Abstract

"MéTODO PARA PRODUZIR UM áCIDO HIALURÈNICO, CéLULA HOSPEDEIRA DE BACILLUS, CONSTRUçãO DE áCIDO NUCLéICO SEQüêNCIA DE áCIDO NUCLéICO ISOLADA, VETOR DE EXPRESSãO, VETOR DE EXPRESSãO RECOMBINANTE, CéLULA HOSPEDEIRA RECOMBINANTE, MéTODO PARA PRODUZIR UM POLIPEPTìDEO, E, POLIPEPTìDEO ISOLADO". A presente invenção diz respeito a métodos para produzir ácido hialurónico, compreendendo: (a) cultivar uma célula hospedeira de Bacillus sob condições adequadas para a produção do ácido hialurónico, em que a célula hospedeira de Bacillus compreende uma construção de ácido nucléico compreendendo uma seqüência codificando a hialuronano sintase operavelmente ligada a uma seqüência promotora estranha à seqüência codificando a hialuronano sintase; e (b) recuperar o ácido hialurónico do meio de cultivo. A presente invenção também diz respeito a uma seqüência de ácido nucléico isolada codificando um óperon de hialuronano sintase que compreende um gene de hialuronano sintase e um gene de UDP-glicose 6-desidrogenase, e opcionalmente um ou mais genes selecionados do grupo consistindo de um gene de UDP-glicose pirofosforilase, um gene de UDP-Nacetilglicosamina pirofosforilase, e um gene de glicose-6-fosfato isomerase. A presente invenção também diz respeito a seqüências de ácido nucléico isoladas codificando uma UDP-glicose-6-desidrocenase, UDP-glicose pirofosforilase, e UDP-N-acetilglicosamina pirofosforilase.
