Description
'Verfahren zur Herstellung von Benzodiazepinen"
Priorität: 19. März 1970, Japan, Nr. 25732/70
Die Erfindung betrifft ein neues Verfahren zur Herstellung von
Benzodiazepinen der allgemeinen Formel I
in der R- ein Wasserstoffatom, ein Alkylrest mit 1 Mg H Kohlenstoffatomen oder eine Cycloalkylrnethylgruppe mit 5 bin 7
Kohlenstoffatomen im Ring ist und Rp und R, Wasserstoff- oder
Halogenatome bedeuten.
Bekanntlich sind die Benzodiazepine der allgemeinen Formel I
wertvolle Arzneimittel mit snalgerisoher, hypnotischer, spaf-molytiRcher
und mußkelre]axierender Wirkung. Diese Verbindungen
sind ferner wertvolle Zwischenprodukte zur Herstellung anderer Benzodiazepine, die ebenfalls wertvolle Arzneimittel mit der
vorgenannten Wirkung sind.

Bisher wurden diese Verbindungen z.B. dadurch hergestellt, daß nan zunächst die in der !-Stellung unc;ubstituierten 1 , "3-Dih-dro-2H-l,4-benzodiazepin-2-one
herstellte, und zwar aus den entsprechenden 2-Amino-benzophenonen durch Umsetzung mit Br-omacetylbromid
und anschließende Behandlung mit Ammoniak oder durch Umsetzung mit GlycinäthT/lester-hydrochlorid, und
anschließend das erhaltene Benzodiazepin in Gegenwart eines Kondensationsmifctels mit einem Alkylierungsmittel umsetzte.
Die nach diesen Verfahren erhaltenen Ausbeuten lassen zu wünschen übrig. Feraer ist das Alkylierungsprodukt immer durch
das unsubstituierte Benzodiazepin verunreinigt, das w^gen seiner
ähnlichen Eigenschaften schwierig abgetrennt werden kann.
Aufgabe der Erfindung war es daher, ein neues, wirtschaftlicheres
Verfahren zur Herstellung dieser Benzodiazepine der allgemeinen Formel I zu schaffen, das in hoher Ausbeute verläuft und
Endprodukte hoher Reinheit liefert. Diese Aufgabe wird er>findungsgemäß
dadurch gelöst, daß man ein Tetrahydrobenzodiazepin der allgemeinen Formel II
in der R., R„ und R-,. die obige Bedeutung haben, mit Licht
bestrahlt.
Vorzugsweise wird die Bestrahlung mit UV-Licht oder Sonnenlicht

und vorzugsv;eise in Gegenwart eines Lösungsmittel, wie eines
aliphatischen Alkohols, z.B. Methanol, Äthanol oder Isopropanol, eines aromatischen Kohlenwasserstoffs, wie Benzol, Toluol oder
Xylol, oder eines anderen organischen Lösungsmittels, wie Aceton, Chloroform, Dioxan, Dimethylsulfoxid, Dimethylformamid
oder Tetrahydrofuran oder deren Gemischen durchgeführt. Das Verfahren der Erfindung v;ird .vorzugsweise bei Raumtemperatur
durchgeführt, es kann jedoch auch bei höheren oder niederen Temperaturen ausgeführt werden.
Beispiele für erfindungsge-näß herstellbare Benzodiazepine sind:
5-Phenyl-l,3-dihydro-2H-l,4-benzodiazepin-2-on,
-l,3-dihydro-2H-1,4-benzodiazepin-2-on,
-l, 3-dih:rdro-?H-l, 4-benzodiazepin-P-on,
l-Kethyl-5-phenyl-7-chlor-l,3-dihydro-2H-»l,4-benzodiazepin-2-on,
l-Cyclopropylmethyl-5-phenyl-7-chlor-l,3-dih?ί■dro-PH-l,4-benzodiazepin-2-on,
l-vethyl-5-(o-fluorphenyl)-7-chlor-l,3-dihydro-2H-l,4-benzodiazepin-2-on,
l-Methyl-5-'o-chlorphenyl)-7-chlor-l,3-dihydro-2H-l,4-benzo-
<iiazepin-2-on und
l-Xthyl-5-phenyl-7-chlor-l,3-dihydro-2H-;,4-benzodiazepin-2-on.

Die verfahrensgemKß eingesetzten Tetrahydrc/liazepine der allgemeinen
Formel II können in einfacher Weise folgendermaßen hergestellt werden:
Ein AminObenzhydrylamin der allgemeinen Formel III
Ein AminObenzhydrylamin der allgemeinen Formel III

in der R1, R? und R, die obige Bedeutung haben, wird mit einem
Halogenessigsäureester zum entsprechenden (2~Aminobenzhydryl)~
aminoessigsäureester umgesetzt. Diese Verbindung wird zur entsprechenden
Carbonsäure verseift und zum Tetrahydrobenzodiazepiri cyclisiert.
Die Beispiele erläutern die Erfindung.
1,0 g l^Methyl-5-phenyl-7-chlor-l,3,4,5-tetrahyclro-2H-T#^-benzodiazepln
wird in 10 ml Dime-thylsulfoxid gelöst. Die Lösung wird
20 Stunden mit einer Queeksilber-Niederdrueklämpe bestrahlt.
Sodann wird das Reaktionsgemisch mit 50 ml Wasser verdünnt, die
ausgeschiedene Fällung abfiltriert und getrocknet, Ausbeute
0,62 g .l-Methyl~5-phenyl-7-chlor-!,jS-dihydro-PH-l^-benzodiazepin-2-on.
Nach Urnkristallisation aus Isoprop^nol schmilzt die
Verbindung bei IJl bis 132°C.
In ähnlicher WeIr^ v/r-rden folgende Verbindungen hergestellt:
l-Cyclopropyirnethyl-5-phenyl-7-chlor-li ."5- dih--rdro-2K-l ,4-benzodiazepin-2-on,
P. 1^5 bis l46°C;
l-Cyclopropylmethvl-fS- 'o-f luorphenyl'i-V-chlor-l, ^-dihvdro-2H-l,4-benzodiazepin-2-on.
Das Jlydrochlorid schmilzt bei 195°C unter
Zersetzung.

1. Verfahren zur Herstellung von Benzodis-zepinen dor allgemeinen

Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man
die Bestrahlung mit UV-Licht, oder Sonnenlicht durchführt.

daß man die Umsetzung in einem Lösungsmittel durchführt.
