Abstract

abstract an object of the present invention is to provide a process for enabling the production of a particulate composition containing crystalline trehalose dihydrate in a higher production yield against starch through consistent steps by using starch as a material. the above object is solved by providing a process for producing a particulate composition containing crystalline a,a-trehalose dihydrate which comprises a,a-trehalose in an amount of 98.0% by weight or more, on a dry solid basis, characterized in that it comprises the steps of allowing to act on liquefied starch an a-glycosyltrehalose-forming enzyme derived from a microorganism of the genus arthrobacter and a trehalose-releasing enzyme derived from a microorganism of the genus arthrobacter along with a starch debranching enzyme and a cyclomaltodextrin glucanotransferase; allowing glucoamylase to act on the resulting mixture to obtain a saccharide solution containing a,a-trehalose; precipitating crystalline a,a-trehalose dihydrate from the above saccharide solution; collecting the precipitated crystalline a,a-trehalose dihydrate by a centrifuge; and ageing and drying the collected crystals; wherein a natural or recombinant cyclomaltodextrin glucanotransferase derived from a microorganism of the genus paenibacillus or a mutant enzyme thereof is used as the cyclomaltodextrin glucanotransferase to increase the a,a-trehalose content in the saccharide solution to over 86.0% by weight, on a dry solid basis, without passing through a fractionation step by column chromatography. tradução do resumo resumo patente de invenção: "método de produção de pó contendo diidrato de alfa,alfa-trealose cristalino". é fornecido um método de produção, por meio do qual é possível produzir um pó contendo diidrato de trealose cristalino com amido como um material de partida com rendimentos elevados de pó para amida. mais especificamente, é fornecido um método de produção para um pó contendo diidrato de a,a-trealose cristalino e tendo pelo menos 98,0 % em massa de a,a-trealose sobre uma base anidra, dito método incluindo: uma etapa em que o amido liquefeito é submetido às ações de uma enzima de desramificação do amido, ciclomaltodextrina glucanotransferase, uma enzima de produção de a-glicosil trealose derivada de um microorganismo que pertence ao gênero arthrobacter e uma enzima de liberação de trealose derivada de um microorganismo que pertence ao gênero arthrobacter, e depois à ação da glicoamilase, para obter uma solução de sacarídeo contendo a,a-trealose; uma etapa em que o diidrato de a,a-trealose é cristalizado a partir da solução de sacarídeo; e uma etapa em que o diidrato de a,a-trealose cristalizado é extraído através da centrifugação, amadurecido e secado. o método de produção é caracterizado pelo fato de que uma enzima natural ou recombinante derivada de um microorganismo que pertence ao gênero paenibacillus ou uma variante de uma tal enzima é utilizada como o ciclomaltodextrina glucanotransferase de tal modo que o teor de a,a-trealose na solução de sacarídeo é mais de 86,0 % em massa em uma base sólida seca, sem ter que passar através de um etapa de fracionamento por cromatografia de coluna.
