Die Erfindung betrifft ein Verfahren zur Gewinnung des Enzyms Creatinin-amidohydrolase aus Mikroorganismen.
In der klinischen Chemie, besonders für die Funktionsdiagnostik der Niere, spielt die Bestimmung der
Zwischen- und Endprodukte des Eiweißstoffwechsels eine wichtige Rolle. Hierzu zählen auch Creatinin und
Creatin. Die Bestimmung von Creatinin wurde daher schon wiederholt beschrieben. Die meisten der bekannten
Methoden basieren auf der nichtenzymatischen laffe-Reaktion, die jedoch den Nachteil hat, unspezifisch
zu sein.
Weiter wurde bereits eine spezifische mikrobiologische Bestimmungsmethode .nit isolierten Bakterien
beschrieben, wobei gewaschene Zellsuspensionen zur Creatinin-Messung verwendet wurden und die Bildung
von Harnstoff und Ammoniak als Maß der Enzymwirkung herangezogen wurde. Lösliche, zum Creatininabbau
befähigte En;:ymextrakte konnten dabei jedoch nicht erhalten werden. Voraussetzung für die Schaffung
einer spezifischen Creatinin-Bestimmung mit Hilfe von Enzymen war jedoch die Auffindung von geeigneten,
löslichen Enzymen, welche spezifische und meßbare Umsetzungen des Creatinins katalysieren können.
Roche, Lacombe und Girard (BBA 6, 210, 1950) charakterisierten in zwei Pseudomonas-Arten
Creatinase, Creatininase und eine Glycocyaminase als spezifische Enzyme, die aus ihren Substraten aus der
Guanidinogruppe Harnstoff in Freiheit setzten. Weiter wurde von A k a m a t s u und Mitarbeiter (Enzymologia,
15, Seiten 122, 158, 173, 1951) in Erdbakterien ein als Creatinmutase bezeichnetes Enzym, welches die
Gleichgewichtseinstellung zwischen Creatinin und Creatin bewirken soll, festgestellt. Hierbei wird
folgender Abbauweg des Creatinins vermutet:

In der gleichzeitig unter der internen Nummer 1749 eingereichten Patentanmeldung P 2i 22 298-41 wird ein
Verfahren zur Trennung und Gewinnung von zwei Enzymen beschrieben, die erstmals aufgefunden und isoliert
winden, welche an diesem Abbauweg entscheidend beteiligt sind. Die Enzyme wurden aus Mikroorganismen
isoliert. Gegenstand der vorliegenden Erfindung ist die Auffindung von Mikroorganismen, welche sich
besonders als Ausgangsmaterial für die Gewinnung des dort beschriebenen neuen Enzyms Creatinin-amidohydrolase
eignen.
Es wurde schon wiederholt versucht, mit Hilfe von Mikroorganismen Creatinin enzymatisch abzubauen. Es
gelang jedoch nicht wasserlösliche Enzymextrakte zu gewinnen, die Creatinin in ausreichendem Maße abbauen
konnten. Derartige Versuche wurden beispielsweise mit Corynebakterium, Pseudomonas aeruginosa,
Pseudomonas ovalis und Pseudomonas eisenbergii und Clostridien durchgeführt
Aufgabe der Erfindung war es daher, Mikroorganismen zu finden und zu züchten, welche eine zur Gewinnung
des obenerwähnten neuen Enzyms geeignete wasserlösliche Aufschlußfraktion liefern.
Gelöst wird diese Aufgabe erfindungsgemäß dadurch, daß zur Gewinnung von Creatinin-amidohydrolase die
Mikroorganismen Alcaligenes spec. WS 51 400 oder Penicülium WS 90 001 verwendet werden, die bei der
DSM nachhinterlegt wurden.
Alcaligenes spec. WS 51 400 besitzt folgende Eigenschaften:
strikt oxydative gramnegative Kurzstäbchen mit peritricher Begeißelung. Die Keime sind schwach
oxydase-positiv, alkalisieren Lackmusmilch und sind zur Denitrifikation befähigt Ferner wurden folgende Eigenschaften
festgestellt:

Nach den gegenwärtigen Kenntnissen gehören die Keime zur Familie Achromobacteriaceae und sind mit
großer Wahrscheinlichkeit zur Gattung Alcaligenes zu stellen.
Bei WS 90 001 handelt es sich um einen Pilz der Gattung Penicülium.
UiTi das gewünschte Enzym in den erfindungsgemäß
zu verwendenden Mikroorganismen adaptiv anzureichern, werden diese vorzugsweise unter Zusatz von
Creatinin zum Wachstumsmedium gezüchtet. Beide Mikroorganismen werden vorteilhaft auf einem Nährboden
wachsengelassen, der Glucose oder Glycerin als
Kohlenstoffquelle, Creatinin sowie Salze und Vitamine in der in der Mikrobiologie bekannten Menge und Zusammensetzung
enthält Ein besonders geeignetes Nährmedium weist folgende Zusammensetzung auf:

gelöst in m/10 Kalmmphosphat-Puffer pH 6 (Pilz) bzw.
pH 7 (Alcaligenes).
Die Stammerhaltung der erfindungsgemäßen Mikroorganismen geschieht in üblicher Weise auf Agarschrägröhrchen
durch Zusatz von 2% Agar zu einem geeigneten Nährboden, vorzugsweise dem oben angegebenen
Nährboden, dem noch 5 ml/1 einer O,O5°/oigen Bromthymolblaulösung zugesetzt wird. Unter den bevorzugten
Wachstumsbedingungen schlägt dieser Indikator bei dem erfindungsgemäßen Pilz nach 4 bis 6
Tagen, bei dem Baktenum in 2 bis 3 Tagen deutlich ins alkalische Gebiet um, bedingt durch den Creatinin-Creatin-Abbau.
Creatinin verbrauchende Mikroorganismen, die den Abbau nicht nach dem obengenannten
Stoffwechselschema mit den obengenannten Enzymen bewerkstelligen, zeigen diese Alkalisierung nicht.
Die erfindungsgemäß zu verwendenden Mikroorganismen ermöglichen die wirtschaftliche Gewinnung der
Creatininamidohydrolase aus der aus ihrem Aufschluß erhaltenen wasserlöslichen Eiweißfraktion, wie in der
oben erwähnten, gleichzeitig eingereichten Patentanmeldung näher beschrieben ist.
Die folgenden Beispiele erläutern die Erfindung weiter.
Anreicherung von aktivem Pilzmycel in
Submers-Schüttelkultur

2 1 eines Nährbodens mit 1 Gew.-% Glucose, 0,5 Gew.-% Creatinin, 0,08 Gew.-% Ammoniumsulfat, 0,02
Gew.-% Magnesiumsulfatheptahydrat, 0,05 Gew.-% Hefeextrakt, je 1 mg Nicotinsäure, Thiamin-p-aminobenzoesäure
und Vitamin B6, 0,1 mg Biotin, Eisensulfat, Calciumchlorid und Mangansulfat in Spuren, in 0,10 M
Kaliumphosphatpuffer pH 6 werden in einem geeigneten Schüttelkolben mit 200 ml einer 48 Stunden alten
j Vorkultur von Penicillium WS 90 001 beimpft und in einem Schüttelapparat 4 Tage kultiviert. Der Creatinin-Gehalt
sinkt in dieser Zeit von 0,5% auf knapp 0,1%, und die Glucose ist bis zu diesem Zeitpunkt nahezu verbraucht.
Der pH-Wert steigt dabei auf 7,5 bis 8,0. Man ίο erhält so 3 bis 4 g WS 90 001 Trockengewicht/l Kulturlösung
mit 45 bis 60 IU/g Trockengewicht Creatinonamidohydrolase.
In einem geeigneten Kulturgefäß werden 15 1 des in
Beispiel 1 beschriebenen Mediums mit 1,51 gut bewachsender
Vorkultur von Alcaiigenes spec. WS 51 400 bei kräftiger Durchlüftung kultiviert Dabei wird während
der gesamten Kulturdauer der pH-Wert konstant auf 7,0 gehalten. Der Creatinin- und der Glucose-Gehalt
werden laufend verfolgt. Der Creatinin-Verbrauch vollzieht sich hauptsächlich im zweiten Drittel der Logphase, während Glucose zunächst abgebaut wird. Die
2r> Kultur wird bei 30° gehalten. Nach 35 Stunden können
1,5 g/l Kulturlösung Bakterientrockenmasse geerntet werden. Die spezifische Aktivität an Creatinin-amidohydrolase
beträgt 1600 IU/g Trockengewicht.
Es wird wie in Beispiel 2 beschrieben Alcaiigenes spec. WS 51 400 im Arbeitsvolumen von 601 kultiviert.
Sobald das gewünschte Wachstumsstadium erreicht ist, wird begonnen, frischen Nährboden dem Kulturgefäß
zuzuführen und in gleichem Maß bewachsene Kulturlösung dem Kulturgefäß zu entnehmen. Die Verdünnungsrate
(Fließgeschwindigkeit/Arbeitsvolumen) beträgt hierbei 0,13 bis 0,18, vorzugsweise 0,16. Pro Stunde
werden 500 i Luft durchgeleitet. Man erhält so in kontinuierlicher Züchtung eine Ausbeute von 3 bis 5 g/l
Bakterientrockenmasse mit einer spezifischen Aktivität wie sie nach dem in Beispiel 2 beschriebenen batch-Verfahren
erhalten wird.
Der Alcaiigenes spec. WS 51 400 eignet sich gut zur kontinuierlichen Züchtung, wie vorstehend beschrieben
wurde.
Beide Mikroorganismen sind in der Sammlung des Bakteriologischen Instituts der Technischen Universität
München in Weihenstephan hinterlegt.
