技术领域
本发明涉及一种含有多酚类物质作为活性成分的α-淀粉酶活性抑制剂,所述多酚类物质来自于番石榴(Psidium guajava Linn.)(此后也称为番石榴多酚)。还涉及含有该抑制剂的食品和饮料。
技术背景
近年来消费者越来越关注对能量摄入的限制,因为能量摄入过多是很多由健康习惯引起的成人疾病的主要原因。如果有某种物质能够在生物体内抑制或阻遏所摄入食物向能量转化,这些物质对那些需要节食的人来说就很有意义,因为这些物质可以使这些人不用减少其食物摄入。特别是,认为抑制以淀粉为代表的碳水化合物的消化对于预防和治疗肥胖症意义更大。因此,近年来能够抑制淀粉消化酶α-淀粉酶活性的物质便越来越引起了人们的关注。
番石榴是一种起源于中美洲的灌木,它的果实、根和叶子一直被民间用于治疗糖尿病和腹泻。依最近的研究,用水或者一种亲水性溶剂提取番石榴叶子得到的一种提取物抑制α-淀粉酶活性。日本专利公告第60-36746号公开了该提取物可以作为健康饮料的成分,日本专利公开第7-59539号公开了该提取物可以作为减肥食品和饮料的成分。
但是传统的番石榴提取物还含有倍半萜、单宁及其他成分,并且它们的淀粉酶抑制剂活性也达不到上述节食作用的要求。
因此,本发明的目的之一是从番石榴提取物中分离一种组分,该组分具有很强的α-淀粉酶抑制活性,因此可能产生一种更有效的节食效果。本发明的另一个内容是含有此组分的食品和饮料。

发明公开
如前文所述,本发明人对番石榴提取物中的高分子量组分进行了广泛研究,发现用本发明的方法获得的一组多酚类物质具有很高的α-淀粉酶抑制活性。简言之,依本发明,对番石榴叶子或果实中获得一种水性提取物进行超滤,将分子量在5000或5000以上的级分用特定疏水色谱的方法进行分级分离。本发明就是建立在上述发现的基础之上。
因此,本发明提供了一种α-淀粉酶活性抑制剂,此抑制剂含有多酚类物质作为活性成分,所述多酚类物质是通过下面的工艺获得:用选自水和其他亲水溶剂的溶剂对番石榴叶子和/或果实进行提取;对所得提取物进行超滤,由此去掉分子量小于5000的物质;剩余级分上样于丁基支持物疏水层析柱,用0.02mol/L磷酸二氢钠水溶液与0.02mol/L磷酸钠水溶液组成的水性溶液在两溶液间的pH梯度下进行洗脱(流速:1ml/分钟);从洗脱的物质中回收一个级分,该级分相应于当在260nm下检测物质的光吸收时洗脱曲线中的第三个单峰。
本发明还提供了一种α-淀粉酶活性抑制剂,其含有多酚类物质作为活性成分,这些多酚类物质具有以下物理化学特性:
(a)含有碳(49.6%)、氢(4.6%)和氮(0.6%);
(b)分子量在5000--100000;
(c)在3,428cm-1,1705cm-1,1615cm-1和1220cm-1处有强红外吸收;
(d)固态碳核磁共振光谱显示与糖类信号(76ppm附近)、芳香族化合物信号(115.0ppm附近)、酚类化合物信号(144和156ppm附近)以及酯羰基信号(168ppm附近)相吻合;
(e)在下述条件下进行液相色谱时10分钟左右出现一单峰:
柱:填充物为硬质合成多聚反相分配胶,丁基以化学键与之结合(型号:Shodex Asahipak C4P-50 4D,Showa Denko产品),内径:4.6mm,长:150mm,或类似于此柱的柱)。
流速:1.5ml/分钟;
柱温:40℃左右;
检测:紫外光吸收仪(波长260nm);
流动相:溶液A:乙腈和水的混合物,pH4.6,含有磷酸二氢钠(0.02mol/L),其中乙腈和水的体积比为15∶85;溶液B:乙腈和水的混合物,pH11.4,含有磷酸钠(0.02mol/L),其中乙腈和水的体积比为15∶85;
分析方法:按下表所列数据进行梯度分析。

实施本发明的最佳方案:
本发明多酚类物质可用下述步骤1-3生产。
1)步骤1
用选自水和亲水性溶剂的一种或多种溶剂提取番石榴的叶子和/或果实,以得到番石榴提取物。
本发明中使用的番石榴为Psidium guajava Linn.,对其种植地没有特别限制。本发明中,番石榴是东南亚、南亚、南美或北美等热带或亚热带地区天然或人工栽培的。本发明中使用的是番石榴的叶子或果实。可以使用新鲜、半干或者干燥番石榴,但优选干燥的番石榴叶子。使用番石榴果实时,优选未成熟果实。这些番石榴叶子或者果实作为原料时在使用前优选地经粉碎。比如,当使用番石榴叶时,番石榴叶子最好切成约3-5mm的小片。另外为了改进味道,可以使用在干燥后再经焙烤的干燥番石榴。
选自水和亲水性溶剂的一种或多种溶剂用作提取溶剂。亲水溶剂的例子包括甲醇、乙醇、正丙醇、丙酮和丙二醇。上述溶剂可以两种或多种组合使用,也可以与水按任何比例组成含水溶剂使用。为了操作方便并提高安全性,优选用水作为提取溶剂。
对所用溶剂的量没有特别限制。当使用番石榴叶子时,叶子和溶剂的重量比优选1∶20;当时用未成熟番石榴果实时,溶剂的重量优选约为果实的约10倍。
对番石榴叶进行提取时,提取条件因使用的溶剂不同而不同。以水为溶剂时,提取在50-100℃进行,优选80-100℃;时间为5-60分钟,优选5-25分钟。
当使用未成熟番石榴果实时,提取优选在60-100℃进行10-60分钟。
进行提取时,可在提取溶剂中加入如碳酸钠的碱以增加溶剂的pH值。也可以向提取溶剂中加入稀无机酸(如稀盐酸)或者有机酸(如柠檬酸、琥珀酸、乳酸、苹果酸或酒石酸)使溶剂变为弱酸性。
提取步骤完成后,优选将所得提取物冷却然后离心,除去杂质。这样得到的番石榴提取物可经浓缩或者稀释以达到合适浓度。
2)步骤2
对步骤1得到的番石榴提取物进行超滤,以此除去分子量小于5000的组分。
对于番石榴提取物的超滤,可以使用加压过滤装置、透析和离心等、也可以使用上述方法和技术的组合。
当进行透析时,将番石榴提取物装入透析管。透析管由一种半透膜材料比如纤维素酯或者塞璐玢制成,其允许分子量小于5000的物质通过。然后对水或者对稀缓冲液(此后称透析液)透析1-7天,最好2-3天,期间定期更换新鲜透析液或水。通过上述步骤,从番石榴提取物中除去小分子量物质。
3)步骤3
对步骤2获得的分子量为5000或大于5000的级分进行丁基支持物疏水层析。将样品上柱,用磷酸二氢钠(0.02mol/L)到磷酸钠(0.02mol/L)pH梯度进行洗脱(流速1.0ml/分钟)。
对疏水层析中使用的固定相填料没有特别限制,只要其含有作为功能基团的丁基和具有足够的机械强度即可。填料的例子包括聚苯乙烯、羧化聚苯乙烯、聚甲基丙烯酸酯以及纤维素衍生物。
以磷酸二氢钠(0.02mol/L)水溶液和磷酸钠(0.02mol/L)水溶液为流动相,在二者之间的浓度梯度下进行洗脱。
溶剂以1.0ml/分钟的流速通过柱,检测洗脱液在260nm处的光吸收值获得被洗脱物质的分离模式。回收相应于第3个单一峰的级分,将所得级分真空或者冷冻干燥,得到本发明中的多酚类物质。
如此得到的水溶性级分的物理化学性质将在下面给出。
元素分析
用通常方法进行元素分析,结果如表2所示。

分子量
用通常的GPC-HPLC方法用多角度光散射检测仪测定该组分分子量,结果表明该级分的分子量在5000-100000范围内,平均分子量为70000。

该级分的糖含量用下述方法测定:将样品在4mol/L三氟乙酸溶液中保持100℃2小时使之加热并分解,依常规方式形成糠醛乙烯氰衍生物,然后用气相色谱法计算糖含量。结果:葡萄糖(2.1%),阿拉伯糖(2.0%),半乳糖(1.3%),甘露糖(0.4%),鼠李糖(0.3%),木糖(0.3%)。
没食子酸含量、联二没食子酸含量和原花色素含量
将样品在含有硫酸铁(III)铵的丁醇-盐酸混合液(95∶5,V/V)中于100℃加热24小时以使样品分解,然后用HPLC方法分析分解的样品中没食子酸和联二没食子酸含量。没食子酸和联二没食子酸的含量分别为0.6%和5.2%。用上述溶剂于95℃加热样品40分钟分解样品,根据分解溶液的光吸收值计算确定样品中原花色素含量。原花色素以还原为花色素计含量为6.8%。
红外吸收光谱
将样品夹在两层二氟化钡晶体中间,加压滚压。用透射法测定光吸收。所获得的红外吸收光谱中,此组分的强吸收峰大约在3428cm-1,1705cm-1,1615cm-1和1220cm-1处。红外吸收光谱见图1。
固态碳核磁共振谱(固态
13
C-NMR)
采用通常的正交极化幻角自旋(CP-MAS)方法。获得的CP-MAS光谱中可见糖类、芳香族化合物类、酚类和酯羰基化合物的信号。光谱证明样该级分由主要是含有一个鞣花单宁基团的多酚。CP-MAS光谱见图2。
从以下的实施例中将会看到,上述番石榴多酚具有很强的α-淀粉酶抑制剂活性,同时也有α-葡萄糖苷酶抑制剂活性。因此,当摄入添加了这类多酚类物质的食物时,淀粉降解为糊精和麦芽糖的过程将被抑制,同时二糖(麦芽糖、异麦芽糖和蔗糖等)降解成葡萄糖的过程也将被抑制。这样,多酚类物质的添加可赋予食物和饮料防止血糖升高和抵抗肥胖的作用。
本发明涉及到的食品和饮料的类型包括固态食品、半液态食品、胶质食品和各种饮料。具体的例子包括片剂、胶囊类产品和颗粒;糖果类食品如小甜饼、果冻和快餐食品;颗粒状调味品、面包和面条;以及饮料类如提神饮料、果汁、以及由乳酸菌制备的饮料等。
除了本发明中的多酚类物质外,还有很多常用的食品添加剂也可以添加到上述食品和饮料中。这类添加剂包括糖醇类(如葡萄糖、蔗糖、果糖、山梨糖醇、木糖醇、赤鲜糖醇、乳糖醇和palatinit);乳化剂如蔗糖脂肪酸酯、多聚甘油脂肪酸酯和卵磷脂);稳定剂(如果胶、羧甲基纤维素、水性大豆多糖、古兰糖胶、阿拉伯树胶、黄原胶、角叉藻聚糖和瓜耳豆胶)。其他可掺入的物质的例子包括维生素类(如维生素A、B族维生素、维生素C和维生素E);矿物质(如乳酸钙、葡萄糖酸钙、泛酸钙、镁化合物和锌化合物);和植物提取物。
当将本发明中的多酚类物质添加到食品和饮料中时,多酚的总量随食品和饮料的形态而异,优选在0.005-0.5%(重量百分比)。每日摄入这类食品和饮料的量折算成该多酚类物质应在5-500mg,优先选择5-100mg。
实施例
下面通过实例对本发明进一步描述,但本发明并不仅限于这些例子。
实施例1:番石榴提取物制备
(1)番石榴叶子(生长于中华人民共和国,广西)干燥,121℃焙烤15分钟,仔细切碎至约5mm大小。取这种切碎的番石榴叶子100公斤,置于2000公斤80℃水中浸提25分钟。将所得提取液冷却至30℃以下,1500rpm离心10分钟以澄清溶液,得到番石榴叶子提取物。
(2)未成熟番石榴果实的提取通过用提取液(约10重量份)与果实(约1重量份)于90℃提取25分钟。
(3)将未成熟果实提取物按约0.1-0.2%的比例加入到番石榴叶子提取物中,得到番石榴提取物。
实施例2:番石榴多酚的制备
用超滤方法对实施例1中制备的番石榴提取物进行过滤(除去分子量小于5000的级分),得到分子量大于5000的级分。将得到的级分用透析膜透析(除去分子量6000-8000的级分),将透析袋内的级分冷冻干燥,得到粗多酚制品。将粗多酚制品溶于0.02mol/L磷酸二氢钠水溶液中,再用疏水层析方法对上述溶液进行纯化(含有丁基相联的填料之大制备柱)。具体地,用二倍于柱体积的0.02mol/L磷酸二氢钠溶液洗涤填料,然后用二倍于柱体积的0.02mol/L磷酸二氢钠溶液和0.02mol/L磷酸钠溶液混合液在两溶液之间的浓度梯度下(即:浓度按比例改变)进行洗脱。随后用0.02mol/L磷酸钠溶液洗脱。检测剩余级分在260nm处的光吸收,以此绘出洗脱曲线,分析淀粉酶抑制剂活性情况(见图3)。从图3的曲线中可见,洗脱液的最后一个单峰与抑制活性峰重合。收集相应于此单峰的级分,进行电透析,冷冻干燥,得到纯化的番石榴多酚之产物。
用上述检测条件(e)中的条件对纯化的番石榴多酚进行层析分析,结果见图4。从图4可见,本发明纯化的番石榴多酚在上述条件下用层析谱色层检测表现为单一峰。
实施例3:α-淀粉酶抑制剂活性
对实施例1和实施例2中得到的番石榴提取物和纯化的番石榴多酚对α-淀粉酶活性的抑制作用进行了研究。
α-淀粉酶抑制活性用常规方法测定。具体的,将来自猪胰液α-淀粉酶与0.02M磷酸钠缓冲液(pH6.5)混合制备淀粉酶溶液。用8%的可溶性淀粉和0.08M的磷酸钠缓冲液(pH6.5)制备底物溶液。将酶、底物溶液和测试溶剂混合物37℃反应7分钟,以酶、底物溶液和水的混合物为对照。加热至100℃中止反应。用HPLC方法检测形成的麦芽糖,以阿拉伯糖为内对照。用下面公式计算抑制剂活性。结果见表3。

如表3所示,本发明的纯化番石榴多酚对淀粉酶的抑制作用比番石榴提取物高大约30%。
实施例4:α-葡萄糖苷酶抑制活性
对实施例1和实施例2中得到的番石榴提取物和纯化的番石榴多酚对麦芽糖酶(α-葡萄糖苷酶的一种)活性的抑制作用进行了研究。
麦芽糖酶抑制活性测定用常规方法。将大鼠肠丙酮粉末(SIGMA)于56mM顺丁烯二酸缓冲液(pH6.0)中匀浆,其中顺丁烯二酸缓冲液约为丙酮粉末的9倍,收集离心上清液得到麦芽糖酶粗制液。用224mM顺丁烯二酸缓冲液(pH6.0)制备底物溶液,使麦芽糖终浓度为1%。将酶、底物溶液和测试溶液的混合物37℃反应5分钟,以酶、底物溶液和水的混合物为对照。加热至100℃中止反应。用HPLC方法检测形成的葡萄糖的量,以阿拉伯糖为内对照。用下面公式计算抑制剂活性。结果见表4。

实施例5:
按下述配方,用实施例2所制备的纯化番石榴多酚制作面包。

烤制后,本面包具有草药香气和很好的味道,与市场上常见的面包产品的口味相当。
工业实用性
本发明α-淀粉酶抑制物与番石榴提取物相比具有较强的α-淀粉酶抑制活性,同时具有α-葡萄糖苷酶抑制活性。因此,将此多酚类物质加到食品和饮料中时,可赋予这些食品和饮料抑制血糖水平和抗肥胖症的作用。具体来说,将此多酚类物质加到富含淀粉的食物中,如调味粉(condiment flour)、面条、面包、甜点等食品中,可提供适合于高血糖和高血脂人群的减肥食品和饮料。
Claims (5)
1.一种α-淀粉酶活性抑制剂,其包含作为活性成分的多酚类物质,所述多酚类物质是通过以下工艺获得的:用一或多种溶剂对番石榴叶子和/或果实进行提取,所用溶剂选自水和亲水性溶剂;对所得提取物进行超滤以除去分子量小于5000的物质;将剩余级分上样于丁基支持物疏水层析柱,用0.02mol/L磷酸二氢钠水溶液和0.02mol/L磷酸钠水溶液组成的水性溶液在pH梯度下以1.0ml/分钟的流速洗脱;回收洗脱级分,所述级分相应于260nm下检测物质的光吸收时洗脱曲线的第三个单峰。
2.权利要求1中的α-淀粉酶活性抑制剂,其中多酚类物质具有以下理化性质:
(a)含有碳49.6%、氢4.6%和氮0.6%;
(b)分子量在5000-100000;
(c)在3,428cm-1,1,705cm-1,1,615cm-1和1,220cm-1处有强红外吸收;
(d)表现稳定的固态碳核磁共振光谱,与76ppm附近的糖信号、115.0ppm附近的芳香族化合物信号、144和156ppm附近的酚类化合物信号以及168ppm附近的酯羰基信号相吻合;
(e)在下述条件下进行液相色谱约10分钟后显示单一峰:
柱:填充物为硬质合成多聚反相分配胶,丁基以化学键与之结合,型号:Shodex Asahipak C4P-504D,Showa Denko产品,内径:4.6mm,长:150mm,或类似于此柱的柱
流速:1.5ml/分钟;
柱温:40℃左右;
检测:紫外光吸收仪,波长260nm;
流动相:溶液A:乙腈和水的混合物,pH4.6,含有0.02mol/L磷酸二氢钠,其中乙腈和水的体积比为15∶85;溶液B:乙腈和水的混合物,pH11.4,含有0.02mol/L磷酸钠,其中乙腈和水的体积比为15∶85;

3.权利要求1到2中任一项的α-淀粉酶活性抑制剂,其还具有α-葡萄糖苷酶抑制剂活性。
4.含有权利要求1到2中任一项所述的α-淀粉酶活性抑制剂的食品和饮料。
5.权利要求1到2中任一项所述的α-淀粉酶活性抑制剂在制备减肥食品和饮料中的应用。
