Die Erfindung betrifft Verfahren zur Herstellung neuer
Reihen von lj^-disubstituierten Piperazin- und Diazepinderivaten und ihrer therapeutisch wertvollen SKureadditionssalze.
Die neuen Verbindungen werden durch die nachfolgende allgemeine Formel gekennzeichnet:

Das niedere Alkyl und niedere,Alkoxy besitzt von 1 bis 5
Kohlenstoffatome und umfasst gerade oder verzweigte gesättigte aliphatisch^ Ketten, wie z.B. Methyl, Äthyl, Propyl, Isopropyl,
Butyl, Isobutyl, tert. Butyl, Pentyl und dergleichen
und dementsprechend Methoxy, Äthoxy, Propoxy, Isopropoxy usw.
und Halogen umfasst Chlor, Brom, Fluor und Jod, zweokraässig
Fluor und Chlor.
Die Verbindungen nach dem Verfahren der vorliegenden Erfindung können in ihre therapeutisch wertvollen Säureadditionssalze
durch Reaktion mit einer entsprechenden Säure, beispielsweise einer anorganischen Säure, wie Halogenwasserstoff
säure, d.h. Chlorwasserstoff-, Bromwasserstoff- oder Jodwasserstoffsäure; Schwefel-, Salpeter- oder Thiocyansäure;
einer Phosphorsäure; einer organischen Säure, wie Essig-, Propion-,
Glycol-, Milch-, Brenztrauben-, Oxal-, Malon-, Sucoin-,
Malein-, Fumar-, Apfel-, Wein-, Zitronen-, Benzoe-, Zimt-,
Mandel-, Methansulfön-, Äthansulfon-, Hydroxyäthansuifon-,
p-Toluolsulfon-, Salicyl-, p-Aminosallcyl-, 2-Phenoxybenzoe-
oder 2-Acetoxybenzoesäure umgewandelt werden.
Die Verbindungen gemäss der Erfindung können hergestellt
werden durch Reaktion einer Verbindung der allgemeinen Formelt

Diese Reaktionen werden zweckmässig unter Erhitzen der Reaktionsteilnehmer in Gegenwart eines geeigneten inerten organischen
Lösungsmittels, wie nachfolgend beschrieben, unter Rückflussbedingungen durchgeführt.
Die Verbindungen der Formel II und der Formel V werden
durch Reaktion eines wasserfreien Piperazins oder Perhydro-1,4-diazepins
der allgemeinen Formel VI mit einer Verbindung der Formel IV oder Formel III erhalten:

Die Verbindungen der Formel II und V können zweckeässig
auch durch Reaktion einer Verbindung der Formel VI hergestellt werden, die eine geeignete Blockierungsgruppe, wie
eine Benzylgruppe, an einem ihrer Ringstickstoffatorae trägt,
mit einer Verbindung der Formeln IV oder III entspreohend den vorgenannten Reaktionsschemen und anschliessender Debenzylierung
der erhaltenen Verbindung. Eine solche Debenzylierung
wird zweckmässig mit Hilfe von Wasserstoff durchgeführt, der durch einen geeigneten Katalysator, beispielsweise Palladiumauf-Holzkohle,
in einem alkoholischen Lösungsmittel, wie Äthanol, aktiviert ist. Eine solche Verwendung ist auch zweckmässig,
wenn m des Reaktionsteilnehmers VI gleich 3 ist, d.h. bei der Herstellung neuer Diazepinderivate gemäss der Erfindung.
Die Verbindungen gemäss der Erfindung haben antigefässverengende
und antihistaminische Wirksamkeit, wie am isolierten Meerschweinchendarm bestimmt worden ist. Zusätzlich haben
sie gezeigt, dass sie eine oder mehrere der nachfolgenden Eigenschaften besitzen:
1.) Eine Koronarvasodilation von langer Dauer, beobachtbar
bei oraler, parenteraler und intravenöser Anwendung der Drogen bei Hunden, besonders, wenn Ar, und/oder Ar« in der
vorgenannten Formel für Fluorphenyl oder Phenyl steht, in 2 oder 3 ist, η zweckmässig 1 1st, R Wasserstoff oder Methyl

Wirksamkeit werden gezeigt in einer zweckmässigen Unterklasse
der obengenannten Formel, in der Ar1 für Fluorphenyl, Alkylphenyl,
Phenyl oder Thienyl steht, Ar2 Pluorphenyl oder Phenyl,
m 2 oder 3, η zweckmässig 1, R Wasserstoff oder Methyl,
X Methylen, R-, Wasserstoff oder niederes Alkyl und Ar, Phenyl
oder substituiertes Phenyl, beispielsweise Halogenphenyl oder Alkylphenyl, darstellt.
Wahlweise können die Verbindungen, in denen X Methylen ist, durch Reaktion einer Verbindung der Formel II:

aktionsfähigen Ester Y mittels einer starken anorganischen oder organischen Säure nach bekannten Methoden überführt,
worauf die Verbindung VIII mit einer Verbindung der Formel?

Die Acylierung der Verbindungen der allgemeinen Formel I, in der R, Wasserstoff ist, wird durch Erhitzen einer solchen
Verbindung mit einem Acylierungsmittel, wie einem Anhydrid einer niederen Fettsäure oder einem niederen Alkylcarbonylhalogenid
unter Rückflussbedingungen in Gegenwart eines nichtwässrigen inerten organischen Lösungsmittels durchgeführt.
Schliesslich können die Verbindungen der allgemeinen Formel I, in der X Carbonyl und/oder R1 niederes Alkylearbonyl
ist, umgewandelt werden in ihre reduzierten Gegenstücke, d.h. in dem Falle, in dem X Methylen und/oder R1 niederes Alkyl
ist, durch Reaktion mit einem überschuss eines gemischten Metallhydrids, wie Lithiumaluminiumhydrid oder dergleichen, in
einem geeigneten inerten organischen Lösungsmittel, wie Tetrahydrofuran, Dioxan oder Diäthyläther. Unter solchen Bedingungen
wird das Carbonyl von R^+ zu dem entsprechenden Carbinoi
reduziert. Die Reduktion der R^-Carbonylgruppe zu einer Methylengruppe
wird durch amalgamlertes Zink und Chlorwasserstoffsäure (Clemmensen-Reduktion) oder durch alkalische Zersetzung
der Hydrazone, Semicarbazone oder Azine (Wolff-Kishner-Reduktion)
durchgeführt.
Abhängig von den während des Reaktionsverlaufs angewendeten Bedingungen werden die neuen Verbindungen entweder in Form
der freien Basen oder der Salze derselben erhalten. Die Salze werden in üblicher Weise in die freien Basen umgewandelt, beispielsweise
durch Reaktion mit Alkali, wie Natrium- oder Kaliumhydroxyd.
Die Basen können in ihre therapeutisch wertvollen Säureadditionssalze durch Reaktion mit einer entsprechenden
organischen oder anorganischen Säure umgewandelt werden.
Vorteilhafte organische Lösungsmittel zur Durchführung
der erforderlichen Reaktionen gemäss der Erfindung umfassen
Ketone, wie Aceton oder 4-Methyl-2-pentanonj aromatische und
aliphatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylol oder Heptan; Äther, wie Tetrahydrofuran, Dioxan oder Diäthyläther.
In entsprechenden Pillen werden Lösungsmittel, wie

dere Alkanole, beispielsweise Äthanol, 2-Propanol oder Buta
nol, verwendet.
Die nachfolgenden Beispiele sollen die Erfindung erläu tern, aber den Schutzbereich derselben nicht begrenzen.
1.) Einer warmen Mischung von 121 Teilen 2,6-Dimethylanilin
in 600 Teilen Äthanol (4o° C) wird tropfenweise eine
Lösung von 55 Teilen Äthylenoxyd in 4oo Teilen Äthanol unter
Rühren zugesetzt. Nach Beendigung des Zusatzes wird das Ganze Über Nacht bei Raumtemperatur gerührt. Die Mischung wird gerührt
und 5 Stunden unter Rückfluss gehalten und dann eingedampft. Der ölige Rückstand wird im Vakuum destilliert und
führt zu N-(2-Hydroxyäthyl)-2,6-dimethylanilin; Kp. 100 bis 125° C bei 0,15 mm Druck.
2.) Eine Mischung von 4} Teilen N-(2-Hydroxyäthyr)-2,6-dimethylanilin

Temperatur so eingestellt, dass nach 15 Stunden das ganze Was ser destilliert ist (Temperatur: 150° C). Dann wird ein dritter
Anteil von 150 Teilen Bromwasserstoffsäurelösung, 48 #ig,
zugesetzt und die Temperatur des Ölbads so eingestellt, dass nach 3 Stunden' das ganze Wasser destilliert ist (Temperatur
etwa 16O° C). Der feste Rückstand wird in 160 Teilen Äthanol gekocht, nochmals heiss filtriert, mit 2-Propanol gewaschen,
getrocknet und führt zu N-(2-Brom&thyl)-2,6-dimethylanilinhydrobromidj Smp. 230 bis 237° C*
3.) Eine Mischung von 94 Teilen 4-ChlQr-l,l-di-(4-methylphenyl)-l-buten und 240 Teilen 2-Propanol wird bei normalem
Druck und bei einer Temperatur von etwa 135° C in Gegenwart

hydriert. Nachdem die berechnete Menge Wasserstoff aufgenommen worden ist, wird die Hydrierung abgebrochen. Der Katalysator
wird abfiltriert und nach dem Kühlen des Filtrate auf -10° C wird die niedergeschlagene feste Masse abfiltriert und
getrocknet und führt zu l-Chlor-4,4-di-(4-methylphenyl)-butan;
Smp. 44 bis 46° C.
4.) Einer gekühlten Lösung (etwa 10° C) von 4g,5 Teilen
l-/2T,4-Di-(4-fluorphenyl)-butyi7-piperazin in 240 Teilen Methanol
wird eine Lösung von 20 Teilen Äthylenoxyd in 40 Teilen
Methanol zugesetzt. Nach Beendigung des Zusatzes wird das Ganze auf 40° C erhitzt und beim Halten auf dieser Temperatur
wird ein Überschuss an gasförmigem Äthylenoxyd im Verlauf von 20 Minuten eingeleitet. Das Ganze wird eingedampft. Der Rückstand
wird in einer Mischung von 2-Propanol und Diisopropyläther gelöst und gasförmiger Chlorwasserstoff eingeleitet. Das
niedergeschlagene Salz wird abfiltriert und getrocknet und führt zu l-/4",4-Di-(4-fluorphenyl)-butyl7-^-(2-hydrox|räthyl)-piperazindihydrochlorid;
Smp. 175 bis 186° C.
Einer Mischung von 80 Teilen Thionylchlorid und 75 Teilen Chloroform werden portionsweise 19,5 Teile 1-/5,4-Di-(4-fluorphenyl)-butyl7-4-(2-hydroxyäthyl)-plperazindihydrochlorid
zugesetzt. Nach Beendigung des Zusatzes wird das Ganze 3 Stunden unter Rückfluss gehalten. Die Reaktionsmischung wird abgedampft.
Der feste Rückstand wird aus kochendem Aoeton Ußikristallisiert,
abfiltriert und getrocknet und führt zu l~/¥#4-Di-(4-fluorphenyl)-butyl7-4-(2-chlorKthyl)-piperazindihydfoohlorid;
Smp. 210 bis 212° C (Zers.).
5.) Einer gekühlten (10° Cs Eiebad) Misohung yott 105
Teilen Essigsäure und l8 Teilen 2-Propionylänilin werden tropfenweise
17 Teile dhloracetylqhlorid *(ejtotherme Reaktion) zugesetzt.
Nach Beendigung des Zusatzes wird das Ganze iSj Minu*
ten b·! 10° C gerührt. Dann werden tropfenweise 152 Tfile \u,
einer Natriumacetat-Wasser-Lösung (vorher hergestellt jaus $%
Teilen Natriumacetat und 128 Teilen Wasser) zugesetzt. Nach
Beendigung des Zusatzes wird das niedergeschlagene Produkt abfiltriert, mit Wasser gewaschen und getrocknet und führt zu
N-(2-Chloracetyl)-2-propionylanilin; Stnp. 76,5 bis 77*5° C
Wenn man in diesem Beispiel 2-Propionylanilin durch 3-Propionylanilin
ersetzt und das vorgenannte Verfahren wiederholt, erhält man N-CChloracetylJ^-propionylanilin; Smp. 69,5
bis 70,5° C.
6.) Eine Mischung aus 23,8 Teilen N-(2-Chloracetyl)-2,6-dimethylanilin,
ig Teilen dl-l-Benzyl-2-methylpiperazin,
32 Teilen Natriumcarbonat und wenigen Kristallen Kaliumjodid
in 520 Teilen 4-Methyl-2-pentanon wird gerührt und 48 Stunden
unter Rückfluss gehalten. Nach dem Kühlen werden 300 Teile
Wasser zugesetzt. Die organische Schicht wird abgetrennt,
über Magnesiumsulfat getrocknet und eingedampft. Der ölige Rückstand wird in Diisopropyläther gelöst. Die Lösung wird
bis zur Klarheit filtriert und es wird gasförmiger Chlorwasserstoff
in das FiItrat eingeleitet. Der feste Niederschlag wird abfiltriert, in 2-Propanol gekocht, wieder abfiltriert
und getrocknet und führt zu einem klebrigen Salz. Dieses wird in Aceton gerührt, wieder getrocknet und führt zu dl-1-Benzyl-2~methyl-4-/l["2,6-dirae
thylanilincarbonyl) -methyl7-pip©r&zindihydrochloridj

in 500 Teilen 4-Methyl~2-pentanon wird tropfenweise eine Lösung von 3.3,7 Teilen l-Chlor-4,4-di-(4-fluorphenyl)-butan in
50 Teilen 4-Methyl-2-pentanon zugesetzt. Nach Beendigung des Zusatzes wird das Ganze weiter gerührt und 46 Stunden unter
Rückfluss gehalten. Nach dem Kühlen werden 300 Teile Wasser zugesetzt. Die organische Schicht wird abgetrennt, über
Magnesiumsulfat getrocknet, filtriert und eingedampft. Der ölige Rückstand wird in 800 Teilen Diisopropyläther gelöst.
Die trübe Lösung wird mehrere Male bis zur Klarheit filtriert und dann wird gasförmiger Chlorwasserstoff in das Filtrat
eingeleitet. Das niedergeschlagene feste Salz wird abfiltriert und aus kochendem 2-Propanol umkristallisiert. Nach
dem Kühlen auf Raumtemperatur wird die feste Masse abfiltriert und getrocknet und führt zu dl-l-/4",4-Di-(4-fluorphe-'
nyl)-butyl^-^-methyl-^-benzylpiperazindihydrochlorid; Sap.

Wenn man dieses Beispiel wiederholt, aber dl-l-Benzyl-2-methylpiperazin

9.) 202 Teile N-(2-Bromäthyl)-anilinhydrobromld werden portionsweise 494,5 Teile Piperazin in 2.000 Teilen 2-Propanol
bei Raumtemperatur in einem Zeitabschnitt von 3 Stunden zugesetzt. Das Ganze wird dann über Nacht gerührt, worauf die
niedergeschlagene feste Masse abfiltriert wird. Das Filtrat wird eingedampft und der Rückstand alkalisch gehalten. Das
Ganze wird mit Chloroform extrahiert. Der Extrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet, filtriert
und eingedampft. Der ölige Rückstand wird im Vakuum destilliert und führt zu 4-(2-Anilinäthyl)-piperazinj Kp. I4o bis
l60° C bei 0,4 bis 0,5 mm Druck.
Bei Wiederholung des Verfahrens unter Ersatz von N-(2-Bromäthyl)-anilinhydrobromid
durch eine äquimolare Menge von N-(2-Chloracetyl)-2,6-dimethylanilin erhSlt man die Verbindung 4-^2,6-Dimethylanilincarbonyl) -methyl7-piperazin; Smp.

umkristallisiert. Nach dem Kühlen über Nacht auf Raumtemperatur wird die feste Masse abfiltriert., im Vakuum getrocknet und
führt zu l-(4/4-Diphenylbutyl)-4-(2-anilinäthyl)-piperazintrihydroehlorld;
Smp. 227 bis 229° C.

15·) Eine Mischung von 20,8 Teilen l-(4,4-Diphenylbutyl)-4-(2-anilinäthyl)-piperazintrihydrochlorid,
20 Teilen Kssigsäureanhydrid, 56 Teilen Triethylamin und 6OO Teilen
Chloroform wird gerührt und 2 Stunden unter Rückfluss gehalten. Nach-dem Kühlen werden 500 Teile Wasser zugesetzt und das
Ganze wird durch Zusatz von Natriumhydroxydlösung stark alkalisch gemacht. Die organische Schicht wird dekantiert, dreimal
mit 50, Teilen Wasser gewaschen, über Magnesiumsulfat getrocknet
und eingedampft. Der ölige Rückstand wird in wasserfreiem Äther gelöst und es wird gasförmiger Chlorwasserstoff in die
Lösung eingeleitet. Das niedergeschlagene Salz wird abfiltriert und zweimal umkristallisiert: zunächst aus 240 Teilen 2-Propanol
und dann aus einer Mischung von 36O Teilen 2-Propanol und
240 Teilen Aceton und man erhält l-(4,4-Diphenylbutyl)-4-/2-(N-acetylanilin)-äthy_l7-piperazindihydrochlorid;
Smp. 221 bis 223° C.
Bei Wiederholung des vorgenannten Verfahrens und Ersatz des 1-(4,4-Diphenylbutyl)-4-(2-anilinäthyl)-piperazintr!hydrochloride
durch äquimolare Mengen des entsprechenden 1-(4,4-Diarylbutyl)-4-f2-anilinäthyl)-piperazins
und Acylierung dieser Verbindung mit einer entsprechenden Menge eines geeigneten Säureanhydrids
erhält man die in der Tabelle IV aufgeführten Verbindungen:

16.) Eine wässrige Lösung von 14 Teilen dl-l-/4~-Phenyl-4-(4-fluorphenyl)-buty17-4-/2-(anilinearbonyl)-äthy17-plperazindihydrochlorid
wird mit Natriumhydroxyd alkalisch gemacht und mit Chloroform extrahiert. Der Extrakt wird über Magnesiumsulfat
getrocknet, filtriert und eingedampft. Der Rückstand wird in 100 Teilen Tetrahydrofuran gelöst. Dieser Lösung wird
eine Suspension von 2,1 Teilen Lithiumtetrahydridaluminat in
100 Teilen Tetrahydrofuran zugesetzt, während die Temperatur auf etwa 30° C (leicht exotherme Reaktion) gehalten wird. Das
Ganze wird gerührt und 3 Stunden unter Rückfluss gehalten. Nach dem Kühlen der Reaktionsmischung unterhalb 15° C wird sie
zersetzt durch aufeinander folgenden Zusatz von 7 Teilen Wasser, 17,5 Teilen Natriumhydroxydlösung, 20 1StIg, und 7 Teilen
Wasser. Die Mischung wird filtriert und der gelatinöse Niederschlag wird zweimal mit Äther gewaschen. Die organischen
Schichten werden über Kaliumcarbonat getrocknet, abfiltriert und eingedampft. Der Rückstand wird in 56O Teilen fither gelöst.
Die Lösung wird über Kaliumcarbonat getrocknet, filtriert
und es wird gasförmiger Chlorwasserstoff in das FiI-trat
eingeleitet. Das niedergeschlagene Salz wird abfiltriert und aus einer Mischung von 400 Teilen Methanol und 20 Teilen
Wasser umkristallisiert und führt zu dl-l-/4"-Phenyl-4-(4-fluorphenyl)-butyl7-4-(5-anilinpropyl)-piperazintrihydrochloridj
Smp. 247 bis 250,5° C.
Bei Wiederholung des Verfahrens unter Ersatz des dl-1-A-Phenyl-4-(4-f
luorphenyl) -bu tyl7-4 -/5- (ani lincarbonyl) äthy!7-piperazindihydrochlorids
durch geeignete Mengen des entsprechenden Anilids werden die folgenden Verbindungen erhalten:
1-/4", 4-Di-(4-f luorphenyl )-butyjL7-4-( 3-anilinpropyl)-piperazintrihydrochlorid;
Smp. 235,5 bis 236,5° C.
dl-l-/4",4-Di-(4-f luorphenyl)-butyl/-4-( l-anilin-2-propyl)-piperazindioxalatj
Smp. 224 bis 225° C.

17.) Einer Mischung von 1,28 Teilen Lithiumtetrahydridaluminat
und 45 Teilen Tetrahydrofuran wird tropfenweise eine
Lösung von 8 Teilen l-(4,4-Diphenylbutyl)-4-/?-(N-propionylanilin)-äthy.l7-piperazin
in 45 Teilen Tetrahydrofuran (leicht exotherme Reaktion) zugesetzt. Nach Beendigung des Zusatzes
wird das Ganze gerührt und 3 Stunden unter Rückfluss gehalten. Nach dem Kühlen wird die Reaktionsmischung durch nacheinander
folgenden Zusatz von 5 Teilen Wasser, 5 Teilen Natriumhydroxydlösung,
20 iClg, und 10 Teilen Wasser zersetzt.
Das Ganze wird von dem Unlöslichen abfiltriert und das Filtrat
wird eingedampft. Der ölige Rückstand wird in wasserfreiem Sther gelöst und es wird gasförmiger Chlorwasserstoff in die
Lösung eingeleitet. Das niedergeschlagene Salz wird abfiltriert und aus 120 Teilen Methanol amkristallisiert und führt
zu l-(4,4-Diphenylbutyl)-4-i/2-(N-propylanilinJ-äthyj;7-plpe.razindihydrochlorid;
Smp. 210 bis 213,5° C.
Bei Wiederholung des Verfahrens unter Ersatz des l-(4,4-Diphenylbutyl)-4-/2-(N-propionylanilin)-äthy_l7-piperazins
durch eine äquimolare Menge der entsprechenden i-i/4~,4-Di-(4-fluorphenyl)-butyJL7-4-/2"-(N-acylanilin)-äthyJ;7-p:perazine
werden die folgenden Verbindungen erhalten:
