Abstract
Die Erfindung betrifft die Verwendung eines Bakterizids, ausgewählt aus Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, beispielsweise in einem Kühlschmierstoff.
Description
- Die vorliegende Erfindung betrifft die Verwendung von Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, beispielsweise in einem Kühlschmierstoff.
- Von Formaldehyd ist allgemein eine mykobakterizide Wirkung bekannt (siehe u.a. S.S. Block, Hrsg., Desinfection, Sterilization, und Preservation, 5. Auflage, 2001, Lippincott, Williams & Wilkins). Außerdem beschreiben die
DE 198 42 116 A1 ,DE 199 61 621 A1 undDE 197 22 858 A1 die Verwendung von Formaldehyd- und formalhaltigen Formulierungen in Kühlschmierstoffen. - Es hat sich allerdings herausgestellt, dass beispielsweise in der metallbearbeitenden Industrie durch Prozessflüssigkeiten und Kühlschmierstoffemulsionen verursachte Berufskrankheiten auftreten, die zur Gruppe der exogenen-allergischen Alveolitiden (hypersensitivity pseumonitis (HP)) gehören. Exogenallergische Alveolitiden sind akute, subakute und chronische Lungenentzündungen, die durch eingeatmete Antigene verursacht werden. Hierzu gehören die Farmerlunge, die Vogelhalterlunge und die Befeuchterlunge sowie eine Reihe seltener beobachteter Erkrankungen. Die Exposition gegenüber Kühlschmierstoffen wird ferner mit Asthma und möglicherweise auch Krebs in Zusammenhang gebracht.
- Als mögliche Ursache für exogen-allergische Alveolitiden werden Mykobakterien diskutiert. Mykobakterien sind vergleichsweise widerstandsfähig gegen biozide Wirkstoffe, insbesondere in Konzentrationsbereichen, wie sie üblicherweise in Prozessflüssigkeiten zum Einsatz kommen. Gesucht werden deshalb wirksaure Zubereitungen, die beim Einsatz in üblichen Konzentrationen das Wachstum von Mykobakterien in Prozessflüssigkeiten wirksam unterdrücken und das Risiko von betriebsbedingten Erkrankungen im Umgang mit diesen Flüssigkeiten deutlich mindern. Dafür sind bisher u.a. Bakterizide auf Basis von Phenolen vorgeschlagen worden.
- Weil in den Kühlschmierstoffen als biozider Wirkstoff überwiegend 2,2',2''-(Hexahydro-1,3,5-triazin-1,3,5-triyl)triethanol (HHT) eingesetzt worden ist, hat man eine unzureichende Wirksamkeit gegenüber Mykobakterien und sogar eine spezifische Wachstumsförderung von Mykobakterien bei Überdosierung dieses Bakterizids vermutet. Deshalb hat mindestens ein weltweit agierendes Unternehmen der Automobilindustrie festgelegt, dass Triazine und andere Formaldehyd freisetzende Verbindungen nicht mehr als Bakterizide in den Formulierungen von Metallbearbeitungsfluiden (metal working fluids, MWFs) eingesetzt werden dürfen. Die Suche nach Alternativen hat sich dementsprechend auf Bakterizide beschränkt, deren Wirkung nicht auf der speziellen bioziden Wirksamkeit von Formaldehyd-Kondensationsprodukten basiert (es wird angenommen, dass die Hydrolyse von Formaldehyd-Kondensationsprodukten an der Zellwand des Mykobakteriums dessen Peptidoglucanstruktur wirksam denaturiert und somit zum Zelltod führt).
- Aufgabe der Erfindung war es, Bakterizide bereitzustellen, die beim Einsatz in üblichen Konzentrationen das Wachstum von Mykobakterien in Prozessflüssigkeiten wirksam unterdrücken und das mit HHT beobachtete Risiko von betriebsbedingten Erkrankungen im Umgang mit diesen Flüssigkeiten deutlich mindern.
- Es wurde nun überraschend gefunden, dass durch Verwendung eines Bakterizids ausgewählt aus Formaldehyd und Formaldehyd freisetzende Verbindungen die o.g. Aufgabe gelöst und die Probleme des Standes der Technik überwunden werden. Die Erfindung beruht somit auf der Erkenntnis, dass Formaldehyd und (von HHT verschiedene) Formaldehyd freisetzende Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien
- i) auch bei Überdosierung das Wachstum von Mykobakterien nicht fördern und
- ii) gegen Mykobakterien wirksamer sind, also bei gleicher Einsatzkonzentration wirksamer sind als HHT und/oder die gleiche Wirksamkeit gegen Mykobakterien wie mit HHT bei einer geringeren Einsatzkonzentration erreicht wird.
- Somit besteht die Erfindung in der Verwendung eines Bakterizids ausgewählt aus Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, die dadurch gekennzeichnet ist, dass bei Überdosie rung des Bakterizids keine Förderung des Wachstums von Mykobakterien auftritt.
- Die Erfindung besteht ferner in der Verwendung eines Bakterizids ausgewählt aus Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, die dadurch gekennzeichnet ist, dass die Wirksamkeit des Bakterizids gegen Mykobakterien bei gleicher Einsatzkonzentration größer ist als die Wirksamkeit von 2,2',2''-(Hexahydro-1,3,5-triazin-1,3,5-triyl)triethanol oder die gleiche Wirksamkeit gegen Mykobakterien wie mit 2,2',2''-(Hexahydro-1,3,5-triazin-1,3,5-triyl)triethanol bei einer geringeren Einsatzkonzentration des Bakterizids erreicht wird.
- Erfindungsgemäß werden Mykobakterien vorzugsweise in technischen Produkten, wie Kühlschmierstoffemulsionen, wasserbasierten Prozessflüssigkeiten, Reinigungsmitteln und Spülflüssigkeiten bekämpft.
- Die erfindungsgemäß eingesetzte Formaldehyd freisetzende Verbindung ist vorzugsweise ausgewählt aus 3,3'-Methylenbis[5-methyloxazolidin], 1,3-Bis(hydroxymethyl)harnstoff, (Ethylendioxy)dimethanol, 2-(2-Butoxyethoxy)ethoxymethanol; Tetrahydro-1,3,4,6-tetrakis(hydroxymethyl)imidazo[4,5-d]imidazol-2,5(1H,3H)-dion, α,α',α''-Trimethyl-1,3,5-Triazin-1,3,5-(2H,4H,6H)-triethanol und Mischungen davon. Häufig liegen Substanzgemische und nicht eindeutig identifizierte Substanzen, die sich im dynamischen Gleichgewicht befinden, vor. Die erfindungsgemäß vorzugsweise eingesetzten Formaldehyd freisetzenden Verbindungen sind Kondensationsprodukte von Formaldehyd (oder Paraformaldehyd) und Alkanolaminen und/oder Glykolen und/oder Harnstoff oder Harnstoffderivaten. Demzufolge werden erfindungsgemäß beispielsweise eingesetzt:
- a) Das Kondensationsprodukt aus Isopropanolamin und Paraformaldehyd (Wirkstoff: 3,3'-Methylenbis[5-methyloxazolidin]), gegebenenfalls stabilisiert mit Antioxidantien,
- b) das Kondensationsprodukt aus Isopropanolamin, Paraformaldehyd und Harnstoff, gegebenenfalls stabilisiert mit Antioxidantien,
- c) das Kondensationsprodukt aus Isopropanolamin, Paraformaldehyd, Harnstoff und Ethylenglykol, gegebenenfalls stabilisiert mit Antioxidantien,
- d) das Kondensationsprodukt aus Paraformaldehyd, Ethylengly kol, Butyldiglykol und Harnstoff (Wirkstoffe: (Ethylendioxy)dimethanol, 2-(Butoxyethoxy)ethoxymethanol und 1,3-Bis(hydroxymethyl)harnstoff),
- e) das Kondensationsprodukt aus Paraformaldehyd, Ethylenglykol und Harnstoff (Wirkstoffe: (Ethylendioxy)dimethanol und 1,3-Bis(hydroxymethyl)harnstoff),
- f) das Kondensationsprodukt aus Isopropanolamin und Paraformaldehyd (Wirkstoff: α,α',α''-Trimethyl-1,3,5-triazin-1, 3, 5- (2H, 4H, 6H) -triethanol),
- g) das Kondensationsprodukt aus Formaldehyd (oder Paraformaldehyd) und Glyoxal und Harnstoff bzw. aus Formaldehyd (oder Paraformaldehyd) und Glycoluril, (Wirkstoff: Tetrahydro-1,3,4,6-tetrakis(hydroxymethyl)imidazo[4,5-d]-imidazol-2,5(1H, 3H)-dion).
- Darüber hinaus sind Benzylalkoholhemiformal (das Kondensationsprodukt aus Benzylakohol und Formaldehyd) und 4,4-Dimethyloxazolidin, 1-(3-Chlorallyl)-3,5,7-triaza-1-azo niatricyclo[3.3.1.13,7]decanchlorid, Bronopol, 7a-Ethyldihydro-1H,3H,5H-oxazolo[3,4-c]oxazol bevorzugt.
- Die überraschend beobachteten Effekte treten auch bei Verwendung eines Gemischs des erfindungsgemäß eingesetzten Bakterizids mit einem oder mehreren Wirkstoffen auf. Dies zeigt sich bei Vergleich der Wirksamkeit eines solchen erfindungsgemäßen Gemischs mit der Wirksamkeit von HHT und gegebenenfalls auch im Vergleich mit der Wirksamkeit eines Gemischs von HTT mit dem (oder den) Wirkstoff(en). Erfindungsgemäß erfolgt der Einsatz des Bakterizids somit vorzugsweise zusammen mit einem oder mehreren weiteren Wirkstoffen ausgewählt aus 3-Iod-2-propinylbutylcarbamat, N-Cyclohexyl-N-nitrosohydroxylamin-Salzen (bevorzugt dem Kaliumsalz), Phenolen wie o-Phenylphenol, p-Chlor-m-kresol, p-Chlor-m-xylenol, Alkoholen wie Phenoxyethanol, Phenoxypropanole, Thiabendazol oder dessen Salzen, p-[(Diiodmethyl)sulfonyl]toluol, 2-Brom-2-(brommethyl)pentandinitril, Isothiazol-3-onen wie 5-Chlor-2-methyl-2H-isothiazol-3-on und/oder 2-Methyl-2H-isothiazol-3-on, 1,2-Benzisothiazol-3(2H)-on, n-Butyl-1,2-benzisothiazol-3(2H)-on und 2-Octyl-3(2H)isothiazol-3-on sowie Salzen von 2-Mercaptopyridin-N-oxid wie dem Natrium- oder Zinksalz (Pyrion-Na, Zn-pyrithion). Erfindungsgemäß werden besonders bevorzugt Gemische mit Pyrion-Na und/oder Isothiazolonen eingesetzt, wie beispielsweise Gemische mit 1,2-Benzisothiazol-3(2H)-on oder 2-Octyl-3(2H)-isothiazol-3-on.
- Die Erfindung betrifft ferner die Verwendung von Isothiazol-3-onen zur Erhöhung der mykobakteriziden Wirksamkeit von Formaldehyd und Formaldehyd freisetzenden Verbindungen. Wie sich insbesondere aus den Beispielen ergibt, erhöhen Isothiazol-3-one, die selbst nicht als typische mykobakterizide Wirkstoffe bekannt sind, überraschend die mykobakterizide Wirksamkeit von Formaldehyd und Formaldehyd freisetzenden Verbindungen.
- Erfindungsgemäß erfolgt der Einsatz des Bakterizids vorzugsweise zur Bekämpfung von M. chelonae, M. immunogenum, M. abscessus, M. avium und M. terrae, bevorzugter nichttuberkulösen schnellwachsenden Mykobakterien, wie M. immunogenum und M. chelonae.
- Gemäß der vorliegenden Erfindung ist es möglich, Mykobakterien insbesondere in Kühlschmierstoffen wirksamer zu bekämpfen. Dazu gehört, dass die erfindungsgemäß eingesetzten Bakterizide überdosiert werden können, ohne dass die Gefahr der Förderung des Wachstums von Mykobakterien auftritt. Dabei bezeichnet der Ausdruck "Überdosierung" eine Konzentration des Bakterizids, die höher liegt als die Mindesteinsatzkonzentration des Bakterizids. Die "Mindesteinsatzkonzentration" des Bakterizids ist diejenige Konzentration, die ausreicht, die Zahl der Mykobakterien einer Probe deutlich zu verringern, vorzugsweise um den Faktor 10, bevorzugter um den Faktor 100 und besonders bevorzugt um den Faktor 1000, oder das Wachstum zu hemmen.
- Dabei haben sich bestimmte Konzentrationsbereiche für das erfindungsgemäß eingesetzte Bakterizid als überraschend wirksam erwiesen. So ist beispielsweise das Kondensationsprodukt aus Paraformaldehyd, Ethylenglykol, Butyldiglykol und Harnstoff (Wirkstoffe: (Ethylendioxy)dimethanol, 2-(Butoxyethoxy)ethoxymethanol und 1,3-Bis(hydroxymethyl)harnstoff) bereits in einer Einsatzkonzentration von 0,15 Gew.-% gegen M. chelonae wirksam, während dafür 0,3 Gew.-% HHT notwendig sind. Die notwendige Menge Formaldehyd freisetzende Verbindung kann für die gleiche Wirksamkeit also geringer gewählt werden.
- Bei Einsatz eines erfindungsgemäßen Bakterizids (oder mehrerer erfindungsgemäßer Bakterizide in Kombination), gegebenenfalls im Gemisch mit einem oder mehreren weiteren Wirkstoffen, beträgt die empfohlene Einsatzkonzentration somit vorzugsweise weniger als 0,3 Gew.-%, wie weniger als 0,25 Gew.-%, beispielsweise 0,1 bis 0,2 Gew.-%, vorzugsweise 0,1 bis 0, 15 Gew.-%.
- Die Vorteile der vorliegenden Erfindung ergeben sich insbesondere aus den folgenden Beispielen.
- Methode zur Bestimmung der Wirksamkeit von bakterizidhaltigen Lösungen gegen Mykobakterien
- – Anzucht der Mykobakterien gemäß DGHM (2001),
- – 5 ml doppelt konzentrierte Testlösung werden mit 5 ml doppelt konzentriertem Flüssigmedium (7H9-Medium) versetzt,
- – Zugabe von 0,1 ml Mykobakteriensuspension (Titer: 1 × 109 – 8 × 109 KBE/ml; Herstellung der Mykobakteriensuspension gemäß DGHM (2001),
- – Inkubation der Teströhrchen bei 28 °C oder 37 °C (entsprechend Mycobakterien-Teststamm) für 3 Wochen (Standkultur),
- – Überprüfung der Hemmwirkung der einzelnen Verdürtnungen durch Untersuchung auf optische Trübung, hervorgerufen durch bakterielles Wachstum sowie zusätzlichen Ausstrich auf 7H10-Argar,
- – Inkubation der Agarplatten für 3 Wochen bei 28 °C oder 37 °C (je nach Teststamm),
- – Kontrolle der Agarplatten auf mykobakterielles Wachstum
- Alle Prozentangaben beziehen sich auf das Gewicht (Gew.-%).
- In den Beispielen bedeuten:
M. terrae: ATCC 15755 (37°C), M. chelonae: ATCC 35725 (28°C), M. chelonae: ATCC 19236 (37°C), M. immunogenum: ATCC 700505 (28°C, 37°C), *: Konzentration in unverdünnter MHK-Testlösung (0,6%ig), MHK: Minimale Hemmkonzentration.
Claims (6)
- Verwendung eines Bakterizids ausgewählt aus Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, dadurch gekennzeichnet, daß bei Überdosierung des Bakterizids keine Förderung des Wachstums von Mykobakterien auftritt.
- Verwendung eines Bakterizids ausgewählt aus Formaldehyd und Formaldehyd freisetzenden Verbindungen in einer Zusammensetzung zur Bekämpfung von Mykobakterien, dadurch gekennzeichnet, daß die Wirksamkeit des Bakterizids gegen Mykobakterien bei gleicher Einsatzkonzentration größer ist als die Wirksamkeit von 2,2',2''-(Hexahydro-1,3,5-triazin-1,3,5-triyl)triethanol oder die gleiche Wirksamkeit gegen Mykobakterien wie mit 2,2',2''-(Hexahydro-1,3,5-triazin-1,3,5-triyl)triethanol bei einer geringeren Einsatzkonzentration des Bakterizids erreicht wird.
- Verwendung nach Anspruch 1 oder Anspruch 2, dadurch gekennzeichnet, daß die Formaldehyd freisetzende Verbindung ausgewählt ist aus 3,3'-Methylenbis[5-methyloxazolidin], 1,3-Bis(hydroxymethyl)harnstoff, (Ethylendioxy)dimethanol, 2-(2-Butoxyethoxy)ethoxymethanol, Tetrahydro-1,3,4,6-tetrakis(hydroxymethyl)imidazo[4,5-d]imidazol-2,5(1H,3H)-dion, α,α',α''-Trimethyl-1,3,5-Triazin-1,3,5-(2H,4H,6H]-triethanol und Mischungen davon.
- Verwendung nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, daß das Bakterizid im Gemisch mit einem oder mehreren weiteren Wirkstoffen ausgewählt aus 3-Iod-2-propinylbutylcarbamat, N-Cyclohexyl-N-nitrosohydroxylamin-Salzen (bevorzugt dem Kaliumsalz), Phenolen wie o-Phenylphenol, Isothiazol-3-onen wie 5-Chlor-2-methyl-2H-isothiazol-3-on und/oder 2-Methyl-2H-isothiazol-3-on, 1,2-Benzisothiazol-3(2H)-on, n-Butyl-1,2-benzisothiazol-3(2H)-on und 2-Octyl-3(2H)isothiazol-3-on sowie Salzen von 2-Mercaptopyridin-N-oxid wie dem Natrium- oder Zinksalz (Pyrion-Na, Zn-pyrithion) eingesetzt wird, vorzugsweise im Gemisch mit Pyrion-Na, 1,2-Benzisothiazol-3(2H)-on oder 2-Octyl-3(2H)-isothiazol-3-on.
- Verwendung nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, daß das Mykobakterium ausgewählt ist aus M. chelonae, M. immunogenum, M. abscesses, M. avium und M. terrae, vorzugsweise M. chelonae und M. immunogenum.
- Verwendung von Isothiazol-3-onen zur Erhöhung der mykobakteriziden Wirksamkeit von Formaldehyd und Formaldehyd freisetzenden Verbindungen.
