6-methoxy-3-[pyridyl-(4)]-indol;
F.:196bis198°C.
Summenformel: Ci5H I4N2O; M = 238.29
Berechnet: C 75,60, H 5,92, N 11,76%;
gefunden: C75.52, H 6,13, N 11,91%.

b) 1 -Benzyl-4-[2'-methyl-6'-methoxyindolyl-(3')]-pyridiniumbromid
3.4 g 2-Methyl-6-methoxy-3-[pyridyl-(4)]-indol und 2.65 g Benzylbromid werden in 30 ml Äthylacetat
gegeben. Man erhitzt 4 Stunden lang unter Rückfluß zum Sieden. Man kühlt ab, saugt bzw. schleudert die
gebildeten Kristalle ab und wäscht diese mit Äthylacetat. Nach dem Trocknen erhält man 5.7 g 1 -Benzyl-4-[2'-methyl-6'-methoxy-indolyl-(3')]-pyridiniunibrornid;
F.: 244 bis 246° C.
Summenformel:C22H2|BrN2O; M = 409,34
Berechnet: C 64,55, H 5,17, N 6,84%;
gefunden: C 64,36, H 5,33, N 6.75%.

c) 3-[l'-BenzyI-1',2',3',6'-tetrahydropyridyl-(4')]-2-methyl-6-methoxy-indol
20,1 g 1-BenzyI-4-[2'-methyl-6'-methoxy-indolyl-(3')]-pyridiniumbromid
werden bei 35 bis 400C in 165 ml Methanol und 70 ml Wasser gegeben. Man kühlt auf
25°C ab und gibt in kleinen Portionen, ohne daß die Temperatur 30 bis 35°C übersteigt, 4 g Natriumborhydrid
hinzu.
Die Mischung wird 2 Stunden lang bei Raumtemperatur gerührt bzw. bewegt und dann mit Wasser versetzt
und der gebildete Niederschlag abgesaugt bzw. abgeschleudert und mit Wasser gewaschen. Nach
Trocknen unter vermindertem Druck erhält man 15,4 g 3-[r-Benzyl-1',2',3',6'-tetrahydro-pyridyl-(4')]-2-methyl
6-methoxy-indol; F.: 142 bis 143°C.
Summenformel:C22H24N2O; M = 332,45
Berechnet: C 79,48, H 7,28, N 8,43%;
gefunden: C 79,40, H 7,44, N 8,35%.

d) 2-Methyl-iS-methoxy-3-[piperidyl-(4)]-indol
24 g 3-[r-Benzyl-r,2',3',6'-tetrahydro-pyridyl-(4')]-2
κι methyl-6-methoxy-indol und 4 g Palladium auf Kohle
(10%ig) werden in 300 ml absolutes Äthanol gegeben Man erwärmt auf 50°C und leitet Wasserstoff ein. In 12
Stunden werden 2'45Ocm3 Wasserstoff absorbiert. Dei
Katalysator wird abfiltriert und erneut 3,0 g Palladiurr auf Kohle (10%ig) hinzugegeben. Die Behandlung mil
Wasserstoff wird etwa 3' Ii Stunden lang fortgesetzt
wonach 3250 cm3 Wasserstoff absorbiert sind. Mar filtriert den Katalysator ab, verdampft das Äthano
unter vermindertem Druck und erhält 16,6 g 2-Methyl· 6-methoxy-3-[piperidyI-(4)]-indol;F.: 166°C.
Summenformel: C5H20N2O; M =244,34
Berechnet: C73;,73, H 8,25, N 11,47%;
gefunden: C 73,6, H 8,3, N 11,5%.
gefunden: C 73,6, H 8,3, N 11,5%.
Claims (3)
1. 3-[Piperidyl-(4)]-indole der allgemeinen Formel!

mit R = H oder Methoxy und r>
R11R2 = H oder Methyl,
wobei R, Ri und R2 nicht zugleich H bedeuten.
2. Verfahren zur Herstellung der 3-[Piperidyl-(4)]-indole der Formel 1 nach Anspruch 1, dadurch
gekennzeichnet, daß man in an sich bekannter Weise >
<> ein 3-[Pyridyl-(4)]-indol der allgemeinen Formel IV
Verbindung der allgemeinen Formel Vl
in der R und R2 wie oben definiert sind —
gegebenenfalls nach Methylierung des Indol-Stickstoffatoms mit einem Methylhalogenid in Gegenwart von Natriumhydrid — mit Wasserstoff in
Gegenwart eines Pd-Katalysators hydriert
3. Verwendung der 3-[Piperidyl-(4)]-indoIe der Formel I nach Anspruch I zur Herstellung von
4-[Indolyl-(3)]-piperidinoalkyl-arylketonen.
