Abstract

derivados de amina espiro-cíclicos como moduladores s1p. a presente invenção refere-se aos derivados de amina espiro-cíclica da fórmula (i) em que r1 é selecionado de ciano, (2-4c) alquenila, (2-4c) alquinila, (1-4c) alquila cada uma opcionalmente substituída com cn ou um ou mais átomos de flúor, (3-6c) cicloalquila, (4-6c) cicloalquenila ou um grupo (8-10c) bicíclico, cada uma opcionalmente substituído com halogêneo ou (1-4c) alquila, fenila, bifenila, naftila, cada uma opcionalmente substituída com um ou mais substituintes independentemente selecionados de halogêneo, ciano, (1-6c) alquila opcionalmente substituída com um ou mais átomos de flúor, (1-6c) alcóxi opcionalmente substituído com um ou mais átomos de flúor, amino, di(1-4c) alquilamino e (3-6c) cicloalquila opcionalmente substituída com fenila que pode ser substituída com (1-4c) alquila ou halogêneo, fenila substituída com fenóxi, benzila, benzilóxi, feniletila ou heterocido monocíclico, cada uma opcionalmente substituído com (1-4c) alquila opcionalmente substituída com um ou mais átomos de flúor, heterocido monocíclico de forma opcional independentemente substituído com halogêneo, (1-6c) alquila opcionalmente sibstituída com u ou mais átomos de flúor, (3-6c) cicloalquila, ou fenila opcionalmente substituída com (1-4c) alquila ou halogêneo, e heterociclo bicíclico opcionalmente substituído com halogêneo ou (1-4c) alquila opcionalmente substituída com um ou mais átomos de flúor; -y-(c~ n~-alquileno)-x- é um grupo de ligação em que y é ligado a r1 e selecionado de uma ligação, -o-, co-, -s-, -so-, -so~ 2~, -nh-, -ch=ch-, -c(cf~ 3~)= ch-, -c<sym>c-, -ch~ 2~-o-, -o-co-, -co-o-, -co-nh-, -nh-co-, e trans-ciclopropileno; n é um número inteiro de 0 a 10; e x é ligado ao componente de fenileno/piridila e selecionado de uma ligação, -o-, -s-, -so-, -so~ 2~- , -nh-, -co-, -ch=ch-, e trans-ciclopropileno; r2 é h ou independentemente selecionado de um ou mais substituintes selecionados de halogêneo, (1-4c) alcóxi e (1-4c) alquila opcionalmente substituída com um ou mais átomos de flúor; e r3 é (1-4c) alquileno-r4 em que o grupo de alquileno pode ser substituído com um ou mais átomos de halogêneo ou com (ch~ 2~)~ 2~ para formar um componente de ciclopropila, ou r3 é (3-6c) cicloalquileno-r4, -ch~ 2~-(3-6c)cicloalquilen-r4, (3-6c) cicloalquileno-ch~ 2- r4 ou -co-ch~ 2~-r4, em que r4 é -oh, -po~ 3~ h~ 2~-r4, em que r4 é -oh, -po~ 3~h~ 2~, opo~ 3~h~ 2~, -cooh, -coo(1-4c) alquila ou tetrazol-5-ila; q é uma ligação ou -o-; r5 é h ou independentemente selecionado de um ou mais halogêneos; z é ch, cr2 ou n; e a representa uma estrutura de anel de morfolina ou uma amina cíclica de 5,6 e 7 membros; ou um sal, solvato ou hidrato farmaceuticamente aceitável destes ou um ou mais de seus n-óxidos. oc compostos da invenção possuem afinidade aos receptores de s1p e podem ser usados no tratamento, alívio ou prevenção de doenças ou condições em que (a) o(s) receptor(es) de s1p está (estão) envolvido(s).
