发明背景
在过去的几年中,基于抗体的治疗方法的发展已取得显著的进步。例如,研究者不仅已鉴定出多种癌症特异的标记而且还鉴定出与这些标记特异结合的多种抗体。抗体可用于将特定分子,例如毒素或免疫刺激部分,例如细胞因子,递送到表达标记的癌细胞以选择性杀伤癌细胞(参见如,美国专利Nos.5,541,087和5,650,150)。
KS-1/4抗体为指向人上皮细胞粘附分子(EpCAM)的鼠源单克隆抗体。EpCAM在某些上皮细胞的顶端面以非常低的水平表达。例如,EpCAM在肠细胞面向摄入的食物且远离循环的细胞表面进行表达,其在此处将不会接触到免疫系统的多数蛋白质和细胞(Balzar等.[1999]分子医学杂志(J.Mol.Med.)77:699-712)。
但在某些情形下,EpCAM在某些细胞如上皮来源的肿瘤细胞上高度表达。通常,这些肿瘤细胞已经失去极性,这导致EpCAM表达于细胞的整个表面。因此,EpCAM是将基于抗体的免疫刺激部分定向到肿瘤细胞的方便的肿瘤特异性标记(Simon等.[1990]Proc.Nat.Acad.Sci.USA 78:2755-2759;Perez等.[1989]免疫学杂志(J Immunol.)142:3662-3667)。

尽管已发展了多种途径,包括例如嵌合抗体、人源化抗体和贴面化(veneering)抗体,但,本领域内仍存在对可结合癌症特异标记且当施用给人类时具有减低免疫原性的抗体的需要。此外,本领域内存在对这样抗体的需要,所述抗体可以将毒素或免疫刺激部分以例如,融合蛋白质或免疫缀合物的形式递送到癌症特异标记处以选择性杀伤肿瘤细胞。
发明概要
本发明部分基于对重组抗体的鉴定,所述抗体特异结合人EpCAM但在人类中其免疫原性低于模板,即鼠抗-EpCAM抗体的免疫原性。具体地,本发明提供了这样的重组KS抗体,在该抗体中定义一个或多个构架区和/或互补决定区的氨基酸序列已经经过了修饰从而降低了其在人类中的免疫原性。
如此处所应用,术语“抗体”和“免疫球蛋白”应理解为(i)完整抗体(例如,单克隆抗体或多克隆抗体),(ii)其抗原结合部分,包括,例如,Fab片段、Fab′片段、(Fab′)2片段、Fv片段、单链抗体结合位点、sFv,(iii)双特异性抗体和其抗原结合部分,和(iv)多特异性抗体和其抗原结合部分。
如此处所应用,术语“特异结合”、“特异地结合”和“特异的结合”意指抗体对特定抗原的结合亲合力为至少约106M-1、更优选地至少约107M-1、更优选地至少约108M-1、且最优选地至少约1010M-1。

本发明提供特异结合人上皮细胞粘附分子(EpCAM)的重组抗体。本发明优选的抗体具有导致在人类中免疫原性降低的改变的可变区。
本发明的抗体可变区特别可用于将抗体和抗体融合蛋白质靶向到在人类患者中过表达EpCAM的肿瘤组织处。在优选的实施方案中,本发明的抗体融合到细胞因子上以形成免疫-细胞因子。
本发明的蛋白质序列

在另一优选的实施方案中,本发明的重组抗-EpCAM抗体具有限定免疫球蛋白重链FR3的氨基酸序列,在此FR3区具有至少一个以下的氨基酸:Xaa68位F或V;Xaa73位E或D;Xaa84位N或S;Xaa85位N或S;Xaa88位N或A;以及Xaa93位T或V。更优选地,重组抗-EpCAM抗体具有限定免疫球蛋白重链FR3的氨基酸序列,在此FR3区具有至少一个以下的替换:Xaa68位V;Xaa73位D;Xaa84位S;Xaa85位S;Xaa88位A;以及Xaa93位V。
在一优选实施方案中,重组抗-EpCAM抗体具有限定免疫球蛋白重链FR4的氨基酸序列,所述序列可由SEQ ID NO:6的106-116位残基表示,即W-G-X-G-T-S-V-T-V-S-S。更特别地,重组抗-EpCAM抗体在此FR4区具有至少一个以下的氨基酸,例如Xaa108位Q或T。更优选地,重组抗-EpCAM抗体在此FR4区具有氨基酸替换,例如,Xaa108位T。
因此,相应于鼠KS-1/4可变区,优选的V区在VH和/或VK区的FR结构域中包含替换。此外,本发明优选的V区相对于鼠KS-1/4可变区不包括氨基酸插入或缺失。
优选的变体包括所具有的可变区与鼠KS-1/4的同一性/同源性高于80%的蛋白质。鼠KS可变区或其部分的氨基酸序列可用作参考序列,以确定候选序列是否具有足够的氨基酸相似性以致在本发明的方法中具有合理预期的成功。优选地,变体序列与鼠KS可变重链或轻链FR或CDR至少有70%类似或60%相同,更优选地至少75%类似或65%相同,且最优选地80%类似或70%相同。
为测定候选肽序列与鼠KS序列是否具有所需的相似性或同一性百分比,候选氨基酸序列和鼠KS序列首先应用动态程序运算法则(Smith和Waterman(1981)分子生物学杂志(J.Mol.Biol.)147:195-197)并联合在Henikoff和Henikoff(1992)PNAS 89:10915-10919的图2中描述的BLOSUM62替换矩阵进行比对(align)。对于本发明,空位(gap)插入罚分的合适数值为-12,且空位延伸罚分的合适数值为-4。应用Smith-Waterman运算法则和BLOSUM62矩阵进行比对的计算机程序,如GCG程序包(oxford Molecular Group,oxford,英国)可从厂商获得并且是本领域内的技术人员广泛应用的。一旦完成候选和参考序列间的比对,即可计算出相似性百分比得分。根据其彼此的相似性,每一序列的各氨基酸进行顺序比较。如果BLOSUM62矩阵中对应于两个比对的氨基酸的数值为零或负数,则成对相似性得分为零;否则成对相似性得分为1.0。粗相似性得分为比对氨基酸的成对相似性得分的总和。然后将粗得分除以较小的候选或参考序列中的氨基酸数目进行标准化。标准化的粗得分为相似性百分比。备选地,为计算百分比同一性,每一序列的比对氨基酸再次顺序比较。如果氨基酸不相同,则成对同一性得分为零;否则成对同一性得分为1.0。粗同一性得分为相同比对氨基酸的总和。然后将粗得分除以较小的候选或参考序列中的氨基酸数目进行标准化。标准化的粗得分为同一性百分比。用于计算相似性和同一性百分数时,忽略插入和缺失。相应地,空位罚分不用于此计算,尽管其用于初始的比对中。
本发明也公开了用于测试KS抗体在细胞中的表达的方法,所述细胞如哺乳动物细胞、昆虫细胞、植物细胞、酵母细胞、其它真核或原核细胞(参见实施例1)。在一优选的方法中,KS抗体V区作为完整的人抗体的组分进行表达,且真核细胞系的抗体表达通过检测人Fc区的ELISA进行分析。为精确定量KS抗体与EpCAM的结合,可应用Biacore试验。
用KS抗体融合蛋白质治疗人类疾病
本发明也公开了可用于治疗人类疾病的KS抗体-IL2融合蛋白质的序列,所述疾病如癌症。某些KS抗体-IL2融合蛋白质,例如KS-1/4-IL2(参见如,实施例X中的构建体3),可用于治疗人类癌症患者,且针对抗体的免疫反应小得令人惊异。
已发现,在应用KS-1/4(VH2/VK1)-IL2治疗人类癌症的过程中观察到的免疫原性甚至小于KS-1/4(构建物3)-IL2使用时的免疫原性。特别是,在临床试验过程中,观察到具有抗独特型抗体和抗抗体-IL2连接处或抗IL-2部分的抗体的患者的频率甚至低于使用KS-1/4(构建物3)-IL2时的频率。本发明的抗体可变区也可与其它细胞因子融合,例如白细胞介素1、2、6、10或12;α和β干扰素;TNF,和γINF。可通过参考以下非限定的实施例更为充分地理解本发明。
实施例
实施例1.用于表达KS抗体和分析其抗原结合活性的方法和试剂
1A.细胞培养和转染
以下的普通技术用于随后的实施例中。对于瞬时转染,通过质粒DNA与磷酸钙共沉淀将质粒DNA导入到人肾293细胞中[Sambrook等.(1989)分子克隆:实验室指南(Molecular Cloning:A Laboratory Manual),ColdSpring Harbor,NY]。
为获得稳定转染的克隆,质粒DNA通过电穿孔导入小鼠骨髓瘤NS/0细胞。NS/0细胞培养在补充有10%胎牛血清的DMEM(Dulbecco改良的Eagle培养基)中。约5 x 106个细胞用PBS洗一次并重悬在0.5ml磷酸盐缓冲液(PBS)中。10μg的线性化质粒DNA与细胞在Gene脉冲杯(Gene PulserCuvette,0.4cm电极间距,BioRad)中冰上孵育10分钟。应用Gene脉冲仪(Gene Pulser,BioRad)进行电穿孔,操作设置为0.25V和500μF。细胞在冰上复苏1omin,之后重悬在生长培养基中并置于两个96孔板上。通过在100nM氨甲蝶呤(MTX)存在条件下生长筛选稳定转染的克隆,所述氨甲蝶呤在转染后两天引入。每3天饲喂细胞2-3次以上,抗MTX的克隆在2-3周内出现。通过抗-人Fc的ELISA测试克隆上清以鉴定出高产的克隆[Gillies等(1989)免疫学方法杂志(J.Immunol.Methods)125:191]。分离高产克隆并在含100nM MTX的生长培养基中增殖。
1B.ELISA
应用三种不同的ELISA来测定MTX抗性克隆的上清中及其它测试样本中蛋白质产物的浓度。应用抗-huFc的ELISA测定含人Fc的蛋白质,例如嵌合抗体的数量。应用抗-huκ ELISA测量κ轻链(嵌合或人免疫球蛋白的)的数量。应用抗-muFc ELISA测量测试样本中含muFc的蛋白质的数量(参见以下的实施例1C)。
抗-huFc ELISA在以下详细描述。
A.包被平板
用AffiniPure山羊抗-人IgG(H+L)(Jackson Immune Research)包被ELISA平板,所述抗体为5μg/ml溶于PBS中,且以100μl/孔置入96孔板(Nunc-Immuno plate Maxisorp)。给包被的平板盖上盖子并在4℃过夜孵育。然后用溶于PBS的0.05% Tween(Tween20)洗涤4次,并用200μl/孔的含1% BSA/1%山羊血清的PBS封闭。用封闭缓冲液37℃孵育2小时后,平板用溶于PBS的0.05% Tween洗涤4次并在纸巾上拍干。
B.测试样本与二抗的孵育
将测试样本在样本缓冲液中稀释成适当的浓度,所述样本缓冲液为含1% BSA/1%山羊血清/0.05% Tween的PBS。用浓度已知的嵌合抗体(具有人Fc)制备标准曲线。为制备标准曲线,在样本缓冲液中进行系列稀释以得出范围从125ng/ml到3.9ng/ml的标准曲线。稀释的样本和标准品以100μl/孔加到平板中,且平板在37℃孵育2小时。
孵育过后,平板用含0.05% Tween的PBS洗涤8次。然后每孔加入100μl的二抗,所述抗体为辣根过氧化物酶(HRP)-缀合的抗人IgG(JacksonImmuno Research),在样本缓冲液中约1:120,000稀释。对于每一批HRP-缀合的抗-人IgG,必须确定二抗的精确稀释。37℃孵育2小时后,平板用含0.05% Tween的PBS洗涤8次。
C.显色
底物溶液以100μl/孔加入平板。将30mg的邻苯二胺二盐酸盐(OPD)(1片)溶解到15ml的0.025M柠檬酸/0.05M Na2HPO4缓冲液(pH5)中,所述缓冲液含0.03%新鲜加入的H2O2,以制备底物溶液。室温置黑暗中30min以允许产生颜色。显色时间根据包被的平板、二抗等的批次差异而改变。观察标准曲线中的颜色生成以确定终止反应的时间。通过加入100μl/孔的4N H2SO4终止反应。通过平板读取器读取平板,所述读取器在490nm和650nm处均进行设定,并通过程序化从490nm的OD值中减去650nm的背景OD。
抗-人κ ELISA根据如以上描述的程序进行,不同之处为二抗是辣根过氧化物酶缀合的山羊抗-人κ(Southern Biotechnology Assoc.Inc.,Birmingham,AL),应用时1:4000稀释。
抗-muFc ELISA的程序也是类似的,不同之处在于ELISA平板用5μg/ml的AffiniPure山羊抗-鼠IgG(H+L)(Jackson Immune Research)的PBS溶液以100μl/孔进行包被;且二抗为辣根过氧化物酶缀合的山羊抗-muIgG,Fcγ(Jackson ImmunoResearch),应用时1:5000稀释。
1 C.克隆KS抗原(KSA,EpCAM)并以人EpCAM-鼠Fc的可溶性形式进行表达
应用Dynabeads mRNA Direct试剂盒(Dynal,Inc.,Lake Success,NY)根据生产商的指南从LnCAP细胞制备信使RNA(MRNA)。用寡聚(dT)和逆转录酶合成第一链cDNA后,通过聚合酶链反应(PCR)克隆编码上皮细胞粘附分子(也称为KS抗原或KSA)的全长cDNA。PCR引物的序列基于在Perez和Walker(1989)免疫学杂志(J.Immunol.)142:3662-3667中描述的发表序列。有义引物的序列为
TCTAGAGCAGCATGGCGCCCCCGCA(SEQ ID NO:27),且反义引物的序列为CTCGAGTTATGCATTGAGTTCCCT(SEQ ID NO:28),其中黑体处表示翻译起始密码子和翻译终止密码子的反密码子,且划线处为XbaI(TCTAGA)和XhoI(CTCGAG)限制性位点。克隆PCR产物并测定几个独立克隆的序列以确认正确的KSA序列。来自LnCAP的KSA的cDNA序列与来自UCLA-P3细胞的KSA的发表序列(Perez和Walker,1989)基本相同。但是,在115位氨基酸残基处,来自LnCAP的核苷酸序列为ATG而不是ACG(Met代替Thr),以及在277位氨基酸残基处,来自LnCAP的核苷酸序列为ATA而不是ATG(Ile代替Met)。
KS-1/4抗体与重组KSA的结合通过免疫染色进行证明。KSA的表面表达通过用在适宜的哺乳动物表达载体(pdC,如在美国专利号5,541,087中所描述的)中的全长KSA转染细胞(例如,CT26、B16等)而获得,随后用KS-1/4抗体进行免疫染色。KSA作为可溶性抗原表达时,将编码KSA的跨膜结构域的cDNA部分缺失。为促进表达、缺失和纯化,可溶性KSA表达为KSA-muFc,其构建在以下描述。编码可溶性KSA的780bp的XbaI-EcoRI限制性片段与编码muFc(美国专利号5,726,044)的AflII-XhoI片段连接,所述连接通过如下接头-衔接子进行:

编码可溶性KSA-muFc的XbaI-XhoI片段与pdC载体连接。导致产生的pdCs-KSA-muFc表达载体用于转染细胞,并通过抗-muFc的ELISA鉴定表达KSA-muFc的稳定克隆。
1D.抗原结合的测定
首先根据供应商的操作指南(Repligen,Cambridge,MA)通过蛋白质A层析来纯化条件培养基中的KSA-muFc。纯化的KSA-muFc用于包被96孔板(Nunc-Immuno plate,Maxisorp),所述KSA-muFc为5μg/ml的PBS溶液,且以100μl/孔进行包被。试验与在实施例1B中描述的ELISA方法类似。简言之,包被的平板加盖在4℃过夜孵育。然后洗涤并封闭平板。测试样本在样本缓冲液中稀释至适当的浓度,以100μl/孔加到平板中,且平板在37℃孵育1小时。孵育过后,平板用含0.05% Tween的PBS洗涤8次。然后每孔加入100μl的二抗,所述二抗为辣根过氧化物酶缀合的抗-人IgG(Jackson Immune Research),在样本缓冲液中约1:120,000稀释。然后如在实施例1B中描述的平板显色并进行读数。
IE.应用Biacore试验测定KS-1/4抗体与EpCAM的结合速率(on-rate)和解离速率(off-rate)。
KS-1/4和KS-IL2分子对抗原EpCAM的亲合力通过抗体-抗原交互作用的表面等离子体共振分析(surface plasmon resonance analysis)进行测定,所述测定应用Biacore仪器(Biacore International AB,Uppsala,Sweden)。应用生产商提供的胺偶联方案将EpCAM-鼠Fc偶联到CM5传感器芯片上。然后使浓度在25nM和200nM之间变化的KS-1/4和KS-IL2通过芯片,由此观察与芯片的结合。应用Biacore软件的内置曲线拟合程序,计算出结合速率、解离速率、结合及解离常数。
IF.应用表达EpCAM的细胞系测定KS-1/4抗体的结合亲合力
应用标准技术以125I碘化纯化的KS-1/4抗体,然后将浓度不断提高的标记蛋白质与EpCAM阳性细胞系PC-3一起孵育。产生饱和结合曲线并通过Scatchard分析确定解离常数。
实施例2.编码小鼠KS-1/4的VH和VK的cDNA的克隆以及用于表达KS-1/4杂交瘤来源的抗体的载体的构建
从表达小鼠KS-1/4的杂交瘤细胞(从R.Reisfeld,Scripps ResearchInstitute获得)制备的信使RNA以寡聚(dT)进行反转录并随后用作PCR模板以扩增编码重链可变区(VH)和轻链可变区(VK)的序列。根据已发表序列(Beavers等,同上引文)设计PCR引物。用于VH的PCR引物具有以下的序列:

其中TCTAGA和AGATCT序列分别为XbaI和BglII限制性位点,用于将VK与表达载体连接(参见以下);ATG为轻链的翻译起始密码子;且反向引物中的TAC将在PCR产物的有义链中引入剪接供体共有序列GTA。
将编码小鼠KS-1/4抗体的VH和VK的PCR产物克隆入pCRII载体(Invitrogen,Carlsbad,CA)。测定几个VH和VK克隆的序列且确定各共有序列。将VH和VK序列逐步插入表达载体pdHL7。VH的连接利用唯一的XhoI和HindIII位点,且VK的连接利用唯一的XbaI和BglII/BamHI位点(VK插入片段中的唯一BglII和载体中的唯一BamHI具有相容的突出端)。导致产生的构建体称作pdHL7杂交瘤chKS-1/4,其已经包含用于表达嵌合抗体的转录调控元件和人Ig恒定区序列(Gillies等.(1989)免疫学方法杂志(J.Immunol.Methods)125:191)。
表达载体pdHL7来自pdHL2[Gillies等.(1991)杂交瘤(Hybridoma)10:347-356],具有以下的修饰:在表达载体pdHL2中,用于轻链和重链-细胞因子的转录单元由重链免疫球蛋白基因的增强子和金属硫蛋白启动子组成。在pdHL7中,这两个转录单元由CMV增强子-启动子[Boshart等.(1985)细胞(Cell)41:521-530]组成。编码CMV增强子-启动子的DNA源自可商业获得的pcDNAI(Invitrogen Corp.,San Diego,CA)的AflIII-HindIII片段。
实施例3.鼠KS-1/4抗体的表达研究
该实施例讨论了应用编码V区序列的抗体表达质粒进行的表达研究,其中所述V区序列在美国专利No.4,975,369中公开。
3A.质粒构建
为直接比较由杂交瘤KS-1/4序列和美国专利No.4,975,369中公开的序列所编码的嵌合抗体,合成了编码美国专利No.4,975,369中描述的VH序列的cDNA。然后该cDNA与已含有KS-1/4的VK的pdHL7表达载体连接。
为构建在美国专利No.4,975,369中描述的VH序列,通过全化学合成获得编码部分VH序列的NdeI-HindIII片段。化学合成重叠寡核苷酸并进行连接。然后将连接的双链体亚克隆到XbaI-HindIII pBluescript载体(Stratagene,LaJolla,CA)中。
该DNA编码美国专利No.4,975,369的蛋白质序列IQQPQNMRTM。编码序列紧3′端的是以gta起始的剪接供体位点。上链5′末端的ctag为用于XbaI克隆位点的突出端。产生的XbaI位点仅用于克隆到pBluescript载体的多聚接头中。紧随该位点之后为NdeI限制性位点(CATATG)。下链5′末端的agct为HindIII克隆位点的突出端。该HindIII粘端随后与Cγl基因[Gillies等.(1991)杂交瘤(Hybridoma)10:347-356]前的内含子中的HindIII位点连接。
进行序列确认后,分离NdeI-HindIII限制性片段。然后将该片段以及编码VH的N端半分子的XhoI-NdeI片段与XhoI-HindIII消化的pdHL7表达载体连接,其中所述表达载体包含KS-1/4的VK。导致产生的构建体pdHL7-′369chKS-1/4包含所述VK及在美国专利No.4,975,369中描述的VH(称为US4,975,369chKS-1/4)。
3B.杂交瘤chKS-1/4和US4,975,369chKS-1/4抗体的比较
通过以上提及的磷酸钙共沉淀方法将质粒DNA pdHL7-杂交瘤chKS-1/4和pdHL7-′369chKS-1/4平行导入人肾293细胞。转染后5天,用抗-huFc ELISA和κ ELISA(参见实施例1的ELISA方法)测试条件培养基且结果总结在表1中

结果表明杂交瘤chKS-1/4正常表达和分泌,且在两种不同ELISA的精确度范围内,分泌的抗体由基本等克分子数的重链和轻链组成。另一方面,对于US4,975,369chKS-1/4抗体在条件培养基中仅检测到低水平的重链,且无κ轻链与之结合。
对这两种瞬时转染的细胞系的全细胞裂解物以及条件培养基进行Western印迹分析。用于Western印迹分析的方法在Sambrook等.(1989),前述引文中描述。为分析全细胞裂解物,将转染细胞裂解,离心以移除碎屑,且每一泳道应用相当于5 x 105个细胞的裂解物。为分析条件培养基,来自300μL条件培养基的蛋白质产物在还原条件下进行SDS-PAGE前,首先通过蛋白质A琼脂糖凝胶层析进行纯化。Western印迹转移之后,印迹与辣根过氧化物酶缀合的山羊抗-人IgG,Fcγ(JacksonImmunoResearch)杂交,所述抗体应用时1:2000稀释。
Western印迹转移显示,在应用的条件下,当使用pdHL7-杂交瘤chKS-1/4进行转染时于条件培养基和裂解细胞二者中均检测出重链。该结果表明chKS-1/4抗体的重链在细胞中产生并有效地分泌(与轻链一起)。另一方面,用pdHL7-′369chKS-1/4转染时仅在细胞裂解物中检测出重链,而条件培养基中则不能检测到。该结果表明尽管相当水平的重链在细胞内产生,但重链并不能分泌。该发现与ELISA的数据一致,表明在US4,975,369 chKS-1/4抗体中少量分泌的重链没有κ轻链与之相连。应当理解,典型地免疫球蛋白重链在缺乏免疫球蛋白轻链时一般不进行分泌[Hendershot等.(1987)今日免疫学(Immunology Today)8:111]。
除前述之外,NS/0细胞用质粒pdHL7-杂交瘤chKS-1/4和pdHL7-US4,975,369 chKS-1/4通过电穿孔进行平行转染。在100nM MTX存在条件下筛选稳定的克隆,如在实施例1中描述的,且通过抗-huFcELISA测试96孔板中MTX抗性克隆的条件培养基,如在实施例1中所描述。结果在表2中总结。

在96孔板筛选阶段,用pdHL7-杂交瘤chKS-1/4构建体获得的大多数克隆产生约100ng/mL到500ng/mL的抗体,最佳克隆的产量约为10-50μg/mL。另一方面,用pdHL7-′369 chKS-1/4构建体获得的大多数克隆产生约0ng/mL到10ng/mL的抗体,最佳克隆的产量约为300-400ng/mL。为检测US4,975,369 chKS-1/4抗体的组成和结合特性,必须培养生长300-400ng/mL抗体的克隆。挑选这些克隆中的两个用于扩大培养。但是,发现这些克隆的表达水平非常不稳定。培养物生长到200mL时,通过抗-FcELISA测试,两克隆的表达水平均降到约20ng/mL。当用抗-κ ELISA测试相同的条件培养基时,没有检测出κ轻链,这与在293细胞中的瞬时表达相同。
以下的实验表明不存在可检测水平的与US4,975,369 chKS-1/4重链连接的κ轻链。简言之,来自每一克隆的各50mL条件培养基用蛋白质A层析浓缩。通过抗-Fc ELISA和抗-κELISA测试洗脱物。作为对照,来自产生杂交瘤chKS-1/4的克隆的条件培养基用同样方法处理并在相同的时间测试。ELISA结果在表3中总结。

结果表明的确不存在可检测水平的与US4,975,369 chKS-1/4重链连接的κ轻链。此外,杂交瘤chKS-1/4抗体在10-20ng/mL之间显示可结合KS抗原,而来自两克隆的US4,975,369抗体一直浓缩到253和313ng/mL仍不结合KS抗原(参见用于测定与KS抗原的结合的实施例9)。
实施例4.KS抗体变体的表达和表征
显著降低抗体的表达或抗体对靶分子的亲合力的突变预期将降低其用于人类治疗目的的功效。一些降低免疫原性的方法,例如"贴面化(veneering)"、"人源化"和"去免疫化(deimmunization)"涉及导入多个氨基酸替换,并可能破坏抗体与抗原的结合(参见如,美国专利Nos.5,639,641;5,585,089;和PCT公开号WO 98/52976;WO 00/34317)。本领域内存在对如下类型的抗体序列的需要,所述序列可与上皮细胞粘附分子结合,但其不同于识别该抗原的起始鼠单克隆抗体。
测试了KS-1/4重链和轻链可变("V")区的多种组合的表达能力以及它们与EpCAM结合的能力。
这些结果总结在表4-6中并在以下进行描述。

在第2和第3组中,每一种蛋白质的相对结合活性用该组第一行显示的对照进行标准化.ELISA试验主要反映出解离速率(off-rates),以几轮洗涤后结合的蛋白质的量为基础。该试验用作排除弱结合者的快速筛选,但并非是对亲合力的精确测定。在第3组中,将VH2变体VH2.1-VH2.4与VH1进行比较以确定氨基酸的插入是否导致相对结合的提高。
序列的关系在以下叙述。如在实施例中所描述,VH0和VK0序列通过PCR扩增来自表达起始的鼠源KS-1/4的杂交瘤细胞系(SEQ ID NO:1和SEQ ID NO:2)。VH‘369为在美国专利No.4,975,369中公开的VH序列。序列VH1、VH2、VH2.1-2.4 VK1和VK2或通过应用去免疫化(deimmunization)技术(所述技术中通过引入可降低肽表位与II类MHC分子结合的突变来去除或减弱潜在的T细胞表位)或通过改变非人T细胞表位以使其相应于存在于人类抗体中的人自体表位(self-epitopes)而获得。这些构建体的设计在以下进一步描述和分析。表6中构建体的产生通过将表达相应重链和轻链V区的核酸组合用于转染哺乳动物细胞而实现。序列VH6、VH7、VK6、VK7和VK8通过将杂交瘤KS-1/4的表面残基改变为以下描述的人类对应残基而产生,所述改变的目的在于移除潜在的人B细胞表位。构建体1到3的产生通过将表达重链和轻链V区VH6、VH7、VK6、VK7和VK8(见表4和以下描述)的核酸组合用于转染哺乳动物细胞而实现。
4A.具有较少人T细胞抗原表位的KS抗体的表征
制备序列VH2.1-VH2.5用于测试是否能够容忍在KS-1/4重链CDR3区中某些氨基酸的插入和替换。构建出用于轻链和重链组合VK0/VH1、VK1/VH7、VK1/VH1、VK1/VH2、VK1/VH1-IL2、VK1/VH2-IL2和VK1/VH2.5-IL2的表达载体,且根据在前述实施例中描述的方法表达和测试相应抗体和抗体-IL2融合蛋白质。
具体地,序列VH1、VH2、VK1和VK2通过全化学合成获得。对于这些序列中的每一段序列,化学合成覆盖这些区的整个编码和互补链的一系列重叠寡核苷酸,对其磷酸化并进行连接。然后用针对片段末端的适宜引物PCR扩增连接的双链体分子,导入pCRII载体(Invitrogen,Carlsbad,CA)并进行序列验证。然后将这些DNA片段导入表达载体pdHL7的合适位点以分别产生完整的重("H")链和轻("L")链。
应用标准分子生物学技术,通过单密码子修饰以在108位获得Thr而不是Gln(表4),由此从VH2产生序列VH2.5。
通过ELISA(表6)并应用表面等离子体激光共振(Biacore仪器和软件)检测这些抗体以比较它们与EpCAM结合的能力。ELISA实验的结果被认为主要反映了解离速率而非结合速率,且通常精确性较低,因此一般利用不佳的ELISA结果将某些构建体排除在进一步的实验考虑之外。但是,ELISA实验显示有良好结合的抗体仍需要进行进一步的表征。

由于VK1/VH1-IL2的解离速率远远快于VK1/V2-IL2或VK8/VH7-IL2的解离速率,因此认为VK1/VH1-IL2是一种有用性较低的融合蛋白质。
鉴于VK1/VH1-IL2和VK1/VH1-IL2仅在CDR3中VH的第100位存在蛋氨酸/异亮氨酸的差异,因此VK1/VH1-IL2的解离速率高于VK1/VH2-IL2提示此差异位置造成了与EpCAM的疏水性接触,且提示略长的蛋氨酸侧链导致接触效率降低。在蛋白质-蛋白质交互作用领域,一般认为疏水交互作用在决定解离速率中起到主要作用,而在决定结合速率中起到的作用要低得多。
4B.具有单核苷酸插入的KS-1/4变体的表征
重链V区的CDR3序列对于KS抗体与EpCAM的亲合力的重要性通过含有位于该区的氨基酸插入或替换的一系列变体进行确定。应用标准DNA重组技术,通过操作编码VH2和VK1的表达载体产生序列VH2.1、VH2.2、VH2.3和VH2.4。用产生的表达载体转染NS/0细胞且纯化分泌的抗体蛋白质,方法如在前述实施例中所描述的。
发现与VH2变体相比VH1变体未达到最佳,这表明CDR3中的异亮氨酸不能用蛋氨酸替换。下一目标是测试CDR3中氨基酸的插入是否可产生结合特性优于VH1的KS-1/4重链V区。表6的数据将VKI/VH2.1、VK1/VH2.2、VK1/VH2.3和VK1/VH2.4的结合与VK1/VH1的结合进行了比较。发现在KS-1/4VH的CDR3中带有氨基酸插入的构建体与VH1相比无一显示出抗原结合的提高,相反,插入突变体的抗原结合活性或是略微降低或是显著降低。
这些结果表明CDR3中氨基酸的插入对于KS-1/4重链V区的抗原结合活性是不利的。当分析该数据时,可获得一些一般性结论。具体地,KS-1/4VH中84到108位氨基酸的片段对于KS-1/4的抗原结合是重要的,所述片段由以下氨基酸组成:
Asn-Asn-Leu-Arg-Asn-Glu-Asp-Met-Ala-Thr-Tyr-Phe-Cys-Val-Arg-Phe-Ile-Ser-Lys-Gly-Asp-Tyr-Trp-Gly-Gln。该片段包括一段构架片段,Asn-Asn-Leu-Arg-Asn-Glu-Asp-Met-Ala-Thr-Tyr-Phe-Cys-Val-Arg,该片段一般可容忍单个和多个氨基酸替换,但不能容忍氨基酸插入,所述氨基酸插入可能对表达和组装具有有害的效应。此外,数据提示,对于86、91、93、94和95位的氨基酸,就进行有效表达并能与EpCAM进行结合的抗体而言,优选抗体具有疏水性氨基酸。
尽管可忍受仅导致部分活性丧失的一些插入,但VH CDR3片段(由Phe-Ile-Ser-Lys-Gly-Asp-Tyr组成)中氨基酸的插入一般对KS-1/4抗体的EpCAM抗原结合功能是不利的。类似地,尽管可忍受一些仅导致部分活性丧失的插入,但这些位置的替换一般对EpCAM抗原的结合也是不利的。
4C.构建其中的鼠表面残基转换为其人类对应残基的KS-1/4抗体的活性衍生物
用通常存在于人类抗体中的氨基酸替换KS-1/4抗体内的氨基酸来制备抗体,以最小化鼠源V区的免疫原性。优选的KS衍生物也保留对人类EpCAM的特异性结合亲合力。
构建体1.已发现KS-1/4轻链与人类的共有III亚群最为类似,且重链与I亚群最为类似。基于这些相似性,产生了由人类共有亚群氨基酸和KS-1/4来源的CDR以及非共有氨基酸组成的概念性序列。对于此构建体以及以下的构建体,应用硅图形工作站(Silicon Graphics Workstation)和BioSym分子建模软件(BioSym molecular modeling software)构建出三维模型。
对三维模型的观察显示,某些人源的氨基酸与CDR接近且可能影响其构象。基于此分析,在轻链中将人Ser22、Arg44和Phe66分别更改回原来的Thr、Lys和Tyr。在重链中,这些更改被认为是非必须的。在最终的用于构建体1的设计中,轻链具有18个不存在于鼠轻链中的人氨基酸,且重链具有22个不存在于鼠重链中的人氨基酸。
应用合成的寡核苷酸产生用于表达构建体1的DNA。构建体1蛋白质得以有效表达,但发现其在EpCAM结合试验中的活性低了10倍以上。
构建体2.然后采用较为保守的方法,其中仅导入以下的变化:
轻链:K18R、A79P
重链:P9A、L11V、A76T、N88S、M91T
应用合成寡核苷酸和标准重组DNA技术生成用于表达构建体2的DNA。构建体2蛋白质未能有效表达。进一步发现构建体2轻链和鼠KS-1/4重链的组合也不能有效表达,而构建体2重链和鼠KS-1/4轻链的组合得以有效表达。因此,似乎表达缺陷存在于构建体2轻链中。
构建体3.基于构建体2轻链中明显的表达缺陷,通过将构建体1轻链的N端部分与鼠轻链的C端部分融合以构建出新的轻链。应用编码35位和36位氨基酸的KpnI位点。当此轻链与构建体2重链组合时,观察到有效表达且结合并未显著丧失。
由于构建体3导致产生的抗体与构建体1或2相比在蛋白质表达和抗原亲合力方面具有更佳的特性,因此将构建体3的DNA序列插入到pdHL7s-IL2中,产生在SEQ ID NO:40中公开的pdHL7s-VK8/VH7-IL2。用于表达目的时,用电穿孔将该质粒DNA导入小鼠骨髓瘤细胞NS/0中以产生如在实施例1A中描述的稳定转染的细胞系。然后在包被有人Fc的ELISA中测试取自稳定克隆的培养基的抗体表达,见实施例1B中的描述。用于该抗体融合蛋白质的重链和轻链的氨基酸序列分别在SEQ ID NO:41和SEQ ID NO:42中显示。
此外,应用在实施例1F中描述的方法,将碘化的VK8/VH7和VK8/VH7-IL2和表达于PC-3肿瘤细胞表面的EpCAM的结合与碘化的VK0/VH0-IL2的结合进行比较。在实验误差的范围内,发现对于VK8/VH7和VK0/VH0以及对于VK8/VH7-IL2和VK0/VH0-IL2有基本相同的结合亲合力。
4D.用于构建活性KS-1/4抗体的结构-功能关系
总之,具有公开的V区序列的KS-1/4抗体以及融合蛋白质的抗原结合活性为设计针对EpCAM的KS-1/4抗体序列以及为KS-1/4抗体的正确表达和分泌提供了指导。具体地,如果氨基酸替换位于CDR之外,则KS-1/4重链和轻链V区可容忍多个氨基酸替换并保留活性。KS-1/4重链和轻链V区一般似乎不能容忍氨基酸插入,特别是当插入位于CDR内或位于CDR间的构架区内时。
例如,当杂交瘤KS-1/4序列用作起始的、"野生型"序列时,数据表明重链V区可在9、11、16、17、38、40、69、70、71、72、76、79、80、83、88、91和111位容忍氨基酸替换而不丧失或几乎不丧失活性。类似地,轻链可在1、3、10、11、12、13、17、18、19、21、41、42、59、71、73、75、77和103位容忍氨基酸替换而不丧失或几乎不丧失活性。这些改变位于KS-1/4重链和轻链V区的CDR之外。这17个明显可接受的重链氨基酸替换占据了CDR之外的约21%的氨基酸位置,且占CDR之外尝试过替换的氨基酸位置的约68%。类似地,这18个明显可接受的轻链氨基酸替换占据了CDR之外的约23%的氨基酸位置,且占CDR之外尝试过替换的氨基酸位置的约72%。仅在两个例子中CDR之外的氨基酸替换导致了蛋白质的有用性显著降低:轻链中的Ala79Pro替换呈现出对表达有负面影响;以及重链中的Q108T替换对抗原结合具有负面影响。因此,可将氨基酸替换导入到CDR之外的KS-1/4抗体重链或轻链序列中,且所述替换有很大的可能性将导致产生活性蛋白质。
涉及到CDR内的氨基酸替换的突变对抗原结合常常具有负面影响。例如,在重链中的I100M替换将结合减小了约8倍。涉及到氨基酸插入的突变通常对KS-1/4序列的效用具有负面影响。例如,VH-′369重链V区不能与轻链组装成正确抗体,如在此所描述的。VH2.1到2.4突变具有在重链V区CDR3中的氨基酸插入,且这些突变中的每一个均对抗原结合具有负面影响。
实施例5.KS抗体(构建体3)-IL2融合蛋白质在人类中的免疫原性
在人类临床试验中,22位患者接受了一个或多个疗程,每一疗程包括连续3天的每天4小时静脉输注KS抗体(构建体3)-IL2。疗程之间间隔一个月(Weber等.(2001).Proc.Am.Soc.Clin.肿瘤学(Oncology)20:259a.)。在每一疗程之前和之后收集每位患者的血清样本并测试其对完整KS抗体(构建体3)-IL2分子或Fc-IL2成分(不含Fv区)的抗体反应性。在免疫前的血清中均未观察到任何反应性。结果表明仅4位患者产生了对单独Fv区或对Fv区和Fc-IL2成分两者的显著免疫反应。此外,在随后接触huKS-IL2后这些反应并未表现出被加强。
抗体-IL2融合蛋白质的用途被认为在于对V区免疫原性进行特别严格的测试,因为白细胞介素2部分具有佐剂效应。因此,这些结果表明可以将KS抗体(构建体3)施用给人类且仅有少数的受者会明显产生对KS抗体(构建体3)-IL2融合蛋白质的抗体反应。考虑到KS抗体(构建体3)含有几乎完全鼠源的可变区(其中仅少数的氨基酸残基被替换为相应的人氨基酸残基),前述的这些结果尤为令人鼓舞。

Claims (8)
1.重组抗-EpCAM抗体,所述抗体包含:
(i)定义部分轻链CDR1的SEQ ID NO:1的第24-31位氨基酸残基;
(ii)定义轻链CDR2的SEQ ID NO:1的第49-55位氨基酸残基;
(iii)定义轻链CDR3的SEQ ID NO:1的第88-96位氨基酸残基;
(iv)定义重链CDR1的SEQ ID NO:2的第26-35位氨基酸残基;
(v)定义部分重链CDR2的SEQ ID NO:2的第50-62位氨基酸残基;
(vi)定义重链CDR3的SEQ ID NO:2的第101-105位氨基酸残基;
(vii)定义修饰的轻链FR1的SEQ ID NO:9的第1-23位氨基酸残基;
(viii)定义修饰的重链FR1的SEQ ID NO:18的第1-25位氨基酸残基;
和
(ix)定义重链FR3的SEQ ID NO:18的第67-98位氨基酸残基。
2.权利要求1的重组抗-EpCAM抗体,其中所述抗体包含完整的抗体轻链和完整的抗体重链,由此轻链如SEQ ID NO:9的第1-106位残基定义的氨基酸序列基所示,而重链如SEQ ID NO:18的第1-116位残基定义的氨基酸序列所示。
3.权利要求1或2的重组抗-EpCAM抗体,其中抗体对EpCAM的KD结合亲合力为至少10-8M。
4.融合蛋白,其包含权利要求1或2的重组抗-EpCAM抗体和细胞因子。
5.权利要求4的融合蛋白,其中细胞因子为IL-2。
6.编码权利要求1-3中任一项的重组抗-EpCAM抗体或权利要求4-5中任一项的融合蛋白的核酸。
7.包含权利要求6的核酸的表达细胞。
8.权利要求1-3中任一项的重组抗-EpCAM抗体或权利要求4-5中任一项的融合蛋白在制备治疗与EpCAM过表达有关的疾病的药物中的用途。
