steht; worin einer, zwei oder drei der Substituenten
A2, A3, A4 und A5 ausgewählt sind aus drei
Halogenatomen, drei Methylradikalen, zwei Methoxyradikalen, zwei Hydroxyradikalen und einem
Äthyl-, Isopropyl-, t-Butyl-, Äthoxy-, n-Butoxy-, Methylthio-, Alkenyl-, Nitro-, Acetyl-, Benzoyl-,
Trifluoromethyl-, Phenyl-, Phenoxy-, Tolyloxy-, Phenylthio-, Phenylsulfonyl-, Anilino-, Benzyl-, α,Λ-Dimethylbenzyl-
und Benzyloxyradikal, wobei drei, zwei bzw. einer der Substituenten A2, A3, A4 und A5
Wassterstoff sind bei der Blockierung der adenergischen /?-Rezeptoren.

Die Erfindung betrifft die Verwendung der im Anspruch näher gekennzeichneten Verbindungen bei
der Blockierung der 0-Rezeptoren des Sympathikus, weiche u. a. zur Behandlung oder Prophylaxe von
Herzkranzarterienerkrankungen brauchbar sind.
Es sind bereits bestimmte l-Amino-S-phenoxy^-propanolderivate
bekannt; es war jedoch nicht bekannt, daß Verbindungen dieses Typs die Aktivität der
ß- Rezeptoren des Sympathikus blocken, vgl. J. Pharm. & Pharmacology (1956), S. 666-667 und (1958)
S. 47-53, wonach lokalanasthetische bzw. analgetische Eigenschaften untersucht wurden.
Geeignete Salze der Alkanolamine und Alkanolaminester,
die erfindungsgemäß verwendet werden können, sind Salze, die sich z. B. von anorganischen Säuren
ableiten, wie Salzsäure, Bromwasserstoffsäure, Phosphorsäure oder Schwefelsäure, oder Salze, die sich von
organischen Säuren ableiten, wie Oxalsäure, Milchsäure, Weinsäure, Essigsäure, Salicylsäure, Zitronensäure,
Benzoesäure, 0-Naphthoesäure, Adipinsäure oder 1,1'-Methylen-bis-(2-hydroxy-3-naphthoesäure),
oder Salze von sauren Kunstharzen, wie sulfonierten Polystyrolharzen. Verhältnismäßig unlösliche Salze, z. B. die Salze
der l,l'-Methylen-bis-(2-hydroxy-3-naphthoesäure), haben den Vorteil, daß sie über einen längeren Zeitraum
einen höheren Blutspiegel des Arzneistoffes ermöglichen.
Gemäß der Erfindung können in Form von Tabletten, Kapseln, wäßrigen oder öligen Lösungen, wäßrigen ao
oder öligen Suspensionen, Emulsionen, sterilen injizierbaren wäßrigen oder öligen Lösungen oder Suspensionen,
oder dispergierbaren Pulvern werden.
Geeignete Tabletten können durch Vermischen des oder der Wirkstoffe mit bekannten pharmazeutischen b5
Trägern, z. B. inerten Verdünnungsmitteln, wie Calciumcarbonat, Calciumphosphat oder Milchzucker, Sprengmitteln,
wie Maisstärke oder Alginsäure, Bindemitteln, wie Stärke, Gelatine oder Gummi arabicum, und
Gleitmitteln, wie Magnesiumstearat, Stearinsäure oder Talcum, hergestellt werden. Solche Tabletten können
gegebenenfalls nach an sich bekannten Methoden mit Deckschichten überzogen werden, um den Zerfall im
Magen zu verzögern und damit eine verzögerte Wirkstoffabgabe während längerer Zeit zu ermöglichen.
Die wäßrigen Suspensionen, Emulsionen, öligen Lösungen und Suspensionen enthalten im allgemeinen
ein Süßmittel, z. B. Glycerin, Traubenzucker oder Rohrzucker und einen Geschmackskorrigens, z. B.
Vanillin oder Orangenextrakt, um ein schmackhaftes Produkt zur Verfügung zu stellen. Die wäßrigen
Suspensionen der Erfindung können auch Suspendieroder Dickmittel enthalten, z. B. Natriumcarboxymethylcellulose,
Netzmittel, z. B. Kondensationsprodukte von Fettalkoholen mit Äthylenoxyd, und Konservierungsstoffe, wie p-Hydroxybenzoesäuremethyl- oder -propylester.
Die Emulsionen können den oder die Wirkstoffe gelöst in einem pflanzlichen oder tierischen Öl
enthalten, z. B. Erdnußöl oder Lebertranöl, und sie können Süßmittel und Geschmackskorrigentien enthalten,
die vorteilhafterweise ätherische öle sein können. Die genannten Emulsionen können auch Emulgier- und
Dispergiermittel enthalten, z. B. Sojabohnenlecithin, Polyoxyäthylensorbitanmonooleat, Gummi arabicum,
Tranganth oder Kasein, sowie Konservierungsmittel, wie p-Hydroxybenzoesäuremethyl- oder -propylester,
und Antioxydationsmittel, wie Gallussäurepropylester.
Die öligen Lösungen enthalten ebenfalls den oder die Wirkstoffe in Lösung in einem pflanzlichen oder
tierischen öl, und sie können gegebenenfalls Geschmackskorrigentien
enthalten, um den Geschmack zu überdecken und zu verbessern. Derartige ölige Lösungen
können vorteilhaft in weiche Gelatinekapseln abgefüllt werden. Die öligen Lösungen können auch
Süßstoffe, wie Staubzucker, enthalten. In diesem Fall kann die ölphase außerdem ein Suspendiermittel, z. B.
Bienenwachs, enthalten, um die Suspension stabil zu halten.
Präparate zur oralen Verabreichung in Form von Gelatinekapseln können aus Kapseln bestehen, welche
den oder die Wirkstoffe allein enthalten. Die Kapseln können auch den oder die Wirkstoffe im Gemisch mit
inerten Verdünnungsmitteln, wie Milchzucker oder Sorbit, enthalten.
Die sterilen injizierbaren wäßrigen Suspensionen können ein Suspendier- oder Dickmittel enthalten, z. B.
Natriumcarboxymethylcelluiose, sowie ein Netzmittel oder Dispergiermittel, z. B. ein Phenol-Polyäthylenoxyd-Kondensat,
z. B. das Kondensationsprodukt von Octylcresol mit etwa 8 bis 10 Molanteilen Äthylenoxyd.
Die sterilen injizierbaren öligen Lösungen der Erfindung können aus einem ungiftigen injizierbaren öl, z. B.
Erdnußöl, oder Ölsäureäthylester hergestellt werden, und sie können außerdem Geliermittel, z. B. Aluminiumstearat
enthalten, um die Absorption im Körper zu verzögern. Diese wäßrigen und öligen injizierbaren
Präparate können ein oder mehrere Konservierungsmittel, z. B. p-Hydroxybenzoesäuremethyl- oder -n-Prcpylester
oder Chlorbutanol enthalten.
Die erfindungsgemäß verwendeten Alkanclaminderivate können durch in der Technik an sich bekannte
Verfahren hergestellt werden.
Die Erfindung wird durch die folgenden Beispiele näher erläutert.
Ein Gemisch von 50 Teilen 1 -(2-Chlorphenoxy)-3-isopropylamino-3-propanol,
125 Teilen Maisstärke, 270 Teilen Calciumphosphat und 1 Teil Magnesiumstearat wird verpreßt und das verpreßte Material wird dann
durch ein Sieb der Maschenweite 1 mm granuliert Das erhaltene Granulat wird in an sich bekannter Weise zu
Tabletten verpreßt Man erhält Tabletten zur oralen Verabreichung.
89 Teile spritzgetrockneter Milchzucker werden durch ein Sieb der Maschenweite 0,25 mm passiert und
dann mit 5 Teilen Maisstärke und 1 Teil Magnesiumstearat vermischt Das Gemisch wird durch ein Sieb der
Maschenweite 0,25 mm passiert und dann gründlich mit 5 teilen l-Isopropylamino-3-(3-tolyloxy)-2-propanoI-hydrochlorid
vermischt Das Gemisch wird wieder durch ein Sieb der Maschenweite 0,25 mm passiert und dann in
an sich bekannter Weise zu Tabletten verpreßt. Man erhält Tabletten zur oralen Verabreichung.
Ein Gemisch von 50 Teilen 1 -(4-Chlorphenoxy)-3-isopropylamino-2-propanol
und 150 Teilen Maisstärke wird in Hartgelatinekapseln abgefüllt. Das erhaltene Arzneipräparat eignet sich zur oralen Verabreichung.
200 Teile Kokosnußöl, 780 Teile Erdnußöl und 20
Teile Bienenwachs werden miteinander bei höchstens 100° C verschmolzen und dann mit 0,75 Teilen
Gallussäurepropylester unter Rühren versetzt. Die ölige Lösung wird unter Rühren auf Raumtemperatur
abkühlen gelassen. 0,4 Teile l-IsopropyIamino-3-(3-tolyloxy)-2-propanol-hydrochlorid,
40 Teile Puderzucker und 0,6 Teile Natriumsalz des Sacharin werden miteinander 10 Minuten vermischt und dann mit 58,6
Teilen der vorgenannten öligen Lösung unter fortwährendem Rühren versetzt. Danach gibt man ein
geeignetes Geschmackskorrigens zu und mischt weitere 30 Minuten. Man erhält eine ölige Suspension zur oralen
Verabreichung.
Eine Lösung von 1 Teil l-Isopropylamino-3-(3-tolyloxy)-2-propanol-hydrochlorid
in 100 Teilen destilliertem V/asser wird in Ampullen abgefüllt. Die Ampullen werden zugeschmolzen und dann 30 Minuten auf 1150C
erhitzt Man erhält Ampullen, die eine sterile wäßrige Lösung für die parenterale Verabreichung enthalten.
Das vorgenannte Verfahren wird wiederholt, jedoch wird eine Lösung von 0,1 Teilen Natriummetabisulfit
und 1 Teil l-IsopropyIamino-3-(3-tolyloxy)-2-propanolhydrochlorid in 100 Teilen destilliertem Wasser in
Ampullen abgefüllt und dann auf die vorstehend beschriebene Weise behandelt. Man erhält ebenfalls
Ampullen, die eine sterile wäßrige Lösung für die parenteral Verabreichung enthalten.
Eine Lösung von 1 Teil l-Isopropylamino-J-(3-tolyloxy)-2-propanol-hydrochlorid
in 100 Teilen destillier-
tem Wasser wird durch ein Bakterienfilter filtriert Das sterile Filtrat wird unter aseptischen Bedingungen in
sterile Ampullen abgefüllt. Man erhält Ampullen, die eine sterile wäßrige Lösung zur parenteralen Verabreichung
enthalten.
Ein Gemisch von 1 Teil Di-[I-isopropylamino-3-(3-tolyloxy)-2-propanol]-l
,1 '-methylen-bis-(2-hydroxy-3-naphthoat), 40 Teilen Rohrzucker, 0,5 Teilen eines
Cetylalkohol-Polyäthylenoxyd-Kondensats, 1 Teil Polyvinylpyrrolidon,
0,25 Teilen p-Hydroxybenzoesäuremethylester und 100 Teilen Wasser wird mehrere Stunden
in der Kugelmühle vermählen. Nach dem Zusatz eines geeigneten Farbstoffs und Geschmackskorrigens erhält
man eine zur oralen Verabreichung geeignete Suspension.
200 Teile Di-[l-isopropyJamino-3-(3-tolyi-
oxy)-2-propanol]-l,r-methylen-bis-(2-hydroxy-3-naphthoat)
werden zu einem feinen Pulver vermählen, durch dreistündiges Erhitzen auf 120° C sterilisiert und dann
mit 20 Teilen steriler, feingepulverter Natriumcarboxymethylcellulose
vermischt. Das erhaltene Pulver wird mit einer sterilen 2%igen Lösung von Polyoxyäthylensorbitanmonooleat
in Wasser gründlich vermischt und das erhaltene Gemisch wird getrocknet. Das trockene,
sterile Produkt wird in Gläschen abgefüllt, die dann verschlossen werden. Durch Zusatz von sterilem
Wasser zum Pulver und anschließendes Schütteln erhält man eine Suspension zur intramuskulären Injektion.
Ein Gemisch von 2 Teilen Aluminiumstearat und 98 Teilen Erdnußöl wird geliert und durch Erhitzen auf
150°C unter Rühren sterilisiert. Das Gemisch wird dann abgekühlt, mit 1 Teil sterilem l-Isopropylamino-3-(3-tolyloxy)-2-propanolhydrochlorid
versetzt und unter sterilen Bedingungen in der Kugelmühle vermählen, um die
Teilchengröße des Wirkstoffes auf unter 50 zu verringern. Man erhält eine sterile ölige Suspension zur
intramuskulären Injektion.
Beispiel 10
Ein Gemisch aus 40 Teilen 1 -(2-AUylphenoxy)-3-isopropylamino-2-propanol-hydrochlorid
und 150 Teilen Lactose wird durch ein Sieb der Maschenweite 0,252 mm hindurchgeschickt. 2 Teile Gelatine werden
zugesetzt, und das Gemisch wird granuliert, indem es durch ein Sieb der Maschenweite 1,003 mm hindurchgeschickt
wird. Die Granalien werden getrocknet und durch ein Sieb der Maschenweite 0,82 mm hindurchgeschickt.
6 Teile Maisstärke und 2 Teile Magnesiumstearat werden zugesetzt, und die Granalien werden in
Tabletten gepreßt, wobei jede 200 mg wiegt und 40 mg aktiven Bestandteil enthält. Die Tabletten sind für orale
Verwendung für therapeutische Zwecke geeignet.

Vergleichsversuche zum Nachweis des technischen Fortschritts der anmeldungsgeinäßen Verbindungen
1. Verfahren zur Messung der ß-adrenergetischen
Blockiemngsaktivität, welches den spezifischen konkurrierenden
Antagonismus eines 0-adrenergetischen Blokkierungsrnittels
für die Herz-ß-rezeptorenstelleri de-
monstriert, und zwar anhand der Inhibierung eil durch Isoprenalin induzierten Tachycardie bei anästl
tisierten Katzen.

zwar mit einer Dosis von 80 mg/kg Körpergewicht intravenös, und die Herzgeschwindigkeit und der
Blutdruck wurden kontinuierlich aufgezeichnet. Dann wurde Isoprenalin [l-(3,4-Dihydroxyphenyl)-2-isopropylaminoäthanol]
intravenös in Intervallen von 10 Minuten mit einer Dosis von 0,20 μg/kg Körpergewicht
verabfolgt Jede Injektion von Isoprenalin ergab eine vorübergehende Zunahme der Herzgeschwindigkeit
(eine Tachycardie) und einen Abfall des Blutdrucks. Der M ittelwert aus drei Zunahmen der Herzgeschwindigkei- ι ο
ten (gemessen in Herzschlägen je min), wurde als Vergleichstachycardie genommen. Die zu prüfende
Verbindung wurde dann der Katze durch kontinuierliche intravenöse Infusion mit einer bestimmten Geschwindigkeit
(gewöhnlich im Bereich zwischen 1 und 10μg/kg Körpergewicht/Minute) 30 Minuten lang
verabreicht, wobei die Herzgeschwindigkeit und der Blutdruck der Katze immer noch kontinuierlich
aufgezeichnet wurden (es wird darauf hingewiesen, daß die Verabreichung der Verbindung selbst einen
gewissen Einfluß auf die Herzgeschwindigkeit und/oder den Blutdruck der Katze haben kann). Isoprenalin
wurde wieder intravenös in Abständen von 10 Minuten mit einer Dosis von 0,20 μπι/kg Körpergewicht
verabreicht, und die Tachycardie, die durch die Verabreichung von Isoprenalin 30 Minuten nach Beginn
der Infusion der zu prüfenden Verbindung verursacht wurde, wurde gemessen. (Die Zunahme der Herzgeschwindigkeit
wurde aus der Herzgeschwindigkeit unmittelbar vor der Verabreichung des Isoprenalins
errechnet.) Der Unterschied zwischen dieser Tachycardie und der Vergleichstachycardie wurde dann als
Prozentsatz der Vergleichstachycardie ausgedrückt. Dieser Prozentsatz wurde als »prozentuale Inhibierung
der Vergleichstachycardie« bezeichnet.
Die Geschwindigkeit der Infusion der zu prüfenden Verbindung wurde dann gesteigert, und zwar in
geometrischer Progression. Die zu prüfende Verbindung wurde nach wie vor in Abständen von 30 Minuten
verabreicht. Die durch Isoprenalin induzierte Tachycardie wurde wie oben beschrieben gemessen, bis eine
Dosis der zu prüfenden Verbindung erhalten war, die eine 75°/oige Inhibierung der Vergleichstachycardie
ergab. Die prozentuale Inhibierung der Vergleichstachycardie wurde dann graphisch gegen den Logarithmus
der Gesamtmenge der zu prüfenden Verbindung, die während der 30 Minuten unmittelbar vor der
Messung einer jeden Tachycardie infusiert wurde, aufgetragen, und aus der graphischen Darstellung
wurde diejenige Dosis entnommen, die bei der Infusion während 30 Minuten eine 50%ige Inhibierung der
Vergleichstachycardie ergab. Diese Dosis, weiche mit ED50 bezeichnet ist, und in μg/kg Körpergewicht
ausgedrückt ist, wurde als Maß für die /?-adrenergetische
Blockierungsaktivität der zu prüfenden Verbindung genommen.
2. Resultat
