•Rapamycin und seine Herstellung.
Die Erfindung "betrifft ein Antibiotikum, eine neue Verbindung,
Rapamycin genannt, und ein Verfahren zu seiner Herstellung.
Das erfindungsgemäße Antibiotikum läßt sich leicht anhand
seiner ausgeprägten antifungisehen Wirksamkeit und seiner relativ geringen Toxizität von bekannten Verbindungen
seiner Klasse unterscheiden.
Das in der vorliegenden Beschreibung angegebene Ultraviolett
Spektrum von Rapamycin zeigt an, daß die Verbindung zur Klasse der Antibiotika gehört, die als Trien-Antibiotika
bekannt sind. In dieser speziellen Klasse sind bisher nur fünf Verbindungen beschrieben worden.

Trienin, A. Aszalos et al., J. Antibiotics, j21_, 611 (1968),
ist ein Trien-Antibiotikum mit Antitumorwirksamkeit, das auch "bemerkenswerte Wirksamkeit gegenüber grampositiven
Organismen und nur geringe Wirksamkeit gegenüber Candida-Stämmen
aufweist. Das von J. J. Armstrong et al., Nature, 206, 399 (1965), beschriebene antifungische Trien und das
von C. Coronelli et al., J. Antibiotics, _20, 329 (1967),
beschriebene Mycotrienin sind vermutlich identisch. Beide haben geringe antifungische Wirksamkeit (MIC gegenüber
C. albicans: 5/ig/ml) und weisen hohe Toxizität auf (DD1-Q
bei Mäusen: 15 mg/kg). Die beiden restlichen Antibiotika - Resistaphylin, S. Aezaiva et al., J. Antibiotics, 24,
393 (1971) und Proticin, G. Nesemann et al., Naturwissenschaften,
_59, 81 (1972) - lassen sich leicht von der Verbindung
der vorliegenden Erfindung unterscheiden, da sie antibakterielle aber keine antifungische Wirksamkeit aufweisen.
Rapamycin ist eine chemische Verbindung, die sich durch Züchtung eines Rapamycin erzeugenden Organismus in einem
wässrigen Nährmedium herstellen läßt. Die Verbindung vermag dem Wachstum von Fungi, z.B. von Candida albicans und
Micorsporum gypseum entgegenzuwirken. Rapamycin kann daher zur Verhütung des Wachstums oder zur Verringerung der Zahl
bestimmter Fungi in verschiedenem Milieu verwendet werden.
Der für die vorliegende Erfindung verwendete Rapamycin erzeugende Organismus, Streptomyces hygroscopicus NRRL 5491,
wurde aus dem Boden der Osterinseln gewonnen und Proben davon sind ohne Vorbehalt bei der Northern Utilization
and Research Division, Agricultural Research Service, U.S. Department of Agriculture, Peario, Illinois, USA., hinterlegt
worden.
Es ist klar, daß die Erfindung nicht auf die Verwendung des

hier beschriebenen speziellen Organismus beschränkt ist
sondern auch durch natürliche Selection oder durch Behandlung des MierοOrganismus mit beispielsweise UV-Strahlung,
Eöntgen-Strahlung, U-Methyl-N'-nitro-N-nitroso-guanidin, Manganchlorid,
Kampfer, Stickstofflost und dergleichen erhaltene Varianten und Mutanten sowie Polyploide der verschiedenen
Mutanten umfaßt.
Streptomyces hygroscopicus NRKL 5491 entwickelt sich reichlich
in Kulturmedien, die üblicherweise für die Züchtung anderer Organismen der gleichen Gattung verwendet werden.
Dieser Organismus vermag bei Temperaturen im Bereich von 2Obis35°C, vorzugsweise bei etwa 280C, auf Czapek1 s-Agar,
Glucose-Aspargin-Agar, Glyzerin-Asparagin-Agar, Stärke—
Agar und Pepton-Rindfleisch-Agar zu wachsen. Der Organismus
gedeiht auch, sehr gut auf Hefeextrakt-Agar, Malzextrakt-Agar,
Stärke-anorganische Salze-Agar, Hafermehl-Agar,
Hafermehl-Iomaten-Agar und Bennet1 s-Agar. Auf Kartoffelscheiben
gibt es kein Luftmycel, der- Substratwuchs ist
jedoch gut entwickelt und hautfarben. Auf allen Medien ist der Luftwuchs zuerst weiß, dann graulich, mit schwarzen
Flecken. Die Sporophoren sind oft kompakt und bilden eine Spirale aus mehr als zehn Sporen. Der Substratwuchs ist
hellgelb bis fast farblos und bei einigen Medien blaßbraun. Gelegentlich wird ein gelbliches Pigment gebildet.
Der Organismus ist H^S- und melaninnegativ.
Die Kohlenhydratverwertung durch Streptomyces hygroscopicus
NRRL 5491 wurde bei Kohlenstoff-Verwertungsagar (ISP Medium 9) nach dem durch "International Streptomycee Project (ISP)"
standardisierten Verfahren untersucht.
Die am besten verwerteten Kohlenhydrate waren D-Glucose,
Inosit, D-Eructose und D-Mannit; weniger gut verwertete Kohlenhydrate waren Rhamnose, Raffinose, Xylose, Stärke

und Arabinose. Die Kohlenhydrate Saccharose und Cellulose wurden nicht verwertet.
Die Milieu- und Ernährung st e dingung en für die Fermentierung
von Streptomyces hygroscopicus NRRL 5491 entsprechen denjenigen, die für die Erzeugung von Antibiotika durch andere
aerobe Microorganismen erforderlich sind. Aerobiose kann somit in einem flüssigen Währmedium, angeimpft mit einer
sterilen Kultur, bebrütet in Kolben, die sioh auf Schüttelmaschinen befinden, unterhalten werden. Für die industrielle
Erzeugung können Metalltanks mit Innenbelüftung und Rührung
mit Hilfe von JFlügelrührern verwendet werden. Rapamycin
wird auch von Oberflächenkulturen erzeugt. Der Microorganismus benötigt als Nährstoffelemente assimilierbaren Kohlenstoff
und organische stickstoffhaltige Substanzen. Die Anwesenheit von Mineralsalzen ist wünschenswert. Die
Züchtung geht am besten wenn der Anfangs-pH des Kulturmediums zwischen 6,5 und 7,5 liegt, der günstigste pH
liegt bei 6,8 - 7,3.
Die brauchbaren Quellen für assimilierbaren Kohlenstoff zur Erzeugung des Antibiotikums sind sehr verschieden,
dazu gehören Zucker (z.B. Glucose, D-Fructose, D-Mannit, Maltose, Arabinose, Rhamnose, Raffinose, Xylose und dergleichen),
Dextrin, Stärken verschiedenen Ursprungs, Glyzerin (und andere Polyalkohole), Inosit und tierische
und pflanzliche Fette, sowie deren Ester. Die Quellen für organischen assimilierbaren Stickstoff, die das Wachstum
aktiv stimulieren und die Bildung von Rapamycin begünstigen sind Substanzen» wie Sojaschrot, Baumwollschrot
und andere pflanzliche Schrotarten (Vollschrot oder teilweise oder ganz entfettet), Fleischmehle oder tierische
Eingeweide, verschiedene Peptone, Caseinhydrolysate, Sojabohnenhydrolysate,
Hefehydrolysate, lactalbumin, Weizen-

glutine, lösliclie Brennereirückstände, Maisquellwasser,
Melasse, Harnstoff und Aminosäuren.
Mineralsalze, z.B. die Chloride, Nitrate, Sulfate, Carbonate und Phosphate von Natrium, Kalium, Ammonium und
Calcium sollten in den angemessenen Konzentrationen vorhanden sein. Das Nährmedium sollte auch Spurenelemente,
wie Magnesium, Eisen, Mangan und Zink enthalten.
Das obige Medium wird für die 3?ermentierung mit einer frischen Kultur von Streptomyces hygroscopieus angeimpft.
Unter den beschriebenen Bedingungen und bei der Züchtungstemperatur von etwa 20 - 350C, vorzugsweise bei 250C, erzielt
man eine maximale Bildung von Rapamycin in etwa 2 bis etwa 8 Tagen in lanks.
Anschließend können verschiedene Verfahren für die Isolierung und Reinigung des Rapamycins angewendet werden,
z.B. Lösungsmittelextraktion, Verteilungschromatographie,
Chromatographie an Silicagel, Flüssigkeits-Flüssigkeitsverteilung
in einer Craig-Vorrichtung und Kristallisation aus Lösungsmitteln. LÖsungsmittellextraktionsverfahren
bevorzugt man für die technische Gewinnung,soweit sie einen geringeren Zeitaufwand erfordern und nicht so teuer
sind.
Allgemein gesagt kann' Rapamycin nach einer der folgenden
Methoden gewonnen werden.
a) Die Permentierungsmischung wird mit einem Lösungsmittel
extrahiert, das mit Wasser im wesentlichen nicht mischbar ist, vorzugsweise mit einem Niedrigalkanol,
z.B. mit n-Butanol, n-Pentanol oder dem technischen Pentanol-

gemisch, bekannt als "Pentasol" oder mit n-Hexanol oder
einem im wesentlichen mit Wasser nicht mischbaren Niedrigalkyl-niedrigalkanoat,
z.B. mit Äthylacetat, Butylacetat, Amylacetat oder dem technischen Amylacetatgemisch oder
mit einem mit Wasser.im wesentlichen nicht mischbaren halogenierten aliphatischen Kohlenwasserstoff, z.B. mit
Chloroform, Methylenchlorid oder Dichloräthan. Die Extrakte werden getrocknet und unter vermindertem Druck eingeengt
und ergeben einen öligen Rückstand, der wieder mit einem mit Wasser mischbaren Lösungsmittel, vorzugsweise mit
einem Niedrigalkanol, z.B. mit Methanol oder Äthanol
extrahiert wird. Diese letztgenannten Extrakte werden durch Diatomeenerde ("Celite") filtriert und das Filtrat
wird unter vermindertem Druck eingeengt, wobei man einen öligen Rückstand erhält, der rohes Rapamycin enthält.
b) Die Termentierungsmischung wird, durch ein Kissen
aus Diatomeenerde ("Gelite") filtriert und der das Mycel
enthaltende Filterkuchen wird,wie unten unter (c) beschrieben,
extrahiert. Das Filtrat, das heißt,die mycelfreie Fermentierungsmischung
wird mehrmals mit einem im wesentlichen mit Wasser nicht mischbaren Lösungsmittel extrahiert,
z.B. mit einem Niedrigalkanol, Niedrigalkyl-niedrigalkanoat oder mit einem halogenierten aliphatiüchen Kohlenwasserstoff,
wie oben unter Abschnitt (a) exemplifiziert. Man trocknet die Extrakte und engt unter vermindertem Druck
ein, dabei erhält man einen öligen Rückstand, der mit einem mit Wasser mischbaren Lösungsmittel, vorzugsweise
mit einem Niedrigalkanol, z.B. mit Methanol oder Äthanol extrahiert wird. Die letztgenannten Extrakte werden in
gleicher Weise, wie oben unter (a) beschrieben, behandelt und ergeben einen öligen Rückstand,der das rohe Rapamycin
enthält.

c) Man trennt das Mycel von der Fermentierungsmischung
ab und sxtrahiert mit einem geeigneten mit Wasser mischbaren Lösungsmittel, vorzugsweise mit einem Niedrigalkanol,
z.B. mit Methanol oder Äthanol. Der Extrakt wird durch Verdampfen bis zur wässrigen Phase eingeengt, die
wiederum mit einem im wesentlichen mit Wasser nicht mischbaren Lösungsmittel extrahiert wird, z.B. mit einem Medrigalkyl-niedrigalkanoat,
mit einem halogenierten aliphatischen Kohlenwasserstoff oder einem im wesentlichen mit
Wasser nicht mischbaren Medrigalkanol, wie oben beschrieben,
oder mit einem aromatischen Kohlenwasserstoff, z.B. mit Benzol oder Toluol. Der letztere Extrakt wird unter
vermindertem Druck eingedampft und ergibt einer, öligen
Rückstand, der das rohe Rapamycin enthält.
Das rohe Rapamycin, erhalten nach einem der in den Abschnitten (a), (b) oder (c) beschriebenen Verfahren, wird
dann nach verschiedenen Methoden gereinigt, siehe z.B. die oben genannten Methoden. Zu bevorzugten Methoden gehörten
die Absorption des rohen Rapamycins auf einem Absorptionsmittel, z.B. Kohle oder Silicagel, aus einer Lösung in
einem im wesentlichen nicht polaren ersten Lösungsmittel und die anschließende Eluierung mit einem zweiten Lösungsmittel,
das polarer ist als das erste Lösungsmittel.
Rapamycin ist brauchbar als antifungisch.es Mittel gegen
eine Vielzahl pathogener Fungi; z.B. gegen Candida albicans und andere Candida Spezien, gegen Microsporum gypseum,
Trichophyton mentogrophytes, Aspergillus sp. und Sporotrichum sp..
Die Hemmwirkung von Rapamycin ist besonders ausgeprägt gegenüber Candida albicans und dieser letztgenannte Organismus
kann in vorteilhafter Weiee für Prüfungszwecke verwendet werden.

Die antifungische Wirksamkeit der vorliegenden Verbindung läßt sich in Standardtests, die für diese Zwecke verwendet
werden, demonstrieren, z.B. anhand von Tests, die in "Antiseptics, Disinfectants, Fungicides and Sterilization",
G-.P. Reddish, Ed., 2. Auflage, Lea und Febiger, Philadelphia,
1957 oder von D.O. Grove und W.A. Randall in "Assay Methods of Antibiotics", Med. Encyel., Inc., New York 1955, beschrieben
sind.
Wird das erfindungsgemäße Antibiotikum als antifungisches Mittel bei warmblütigen Lebewesen, z.B. bei Ratten, verwendet,
so kann es alleine oder in Kombination mit pharmazeutisch verträglichen Trägern verwendet werden, deren
Anteil durch die Löslichkeit und chemische Natur der Verbindung, den gewählten Verabreichungsweg und die biologische
Standardpraxis bestimmt wird. Beispielsweise kann eine antifungisch wirksame Menge des Antibiotikums oral
in einer festen Form, die Verdünnungsmittel wie Stärke, Zucker, bestimmte Tontypen usw. enthält, verabreicht werden.
Ebenso kann eine solche Menge auch oral in Form von Lösungen oder Suspensionen verabreicht werden oder das Antibiotikum
kann parenteral injiziert werden. Für die parenterale Verabreichung
kann das Antibiotikum in Form einer sterilen Lösung oder Suspension verwendet werden, die andere gelöste
Substanzen oder Suspendiermittel enthält, z.B. so viel Salz oder Glucose, daß die Lösung isotonisch ist,
Gallensalze ("bile salts"), Akazin, Gelatine, Sorbitanmonoleat, Polysorbat-80 (Sorbit-Oleatester und seine Anhydride
kopolymerisiert mit Ä'thylenoxyd) und dergleichen.
Die Dosis des erfindungsgemäßen Antibiotikums schwankt in Abhängigkeit von der Form der Verabreichung und der gewählten
speziellen Verbindung. Sie richtet sich auch nach dem speziellen zu behandelnden Patienten. Im allgemeinen

wird die Behandlung mit kleinen Dosen, die wesentlich kleiner als die optimale Dosis der Verbindung sind, begonnen.
Danach wird die Dosis in kleinen Schritten erhöht bis der optimale Effekt unter den gegebenen Umständen erreicht ist.
Im allgemeinen wird die Verbindung der vorliegenden Erfindung am liebsten bei einer Konzentration angewendet,
die gute antifungische Ergebnisse liefert,ohne nachteilige
oder schädliche Nebenwirkungen zu verursachen, vorzugsweise wird sie in einer Menge im Bereich von etwa 1,0 mg bis
etwa 250 mg pro kg pro Tag angewendet, obgleich - wie oben
erwähnt - Variationen vorgenommen werden können. Eine Dosis im Bereich von etwa 10 mg bis etwa 100 mg pro kg
pro lag wird jedoch am liebsten zur Erzielung guter Ergebnisse angewendet..
Das Mittel kann außerdem topisch angewendet werden. Pur
die topische Anwendung kann es in die Form von Lösungen, Crems oder Lotionen in pharmazeutisch verträglichen Verdünnungsmitteln,
die 0,1 - 5%, vorzugsweise 2$ von dem
Mittel enthalten, formuliert werden und auf die befallene
Hautstelle topisch aufgebracht werden.
Rapamycin kann auch verwendet werden, um Laboratoriumsgeräte, chirurgische Instrumente, Umkleideräume oder
Duschräume von PungusOrganismen zu reinigen und zu desinfizieren. Pur solche Zwecke verwendet man vorzugsweise
0,1 - 10j£ige Lösungen von Rapamycin in einem Sfiedrigalkanol,
vorzugsweise in Methanol, verdünnt mit 10 - 100 Volumina Wasser, das 0,001 - 0,1$ eines nicht ionischen oberflächenaktiven
Mittels, z.B. Polysorbat 80 USP enthält, unmittelbar bevor man es auf die zu reinigenden und zu desinfizierenden
Gegenstände aufbringt.
Nach einer bevorzugten Ausführungsform der vorliegenden Erfindung
wird Rapamycin auf folgende Weise hergestellt:

Ein geeignetes Fermentationsgefäß wird mit Produktionsmedium 8EM (siehe Beispiel 1) beschickt. Nach dem Sterilisieren
und Abkühlen wird das Medium mit einem Impfpräparat der ersten Stufe von Streptomyces hygroscopicus angeimpft.
Ein maximaler Gehalt von 20 - 100 jug/ml Antibiotikum wird
nach 2-8 Tagen, gewöhnlich nach etwa 5 Tagen, in der Fermentationsmischung erreicht, was mit Hilfe der Becherplattenmethode
und unter Verwendung von Candida albicans als TestOrganismus bestimmt wird. Das Mycel wird durch
Filtration über Diatomeenerde isoliert. Dann wird das Rapamycin mit einem wassermischbaren Lösungsmittel, z.B.
mit einem Niedrigalkanol, vorzugsweise mit Methanol oder Äthanol, aus dem Mycel extrahiert. Der letztere Extrakt
wird dann eingeengt, vorzugsweise unter vermindertem Druck, und die resultierende wässrige Phase wird mit einem mit
Wasser nicht mischbaren Lösungsmittel extrahiert. Ein bevorzugtes mit Wasser nicht mischbares Lösungsmittel für
diesen Zweck ist Methylenchlorid, man kann aber auch Chloroform, Tetrachlorkohlenstoff, Benzol, n-Butanol und dergleichen
verwenden. Der letztere Extrakt wird eingeengt, vorzugsweise unter vermindertem Druck, und ergibt das Rohprodukt
als öl.
Das Produkt kann auf verschiedene Weise weiter gereinigt werden. Eine bevorzugte Reinigungsmethode ist die, das
Rohprodukt in einem im wesentlichen nicht polaren ersten Lösungsmittel, z.B. in Petroläther oder Hexan zu lösen
und die erhaltene Lösung mit einem geeigneten Absorbens, z.B. mit Kohle oder Silicagel, zu behandeln, so daß das
Antibiotikum auf dem Absorptionsmittel absorbiert wird. Das Absorptionsmittel wird dann abgetrennt und mit einem
zweiten Lösungsmittel, das polarer als das erste Lösungsmittel ist, z.B. mit Äthylacetat, Methylenchlorid oder

einer Mischung aus Methylenchlorid und Äther (bevorzugt),
gewaschen oder eiuiert. Danach erhält man beim Einengen der Waschlösung oder des Eluates im wesentlichen reines
Rapamycin. Eine weitere Reinigung wird durch partielle Ausfällung mit einem nicht polaren Lösungsmittel, z.B. mit
Petroläther, Hexan, Pentan und dergleichen, aus einer lösung des Rapamycins in einem polareren Lösungsmittel, z.B.
Äther, Äthylacetat, Benzol und dergleichen, erzielt. Eine noch weitere Reinigung erzielt man durch Kolonnenchromatographie,
vorzugsweise unter Verwendung von Silicagel,, und durch Kristallisation des Rapamycins aus Äther=
a) Gereinigtes Rapamycin ist eine farblose kristalline Verbindung mit einem Schmelzpunkt von 1-33 - 1850C nach
Umkristallisation aus Äther?

Absorptionsmaxima in seinem Ultraviolettabsorptionsspektrum auf (95$ Äthanol):
267 nm (E^m 4H), 277 nm (E^m 541) und 288 nm (E1Jn 416);
f) Das InfrarotabsorptionsSpektrum von Rapamycin
in Chloroform ist in der "beiliegenden Figur 1 wiedergegeben, es zeigt charakteristische Absorptionsbanden bei 3560, 3430,
1730, 1705 und 1630 - 1610 cm"1;
weitere Infrarotabsorptionsbanden sind durch die folgenden Zahlenwerte, die in cm" angegeben sind, gekennzeichnet,
wobei (s) eine starke, (m) eine mittlere und (w) eine schwache Bande bezeichnet. Diese Klassifizierung ist willkürlich
gewählt, so daß eine Bande als stark (s) bezeichnet wird, wenn ihre Spitzenabsorption zu mehr als 2/5 über dem
Untergrund im gleichen Gebiet liegt; als mittelmäßig (m) wird eine Bande bezeichnet, wenn ihr Peak zwischen 1/3 und

i) Rapamycin weist eine ItD1-Q (i«p.» Maus) von
597,3 ± 28,1 mg/kg und eine LD50 (p.o., Maus) von>2500 mg/kg
Bei der Untersuchung der Schutzwirkung wurden Mäuse durch intravenöse Injektion von 0. albicans ATCO 11651 infiziert.
Eine, vier und 24 Stunden nach der Infektion wurde Mäusen 10 mg/kg (s.c.) Rapamycin verabreicht. Mit dieser Dosis
wurden 50$ der Mäuse geschützt. Behandlung mit 25 mg/kg
(s.o.) "bot einen vollständigen Schutz. Wurde Rapamycin

oral in einer Menge von 10 mg/kg verabreicht, so überlebten
4 von 10 Mäusen und mit 25 mg/kg wurde vollständiger Schutz festgestellt.
Eine 1%ige Suspension (0,2 ml) von Rapamycin in Wasser,
das 1,5$ Polysorbat 80 (Tween 80) enthält, verursachte bei
der intradermalen Injektion in das Ohr eines Kaninchens keine Reizung. Ebenso verursachten zwei Tropfen einer 0,5%igen
Suspension, aufgebracht auf das Auge eines Kaninchens, keine Reizung.
Die folgenden Beispiele dienen zur weiteren Veranschaulichung
der vorliegenden Erfindung.
Beispiel 1.
Microorganismus
Streptomyces hygroscopicus NRRL 5491 wurde auf Hafermehl/ Tomatenpaste/Schrägagar (T.G. Pridham, et al., Antibiotic
Annual 1956 - 1957, Medical Encyclopedia Inc., New York, Seite 947) und in Roux-Kolben, die das gleiche Medium enthalten,
gezüchtet und unterhalten. Nach 7-tägiger Brut bei 280C war gutes Wachstum erreicht. Aus einem Roux-Kolben wurden
die Sporen herausgewaschen und in 50 ml sterilem, destilliertem Wasser suspendiert. Diese Suspension wurde zum
Animpfen der ersten Impfkultur verwendet. Das Medium der ersten Impfkultur bestand aus Emerson-Brühe (R.L. Emerson
et al., J. Bacteriol, j>2, 357'(1946)) 0,4%; Pepton, 0,4%;
Natriumchlorid, 0,25%? Hefeextrakt, 0,1%; und Glucose, 1%;
pH = 7,0; Kolben, die das-obige Medium enthielten, wurden mit 1% der oben beschriebenen Sporensuspension angeimpft.
Die angeimpften Kolben wurden 30 Stunden lang bei 280C auf
einer Schüttelvorrichtung, dia auf 65 Upm (Hubläng« 10,16 cm;
4 inch) eingestellt war, bebrütet. '

Die Produktion erfolgte in 250 1 New Brunswiek-Permentern,
Modell 1P-250, ausgestattet mit automatischem Antischaum-Zugabesystem
und pH-Kontrollsehreiber, Die Permenter -wurden
mit 160 1 eines wässrigen Produktionsmediums (8 KM) beschickt,
bestehend aus den folgenden Bestandteilens

Die Permenter wurden bei 1210C 45 Minuten lang sterilisiert,
abgekühlt und mit einem Kolben (2$ inoculum) Impfkultur der ersten Stufe angeimpft. Bruttemperatur: 280Cj Belüftung:
0,5 Volumen/Volumen/Minute; Rührung: 250 Upm.
Innerhalb 5 Tagen wurde ein Gehalt von ca. 20 jug/ml er- reicht,
bestimmt durch microbiologische Prüfung auf Agarplatten mit Candia albicans Saat. Die !fermentation wurde
beendet. :f
Die Extraktion und Isolierung des Antibiotikums erfolgte
nach einer der folgenden Methoden:

a) Die Fermentationsmischung wurde zweimal mit
1 Vol./Vol. n-Butanol extrahiert. Die vereinigten Butanolextrakte
wurden mit 1 VoI./Vol. Wasser gewaschen, mit
wasserfreiem Natriumsulfat getrocknet und unter vermindertem.
Druck zur Trockene eingedampft, wobei man einen Rückstand erhielt. Der ölige Rückstand wurde dreimal mit 2 1 Methanol
extrahiert. Die vereinigten Methanol extrakte wurden durch
Diatomeenerde ("Oelite") geschickt und zur Trockene eingedampft
und ergaben einen öligen Rückstand, der rohes Rapamycin
enthält.
b) Die Fermentierungsmischung wurde über Diatomeenerde
("Celite") filtriert. Das Filtrat wurde zweimal mit
1 Vol./Vol. Äthylacetat extrahiert. Man wusch die Äthylacetatextrakte
mit 1 Volumen Wasser, trocknete mit wasserfreiem Natriumsulfat und dampfte unter vermindertem Druck
zur Trockene' ein. Der Rückstand wurde zweimal mit 1 1 Methanol extrahiert. Die Methanolextrakte wurden unter vermindertem
Druck zu einem öligen Rückstand eingedampft, der rohes Rapamycin enthält. .·>
c) Das gemäß Abschnitt (b) erhaltene Mycel wurde mit 1-2 Volumina Wasser gewaschen. Das gewaschene Mycel
wurde dreimal mit je 5 Volumina Methanol pro Gewichtseinheit
feuchtes Mycel gewaschen. Man vereinigte die methanolischen Extrakte und engte unter vermindertem Druck auf ein
kleines Volumen wässrige Phase ein, die ungefähr 10$ Vol./Vol.
Methanol enthielt. Diese wässrige Phase wurde dreimal mit einem Volumen Methylenchlorid extrahiert; die Methylenchloridextrakte
wurden vereinigt, mit wasserfreiem Natriumsulfat getrocknet und eingedampft und ergaben einen öligen
Rückstand.

Der ölige Rückstand wurde mit 1 Volumen Petroläther verdünnt, dann wurden 30$ (Gewicht/Volumen) Kohle (Darco G60)
zugegeben. Die Mischung wurde 1/2 Stunde läng gerührt und dann filtriert. Die Kohle, die im wesentlichen das gesamte
Produkt zurückhielt, wurde zweimal mit 1 Volumen Petroläther gewaschen, dann wurde die Kohle dreimal mit 5 Volumen
("bezogen auf das Gewicht der Kohle) einer Mischung aus Methylenchlorid und Äther (50:50) eluiert. Die Methylenchlorid/Ätherextrakte
wurden zur Trockene eingedampft und der Rückstand in einer kleinen Menge Äther gelöst. Das rohe
Produkt erhielt man durch Ausfällung aus der Ätherlösung mit kaltem Petroläther.
Andererseits wurde der nach einem der oben beschriebenen Extraktionsverfahren erhaltene ölige Rückstand mit einem
Volumen Hexan verdünnt und durch eine präparative Silicagel G-Kolonne geschickt. Das Produkt wurde auf der Kolonne
adsorbiert.· Das das adsorbierte Produkt enthaltende Silicagel G wurde mit mehreren Volumina Hexan und 50:50 Hexan/
Ä'ther-Gemisehen gewaschen. Das Produkt wurde mit Äther von
der Kolonne eluiert. Das Eluierungsmittäl Äther wurde auf ein kleines Volumen eingedampft und das Rohprodukt durch
Ausfällung mit kaltem Petroläther aus der Ätherlösung erhalten.
Reinigung
Das obige Rohprodukt wurde durch Kolonnenchromatographie an Silicagel G, Merck (50:1 Gew./Vol.) in Hexan/Äther
(50:50) weiter gereinigt. Man eluierte das Produkt mit Äther aus der Kolonne. Das Äthereluat wurde auf ein kleines
Volumen eingedampft und gereinigtes Rapamyoin wurde mit Petroläther ausgefällt. Die Analysenproben wurden durch
Kristallisation aus Äther hergestellt, Fp = 183 - 1850C,

Claims (10)
1. Rapamycin, ein Antibiotikum, eine farblose,
kristalline Verbindung, gekennzeichnet durch
a) einen Schmelzpunkt von 183 - 185°C nach Umkristallisation
aus Äther;
b) Löslichkeit in Äther, Chloroform, Aceton, Methanol
und Dimethylformamid, sehr geringe Löslichkeit in Hexan und Petroläther und praktische Unlösliclökeit in Wasser;
c) einen einheitlichen Pl eck auf Silioagel-Dünnsohichtplatten;
d) folgende durchschnittliche Elemeni;aranalyse:
C, 66,48#; H, 8,84& N, 1,3756;
e) folgende charakteristische Absorptionsmaxima im
UV-Absorptionsspektrum (95%iges Methanol): 267 nm (E^m 417),
277 nm (E^m 541) und 288 nm (p\*m 416);
f) ein Infrarotabsorptionsspektrum gemäß beiliegender
Figur 1;

g) ein NMR-Spektrum gemäß "beiliegender Figur 2j
h) eine Mindesthemmkonzentration von 0,02 - 0,1 /ig/ml
gegenüber Candida albieans; und
i) eine LD50 (i.p., Maus) von 597,3 £ 28,1 mg/kg
und eine ID50 (p.o„, Maus) von>
2500 mg/kg.
2. Pharmazeutisches Mittel enthaltend eine antifungiseh
wirksame Menge Antibiotikum nach Anspruch 1 und einen pharmazeutisch verträglichen Träger.
3. Verfahren zur Herstellung von Rapamyein, dadurch gekennzeichnet, daß man Streptomyces hygroscopicus NRRL
5491 in einem wässerigen Nährmedium, das eine Quelle für
assimilierbaren Kohlenstoff und Stickstoff und Mineralsalze enthält, unter aeroben Bedingungen züchtet, bis die
Fermentationsmischung durch Erzeugung von Rapamycin eine wesentliche antifungische Wirksamkeit aufweist und daß
man das Rapamycin aus der Fermentationsmisohung isoliert.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß die Züchtung bei einer Temperatur im Bereich von 20 bis
350O und bei einem Anfangs-pH zwischen 6,5 und 7,5 durchgeführt
wird.
5. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß die Isolierung von Rapamycin durch Extraktion der Fermentationsmischung
mit einem mit Wasser im wesentlichen nicht mischbaren Lösungsmittel, Einengen des so erhaltenen
Extraktes auf einen Rückstand, Extraktion des Rückstandes mit einem wassermischbaren Lösungsmittel, Filtration des
letzteren Extraktes, Einengen des Filtrates und Reinigen des so erhaltenen rohen Rapamycins erfolgt.

6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß die Reinigung des rohen Rapamycins durch Auflösen des
rohen Rapamycins in einem im wesentlichen nicht polaren ersten lösungsmittel und Behandeln der resultierenden
Lösung mit einem Absorptionsmittel, Abtrennen des Absorptionsmittels,
Eluieren des Rapamycins aus dem Absorptionsmittel mit einem zweiten Lösungsmittel, das polarer
als das erste Lösungsmittel ist, und Einengen des Eluates erfolgt.
7. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß die Isolierung des Rapamycins durch Abtrennen
des Mycels von der Fermentationsmischung, Extrahieren der mycelfreien Fermentationsmischung mit einem mit Wasser
nicht mischbaren Lösungsmittel, Einengen des so erhaltenen Extraktes zu einem Rückstand, Extrahieren des Rückstandes
mit einem wassermischbaren Lösungsmittel, Filtrieren des letzteren Extraktes, Einengen des Filtrates und Reinigen
des so erhaltenen Rapamycins erfolgt.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, daß das rohe Rapamycin gereinigt wird, indem man das
rohe Rapamycin in einem im wesentlichen nicht polaren ersten Lösungsmittel löst, die erhaltene Lösung mit einem
Absorptionsmittel behandelt, das Absorptionsmittel abtrennt, das Rapamycin aus dem Absorptionsmittel mit einem zweiten
Lösungsmittel, das polarer als das erste Lösungsmittel ist, eluiert und das Eluat einengt.
9. Verfahren nach Anspruch 5, dadurch gekennzeichnet,
daß man das Rapamycin isoliert, indem man das Mycel
von der Fermentationsmischung abtrennt, das Mycel mit einem wassermischbaren Lösungsmittel extrahiert, den so erhaltenen
Extrakt auf die wässerige Phase einengt, die wässerige

Phase mit einem mit Wasser nicht mischbaren Lösungsmittel
extrahiert, den letzteren Extrakt einengt und das so erhaltene rohe Rapamycin reinigt.
10. Verfahren nach Anspruch 9, dadurch gekennzeichnet,
daß man das rohe Rapamycin reinigt, indem man das rohe Rapamycin in einem im wesentlichen nicht polaren ersten
Lösungsmittel auflöst, die erhaltene Lösung mit einem Absorptionsmittel behandelt, das Absorptionsmittel abtrennt,
das Rapamycin mit einem zweiten Lösungsmittel, das polarer als das erste Lösungsmittel ist, aus dem Absorptionsmittel
eluiert und das Eluat einengt.
