Description
「クチナシ青色素」は、一般に、アカネ科クチナシ(Gardenia augusta MERRILL var.grandiflora HORT.,Gardenia jasminoides ELLIS)の果実より抽出して得られたイリドイド配糖体(主成分は、ゲニポシド)を、タンパク質分解物等の存在下で、β-グルコシダーゼ処理して得られる。ここで、ゲニポシドはβ-グルコシダーゼ処理によりアグリコンであるゲニピンに変換され、及びゲニピンがタンパク質分解物等と反応してクチナシ青色素の色素成分(色素の本質(coloring principle))が生じる。
従って、「クチナシ青色素」は、通常、単一化合物ではなく、混合物であり、また、通常、原料由来の未反応のゲニポシド、ゲニピン、又はこれらの両方を含有する。少なくとも出願人の知る限り、従来、クチナシ青色素からゲニポシド、及びゲニピンを高度に除去する方法は確立されておらず、精製されたクチナシ青色素であっても、ゲニポシド、ゲニピン、又はこれらの両方を含有する。

従って、「クチナシ黄色素」も、単一化合物ではなく、また、通常、原料由来のゲニポシドを含有する。
しかし、ゲニポシドは、ラットに大量に経口投与すると肝毒性を示し、その毒性発現には、ラットの腸内細菌のβ-グルコシダーゼにより生じたゲニピンが関与している可能性が報告されている(非特許文献1)。
また、当該特許文献1には、活性炭が、クチナシ黄色素の色素成分と、ゲニポシドとを共に非選択に吸着してしまうので、活性炭処理では、クチナシ黄色素の色素成分と、ゲニポシドとを分離することはできないことが記載されている。
しかし、ゲニポシド、ゲニピン、又はこれらの両方を含有する組成物から、ゲニポシド、ゲニピン、又はこれらの両方を除去することに着目した技術の例は少なく、出願人が知る限り、前記特許文献1に記載の技術が唯一の例である。
前記特許文献1に記載の技術は、クチナシ黄色素の色素成分であるクロシンを選択的に吸着させることを原理とするものであり、クチナシ黄色素以外に適用できる技術ではない。
そこで、本発明は、ゲニポシド、ゲニピン、又はこれらの両方を含有する組成物から、ゲニポシド、ゲニピン、又はこれらの両方を除去する新たな方法を提供することを目的とする。
しかし、本発明者らは、鋭意検討の結果、ゲニポシド、ゲニピン、又はこれらの両方を含有する組成物を、特定の活性炭、具体的には(a)メチレンブルー吸着性能が50ml/g以上であり、且つ(b)よう素吸着性能が750mg/g以上である活性炭により処理することで、クチナシ青色素の色素成分、及びクチナシ黄色素の色素成分のような有用な成分の損失を抑制しながら、ゲニポシド、ゲニピン、又はこれらの両方を除去できることを見出した。前述した通り、活性炭処理では、クチナシ黄色素の色素成分と、ゲニポシドとを分離できないことが知られていた(前記特許文献1)ので、これは、驚くべきことである。
本発明者らは、かかる知見に基づき、更なる研究の結果、本発明を完成するに至った。
ゲニポシド、ゲニピン、又はこれらの両方を含有する材料を、(a)メチレンブルー吸着性能が50ml/g以上であり、且つ(b)よう素吸着性能が750mg/g以上である活性炭により処理して、ゲニポシド、ゲニピン、又はこれらの両方を除去する方法。
項2. ゲニポシド、及びゲニピンの総含量が低減された、クチナシ由来色素の製造方法であって、
項1に記載の方法により、前記ゲニポシド、ゲニピン、又はこれらの両方を含有するクチナシ由来色素から、ゲニポシド、ゲニピン、又はこれらの両方を除去する工程を含む製造方法。
項3. クチナシ由来色素が、クチナシ青色素、クチナシ赤色素、又はクチナシ黄色素である項2に記載の製造方法。

項1に記載の方法により、ゲニピンを含有するチブサノキ果汁又はチブサノキ抽出物から、ゲニピンを除去する工程を含む製造方法。
項7. 色価100換算における、ゲニポシド、及びゲニピンの総含量が300ppm以下である、クチナシ青色素含有組成物。
本明細書中、特に記載の無い限り、「色価」は、「E1cm 10%」である。また、本明細書中、特に記載の無い限り、「色価」は、第8版食品添加物公定書(日本国厚生労働省)に記載の方法に従って決定される。
本明細書中、「色価100換算」とは、測定値等の各種数値を、対象となる材料、色素等の色価100当たりの数値に換算することをいう。例えば、色価200の材料に含まれるゲニポシド、ゲニピン、又はこれらの両方の含量が500ppmである場合、色価100換算における、ゲニポシド、ゲニピン、又はこれらの両方の含量は、500ppmに、色価の比率(100/200)を乗じることで、250ppmと算出される。当然のことであるが、例えば、色価100の試料の測定値は、それ自体が「色価100換算」における数値であることができる。
本明細書中、特に記載の無い限り、「室温」及び「常温」は、10~40℃の範囲内の温度を意味する。
本明細書中、「色素成分」とは、色素の本質(coloring principle)を意味する。
本明細書中、「色素」は、色素成分を含有し、又はこれのみからなり;複数種の色素成分(化合物)の混合物であることができ;また、色素成分以外の原料由来の成分、製法に起因する成分、又はその両方を含有してもよい。本明細書中、「色素」は、好ましくは、天然物由来の色素である。また、本明細書中、「色素」は、精製前、又は粗精製の色素(又は色素含有材料)、或いは精製後の色素を意味する場合がある。これは、文脈によって、判断できる。

本発明の、ゲニポシド、ゲニピン、又はこれらの両方を含有する材料から、ゲニポシド、ゲニピン、又はこれらの両方を除去する方法(以下、本発明の除去方法と称する場合がある。)は、
ゲニポシド、ゲニピン、又はこれらの両方を含有する材料を、(a)メチレンブルー吸着性能が50ml/g以上であり、且つ(b)よう素吸着性能が750mg/g以上である活性炭により処理して、ゲニポシド、ゲニピン、又はこれらの両方を除去する方法である。
本発明の除去方法を施される「ゲニポシド、ゲニピン、又はこれらの両方を含有する材料」における「ゲニポシド、ゲニピン、又はこれらの両方」の量は、特に限定されず、その除去が望まれる程度の量であればよい。
当該量の上限は、例えば、当該材料中の固形分全体に対して、ゲニポシド、及びゲニピンの総含量として、2w/w%、1.2w/w%、又は0.6w/w%である。

<クチナシ黄色素の色価測定方法>
色価100に換算して約5gに相当する量の試料を精密に量り、当該試料に0.02mol/L水酸化ナトリウム溶液50mlを加えて50℃の水浴中で20分間加温し、必要があれば振り混ぜながら溶かし、水を加えて正確に100mlとする。その1mlを正確に量りとり、50vol%エタノールを加えて正確に100mlとし、必要があれば遠心分離し、その上澄液を検液とする。50vol%エタノールを対照として、410~425nmの極大吸収部における、液層の長さ1cmでの吸光度Aを測定し、次式により色価を求める。

本発明の除去方法で用いられる活性炭は、(a)メチレンブルー吸着性能が50ml/g以上であり、且つ(b)よう素吸着性能が750mg/g以上である活性炭である。

-195.8℃の液体窒素沸点において試料(活性炭)に窒素ガスを吸着させる。

比表面積がこのような数値であることにより、クチナシ青色素の色素成分に代表される有用な成分の損失を抑制して、「ゲニポシド、ゲニピン、又はこれらの両方」を選択的に除去できる。
<比表面積容積の決定方法>
-195.8℃の液体窒素沸点において試料(活性炭)に窒素ガスを吸着させる。
相対圧0.1以下の範囲で圧力と吸着量の関係を求め、B.E.T理論に基づき比表面積を求める。
商業的に入手できる活性炭の例としては、クラレケミカル株式会社製のクラレコール(商品名)シリーズ、株式会社ユニオンサービス製のSPシリーズ、日本エンバイロケミカルズ株式会社製の白鷺シリーズ、太平化学産業株式会社製の梅蜂印シリーズ及び株式会社ユー・イー・エス製の活力炭シリーズが挙げられる。
ゲニポシド、ゲニピン、又はこれらの両方を含有する材料を活性炭に接触させることは、当該材料が固体の場合、例えば、当該材料を水系溶媒に溶解又は懸濁することにより、液体の形態にし、当該液体の状態の材料と活性炭とを混合することにより実施できる。一方、当該材料が液体の場合、例えば、当該材料をそのまま、又は水系溶媒で希釈して、当該液体の状態の材料と活性炭とを混合することにより実施できる。当該混合は、振とう機、又は攪拌機等を用いる慣用の手段で実施すればよい。
前記水系溶媒の例は、水(例、水道水、イオン交換水、蒸留水)、及び含水アルコール(例、含水エタノール)を包含する。クチナシ青色素の色素成分に代表される有用な成分が活性炭に吸着されてしまうことを抑制する観点からは、前記水系溶媒は、アルコール(例、エタノール)の含有量が小さいことが好ましく、好ましくは水(例、水道水、イオン交換水、蒸留水)である。
前記「液体の状態の材料」のpHを調整する場合、当該調整は、塩酸、又は水酸化ナトリウムを用いる等の慣用の方法により行えばよい。
当該時間が短すぎると「ゲニポシド、及びゲニピン、又はこれらの両方」の除去が不充分になり、一方、当該時間が長すぎても「ゲニポシド、及びゲニピン、又はこれらの両方」の除去率が向上しなくなるので、作業効率の点で不利になる。
本発明の除去方法においては、好ましくは、前記「ゲニポシド、ゲニピン、又はこれらの両方を含有する材料」を、先に膜により処理し、次いで、前記「活性炭による処理」を実施する。
当該膜による処理によって、ある程度、「ゲニポシド、ゲニピン、又はこれらの両方」を除去することができる。これによって、例えば、「活性炭による処理」における活性炭の使用量を低減できる。
当該膜による処理に用いられる膜は、好ましくは、限外濾過膜であり、より好ましくは、分画分子量が2000~5000の範囲内である膜である。このような膜は、商業的に入手可能である。当該限外濾過膜を用いた処理の条件は、好ましくは、例えば、0.3~0.7MPaで、3~24時間である。
当該膜による処理を施した場合、前記「活性炭による処理」を施される「ゲニポシド、ゲニピン、又はこれらの両方を含有する材料」は、当該膜による処理の濃縮液である。当該濃縮液は、必要に応じて、前記「活性炭による処理」の前に、更に、希釈、又は濃縮等の処理を施されていてもよい。
当該色素成分の例は、クチナシ青色素成分、クチナシ赤色素成分、及びクチナシ黄色素成分を包含する。
当該色素成分は、本発明の特に好適な一態様では、クチナシ青色素成分である。
ここで、「色価の残存率」とは、次のように定義される。
色価の残存率(%)=(本発明の除去方法による処理後の色価)/(本発明の除去方法による処理前の色価)×100
前記で説明した本発明の除去方法によれば、ゲニポシド、ゲニピン、又はこれらの両方を含有するクチナシ由来色素から、ゲニポシド、ゲニピン、又はこれらの両方を除去することによって、ゲニポシド、及びゲニピンの総含量が低減された、クチナシ由来色素を製造できる。すなわち、本発明の一態様は、ゲニポシド、及びゲニピンの総含量が低減された、クチナシ由来色素の製造方法である。
当該クチナシ由来色素の例は、クチナシ青色素、クチナシ赤色素、及びクチナシ黄色素を包含する。
本発明の製造方法で得られるクチナシ由来色素は、活性炭による処理後の溶液の形態であってもよく、その濃縮物の形態、又は任意の方法で乾燥(例、真空乾燥、凍結乾燥、噴霧乾燥など)して得られる粉末形態を有するものであってよい。
当該量は小さいほど好ましく、前記の測定方法による測定限界未満であることが特に好ましい。従って、当該量の下限は限定されないが、例えば、当該量が1ppm以上、2ppm以上、又は5ppm以上であるクチナシ由来色素は、使用の目的及び形態によって、許容され得る。
その例は、シュクロース、乳糖、グルコース、デキストリン、アラビアゴム、水、エタノール、プロピレングリコール、グリセリン、及び水飴等を包含する。
本発明は、前記のクチナシ由来色素、又はその色素製剤を用いて着色された食品、香粧品、医薬品、医薬部外品、飼料等の着色組成物を提供する。
当該食品の例は、冷菓、生菓子、和菓子、及び洋菓子等の菓子類;飲料、及びアルコール飲料等の飲料類;乾燥野菜、及び漬け物等の農産加工品;海産物加工品;並びに畜肉加工品等を包含する。
当該香粧品の例は、化粧料(例、アイシャドー、マスカラ、口紅、リップクリーム、及び化粧水等)、石鹸、シャンプー、リンス、洗剤、歯磨き、及び洗口液等を包含する。
当該医薬品の例は、錠剤(例、糖衣錠)、顆粒剤、液剤、及びカプセル剤等を包含する。
通常、これらの着色組成物におけるクチナシ由来色素の含量は、特に制限されないが、例えば、クチナシ青色素の場合、605nm前後である、その極大吸収波長における着色組成物の吸光度が0.01~1となるような量であることができる。
前記で説明した本発明の除去方法によって、ゲニピンを含有するチブサノキ果汁又はチブサノキ抽出物(前述の通り、通常、チブサノキ果汁又はチブサノキ抽出物は、ゲニピンを含有する。)から、ゲニピンを除去することによって、ゲニピンの含量が低減されたブサノキ果汁又はチブサノキ抽出物を製造できる。すなわち、本発明の一態様は、ゲニピンの含量が低減された、チブサノキ果汁又はチブサノキ抽出物の製造方法である。
CV:色価(Color Value)
例えば、「Total G残存率」は、次のように定義される。
Total G残存率(%)=(処理後ゲニポシド量+処理後ゲニピン量)/(処理前ゲニポシド量+処理前ゲニピン量)×100
当該分析において、分析試料の色価が100以上の場合は、分析試料を超純水で色価100に希釈した。

移動相: a)水, b)アセトニトリル(b. 12% in 8 min., 12-15% in 2 min., 15% in 10 min., 15-100% in 5 min. 100% in 10min.)

温度: 40℃
検出: UV-Vis detector 238 nm
注入量: 20μL
種々の活性炭を用いたゲニポシド吸着試験
種々の活性炭(活性炭No.1~16)を用いたゲニポシド吸着試験を行った。使用した活性炭は、いずれも商業的に入手した。このうち、活性炭No.5、及びNo.6は、同じ製品であり、ロット番号のみが異なった。
表1からわかるように、活性炭の種類によって、ゲニポシド吸着能が著しく相違した。
クチナシ青色存在下でのゲニポシド吸着試験
試験例1でゲニポシドをよく吸着した活性炭のうちの9種類(活性炭No.4、6、7、9、11、13、14、15及び16)について、実際にクチナシ青色素中からゲニポシド、及びゲニピンを除去できるかの検討を行うため、クチナシ青色素存在下でのゲニポシド、及びゲニピン吸着試験を行った。ここで、用いたクチナシ青色素からは、ゲニピンが検出されなかったので、実際に活性炭への吸着が確認されたのは、ゲニポシドの吸着性である。しかし、ゲニピンは、構造、及び理化学的性質がゲニポシドと類似するので、前記試験例及び実施例において、ゲニポシドと同様の挙動を示すと合理的に推測される。
結果を表2に示す。表2中、「対照」は、活性炭非処理である。
活性炭の量、及び活性炭による処理時間の検討
活性炭No.7のゲニポシド吸着量は、最大で0.1g/g程度と推測される。表3の結果から、効率よく除去するには過剰量の活性炭を投入することが好ましいことが明らかになった。処理時間については、色価100換算におけるゲニポシド含量(ゲニポシド/CV100)の低下は、処理時間が1時間を超えると緩やかになる傾向がみられた。
ゲニポシド低減クチナシ青色素の製造1
(工程1)
水道水中のクチナシ果実由来の精製ゲニポシド(ゲニポシド含量37.5w/w%)に、タンパク質加水分解物の存在下でβ-グルコシダーゼを作用させて、クチナシ青色素(溶液状態、液量2350g、色価113.3、色価100換算におけるゲニポシド含量1372ppm、ゲニピン含量検出限界未満)を調製した。
当該クチナシ色素を次の濾過条件で濾過し、濾液として、クチナシ青色素(溶液状態、液量2606g、色価100.9)を得た。
[濾過条件]
濾紙:ADVANTEC NO.2、φ125mm(ADVANTEC社)
濾過助剤:ケイソウ土
濾過助剤のプリコート量:20g
濾過助剤のボディフィード量:液量の2w/w%
(工程2)
前記濾紙による濾過後のクチナシ青色素を、限外濾過膜で処理(0.5MPa、15時間40分)して、クチナシ青色素(溶液状態、液量2448g、色価102.6、固形分14.6%、色価/固形分704.2、収率95.5%、色価100換算におけるゲニポシド含量432ppm、ゲニピン含量検出限界未満)を得た。
(工程3)
前記限外濾過処理後のクチナシ青色素に、1w/w%、3w/w%、又は5w/w%の量の活性炭No.9を添加後、常温にてスターラーにて攪拌し、1時間、3時間、及び5時間後の色価、及びゲニポシド含量を測定した。これらから算出した色素残存率(5時間後)を表4に示す。また、これらから算出した色価100換算におけるゲニポシド、及びゲニピンの総含量(Total G/CV100)のグラフを表4及び図3に示す。
アルコールの有無による吸着効果の違いの検討のため、製造試験例1の前記限外濾過処理後のクチナシ青色素80重量部に、水、又はエタノールをそれぞれ20重量部添加した液を調製し、これに、1w/w%の量比で活性炭No.9を添加後、常温にてスターラーにて攪拌し、5時間後の色価、及びゲニポシド含量を測定した。これから算出した色素残存率を表5に示す。
これにより、本発明の目的においては、エタノールを添加しないか、その添加量を小さくすることが望ましいことが明らかになった。
活性炭の性質と、ゲニポシド残存率、及び色価残存率との関係の分析
これらの調査、及び分析のデータを表6に示す。表6中、「対照」は、活性炭非処理であり、及び「ND」は、検出限界未満である。
また、細孔容積が1.12未満である活性炭は、クチナシ青色素の色素残存率が極めて高い傾向がみられた。
また、比表面積は大きいほうがゲニポシドをより多く吸着する点で好ましい。特に、比表面積が、650m2/g以上の場合に、色価100換算におけるゲニポシド残存率が低い結果が得られた。
クチナシ黄色素
クロシン(東京化成工業)5%溶液100gに、ゲニポシド含量約2000ppmとなるようにクチナシ果実由来の精製ゲニポシド(ゲニポシド含量34.8w/w%)を0.57g添加した。50mL容スクリュー管3本に、それぞれ20gはかりとり、それぞれを対照(活性炭非処理)、活性炭No.7添加区、及び活性炭No.8添加区に用いた。活性炭の添加量は、5w/w%添加(各1g添加)であった。
常温にてそれぞれ3時間攪拌後、色価、及びゲニポシド含量の測定を行なった。これらから算出した色価残存率、及びゲニポシド残存率を表7に示す。
市販のクチナシ青色素の分析
本発明のクチナシ青色素との対比のため、商業的に入手可能な4種のクチナシ青色素の分析を行った。
結果を表8に示す。表8中、「ND」は、検出限界未満である。
陰イオン交換樹脂によるクチナシ青色素中のゲニポシド除去の検討
陰イオン交換樹脂によるクチナシ青色素中のゲニポシド除去の可能性を検討した。
バッチ式の予備試験で一定のゲニポシド吸着効果が認められた弱塩基性陽イオン交換樹脂 IRA96SB(商品名、オルガノ社)をカラム式で用いて、ゲニポシド除去試験を実施した。
(樹脂コンディショニング)
IRA96SBをイオン交換水に一晩浸漬して膨潤させ、当該膨潤樹脂20mLをカラムに充填した(3本)。
(吸着試験)
試験液A:ゲニポシド標品の約30ppm溶液、pH6.18
試験液B:クチナシ青色素液(色価82.6)、pH6.04
試験液C:クチナシ青色素液(色価82.6)、pH9.30(48%NaOHにてpH調整した)。
前記試験液A~Cの20.00gを、それぞれ、SV(空間速度)2程度の速度で通液し、イオン交換水を各樹脂の3倍量(60mL)、及びSV4程度の速度で通液することにより水洗した。通液開始時から水洗終了時までの全ての液を回収した(約80g)。
HPLCで各サンプルのゲニポシド含量を測定した。
更に、通液時のpHによる吸着の差はあまり見られなかったが、実際の工程を想定した場合に不都合なことに、pHが低い場合は、樹脂中、及び回収液に析出物が発生しやすくなる傾向がみられた。
従って、当該試験からは、弱塩基性陽イオン交換樹脂によるクチナシ青色素からのゲニポシドの除去は困難であると結論された。
酸析、又は塩析によるクチナシ青色素中のゲニポシド除去の検討
酸析、又は塩析によるクチナシ青色素中のゲニポシド除去の可能性を検討した。
(クチナシ青色素の用意)
タンパク質加水分解物の存在下でクチナシ果実由来の精製ゲニポシド(ゲニポシド含量36.9w/w%)にβ-グルコシダーゼを作用させてクチナシ青色素(溶液状態、色価118.1、ゲニポシド含量2288ppm)を得た。
殺菌後のクチナシ青色素50gを97%硫酸にてpH2.5に調整した。スターラーで攪拌後、常温で一晩静置した。上清と沈殿物を遠心管に移し、3000G×20分間遠心処理し、上清を回収した。残った沈殿物を、pH2.5に調整した硫酸水で2回洗浄し、更にイオン交換水で1回洗浄した。当該洗浄後の沈殿物にイオン交換水を適当量添加し、沈殿物の一部を溶解させた。この際、沈殿物を溶解させるため、47%NaOHを適当量加えた。
得られた上清、及び沈殿物中のゲニポシド含量をHPLCにて測定した。
殺菌後のクチナシ青色素50gに硫酸アンモニウムを23.6g添加した。スターラーで攪拌後、常温で一晩静置した。上清と沈殿物を遠心管に回収し、3000G×20分間遠心処理し、上清を回収した。一方、残った沈殿物を、70%飽和度の硫酸アンモニウム水で2回洗浄し、更にイオン交換水で1回洗浄した。当該洗浄後の沈殿物にイオン交換水を適当量添加し、沈殿物の一部を溶解させた。
得られた上清、及び沈殿物中のゲニポシド含量をHPLCにて測定した。
ゲニポシド低減クチナシ青色素の製造2
(工程1)
水道水中のクチナシ果実由来の精製ゲニポシド(ゲニポシド含量35.7w/w%)に、タンパク質加水分解物の存在下でβ-グルコシダーゼを作用させて、クチナシ青色素(溶液状態、液量1200L、色価105.2、色価100換算におけるゲニポシド含量597.4ppm、ゲニピン含量検出限界未満)を調製した。
当該クチナシ色素を次の濾過条件で濾過し、濾液として、クチナシ青色素(溶液状態、液量1900L、色価65.2)を得た。
[濾過条件]
濾過設備:フィルタープレス
濾過助剤:ケイソウ土
濾過助剤のプリコート量:30kg
濾過助剤のボディフィード量:20kg
(工程2)
前記濾過設備による濾過後のクチナシ青色素を、限外濾過膜(分画分子量3000)で処理(0.5MPa、5時間)して、クチナシ青色素(溶液状態、液量1300L、色価96.0、固形分11.7%、色価/固形分820.5、収率98.9%)を得た。
(工程3)
前記限外濾過処理後のクチナシ青色素に、3w/w%量の活性炭No.9を添加後、15℃に冷却して攪拌機で2時間攪拌し、一晩静置した。
当該クチナシ青色素を次の濾過条件で濾過し、濾液として、クチナシ青色素(溶液状態、液量2800L、色価41.7)を得た。

濾過設備:フィルタープレス
濾過助剤:ケイソウ土
濾過助剤のプリコート量:30kg
濾過助剤のボディフィード量:58kg
(工程4)
クチナシ青色素を含有する前記工程3の濾過処理を経た濾液を減圧濃縮し、クチナシ青色素(色価242.9、液量343kg)を得た。
このクチナシ青色素のゲニポシドとゲニピン含量を測定したところ、ゲニポシド含量は1.6ppm(0.7ppm/CV100)であり、及びゲニピンは検出限界以下であった。
すなわち、色価100換算におけるゲニポシド、及びゲニピンの総含量(Total G/CV100)は、0.7ppm/CV100であった。
クチナシ赤色素
タンパク質加水分解物の存在下で、クチナシ果実由来の精製ゲニポシドのエステル加水分解物にβ-グルコシダーゼを作用させて得られたクチナシ赤色素に、ゲニポシド含量が約2000ppmとなるようにクチナシ果実由来の精製ゲニポシド(ゲニポシド含量36.5w/w%、ゲニピン含量検出限界未満)を添加してゲニポシド添加クチナシ赤色素を調製した。
50mL容スクリュー管3本に、当該ゲニポシド添加クチナシ赤色素(当該色素は、水で色価を100に調整した溶液の形態である。)をそれぞれ20g量りとり、それぞれを対照(活性炭非処理)、活性炭No.7添加区、および活性炭No.8添加区とした。活性炭の添加量は、5w/w%添加(各1g添加)であった。
常温にてそれぞれ3時間攪拌後、0.2μmフィルターでろ過したサンプルを得た。
ろ過したサンプルについて、色価、及びゲニポシド含量の測定を行った。これらから算出した色価残存率、及びゲニポシド残存率を表11に示す。表11中、「ND」は、検出限界未満である。
Claims (7)
- ゲニポシド、ゲニピン、又はこれらの両方を含有する材料から、ゲニポシド、ゲニピン、又はこれらの両方を除去する方法であって、
ゲニポシド、ゲニピン、又はこれらの両方を含有する材料を、(a)メチレンブルー吸着性能が50ml/g以上であり、且つ(b)よう素吸着性能が750mg/g以上である活性炭により処理して、ゲニポシド、ゲニピン、又はこれらの両方を除去する方法。 - ゲニポシド、及びゲニピンの総含量が低減された、クチナシ由来色素の製造方法であって、
請求項1に記載の方法により、前記ゲニポシド、ゲニピン、又はこれらの両方を含有するクチナシ由来色素から、ゲニポシド、ゲニピン、又はこれらの両方を除去する工程を含む製造方法。 - クチナシ由来色素が、クチナシ青色素、クチナシ赤色素、又はクチナシ黄色素である請求項2に記載の製造方法。
- 請求項2又は3に記載の製造方法で製造されたクチナシ由来色素を含有する色素製剤。
- 請求項2又は3に記載の製造方法で製造されたクチナシ由来色素を含有する着色組成物。
- ゲニピンの含量が低減された、チブサノキ果汁又はチブサノキ抽出物の製造方法であって、
請求項1に記載の方法により、ゲニピンを含有するチブサノキ果汁又はチブサノキ抽出物から、ゲニピンを除去する工程を含む製造方法。 - 色価100換算における、ゲニポシド、及びゲニピンの総含量が300ppm以下である、クチナシ青色素。
