Abstract
Die vorliegende Erfindung betrifft, neue, spezifische und effektive Prodrug-Verbindungen der allgemeinen Formel A-B-C, in der A jede Aminosäure, B die chemische Bindung zwischen A und C oder jede Aminosäure vorzugsweise ausgewählt aus Prolin, Hydroxyprolin, Thiazolidincarbonsäure, Dehydroprolin, Pipecolinsäure, Azetidincarbonsäure oder Aziridincarbonsäure, verbunden über Peptidbindungen mit A und C, sein kann, und C instabile Inhibitoren der Serinpeptidase Dipeptidyl Peptidase IV (DP IV) wie z. B. Dipeptidyl Alkylketon-Derivate sein kann. DOLLAR A Derartige Prodrug-Verbindungen von DP IV-Inhibitoren zeichnen sich dadurch aus, daß sie in wässrigen Lösungen, einschließlich biologischer Flüssigkeiten, chemisch stabil sind. Unmittelbar nach chemischer oder enzymatischer Freisetzung der DP IV-aktiven Wirkkomponenten entfalten diese ihre DP IV-inhibotorische Aktivität. Durch die Freisetzung dieser Dipeptidyl-Derivate beginnt deren Zerfall, der ihre Lebensdauer und somit ihre inhibitorische Aktivität begrenzt. DOLLAR A Dieser beschleunigte Zerfall basiert auf der Eigenschaft von Dipeptidyl-Derivaten, vorzugsweise von Aminoacyl-Pyrrolidin-Derivaten, zu zyklisieren. DOLLAR A Solche Prodrug-Verbindungen dienen einem zell-, gewebs- bzw. organspezifischen Transport durch biologische Membranen, einer zeitgerechten und zielgerichteten in vivo Aktivierung mittels der Prodrug chemisch maskierten Inhibitoren der DP IV und einer zeitlich kontrollierten in vivo Deaktivierung der aus der Prodrug-Verbindung ...
Description
Die vorliegende Erfindung betrifft neue, spezifische und effektive Prodrug-Verbindungen
instabiler der Serinpeptidase Dipeptidyl Peptidase IV (DP IV). Es ist bereits bekannt, daß
Aminoacyl Thiazolidide, Aminoacyl Pyrrolidide, N-Dipeptidyl, O-Acyl Hydroxylamine und
andere Verbindungen als Inhibitoren des Serumenzyms Dipeptidyl Peptidase IV (DP IV) und
analoger Enzyme wirken [vgl. DEMUTH, H.-U., J. Enzyme Inhibition 3, 249 (1990); DE-
MUTH, H.-U., HEINS, J., in Dipeptidyl Peptidase IV (B. Fleischer, Ed.) R. G. Landes, Bio
medical Publishers, Georgetown, 1995, 1-37].
Es wurde gefunden, daß mittels stabiler Inhibitoren der Dipeptidyl Peptidase IV die T-Zell
vermittelte Immunantwort z. B. bei Transplantationen beeinflußt werden [vergl. KOROM, S.,
DEMEESTER, I., STADLBAUER, T. H. W., CHANDRAKER, A., SCHAUB, M., SAYEGH,
M. H., BELYAEV, A., HAEMERS, A., SCHARPE, S., KUPIEC-WEGLINSKI, J. W., Inhibi
tion of CD26/dipeptidyl peptidase IV activity in vivo prolongs cardiac allograft survival in rat
recipients. Transplantation 63, 1495 (1997)], rheumathische Arthritis [vergl. TANAKA, S.,
MURAKAMI, T., HORIKAWA, H., SUGIURA, M., KAWASHIMA, K., SUGITA,T.,

19, 15 (1997)] unterdrückt werden kann.
Es wurde gefunden, daß durch Verabreichung von Effektoren der Dipeptidyl Peptidase IV
bzw. DP IV-analoger Enzymaktivität im Blut eines Säugers, durch deren damit verbundene,
temporäre Aktivitätsreduktion, in kausaler Folge die endogenen (oder zusätzlich exogen ver
abreichten) insulinotropen Peptide Gastric Inhibitory Polypeptide 1-42 (GIP1-42) und Gluca
gon-Like Peptide Amide-1 7-36 (GLP-17-36) (o. a. GLP-17-37 oder deren Analoga) durch DP IV- und
DP IV-ähnliche Enzyme vermindert abgebaut werden und damit die Konzentrations
abnahme dieser Peptidhormone bzw. ihrer Analoga verringert bzw. verzögert werden (vgl.
Die durch die Wirkung von DP IV-Effektoren erzielten, erhöhten Stabilität der (endogen vor
handenen oder exogen zugeführten) Incretine oder ihrer Analoga, die damit vermehrt für die
insulinotrope Stimulierung der Incretin-Rezeptoren der Langerhansschen Zellen im Pankreas
zur Verfügung stehen, verändert sich u. a. die Wirksamkeit von körpereigenem Insulin, was
eine Stimulierung des Kohlehydratstoffwechsels des behandelten Organismus nach sich zieht.
Als Resultat sinkt der Blutzuckerspiegel unter die für Hyperglykämie charakteristische Glu
kosekonzentration im Serum des behandelten Organismus. Dadurch können mittels DP IV-Inhi
bitoren Stoffwechselanomalien wie Übergewicht, Glukosurie, Hyperlipidämie sowie
mögliche schwere metabolische Azidosen, Diabetes mellitus, die eine Folge längerer, erhöhter
Glukosekonzentrationen im Blut sind, verhindert bzw. gemildert werden [vgl. DE 196 16 486].
Mit Hilfe von DP IV-Inhibitoren kann auch das Eindringen von HIV in CD 26 (DP IV) posi
tive Zellen experimentell verhindert werden [vergl. WAKSELMAN, M., NGUYEN, C., MA
ZALEYRAT, J.-P., CALLEBAUT, C., KRUST, B., HOVANESSIAN, A. G., Inhibition of
HIV-I infection f CD 26+ but not CD 26- cells by a potent cyclopeptidic inhibitor of the DPP
IV activity of CD 26. Abstract P 44 of the 24th European Peptide Symposium 1996].
Weiterhin wurde gefunden, daß DP IV neuroaktive Peptide wie Neuropeptid Y und CLIP in
ihrer Aktivität modulieren kann [vergl MENTLEIN, R., DAHMS, P., GRANDT, D., KRU
GER, R., Proteolytic processing of neuropeptide Y and peptide YY by dipeptidyl peptidase IV.

Diese vielseitigen Wirkungen von DP IV-Inhibitoren implizieren, daß sich deren Wirkungen
bei Anwendungen in einem bestimmten pathophysiologischen Zustand eines Gewebes auf
andere, normale physiologische Zustände z. B. in anderen Organen auswirken können. Diese
Auswirkungen können positive als auch negative Folgen für den Zielorganismus haben.
Um zwischen Wirkungen von DP IV-Inhibitoren in verschiedenen physiologischen Regelkrei
sen zu differenzieren, ist es demzufolge von besonderem Interesse, einerseits eine hohe Bio
verfügbarkeit von DP IV-Inhibitoren im gewünschten Zielgewebe oder Zielorgan zu errei
chen, andererseits aber eine erwünschte Wirkungsdauer dieser DP IV-Inhibitoren nicht zu
überschreiten.
Beispielsweise reicht für die Verstärkung des Incretin-Effektes bei Diabetes mellitus eine
Wirkungsdauer der Inhibitoren von wenigen Minuten [PEDERSON, R. A., WHITE, H. A.,
SCHLENZIG, D., PAULY, R. P., MCINTOSH, C. H. S., DEMUTH, H.-U., Improved glucose
tolerance in zucker fatty rats treated by oral administration of the dipeptidyl peptidase IV
inhibitor isoleucyl thiazolidide. Diabetes 47, 1253-1258 (1998)], während z. B. die Suppres
sion der DP IV-vermittelten Immunantwort bei Transplantationen eine langfristige Wirkung
der Inhibitoren erfordert [DEMEESTER, I., BELYAEV, A., LAMBEIR, A. M., DEMEYER,

Um mittels DP IV-Inhibitoren in solche physiologischen Regelkreise einzugreifen, die nur
eine kurzzeitige Beeinflussung des Zielenzymes DP IV erfordern, sind Inhibitoren wün
schenswert, die nach definierbaren Halbwertszeiten in inhibitorisch inaktive chemische Ver
bindungen übergehen. Darüber hinaus sollte dieser Zerfallsprozess erst dann einsetzen, wenn
die Verbindung das gewünschte Zielkompartiment, beispielsweise den Blutkreislauf erreicht
und die gewünschte Wirkung entfaltet.
Eine derart manipulierbare Pharmakokinetik und Bioverfügbarkeit von Enzyminhibitoren
wird üblicherweise durch die chemische Maskierung von Enzyminhibitoren durch sogenannte
Prodrugs erreicht [vergl. BUNDGAARD, H. Design of Prodrugs. Elsevier Science Publishers,
Amsterdam, 1985]. Mit Hilfe des Prodrug-Konzeptes lassen sich pharmakokinetische, phar
makodynamische, toxikologische und Stabilitätseigenschaften von Enzyminhibitoren beein
flussen [GANGWAR, S., PAULETTI, G. M., WANG, B., SIAHAAN, T. J., STELLA, V. J.,
BORCHARDT, R. T., Prodrug strategies to enhance the intestinal absorption of peptides. Drug
Discovery Today 2, 148 (1998)]. Die Freisetzung von biologisch aktiven Wirkstoffen (u. a.
Pharmaka, Peptidhormonen oder auch Enzyminhibitoren) erfolgt dabei sowohl durch chemi
sche oder enzymatische Mechanismen. Beispielsweise dienen Esterasen, Proteasen und Pepti
dasen zur Wirkstoff-Freisetzung aus Prodrugs [vergl. WO 97/45117, US 5 433 955, US 5 614 379,
US 5 624 894].
Es ist bekannt, daß N-terminal ungeschützte, C-terminal aktivierte Dipeptidderivate zu intra
molekularen Reaktionen neigen (GAUTSCHI, M., SCHMID, J. P., PEPPARD, T. L., RYAN,
T. P., TUORTO, R. M., YANG, X. G., Chemical characterization of diketopiperazines in beer.
J. Agr. Food. Chem. 45, 3183-3189 (1997)].
Beispielsweise zyklisiert das DP IV-Substrat Histidyl-Prolyl-Amid in Serum mit Halbwerts
zeiten von weniger als 10 Minuten zu His-Pro-Diketopiperazin [BAUER, K., KLEINKAUF,
H., Catabolism of thyroliberin by rat adenohypophyseal tissue extract. Eur. J. Biochem. 106,
107-117 (1980)], was damit seine Eigenschaften als Substrat der DP IV verliert. Diese Re
aktion erfolgt spontan und ist für insbesondere an C-terminaler Position des Peptides Prolin
haltiger Peptide charakteristisch. Diese spontane intramolekulare Reaktion ist auf den nukleo
philen Angriff des N-terminalen Aminostickstoffs auf die C-terminale Carbonylfunktion des
Dipeptid-Derivates zurückzuführen und wird durch die insbesondere in Prolin-haltigen Pepti
den erleichterte cis-trans Isomerisierung um die Aminosäure-Imidbindung erleichtert (REI
MER, U., EL MOKDAD, N., SCHUTKOWSKI, M., FISCHER, G., Intramolecular assistance
of cis/trans isomerization of the histidine-proline moiety. Biochemistry 36, 13802-13808
(1997)]. Aufgrund dieser intramolekularen Zyklisierungsreaktion sind C-terminal carbonyl
aktive Dipeptid-Derivate, wie Dipeptidylchlormethylketone, Dipeptidylboronsäuren und Di
peptidyl cyanide als instabile, kurzlebige Inhibitoren der DP IV bekannt DEMUTH, H.-U.,
HEINS, J., in Dipeptidyl Peptidase IV (B. Fleischer, Ed.) R. G. Landes, Biomedical Publi
shers, Georgetown, 1995, 1-37; PARGELLIS, C. A., CAMPBELL, S. J., PAV, S., GRAHAM
E. T., PITNER, T. P., Inhibition of dipeptidyl peptidase IV (CD 26) by peptide boronic acid
dipeptides. J Enzyme Inhib. 11, 151-169 (1997)]. Diese Eigenschaften von insbesondere
C-terminal Prolin-haltigen, carbonylaktiven Dipeptid-Derivaten kann man erfindungsgemäß
zum Design von derartigen Prodrugverbindungen von DP IV-Inhibitoren nützen, um nach
Freisetzung des inhibitorisch aktiven DP IV-Inhibitors dessen nunmehr erwünschte Deaktivie
rung durch intramolekulare Zyklisierung einzuleiten.
Aufgrund der retardierten Freisetzung des instabilen DP IV-Inhibitors aus der Prodrugfrom
wird neben der gewünschten effektiven Inhibierung des Zielenzyms überraschenderweise eine
verlängerte Wirksamkeit bei deutlich verminderter Konzentration der Prodrug gegenüber den
instabilen DP IV-Inhibitoren beobachtet.
Die Herstellung von instabilen DP IV-Inhibitoren C (entsprechend der allgemeinen Formel
A-B-C) wird durch die Beispiele 1.1 und 1.2 belegt. Die Synthese einer erfindungsgemäßen
Prodrug instabiler DP IV-Inhibitoren durch das Beispiel 1.3. Die Ausgangsverbindungen, die
jeweils korrespondiereden Peptidylchlormethylketone, wurden nach bekannten Verfahren
(WEINSTEIN, B., Chemistry and Biochemistry of Amino Acids, Peptides, and Proteins,
Marcel Dekker, New York, Basel, 1977) hergestellt. Die beispielhaft dargestellten Pyridini
ummethylketone unter 1.1 und 1.2 sind als N-terminal geschützte Dipeptidderivate außeror
dentlich stabil und vollständig charakterisierbar. Die N-terminal deblockierten Dipeptidderi
vate beginnen bei normaler Luftfeuchte den intramolekularen Zersetzungsprozeß unmittelbar
nach Deblockierung, so daß keine Schmelzpunkte bestimmbar sind. Die Charakterisierung der
Produkte erfolgt mittels HPLC und Massenspektrometrie.

Darstellung: 10 mmol Z-Val-Pro-Chlormethylketon werden in Pyridin gelöst
und bei 25°C 5 Tage gerührt. Das überschüssige Pyridin wird
bei 2 mbar Vakuum abdestilliert. Das Z-Val-Pro-Pyri
diniummethylketon wird noch einer HPLC-Reinigung un
terzogen. Die Verbindung ist ein Öl.

form/Methanol wird anfangs (9 : 1 Vol.-Teile) das Produkt und
mit zunehmender Polarität des Eluenten das Chlormethylketon
aufgefangen. Z-Phe-Pro-Pyridiniummethylketon wird abschlie
ßend einer HPLC-Reinigung unterzogen.

Zur Analyse der Stabilität der unter 1.1 und 1.2 dargestellten Inhibitoren wurde diese in wäss
riger Pufferlösung inkubiert und ihre intramolekulare Zykliserungsreaktion mit Hilfe von
MALDI-TOF Massenspektrometrie verfolgt (Abb. 2 und 3). Die Produkte dieser Re
aktion ergeben die jeweiligen Pyrazinderivate (Abb. 1).
Der Abbau von H-Phe-Pro-Pyridinium-Methylketon mit dem Molekulargewicht 337.2 Da
vollzieht sich unter Wasserabspaltung zum zyklischen Pyrazinderivat mit dem Molekularge
wicht 319.2 Da quantitativ innerhalb von 30 Minuten (Abb. 2).
Der Abbau von H-Val-Pro-Pyridinium-Methylketon mit dem Molekulargewicht 291.2 Da
vollzieht sich unter Wasserabspaltung zum zyklischen Pyrazinderivat mit dem Molekularge
wicht 273.2 Da quantitativ innerhalb von 60 Minuten (Abb. 3).
Die während der intramolekularen Reaktion erfolgende Ausbildung des Doppelbindungssy
stems des Pyrazins ermöglicht die quantitative Analyse des Zyklisierungsvorganges mittels
UV-Spektrometrie (Abb. 4). Die daraus ermittelten Geschwindigkeitskonstanten für die
intramolekulare Zyklisierung der instabilen DP IV-Inhibitoren in 0.1 M HEPES-Puffer,
pH = 7.6, 25°C, sind in Tabelle 1 zusammengefaßt.
Im Gegensatz dazu erwies sich die Prodrug des DP IV-Inhibitors H-Val-Pro-CH2-(N⁺C5H5)
H-Gly-Pro-Val-Pro-CH2-(N⁺C5H5) über 24 Stunden unter gleichen Bedingungen völlig stabil.
Inkubiert man das Targetenzym DP IV mit instabilen Inhibitoren in Gegenwart eines Sub
strates, so ist initial eine Hemmung des Enzyms zu beobachten, die durch die parallel verlau
fende intramolekulare Zyklisierung des Inhibitors mit fortschreitender Versuchszeit wieder
nachläßt, da durch die spontan verlaufende chemische Reaktion, die Konzentration des Inhi
bitors in der Reaktionslösung abnimmt. Dieser Effekt ist in den Abb. 5 und 6 darge
stellt. Durch die zeitabhängige Konzentrationsabnahme des Inhibitors nimmt die Geschwin
digkeit der enzymkatalysierten Hydrolyse des Substrates mit fortschreitender Zeit wie erwar
tet wieder zu.
Im Gegensatz zu den unmaskierten DP IV-Inhibitoren erweist sich die Prodrug
H-Gly-Pro-Val-Pro-CH2-(N⁺C5H5) über 24 Stunden in gepufferter wäßriger Lösung in Abwesenheit eines
Enzyms als stabil. Erst durch die Zugabe des Enzyms DP IV (hier exemplarisch auch zur
Freisetzung des DP IV-Inhibitors genutzt) wird unter Abspaltung des N-terminalen Dipeptides
H-Gly-Pro-OH der aktive DP IV-Inhibitor H-Val-Pro-CH2-(N⁺C5H5) freigesetzt. Im Massen
spektrum (Abb. 7) sind demzufolge selbst nach 60 Minuten Inkubationszeit noch deut
lich mehr als 50% der inkubierten Prodrug zu erkennen. Aufgrund dieser retardierten Freiset
zung wird neben der gewünschten effektiven Inhibierung des Zielenzyms überraschenderweise
eine deutlich verlängerte Wirksamkeit bei deutlich verminderter Konzentration der Prodrug
gegenüber den instabilen DP IV-Inhibitoren beobachtet (Abb. 8).

Claims (9)
1. Prodrug-Verbindungen der allgemeinen Formel A-B-C in der A jede Aminosäure, B die
chemischen Bindung zwischen A und C oder jede Aminosäure vorzugsweise ausgewählt
aus Prolin, Hydroxyprolin, Thiazolidincarbonsäure, Dehydroprolin, Pipecolinsäure,
Azetidincarbonsäure oder Aziridincarbonsäure, verbunden über Peptidbindungen mit A
und C, sein kann, und C ein instabiler Inhibitor der Serinpeptidase Dipeptidyl Peptidase IV
(DP IV) wie z. B. ein Dipeptidyl Alkylketon-Derivat sein kann.
2. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zum zielgerichteten zell-, gewebs- und organspezifischen Transport durch biologische
Membranen von mittels der Prodrugform chemisch maskierten Inhibitoren der DP IV.
3. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zur zeitgerechten in vivo Aktivierung von mittels der Prodrugform chemisch maskierten
Inhibitoren der DP IV.
4. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zur zeitlich kontrollierten in vivo Deaktivierung der aus der Prodrug-Verbindung
freigesetzten maskierten Inhibitoren der DP IV.
5. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zur zielgerichteten, d. h. zell-, gewebs- und organspezifischen in vivo Aktivierung von
mittels der Prodrugform chemisch maskierten Inhibitoren der DP IV.
6. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zur Behandlung von Erkrankungen von Säugern, die durch Modulation der DP IV-Akti
vität in verschiedenen Zellen, Geweben und Organen beeinflußt werden können.
7. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 zur Behandlung insbesondere von Stoffwechselerkrankungen des Menschen.
8. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
1 in pharmazeutischen Formulierungen.
9. Verwendung von Prodrug-Verbindungen der allgemeinen Formel A-B-C gemäß Anspruch
7 in pharmazeutischen Formulierungen insbesondere zur enteralen und parenteralen
Verabreichung.
