2. Verwendung der Isothiazolone gemäß Anspruch 1 zur Bekämpfung von Bakterien und Pilzen.
Die Erfindung betrifft neue Isothiazolone gemäß dem Patentanspruch sowie deren Verwendung zur Bekämpfung
von Bakterien und Pilzen.
Die erfindungsgemäßcn Isothiazolone können durch
Umsetzung eines 3-Hydroxyisothiazols mit einem Isocyanat oder Isothiocyanat auf einer im wesentlich
äquimolaren Grundlage hergestellt werden. Das nichtsubstituierte 3-Hydroxyisothiazol kann aus Propiolamid
nach der Methode von W. D. Crow und N. |. Leonard (].
Og. Chem., 30, Seiten 2660 bis 2665 [1965]) synthetisiert werden. Die Herstellung der substituierten 3-Hydroxyisothiazole
kann nach der Methode erfolgen, wie sie in der DF.-OS 16 95 669 vorgeschlagen wird. F.inc allgemeine

worin R, R1, R" und X die in dem Patentanspruch
angegebenen Bedeutungen besitzen.
Es wird zwar ein äquimolares Verhältnis der Reaktionspartner bevorzugt, man kann jedoch auch
einen Überschuß von bis zu 2 Mol oder mehr an Isocyanat oder Isothiocyanat je Mol 3-Hydroxyisothiazo'i
einsetzen. In vielen Fällen kann zur Durchführung dieser Umsetzung ein inertes organisches Lösungsmittel
eingesetzt werden. Vorzugsweise werden aromatische Kohlenwasserstoffe, halogenierte aromatische Kohlenwasserstoffe,
Amide, Ester, aliphatische Kohlenwasser stoffe und Äther als Lösungsmittel eingesetzt. Besonders
geeignet sind Benzol, Toluol, Heptan, Äthyläther und Dioxan. Beim Einsatz einfacher Isocyanate ist die
Umsetzung spontan, im Falle von weniger reaktiven langkettigen oder aromatischen Isocyanaten kann in
vorteilhafter Weise ein Katalysator oder Beschleuniger, wie beispielsweise ein tertiäres Amin, verwendet
werden. Beispiele für derartige tertiären Amine sind l'yndin und Triethylamin. Die Reaktionstemperamr ist
zwar nicht kritisch, gewöhnlich wird jedoch die Umsetzung in einem Bereich von —20 bis +80"C oder
mehr durchgeführt.
Die erfindungsgemäßcn Isolhia/.olonc, mit Ausnahme
der Hydroxyniethylderivate. der 2-Thiocarbamoylderivate
sowie einiger 2-Carbamoylderivate, können durch Umsetzen eines substituierten Disulfidamids mit einem
Halogenierungsmittel hergestellt werden. Dieses Disulfidamid entspricht der Formel

worin X und Z Wasserstoffatome oder niedermolekulare Alkylgruppen bedeuten und Y den in dem
Patentanspruch .ingegebenen Definitionen entspricht. Eine derartige Cyclisierung erfolgt durch Umsetzung
des Disulfids mit einem Halogenicrungsmittel. Typische Halogenierungsmittel sind beispielsweise Chlor, Brom,
Sulfurylchlorid, Sulfurylbromid, N-Chlorsuceinimid oder
N-Bromsuccinimid.
Chlor und Sulfurylchlorid sind die bevorzugten I lalogenicrungsmitiel.
Die Cyclisierung des Disulfidamids erfolgt, wenn drei
moliiquivalcntc Halogenierungsmittel für die Umsei-
zung eingesetzt werden. Durch einen Überschuß an Halogenierungsmitte! kann das Isothiazolon in der A-
und/oder 5-Stellung halogeniert werden. Werden fünf moläquivalente Halogenierungsmittel eingesetzt, dann
findet eine Monohalogenierung statt. Für eine Dihalo- ~. genierung sind sieben moläquivalente Halogenierungsmittel
erforderlich.
Die Herstellung von Isothiazolonen, die in der A- und 5-Stellung mit verschiedenen Halogenatomen substituiert
sind, erfolgt durch Halogenierung eines bereits in κι einer der beiden in Frage stehenden Stellungen
halogenierten Isoihiazolons. Das Ausgangsisotliiazolon
wird in der vorstehend beschriebenen Weise durch Cyclisierung eines Disulfidamids gewonnen. Die einzelnen
Verfahrensvarianten, die man als sekundäre ι ϊ Halogenierung bezeichnen kann, sind bekannt.
Bei diesem Verfahren ist die Temperatur i;icht kritisch, d. h, die Cyclisierung erfolgt bei jeder
Temperatur. Im allgemeinen wird sie jedoch in einem Bereich von 0 bis 100°C durchgeführt. Jn
Die Umsetzung erfolgt in einem inerten nichtwäßrigen Lösungsmittel, wie Benzol, Toluol, Äthylacetat oder
Äthylendichlorid.
Ein anderes Verfahren zur Herstellung einiger erfindungsgemäßer Isothiazolone ist das Verfahren von .'>
Crow und Leonhard, das in der weiter oben angegebenen Literaturstelle »J. Org. Chem.« beschrieben
wird. Dieses Verfahren besteht darin, ein ^-substituiertes
Thiocyanacrylamid oder Thiosulfatacrylamid in das Isothiazolon umzuwandeln. Diese Umsetzung kann κι

Vorzugsweise wird die Umwandlung des substituierten Thiocyanacrylamids oder Thiosulfatacrylamids in
das Isothiazolon durch Behandeln mit einer Säure, wie -,»
beispielsweise Schwefelsäure, durchgeführt. Bedeutet jedoch M die Gruppe -SCN, dann können auch
Übergangsmetallsalze, wie Nickelsulfat, Eisen(ll)-sulfat, Eisen(lll)-sulfat, Kupfersulfat eingesetzt werden. Steht

Weise die Umwandlung mit Jod erfolgen. Die Herstellung des Thiocyanacrylamids und Thiosulfatacrylamids
nach Crow und Leonard besteht darin, daß man substituiertes Propiolairid mit Thiocyansäure oder
Thiosulfat umsetzt. hl>
Die folgenden Beispiele erläutern die Erfindung. In der folgenden Tabelle I sind die nach den vorstehend
beschriebenen Verfahren hergestellten Verbindungen 1 bis 61 aufgeführt. Die Tabelle Il zeigt die Elemcntaranalysen
und Schmelzpunkte für die in Tabelle I h-,
beschriebenen Verbindungen. Nähere Herstellungsmethoden sind nachfolgend für die Beispiele I, 3, 8, 13, 25
und 42 aufgeführt.
Eine Lösung von 5,05 Teilen (0,05 Mol) 3-Hydroxyisothiazol in 88 Teilen trockenem Benzol wurde bei
200C gerührt. Dann wurde zu der obigen Lösung eine 50%ige Benzollösung zugesetzt, die 2,85 Teile (0,05 Mol)
wiethylisoeyanat enthielt. Unmittelbar nach der Zugabe
begann sich ein weißer Niederschiag zu bilden, der beim
Rühren zunehmend dicker wurde. Dieses Rühren wurde über Nacht fortgesetzt. Filtration des Reaktionsgemisches
ergab 2,0 Teile weißes, festes 2-(N-Methylcarbamoyl)-3-isothiazolon
mit einem Schmelzpunkt von 138 bis 1400C. Nach Zugabe von 35 Teilen Hexan ergab das
Filtrat 3,5 Teile zusätzliches Produkt mit einem Schmelzpunkt von 138 bis 139°C. Auf diese Weise
erhielt man eine Gesamtausbeute von 5,5 Teilen (70%) 2-(N-Methylcarbamoyl)-3-iaOthiazolon.

Zu einer gerührten Lösung von 2,9 Teilen (0,029 Mol) 3-Hydroxyisothiazol in 88 Teilen Benzol wurden 2,1
Teile (0,029 Mol) Methylisothiocyanat zugesetzt. Das Reaktionsgemisch wurde auf 50 bis 600C zehn Stunden
erhitzt und das Reaktionsmedium verdampft, um 2,5 Teile weißes, festes 2-(N-Methylthiocarbamoyl)-3-isothiazolon
zu hinterlassen. Das Produkt wurde aus Äthanol umkristallisiert.
Zu einer gerührten Lösung von 3,0 Teilen (0,025 Mol) 3-Hydroxy-5-methyl-isothiazol in 132 Teilen trockenem
Benzol wurden 4,0 Teile (0,025 Mol) 3-Chlorphenylisocyanat
zugesetzt. Die Temperatur der exothermen Reaktion stieg von 27 auf 48°C. Nach zweistündigem
Rühren bildete sich ein weißer, fester Schlamm. Der Feststoff wurde abfiltriert und getrocknet, um 7,0 Teile
5-Methyl-2-(N-3-chlorphenylcarbamoyl)-3-isothiazolon zu ergeben.
Beispiel 13
Zu einer gerührten Lösung von 4 Teilen (0,04 Mol) 3-Hydroxyisothiazol in 88 Teilen trockenem Benzol
wurden 2,84 Teile (0,04 Mol) Äthylisocyanat tropfenweise bei 25°C zugesetzt. Die Lösung wurde bei 25°C zwölf
Stunden gerührt, dann wurde das Lösungsmittel unter Vakuum abgedampft, um 6,0 Teile gelbes, festes
2-(N-Äthylcarbamoyl)-3-isolhiazolon zu ergeben. Reinigung des Produktes erfolgt durch Umkristallisieren aus
Benzol-Hexan.

wurde durch Umsetzung von 3-Hydroxy-5-methylisothiazol und Älhylisothiocyanat gemäß dem Verfahren von Beispiel 3 hergestellt.

Formaldehydlösung in einem Anteil zugesetzt. Es bildete sich bald ein fester Niederschlag, der nach
zweistündigem Rühren abfiltriert wurde, um 1,8 g weißes, festes 2-Hydroxymethyl-3-isothiazoIon zu ergeben.

Ein Teil der erfindungsgemäßen Isothiazolone eignet sich auch für eine Saatgutbehandlung, um Mikroorganismen,
insbesondere Pilze, zu bekämpfen. Bei dieser Bekämpfungsmethode werden die Isothiazolone oder
Zubereitungen, welche diese Verbindungen enthalten, auf die Oberfläche des zu behandelnden Saatguts
aufgebracht, beispielsweise durch Aufschlämmen, Eintauchen, Bestäuben oder Besprühen.
Die erforderliche Wirkstoffmenge hängt von dem Saatguttyp, der Aufbringungsmethode, dem Boden, den
atmopshärischen Bedingungen etc. ab. Im allgemeinen werden 88 bis 1266 g des Wirkstoffs pro 100 kg Saalgut
zur Bekämpfung von Mikroorganismen angewendet, wobei eine Menge von 70 bis 635 g pro 100 kg
bevorzugt wird.
Um die Wirksamkeil der erfindungsgemäßen Verbindungen
für eine Saatgutbehandlung zu ermitteln, wird eine Methode angewendet, die darin besteht, daß man 2
Anteile von je 100 Maiskörnern mit den zu untersuchenden Isothiazolonen in einer Menge von 77 bis 160 g pro
100 1 Saatgut aufschlämmt. Zu Vergleichszwecken wird eine nichtbehandelte Vergleichsprobe getestet. Nach
dem Trocknen wird das Saatgut in einen Mistboden eingebracht, der mit Pythium übersät ist und einen
hohen Feuchtigkeitsgehalt besitzt. Die Maiskörner werden in diesem Boden 1 Woche bei etwa 1O0C
gehalten, um eine Infektion zu ermöglichen. Nach Beendigung dieser Zeitspanne wird die Temperatur
erhöht und auf etwa 21 bis 270C gehalten, um ein Keimen der nichtinfizierten Körner zu ermöglichen. Die
Zahl der keimenden Körner wird nach dem Auftauchen der Pflanze ausgezählt. Diese Werte sind in der Tabelle
IV aufgeführt.

Die erfindungsgemäßen Verbindungen können als Baktericide und Fungicide oder als Kombinationen
davon für beliebige Systeme verwendet werden, insbesondere für wäßrige Systeme, beispielsweise
Wasserkühlsysteme, Schwimmbecken und Papierkraftstoffverfahren. Außerdem können sie als Schutzmittel,
) insbesondere als Gewebeschutzmittel, Seifenzusätze.
sanitäre Mittel sowie Schutzmittel zum Bearbeiten von Metallen eingesetzte Verbindungen, verwendet werden.
Die jeweils einzusetzende Wirkstoffmenge hängt von
dem zu bekämpfenden Mikroorganismus, den jeweils
ίο eingesetzten Isothiazolonen bzw. den Zubereitungen, in
denen diese verwendet werden, ab. In einem flüssigen Medium werden in zweckmäßiger Weise die Isothiazolone
in einer Menge von 0,1 bis 10 000 Teile je Million Teile (ppm) oder 0,00001 bis 1%, bezogen auf das
Gewicht des zu behandelnden Systems, verwendet. Ein Bereich von 1 bis 2000 ppm wird bevorzugt.
Die Bekämpfungswirkung kann in einer vollständigen Abtötung, in einer Ausrottung, in einem Wachstumsstillstand
oder -hemmung, in einer Verminderung der Anzahl der Bakterien oder Pilze oder in einer
Kombination aus diesen Wirkungen bestehen.
Eine sehr gute fungistatische Aktivität durch die erfindungsgemäßen Isothiazolone wird dann erzielt,
wenn man diese Substanzen als Farbschutzmittel und
2> Farbfungistatika einsetzt. Eine mikrobielle Aktivität in
Farbemulsionen auf der Grundlage von Acrylverbindungen wird in wirksamer Weise dann gehemmt, wenn
die Verbindungen in einer Menge von etwa 42 bis 420 g/1000 1 der Farbanstrichmittel eingesetzt werden.
jo Die erfindungsgemäßen Isothiazolone sind außerdem
wertvolle Fungicide für einen Einsatz in der Landwirtschaft, wobei sie für diesen Zweck mit verträglichen
Trägermaterialien und gegebenenfalls oberflächenaktiven Mitteln oder anderen Zusatzstoffen zu Zubereitun-
j > gen formuliert werden.
Die erfindungsgemäßen Verbindungen können in einer mit Wasser mischbaren Flüssigkeit, wie Äthanol.
Isopropanol oder Aceton aufgelöst werden. Diese Lösungen lassen sich mit Wasser verdünnen.
Die Verbindungen können ferner mit einem feinteiligen festen Trägermaterial vermisch; werden, beispielsweise
Tonen, anorganischen Silikaten, Carbonaten oder Kieselsäuren. Man kann auch organische Trägermaterialien
verwenden. Staubkonzentrate werden gewöhn-
-r, lieh mit einem Gehalt der Isothiazolone in einem
Bereich von 20 bis 80% hergestellt. Für die endgültige Verwendung werden diese Konzentrate normalerweise
mit zusätzlichen Feststoffen auf etwa 1 bis 20% verdünnt.
so Benetzbare Pulverzubereitungen kann man durch
Einarbeiten der erfindungsgemäßen Verbindungen in inerte feinteilige feste Trägermaterialien zusammen mit
einem oberflächenaktiven Mittel, das auch einem oder mehreren Emulgatoren, Netzmitteln. Dispergiermitteln
oder Verteilungsmitteln oder Gemischen davon bestehen kann, herstellen. Die Isothiazolone werden dabei
gewöhnlich in einer Menge von 10 bis 80 Gew.-% und die oberflächenaktiven Mittel in einer Menge von 0.5 bis
10 Gew.-% eingesetzt. Verwendbare Emulgatoren und
bo Netzmittel sind beispielsweise polyoxyäthylierte Derivate
von Alkylphenolen, Fettalkohole, Fettsäuren und Alkylamine, Alkylarensulfonate sowie Dialkylsulfosuccinate.
Verteilungsmittel sind beispielsweise Glycerinmannitlaurat
oder ein Kondensat von Polyglycerin mit
b5 einer Phthalsäureanhydrid modifzierten Ölsäure. Dispergiermittel
sind beispielsweise das Natriumsalz des Copolymeren von Maleinanhydrid mit einem Olefin, wie
Diisobutylen, NatriumUgninsulfonat und Natriumform-
aldehydnaphthalinsulfonat.
Eine einfache Methode zur Herstellung von festen Zubereitungen besteht darin, die lsothiazolone auf
festen Trägermaterial!«;.i mit Hilfe eines flüchtigen
Lösungsmittels, wie Aceton, aufzubringen. Auf diese Weise können auch Zusatzstoffe, wie Aktivatoren,
Klebstoffe, Pflanzennährstoffe, synergistische Mittel und verschiedene oberflächenaktive Stoffe, eingearbeitet
werden.
Emulgierbare Konzentrate kann man in der Weise gewinnen, daß man die lsothiazolone in einem in der
Landwirtschaft verträglichen organischen Lösungsmittel löst und ein in dem Lösungsmittel lösliches
Emulgiermittel zusetzt. Geeignete Lösungsmittel sind üblicherweise mit Wasser nicht mischbar. Erwähnt seien
beispielsweise Kohlenwasserstoffe, chlorierte Kohlenwasserstoffe. Ketone, Ester, Alkohole und Amide.
Gewöhnlich werden Mischungen dieser Lösungsmittel verwendet. Die als Emulgatoren brauchbaren oberflächenaktiven
Mitte! können etwa 0,5 bis 10 Gew.-% des
emulgierbaren Konzentrats ausmachen und anionischen, kationischen oder nichtionischen Charakter
haben. Anionische oberflächenaktive Mittel sind beispielsweise Alkoholsulfate oder -sulfonate, Alkylarensulfonate
und Sulfosuccinate. Kationische oberflächenaktive Mittel sind beispielsweise Alkylaminsalze von
Fettsäuren und quatemäre Fettsäurealkylverbindungen. Nichtionische Emulgatoren sind beispielsweise Äthylenoxidaddukte
von Alkylphenolen, Fettalkohole, Mercaptane und Fettsäuren. Die Konzentration der aktiven
Bestandteile kann zwischen 10 und 80% variieren und liegt vorzugsweise im Bereich von 25 bis 50%.
Für die Verwendung als phytopathogene Mittel sollen diese Verbindungen nach an sich bekannten Methoden
in einer wirksamen Menge aufgebracht werden, die ausreicht, die erwünschte biocide Aktivität zu ergeben.
Gewöhnlich bringt man die lsothiazolone auf die zu schützenden Stellen, in einem in der Landwirtschaft
verträglichen Trägermaterial eingearbeitet, in einer wirksamen Menge auf. In einer bestimmten Situation
jedoch kann es erwünscht und vorteilhaft sein, die Verbindungen direkt und ohne wesentliche Menge an
Trägermaterial auf den zu schützenden Stellen aufzubringen. Dies ist eine besonders wirksame Methode,
wenn die Natur der lsothiazolone derart ist, daß sie eine sogenannte »Niedervolumenaufbringung« gestattet,
d. h., wenn die Verbindungen in flüssiger Form oder in höhersiedenden Lösungsmitteln im wesentlichen löslich
sind.
Die Aufbringungsmengen variieren selbstverständlich je nach dem Aufbringungszweck, den benützten
Isothiazolonen, der Häufigkeit der Aufbringung usw.
Bei dem Einsatz der erfindungsgemäßen lsothiazolone als Bakterieide und Fungicide in der Landwirtschaft
können auch verdünnte Sprühflüssigkeiten mit Konzentrationen von 60 bis 2980 g/1000 I der Sprühflüssigkeit
eingesetzt werden. Vorzugsweise werden diese Spriihflüssigkeiten in einer Konzentration von 120 bis
2380 g/1000 I, insbesondere in einer Konzentration von
lü 150 bis 6000/10001, verwendet. Im Falle von konzentrierteren
Sprühflüssigkeiten wird der Wirkstoff um einen Faktor von 2 bis 12 erhöht Verdünnte
Sprühflüssigkeiten werden zur Behandlung von Pflanzen gewöhnlich so lange eingesetzt, bis die Flüssigkeit
abläuft, während im Falle von konzentrierteren Sprühflüssigkeiten diese als Nebel aufgebracht werden.
Die erfindungsgemäßen Verbindungen können allein
als biocide Mittel oder in Verbindung mit anderen Fungiciden, Insekticiden, Miticiden oder vergleichbaren
Pesticiden verwendet werden.
Freilandversuche haben gezeigt, daß die erfindungsgemäßen lsothiazolone zur Bekämpfung von Pilzen, wie
Spargelkohlmehltai1 (Peronospora parasitica), staubartigem
Bohnenmehltau (Erysiphe polygoni), staubartigern Weizenmehle j (Erysiphe graminis), Reismehltau
(Piricularia oryzae), Tomatenmehltau (Phytothora infestans) sowie Alternaria solani, verwendet werden
können.
Die fungicide und baktericide Wirkung der erfin-
i<> dungsgemäßen Isoxazolone ist besser als diejenige
von bekannten Vergleichsverbindungen, wie die nachfolgenden Ergebnisse von Vergleichsversuchen zeigen,
wobei als Vergleichssubstanzen 6-Acetoxy-2,4-dimethyl-m-dioxan und Hexahydro-l,3,5-tris-(2-hydroxyäthyl)-s-triazin
verwendet wurden. Die zuerst genannte Verbindung ist unter der Handelsbezeichnung »Givgard
DXN« und »Dioxin« als Schutzmittel für wäßrige Systeme und Emulsionen im Handel. Die zweite der
genannten Verbindungen ist unter der Handelsbezeichnung »Grotan« als industrielles Bactericid im Handel.
Die Ergebnisse in der nachfolgenden Aufstellung zeigen, daß die erfindungsgemäßen Verbindungen den
bekannten Verbindungen überlegen sind.
Weiterhin wurde für eine Reihe der erfindungsgemä-Ben lsothiazolone die akute orale Toxizitat bei Warmblütern, und zwar bei Mäusen und Ratten, bestimmt. Die ermittelten Werte zeigen eindeutig, daß die Toxizitäten so gering sind, daß keinerlei Einschränkungen hinsichtlich der Verwendung als fung-statische und bakteriostatische Mittel bestehen.
Weiterhin wurde für eine Reihe der erfindungsgemä-Ben lsothiazolone die akute orale Toxizitat bei Warmblütern, und zwar bei Mäusen und Ratten, bestimmt. Die ermittelten Werte zeigen eindeutig, daß die Toxizitäten so gering sind, daß keinerlei Einschränkungen hinsichtlich der Verwendung als fung-statische und bakteriostatische Mittel bestehen.

eine Cyclohexylgruppe, eine gegebenenfalls durch eine niedermolekulare Alkylgruppe oder niedermolekulare
Alkoxygruppe substituierte Aralkylgruppe mit bis zu 10 Kohlenstoffatomen, eine 2- oder
4-Chlorbenzyl-, 2,4- oder 3,4-DichlorbenzyJgruppe,
eine Hydroxymethylgruppe, einen gegebenenfalls durch Chlor, niedermolekulare Alkylgruppen, eine
Nitrogruppe oder Carbäthoxygruppe substituierten Phenylrest oder eine Carbamoylgruppe der allgemeinen
