Description
bei pH 8,0 und 37° C katalysiert und für Creatinin als Substrat bei diesen Bedingungen die Ku 3,3 χ 10-2
M aufweist.
2. Creatinin-amidohydrolase nach Anspruch 1, dadurch gekennzeichnet, daß sie aus Mikroorganismen
stammt
3. Creatinin-amidohydrolase nach Anspruch 2, dadurch gekennzeichnet, daß sie aus Alcaligenes
spec. WS 51 400 oder Penicillum WS 90 001 stammt.
Die Erfindung betrifft das
Enzym Creatinin-amidohydrolase
Enzym Creatinin-amidohydrolase
Weiter wurde bereits eine spezifische mikrobiologische Bestimmungsmethode mit isolierten Bakterien
beschrieben, wobei gewaschene Zellsuspensionen zur Creatinin-M essung verwendet wurden und die Bildung
von Harnstoff und Ammoniak als Maß der Enzymwirkung herangezogen wurde. Lösliche, zum Creatininabbau
befähigte Enzymextrakte konnten dabei jedoch nicht erhalten werden. Voraussetzung für die Schaffung
einer spezifischen Creatinin-Bestimmung mit Hilfe von
ίο Enzymen war jedoch die Auffindung eines geeigneten,
löslichen Enzyms, welches spezifische und meßbare Umsetzungen des Creatinins katalysieren kann.
Roche. Lacombe und Girard (BBA 6, 210, 1950) charakterisierten in zwei Pseudomonas-Arten Creatinase,
Creatininase und eine Glycocyaminase als spezifische Enzyme, die aus ihren Substraten aus der
Guanidinogruppe Harnstoff in Freiheit setzten. Weiter wurde von Akamatsu und Mitarbeiter (Enzymologia, 15,
Seiten 122, 158, 173, 1951) in Erdbakterien ein als Creatinmutase bezeichnetes Enzym postuliert, welches
die Gleichgewichtseinstellung zwischen Creatinin und Creatin bewirken soll. Hierbei wird folgender Abbauweg
des Creatinins vermutet:

In der klinischen Chemie, besonders für die Funktionsdiagnostik der Niere, spielt die Bestimmung
der Zwischen- und Endprodukte des Eiweißstoffwechsels eine wichtige Rolle. Hierzu zählen auch Creatinin
und Creatin. Die Bestimmung von Creatinin wurde daher schon wiederholt beschrieben. Die meisten der
bekannten Methoden basieren auf der nichtenzymatischen Jaffe-Reaktion, die jedoch den Nachteil hat,
unspezifisch zu sein.

Nunmehr ist es gelungen, den hier vermuteten Weg des Creatinin-Abbaus zu bestätigen und erstmals zwei
lösliche Enzyme in Mikroorganismen aufzufinden und in hoher Reinheit zu isolieren, welche an diesem
Abbauweg entscheidend beteiligt sind.
Gegenstand der Erfindung ist eines dieser Enzyme, nämlich Creatinin-amidohydrolase mit den in den
Patentansprüchen definierten Eigenschaften.
Durch das erfindungsgemäße Enzym wird folgende Reaktion katalysiert:
Creatinin + H7O
Creatinin-amidohydrolase
Creatin
Da es sich um eine Gleichgewichtsreaktion unter Wasseraufnahme oder -abgabe handelt, wird das Enzym
als Creatinin-amidohydrolase bezeichnet.
Als Ausgangsmaterial für die Gewinnung des neuen Enzyms eignen sich Mikroorganismen allgemein, in
denen das gewünschte Enzym adaptativ angereichert ist, was gewöhnlich dadurch geschieht, daß in Gegenwart
oder unter Zusatz von Creatinin gezüchtet wird.
Aus den so gewonnenen Mikroorganismen wird durch Aufschluß die wasserlösliche Fraktion gewonnen.
Aus der so erhaltenen wasserlöslichen Fraktion läßt sich die Creatinin-amidohydrolase anreichern, indem man
ihre Eigenschaft, Creatinin in Creatin zu überführen, bo
ausnützt.
Die Reinigung und Gewinnung des Enzyms ist eingehend in der Stammanmeldung P 21 22 298.0-41
beschrieben.
Zwei für die Gewinnung des Enzyms der Erfindung besonders geeignete Mikroorganismen sind Alcaligenes
spec. WS 51 400 aus der Familie Achromo bactericeae und Penicillum WS 90 001.
Creatinin-amidohydrolase verliert bei pH-Werten von 6 und darunter schnell an Aktivität und weist bei
pH-Werten um 8,0 die größte Stabilität auf.
Die Salzfällung, beispielsweise unter Verwendung von Ammoniumsulfat, eignet sich zur Gewinnung des
Enzyms aus seinen Lösungen. Bei der Verwendung von Ammoniumsulfat fällt Creatinin-amidohydrolase bei
einer Konzentration von 2,2-M. Durch Dialyse bei 4° C, zweckmäßig gegen verdünnten Puffer, beispielsweise
0,02-M Diäthanolamin-Puffer, pH 8,0, kann das Enzym von Salzen und anderen niedrigmolekularen Begleitstoffen
befreit werden.
Die so erhaltenen Präparate können in eingefrorenem Zustand monatelang ohne Aktivitätsverlust gelagert
werden.
Das neue Enzym kann für wissenschaftliche Zwecke sowie zur spezifischen Bestimmung des Creatinins
verwendet werden.
Die folgenden Beispiele erläutern die Erfindung weiter.
Beispiel 1 Isolierung hochgereinigter Creatinin-amidohydrolase
Aus einem in Gegenwart von Creatinin gezüchteten Kulturansatz Alcaligenes spec. WS 51 400 werden die
Zellen (1 kg Trockengewicht) abgetrennt, mit 0,1-M Kaliumphosphatpuffer,
pH 8,0, auf 201 aufgenommen und ohne Kühlung in der Hochdruckdispersion bei ca. 800
atü aufgeschlossen. Der Extrakt wird mit 5 Vo!.-% einer
10%igen Polyäthylenimin-Lösung (Mol-Gewicht ca. 1800) von pH 8,0 (ca. 11) versetzt Nach Zugabe von KCl
(0,01-M Endmolarität) und NH4Cl (0,1-M Endmolarität)
werden pro Liter Extrakt innerhalb von 10 Minuten 0,8 Volumen 9O°/oiges wäßriges Isopropanol zugesetzt und
30 Minuten bei Raumtemperatur gerührt
Der reichliche Niederschlag wird abzentrifugiert.
Dem Überstand werden pro Liter weitere 0,45 Volumen 90%iges IsopropanoJ zugegeben.
Der so gebildete Niederschlag, welcher Creatininamidohydrolase und Creatin-amidinohydrolase enthält,
wird abgetrennt und in 400 ml 0,02-M Kaliumphosphatpuffer, pH 8,0, aufgenommen. Ein ungelöster Rückstand
wird abgetrennt und der Überstand auf eine mit dem gleichen Puffer äquilibrierte DEAE-Sephadex-Säule
(3,5 cm χ 1 m) gegeben. Nach dem Aufziehen wird die Säule mit 1 Volumen 0,1-M Kaliumphosphatpuffer, pH
8,0, gewaschen und dann mit 0,2-M Kaliumphosphatpuffer, pH 8,0, eluiert Die Creatinin-amidohydrolase-haltigen
Fraktionen werden vereinigt, bei pH 8,0 mit Ammoniumsulfat auf 2,2-M eingestellt, das ausgefallene
Enzym abzentrifugiert und in 0,02-M Diäthanolaminpuffer, pH 8,0, auf 100 ml gelöst und 4 Stunden bei +40C
gegen gleichen Puffer dialysiert Man erhält so in einer Gesamtausbeute von 64% ein Präparat mit einer
spezifischen Aktivität von 303 U/mg. Die nachstehende Tabelle zeigt zusammenfassend die in den einzelnen
Stufen dieses Verfahrens erhaltenen Aktivitäten und Ausbeuten.

Bei Aufschluß mit Hilfe von Ultraschall anstelle der Hochdruckdispersion konnte der Gehalt an Creatininamidohydrolase
bei etwa gleicher spezifischer Aktivität auf das 2,5fache, bezogen auf eingesetzte Mikroorganismen,
Trockengewicht erhöht werden.
Die wie oben beschrieben erhaltene Creatinin-amidohydrolase weist eine Gleichgewichtskonstante

(37°C; pH 8,0) auf.
Die Michaelis-Konstante für Creatinin als Substrat KM = 3,3 χ 10-2M(37°C;pH8,0).
7 g (Trockengewicht) eines in Gegenwart von Creatinin gewachsenen Alcaligenes spec. WS 51 400
wurden geerntet und bei pH 8,0 mit Ultraschall aufgeschlossen. Die erhaltene Suspension wurde wie in
Beispiel 1 beschrieben mit 0,8 Volumen Isopropanol versetzt, 30 Minuten bei Raumtemperatur gerührt und
zentrifugiert. Der Überstand wird mit weiteren 0,45 Volumen Isopropanol versetzt und erneut zentrifugiert
Der Niederschlag wird wie in Beispiel 1 beschrieben aufgenommen und über eine DEAE-Sephadex-Säule
chromatographiert, wobei lediglich die Creatinin-amidohydrolase eluiert wird. Die nachstehende Tabelle
zeigt die Einzelheiten dieses Verfahrens. Das erhaltene Präparat war zur Creatinin-Bestimmung geeignet

Bei den in den vorstehenden Beispielen beschriebenen Versuchen wurde aus Creatinin in Gegenwart von
Creatinin-amidohydrolase gebildetes Creatin bestimmt durch Zusatz von Adenosintriphosphat (ATP), welches
in Gegenwart von Creatinphosphokinase (CPK) in Creatinphosphat und Adenosindiphosphat (ADP) über-
führt wird. Gebildetes ATP wird mit Pyruvat-Kinase überführt und der Verbrauch an NADH optisch
(PK) unter Lactatdehydrogenase (LDH) sowie Phos- gemessen. Diese bekannten Schritte lassen sich wie folgt
phoenolpyruvat (PEP) unter NADH-Verbrauch in ATP darstellen:
