Abstract

Compuestos inhibidores 11-beta-hidroxiesteroide deshidrogenasa tipo 1. Los inhibidores 11-beta-hidroxiesteroide deshidrogenasa tipo 1 son utiles para tratar, prevenir o disminuir la progresion de enfermedades que requieren la terapia del inhibidor 11-beta-hidroxiesteroide deshidrogenasa tipo 1, tales como diabetes tipo II, enfermedades cardiovasculares, obesidad y osteoporosis. También se divulga una composicion farmacéutica que comprende al compuesto y un proceso de preparacion del mismo. Reivindicacion 1: Un compuesto caracterizado porque tiene la formula (1), enantiomeros, diaestereomeros, solvatos o sales de los mismos en donde: A es un sistema de anillo bi, tri o policíclico no aromático de 5 hasta 20 miembros que opcionalmente contiene 1-3 heteroátomos seleccionados de N, O, S, S(O) y S(O)2, los cuales pueden opcionalmente substituirse con uno o más R4; X es -C(=O)OH, -C(=O)C(=O)OH, -C(=O)NR9R9, tetrazolilo, o -C(=O)NHS(O)2R9; W está ausente, -(CR8aR8b)m, -(CR8aR8b)m-O, - (CR8aR8b)m-N(R14)-, cicloalquilo C3-6, alquenilo o alquinilo, en donde el cicloalquilo o alquenilo puede ser opcionalmente substituido con uno o más R8a; m es 1-3; Z es -CN, alquilo C3-10, cicloalquilo, arilo, arilalquilo, heterociclilo o heterociclilalquilo, todos los cuales pueden opcionalmente substituirse con uno o más R4, y el heterociclilo y heterociclilalquilo contiene 1-4 heteroátomos seleccionados de N, O, y S; R8, cada vez que se presenta, es independientemente alquilo, arilo o heterociclilo; R8a, cada vez que se presenta, es independientemente hidrogeno, alquilo o arilo; R8b, cada vez que se presenta, es independientemente hidrogeno, -C(=O)OH, alquilo, -OH, halo, -CN, -OR10, -C(=O)R10 o -C(=O)NR9R9, en donde el alquilo puede ser opcionalmente substituido con uno o más R9a, o R8a y R8b se toman junto con el carbono al cual ambos se enlazan para formar un anillo de 3 hasta 7 miembros, el cual puede opcionalmente contener 1-4 heteroátomos seleccionados de N, O, y S y ser opcionalmente substituido con 0-3 R9a; R9, cada vez que se presenta, es independientemente hidrogeno, alquilo, alcoxi, cicloalquilo, arilo, arilalquilo, heterociclilo o heterociclilalquilo, en donde el alquilo, cicloalquilo, arilo, arilalquilo, heterociclilo o heterociclilalquilo puede ser opcionalmente substituido con 0-5 R9a, y el heterociclilo o heterociclilalquilo contiene 1-4 heteroátomos seleccionados de N, O, y S; o dos R9 junto con el nitrogeno al cual se enlazan forman un sistema de anillo cíclico insaturado, saturado o parcialmente saturado que contiene desde 2-10 átomos de carbono y desde 0-4 heteroátomos adicionales seleccionados de N, O, S, S(O) y S(O)2, en donde el sistema de anillo puede ser opcionalmente substituido o fusionado con uno o más R9a;
